Abstract

Aim: Strategic Targeting of Registries and International Database of Excellence (STRIDE) is an ongoing, multicenter registry providing real-world evidence regarding ataluren use in patients with nonsense mutation Duchenne muscular dystrophy (nmDMD). We examined the effectiveness of ataluren + standard of care (SoC) in the registry versus SoC alone in the Cooperative International Neuromuscular Research Group (CINRG) Duchenne Natural History Study (DNHS), DMD genotype–phenotype/–ataluren benefit correlations and ataluren safety. Patients & methods: Propensity score matching was performed to identify STRIDE and CINRG DNHS patients who were comparable in established disease progression predictors (registry cut-off date, 9 July 2018). Results & conclusion: Kaplan–Meier analyses demonstrated that ataluren + SoC significantly delayed age at loss of ambulation and age at worsening performance in timed function tests versus SoC alone (p ≤ 0.05). There were no DMD genotype–phenotype/ataluren benefit correlations. Ataluren was well tolerated. These results indicate that ataluren + SoC delays functional milestones of DMD progression in patients with nmDMD in routine clinical practice. ClinicalTrials.gov identifier: NCT02369731. Abstract

Examination of the anorthite + margarite + epidote microinclusions within garnet and rutile in a graphite metapelite from northeast Sardinia reveals the presence of a CO2-rich fluid at an early stage in the prograde pelite metamorphism. Mineral reactions regulate the fluid composition and minute carbonate-rich segregations are the sources of this fluid. The low H2O content of the fluid inhibits the transport process, leading to mosaic equilibria and the fine grain size of the inclusions. Calc-silicate inclusions show an interval in metamorphism and a dehydration process ends this interval by introducing significant quantities of fluid into the rock.

12. Revised Thermobarometry of Alpe Arami and other Garnet Peridotites from the Central Alps

Abstract

The four, currently best constrained, independent thermobarometers for garnet peridotites, namely Taylor's (Neues Jahrbuch für Mineralogie, Abhandlungen 172, 381-408, 1998) pyroxene solvus and Krogh's (Contributions to Mineralogy and Petrology 99, 44-48, 1988) clinopyroxene-garnet Fe-Mg exchange thermometers, and Taylor's (1998) Al-in-orthopyroxene and Nimis & Taylor's (Contributions to Mineralogy and Petrology, 139, 541-544, 2000) Cr-in-clinopyroxene barometers, have been applied to garnet lherzolites from the Central Alps. Analyses from the literature, as well as new in-house analyses, all pertaining to core compositions of first-generation, garnet lherzolite minerals, have been selected for thermobarometric calculations. The P-T data obtained for the three known garnet lherzolite occurrences in the Central Alps are tightly constrained, consistent with one another, and summarized as follows: Alpe Arami, 3·2 GPa and 840°C; Monte Duria, 3·0 GPa and 830°C; Cima di Gagnone, 3·0 GPa and 740°C. These values are consistent with experimental data on pargasite stability and composition in peridotitic systems. Our P-T estimates, along with microstructural and field geological observations, indicate that the garnet lherzolite parageneses form part of the prograde, Alpine, high-pressure metamorphic sequence of the Adula-Cima Lunga unit. Thermobarometry shows that the garnet lherzolites reached a maximum depth of subduction of ∼100 km, leaving little room for an extraordinary high-pressure, Alpine metamorphism at Alpe Arami. The very high pressure estimates obtained by some other workers are ascribed to inaccuracies in thermometric evaluations and to the strong temperature dependence of the Al-in-Opx barometer. Abstract

Rocks of the Cerro del Almirez ultramafic massif (Sierra Nevada, Betic Cordillera, S. Spain) record the high-pressure dehydration of antigorite-olivine serpentinite to form chlorite harzburgite (ol + opx + chl). In the field, these two rock types are separated by a well-defined isograd. Titanian clinohumite (TiC1) and olivine show textural and compositional differences depending on the rock type. OH-TiC1 occurs in the serpentinite as disseminated grains and in veins. F-OH-TiC1 is observed exclusively in the chlorite harzburgite, where it occurs as porphyroblastic grains and within prograde olivine as irregular and lamellar, planar intergrowths at microscopic and submicroscopic scales. Petrological evidence of partial to complete breakdown of TiCl to olivine + ilmenite is preserved in both rock types. Chlorite harzburgite is characterized by a brown pleochroic olivine with abundantally oriented microscopic to submicroscopic oxide particles. The mean Ti-content of the brown olivine is 144 ppm. The brown olivine preserves TiC1 lamellae that sometimes grade into ghost lamellae outlined by the oxide trails. This observation suggests that some of the oxide inclusions in the brown olivine are derived from the breakdown of TiC1 intergrowths. Thermodynamic modelling of selected Almirez bulk rock compositions indicates a temperature increase from 635[degrees]C to 695[degrees]C, at pressures ranging from 1.7 GPa to 2.0 GPa, as the cause for the compositional adjustment of TiCl between the Almirez antigorite serpentinite and chlorite harzburgite. These P-T estimates are in good agreement with the sequence of phase relations observed in the field. The computed phase diagrams in conjunction with the geothermal conditions envisaged for different subduction settings indicate that TiC1 is stable in the vicinity of the antigorite serpentinite/chlorite harzburgite phase boundary in some subduction settings. In these circumstances, clinohumite-olivine intergrowths in chlorite harzburgite may act as a sink for high field strength elements, and probably other elements, that are present in the mantle-wedge fluids.

Abstract

During regional metamorphism of the Malenco serpentinized peridotite (Sondrio, northern Italy), the mineral assemblage pentlandite-awaruite-magnetite-native copper-antigorite-brucite-olivine-diopside is formed. The opaque assemblage indicates very reduced fluids with fO2 values 4 log units below QFM. Primary fluid inclusions were trapped in diopside overgrowth, contemporaneous with the opaque assemblage. These metamorphic fluids are saline aqueous solutions (about 10.4 mol% NaCl equivalent) and contain molecular H2 of approximately 1 mol%, as shown by micro-Raman analysis and microthermometry. The fluids are interpreted to have been formed during deserpentinization at the olivine-in isograd under strong reducing conditions.
