The study objective is to evaluate the impact of CBD on vitamin D3 receptor (VDR) protein expressions, tissue elasticity, anti-inflammatory, and anti-senescence activity in human and rodent cell lines.Methods: Cell viability was estimated by MTT assay. Relative quantification (RQ) of VDR protein expression was measured by RT-PCR. Tissue elasticity was measured by atomic force microscopy (AFM). Cellular senescence and ATP were performed using trypan blue exclusion test and colorimetric assay, respectively.Results: Cell viability assay data showed CBD was safe and nontoxic upto 7.5 μM. The VDR protein expression was significantly increased by 109.71% (p = 0.013), 236.96% (p ≤ 0.001), 170% (p ≤ 0.001), 100% (p = 0.019), 80% (p = 0.021), 427.27% (p ≤ 0.001), 366.67% (p ≤ 0.001), 56.31% (p = 0.016), and 63.84% (p ≤ 0.001) in MG-63, MDA-MB-231, SH-SY5Y, HEK-293, HT-29, EaHy-926, HepG2, A-549, and C2C12 cells, respectively compared to the normal control group. CBD treatment significantly reduced the levels of TNF-α (46.58%; p ≤ 0.049) and IL-6 (43.61%; p ≤ 0.001) at CBD-5 μM compared to the vehicle control group. Tissue elasticity was significantly (p ≤ 0.001) increased by 37.09% and 49.49% in CBD-2.5 and CBD-5 μM, respectively, compared to the vehicle control group. Significantly (p ≤ 0.001) reduced senescence cells by 39.22% in CBD-5 μM than the vehicle control group. The level of ATP was significantly (p ≤ 0.001) increased by 90.55, 117.06, and 153.54% in CBD-1, CBD-2.5, and CBD-5 μM, respectively, compared to the vehicle control group.Conclusion: Overall, data suggest that CBD considerably improved VDR protein expression, inflammation, cell growth, tissue elasticity, and enhanced mitochondrial bioenergetics in multiple cell lines. In this study, for the first time, we showed the evidence suggesting that the VDR plays a critical and multifaceted role in various types of human cells. Angiographic retinal ischemic index (ARI) was also calculated from UWF-FA as the ratio of digitally mapped ischemic retina area against area of optic disc, termed unit of disc diameter (DD).Results: We observed emerging angiographic features common to these patients: pattern of a localized non-perfused retina at early phase of UWF-FA remaining non-perfused at the late phase (black retinal ischemia, black-RI) (100%); presence of retinal microvascular anomalies (RMAs) at the “water-shed-border” of black-RI (100%); site of NVEs observed at either the same “water-shed-border” (42%) or from the main vessel branch within the black-RI (30%), or from both sites (28%); multiple NVEs were observed in all eyes except two with single active NVE. Median ARI size was 114 DD (SD 80 DD), range 5– 354 DD.Conclusion: We report a recurring angiographic pattern common to eyes with active BRVO-NVEs from UWF-FA, and NVEs in this clinical group can develop from varied ARI sizes. Abstract

We demonstrate optical orientation in Ge/SiGe quantum wells and study their spin properties. The ultrafast electron transfer from the center of the Brillouin zone to its edge allows us to achieve high spin-polarization efficiencies and to resolve the spin dynamics of holes and electrons. The circular polarization degree of the direct-gap photoluminescence exceeds the theoretical bulk limit, yielding ~37% and ~85% for transitions with heavy and light holes states, respectively. The spin lifetime of holes at the top of the valence band is found to be ~0.5 ps and it is governed by transitions between heavy and light hole states. Electrons at the bottom of the conduction band, on the other hand, have a spin lifetime that exceeds 5 ns below 150 K. Theoretical analysis of the electrons spin relaxation indicates that phonon-induced intervalley scattering dictates the spin lifetime., Comment: 5 pages, 3 figures

11. Early Fatherhood: A Mapping of the Evidence Base Relating to Pregnancy Prevention and Parenting Support

Abstract

Teenage pregnancy prevention programmes targeted at young women have received considerable attention from researchers and programme developers. However, to date, relatively limited information is available on preventing teenage fatherhood or improving outcomes for young fathers. A notable gap is concerned with understanding the forms of sexual health programmes that are most effective from the perspective of young men. We conducted a systematic mapping to identify studies involving young men aimed at preventing teenage pregnancy, improving outcomes for teenage fathers or exploring the perspectives of young men around pregnancy and fatherhood. We searched a wide range of electronic databases from January 1996 to August 2008. Three quantitative and 15 qualitative studies were identified, of which nine were UK based. Key themes related to the inappropriateness of current sexual health promotion to respond to the needs of young men. While young men often possessed very similar ideals to young women, existing programmes were problematic when they negatively stereotyped young men and ineffectively addressed models of masculinity or the difficulties young men may have forming meaningful relationships. Further investigations are required on programme development for young men, particularly on sexual health promotion interventions for "looked-after" young men and those from unstable childhoods.

Abstract

PURPOSE: To assess repeatability and agreement of a Scheimpflug topographer and a Hartmann-Shack aberrometer with a ray-tracing aberrometer in normal, keratoconus, and corneal cross-linking (CXL) groups. METHODS: In this cross-sectional, observational study, normal, keratoconus, and CXL groups with 125 eyes in each of the three groups were included. All eyes underwent three sets of measurements, at a single visit, using the Pentacam AXL Wave (Oculus Optikgeräte GmbH) and iTrace (Tracey Technologies). Keratometry, pachymetry, objective refraction, and total ocular aberrations including root mean square (RMS) lower order aberrations (LOAs), RMS higher order aberrations (HOAs), RMS coma, and spherical aberrations (SA) were analyzed. Objective assessment of haze after CXL was done using the Pentacam AXL Wave. Repeatability was assessed using within-subject standard deviation (Sw), test–retest variability, within-subject coefficient of variation (COV), and intraclass correlation coefficient (ICC). Bland-Altman analysis assessed 95% limits of agreement and correlation coefficient ( r ) determined the strength of the relationship between measurements. RESULTS: The Pentacam AXL Wave had Sw for keratometry of 0.12 in the normal group and 0.15 in the keratoconus group and lower (poorer) Sw of 0.17 in the CXL group. For pachymetry, Sw was 9.18, 9.53, and 10.11 in the normal, keratoconus, and CXL groups, respectively. Total aberrations had ICCs ranging from 0.88 for RMS HOAs to 0.95 for SA in the normal group, 0.86 for RMS HOAs to 0.92 for SA in the keratoconus group, and 0.72 for RMS HOAs to 0.82 for SA (poorer) in the CXL group. In the normal group, mean differences between the two devices were nonsignificant for all parameters except SA (0.011 ± 0.038 μm, P < .01; limits of agreement = −0.09 to 0.07; r = 0.87). In the keratoconus group, mean differences were significant in all aberrations except RMS LOAs (−0.27 ± 0.85 μm, P = .10; limits of agreement = −3.3 to 3.8; r = 0.92). In the CXL group, all parameters varied significantly ( P < .006). CONCLUSIONS: The Pentacam AXL Wave showed comparable repeatability in the normal and keratoconus groups, but was poorer in the CXL group, more so with increasing corneal haze. 20. Digital health technology for non-motor symptoms in people with Parkinson's disease: Futile or future?

Item does not contain fulltext, INTRODUCTION: There is an ongoing digital revolution in the field of Parkinson's disease (PD) for the objective measurement of motor aspects, to be used in clinical trials and possibly support therapeutic choices. The focus of remote technologies is now also slowly shifting towards the broad but more "hidden" spectrum of non-motor symptoms (NMS). METHODS: A narrative review of digital health technologies for measuring NMS in people with PD was conducted. These digital technologies were defined as assessment tools for NMS offered remotely in the form of a wearable, downloadable as a mobile app, or any other objective measurement of NMS in PD that did not require a hospital visit and could be performed remotely. Searches were performed using peer-reviewed literature indexed databases (MEDLINE, Embase, PsycINFO, Cochrane Database of Systematic Reviews, Cochrane CENTRAL Register of Controlled Trials), as well as Google and Google Scholar. RESULTS: Eighteen studies deploying digital health technology in PD were identified, for example for the measurement of sleep disorders, cognitive dysfunction and orthostatic hypotension. In addition, we describe promising developments in other conditions that could be translated for use in PD. CONCLUSION: Unlike motor symptoms, non-motor features of PD are difficult to measure directly using remote digital technologies. Nonetheless, it is currently possible to reliably measure several NMS and further digital technology developments are underway to offer further capture of often under-reported and under-recognised NMS.

21. The influence of a second ground electrode on hydrogen peroxide production from an atmospheric pressure argon plasma jet and correlation to antibacterial efficacy and mammalian cell cytotoxicity

Abstract

This study investigates how addition of a 2nd ground electrode in an argon plasma jet influences the production of hydrogen peroxide (H2O2) in deionised water (DIW). Briefly, plasma is ignited by purging argon gas through a quartz tube at 1 l min-1 and applying a sinusoidal voltage of 7 kV (peak-peak) at 23.5 kHz to a high voltage stainless steel needle electrode sealed inside the quartz tube surrounded by one or two copper ring(s) that served as the ground electrode(s) situated downstream of the high voltage electrode. The mechanisms of H2O2 production are investigated through the electrical and optical plasma properties and chemical analysis of the treated DIW. We discover that the addition of a 2nd ground electrode results in higher accumulation of charges on the inner wall surface of the quartz tube of the plasma jet assembly resulting in an increase in the discharge current and dissipated power. This further leads to an increase in the electron temperature that more than doubles the H2O2 production through dissociative recombination of water vapour molecules, whilst still maintaining a biological tissue tolerable gas temperature. The double ground electrode plasma jet is shown to be highly effective at reducing the growth of common wound pathogens (Pseudomonas aeruginosa and Staphylococcus aureus) in both planktonic and biofilm states whilst inducing a low level of cytotoxicity in HaCaT keratinocyte skin-like cells under certain conditions. The information provided in this study is useful in understanding the complex physicochemical processes that influence H2O2 production in plasma jets, which is needed to optimise the development of plasma sources for clinical applications.

Abstract

Activated ion mobility measurements provide Insights to the stability of tertiary and quaternary structures of proteins and pairing such approaches with fragmentation can delineate which part(s) of the primary sequence are disrupted from a folded structure. In this work we use 213 nm photodissociation coupled with ion mobility mass spectrometry and collisional activation to determine the conformational landscape of model proteins. UVPD experiments are performed on proteins following in source activation as well as on collisionally activated photoproducts post ion mobility separation. For cytochrome c, there is a significant increase in the fragmentation yield with collisional activation post mobility, for all conformational states. Similar strategies are deployed with the model multimeric proteins, concanavalin a, and haemoglobin. For these complexes’ CID leads to classic asymmetric charge distribution in subunit products, which when preceded by UV irradiation yields fragments from within the sub-unit that can be mapped to the quaternary fold. Data driven, multivariate analysis (MVA) was used to determine the significant differences in UVPD and CID fragmentation pattern following in source activation. This data driven approach reveals diagnostic fragments without a priori assignments limited to predicated backbone cleavage and provides a new approach to map conformation landscapes that may have wider utility.
