13. Functional characterisation of LEA proteins from bdelloid rotifers

Abstract

‘Anhydrobiosis’ or ‘life without water’ is a fascinating phenomenon that was first described by the eminent Dutch microscopist Antony van Leewenhoek in bdelloid rotifers in 1702. Despite being studied for over three hundred years, our understanding of its molecular basis remains largely elusive. Recently, two group 3 LEA (late embryogenesis abundant) proteins, ArLEA1A and ArLEA1B, have been identified in the desiccation-induced gene set in the bdelloid species Adineta ricciae that are hypothesised to protect these animals by preventing protein aggregation and stabilising membranes respectively. In this dissertation, the functional characteristics of bdelloid LEA proteins have been further explored using computational and experimental tools. Analysis of their phylogeny and domain composition reveals that ArLEA1A and ArLEA1B are evolutionarily distinct from other related group 3 LEA proteins. Moreover, unlike some LEA proteins that are unstructured, bdelloid LEA proteins are predicted to form a-helices, with ArLEA1B having an additional propensity to polymerise into tropomyosin like filaments. These proteins are also predicted to localise in the ER (endoplasmic reticulum) and interact with cell signalling molecules. Intracellular localisation analysis of ArLEA1A and ArLEA1B using confocal microscopy confirms that these proteins translocate into the ER as predicted and their distribution within the entire secretory system and the cell exterior is regulated by their N- and C-terminal signals. Preliminary results suggest that over-expressing ArLEA1A fails to provide protection against protein aggregation within the mammalian ER. Lastly, both ArLEA1A and ArLEA1B are found to provide partial protection against desiccation induced damage of fluorescence properties of the red fluorescent protein mCherry. Overall, the results in this dissertation provide important mechanistic insights into the mode of action of bdelloid LEA proteins during anhydrobiosis.

33. A study on chromium accumulation in Labeo rohita in the river Yamuna ecosystem in Mathura-Agra region in Uttar Pradesh, India

Abstract

The present study revealed the chromium toxicity and its health measures in L. rohita from the Yamuna river at Mathura- Agra region. Samples were taken in triplicate from both sites i.e. Vrindavan (Bihar ghat) and Agra (Renuka Ghat). The study was carried out on Four different organs (i.e. gills, muscles, liver, and kidney) of the fish sample. The sampling was done from Oct 2018 to January 2020. Chromium concentration in different organs of the fish was analyzed by Atomic absorption spectrophotometer(AAS). The average Cr concentration in gills was highest (9.64  mg/l) at the Mathura site followed byAgra sites (7.78 mg/l) for the month of April 2019. The concentration of Cr was highest in samples taken in the month of April 2019 and it was lowest in October 2018. The significantly high Cr concentration values were observed in the Mathura region than the Agra region for both seasons. In all samples, Cr concentration was above the standards stated by WHO except in the Kidney. In the present study, the bioaccumulation factor showed the chromium concentration in the tissues followed the order of gill > liver > muscle > kidney. HPI, MQI, and Pearson’s correlation coefficient analysis were also done in which HPI was observed very high and there was a positive correlation between all the samples.   Keywords:  Chromium, BCF, HPI, MQI, AAS, Fish Organs

Abstract

The present study was conducted on the experimental animals, Macrobrachium dayanum (Henderson) (Crustacea: Decapoda) to analyze the seasonal changes in reproductive cycle of female, found at different locations in river Gomti (26°55´; 80°59´ E), in and around Lucknow (U.P. India). The reproductive activities were found in cyclic manner in M.dayanum, which breeds continuously throughout the year but with two distinct peaks one major in the month of May-July and one minor in the month of January – Feburary. Five stages of Oogenesis i.e. proliferative, meiotic, previtellogenic, vitellogenic and resorbtion or atretic oocyte phase have been recorded in M. dayanum. Mature ova were noticed in two peaks i.e. major in month of July – August and September and minor in months of February and March. Histoarchitecture of hepatopancreas showed seasonal changes in accordance with the gonadal development. The reproductive cycle of M.dayanum is reported for the first time and very little work is known on the seasonal changes with respect to reproduction and its correlation with hepatopancreas.
