AGEs, Advanced Glycation End Products, are heterogeneous compounds with pro-oxidant and pro-inflammatory properties. These compounds are formed in the body (particularly under conditions of hyperglycemia and oxidative stress) and in environment, being the diet their main source exogenous. Neutral or alkaline food, subjected heat-treated at temperatures above 100°C with low humidity, for prolonged periods had increased formation of AGEs, conditions frequently encountered in the typical Western diet, which is a source of high levels of AGEs. The heat treatment of food, in addition to form AGEs, contributes to reducing labile vitamin with anti-glycation action and essential for humans, thiamin. The chronic consumption of diet rich in AGEs and deficient in thiamin are conditions that promote oxidative stress, carbonyl stress and persisted inflammation, contributing to the aging process with the development of several chronic diseases. When absorbed, dietary AGEs are added to those produced in the body increasing your body pool, through the kidney and liver, undergo catabolism and degradation to be eliminated. In particular, the liver is the main organ of clearance and catabolism of AGEs, becoming an easy target for the damaging effects of AGEs. In cases of thiamin deficiency, the hepatocyte is more susceptible to glyoxal, a carbonyl highly reactive precursor of AGEs, and AGEs themselves, with consequent loss of glyoxal metabolism in the liver. Supplementation of thiamin results in cytoprotection, restoration of the glyoxal detoxification, reduction of lipid peroxidation and the formation of reactive oxygen species. These elements led to the investigation of the effects of chronic consumption of thermolyzed diet, experimental model of high intake of AGEs, associated with different levels of thiamin in morpho-functional aspects of liver tissue in adult rats. For this purpose, male Wistar rats, five months old, were divided into four experimental groups according to the offered diets: standard commercial diet, control (C); thermolyzed (T), standard diet autoclaved at 121.5°C for 30 min; thermolyzed with level 1 thiamin (TT1), thiamin 6 mg/kg of diet; and thermolyzed with level 2 thiamin (TT2), 120 mg thiamin/kg of diet. After 23 weeks of treatment, animals were anesthetized and proceeded to collect samples of liver and blood, to histological analysis of hepatic tissue and tests serum biochemical of hepatic function and injury. The chronic consumption of thermolyzed diet did not promote the appearance of morphological and functional alterations in adult rat blood and liver. It is possible to consider the composition of base diet used in the experiment may have contributed to the results obtained. This study draws attention to the importance of studies to clarify the involvement of different components food for the formation of AGEs in food, which contribute to the establishment of recommendations for promoting a healthy diet, contributing to the prevention chronic diseases, including those affecting the hepatic tissue. Estes compostos são formados no organismo (especialmente em condições de hiperglicemia e estresse oxidativo) e no ambiente, sendo a dieta sua principal fonte exógena. Alimentos neutros ou alcalinos, submetidos a tratamento térmico com temperaturas superiores a 100ºC com baixa umidade, por tempo prolongado, apresentam formação aumentada de AGEs, condições encontradas com freqüência na dieta típica do ocidente, que constitui fonte de alto teor de AGEs. O tratamento térmico dos alimentos, além de formar AGEs, contribui para a redução da vitamina termolábil com ação antiglicante e essencial para seres humanos, tiamina. O consumo crônico de dieta rica em AGEs e deficiente em tiamina são condições que favorecem o estresse oxidativo, o estresse carbonílico e a inflamação persistentes, contribuindo para o processo de envelhecimento junto ao desenvolvimento de diversas doenças crônicas. Quando absorvidos, os AGEs dietéticos somam-se aos produzidos no organismo aumentando seu pool corporal, passando pelos rins e pelo fígado, sofrem catabolismo e degradação para serem eliminados. Particularmente, o fígado é o principal órgão de clearance e catabolismo de AGEs, tornando-se um alvo fácil para os efeitos prejudiciais dos AGEs. Em situações de deficiência de tiamina, o hepatócito fica mais susceptível ao glioxal, uma carbonila altamente reativa, precursora dos AGEs, e aos próprios AGEs, com conseqüente prejuízo do metabolismo do glioxal no fígado. A suplementação de tiamina resulta em citoproteção, restauração da detoxificação do glioxal, redução da peroxidação lipídica e da formação de espécies reativas de oxigênio. Esses elementos motivaram a investigação dos efeitos do consumo crônico de dieta termolizada, modelo de experimental de elevada ingestão de AGEs, associado a diferentes níveis de tiamina, em aspectos morfo-funcionais do tecido hepático em ratos adultos. Para tanto, ratos machos Wistar, de cinco meses de idade, foram subdivididos em quatro grupos experimentais, segundo as dietas oferecidas: dieta comercial padrão, controle (C); termolizada (T), dieta padrão autoclavada a 121,5ºC, por 30 min; termolizada com tiamina nível 1 (TT1), 6 mg de tiamina/Kg de dieta; e termolizada com tiamina nível 2 (TT2), 120 mg de tiamina/Kg de dieta. Após 23 semanas de tratamento, os animais foram anestesiados e se procedeu à coleta de amostras de fígado e de sangue, para análise histológica do tecido hepático e determinações bioquímicas séricas de função e lesão hepática. O consumo crônico de dieta termolizada não promoveu o aparecimento de alterações morfo-funcionais sanguíneas e hepáticas em ratos adultos. É possível considerar que a composição da dieta de base utilizada no experimento pode ter contribuído para os resultados obtidos. O presente estudo chama a atenção para a importância da realização de estudos que esclareçam a participação dos diferentes componentes alimentares para a formação de AGEs em alimentos, o que contribuiria para o estabelecimento de recomendações dirigidas para a promoção de uma alimentação saudável, concorrendo para a prevenção de doenças crônicas, incluindo as que acometem o tecido hepático.

Epilepsy is a chronic disturbance of brain function characterized by the presence of recurrent and spontaneous seizures, making one of the most frequent and severe neurological diseases, affecting approximately 50 million people worldwide, mainly children. Among these, 40% had antiepileptic drug refractory seizures. Non-drug options as surgery, vagus nerve stimulation and the ketogenic diet are timely. This diet, used since 1921 in treatment of drug-resistant epilepsy is characterized by a high concentration of lipids and often by low concentration of carbohydrates and proteins. Traditionally, the ketogenic diet use long chain triacylglycerols (LCT), however, the medium chain triacylglycerols (MCT) are considered an alternative substrate for promoting faster ketonemia-ketonuria. The oil from coconut (Cocos nucifera L.) is a natural source of MCT, and is used for several purposes, including therapeutic. This dissertation aims to investigate the effects of the ketogenic diet based on coconut oil and soybean oil on epileptic seizures in rats, consists of a review chapter, entitled Drug-resistant epilepsies: an emphasis on treatment ketogenic, and an article from the results, entitled Effects of ketogenic diet based on coconut oil on the seizures of rats with epilepsy induced by pilocarpine. The article deals an experimental study conducted in Wistar rats, divided into three groups (n=10), named according to the diet received, in Control (standard diet AIN-93G), CetoTAGcoco (ketogenic diet based in coconut oil; AIN-93G diet modified to contain 7% soybean oil, 22.79% of extra virgin coconut oil and 40% margarine) and CetoTAGsoja (ketogenic diet based in soybean oil; AIN-93G diet modified to contain 29.79% of soybean oil and 40% margarine). The ratio lipid:carbohydrate+protein of ketogenic diets was 3.5:1 (control diet, 1:11.8). The experimental period lasted 19 days. The animals fed ketogenic diets showed food intake (g) below, however, dispend energetic and weight gain similar to Control group. The behavioral analysis showed that the experimental groups did not differ in frequency and duration of seizures, however, the animals in group CetoTAGcoco had shorter duration of seizures in the 19th day, that the Control group (0.00±0.00 against 22.78±2.95, respectively, P

Dietary lipids determine the incorporation and fatty acid composition of the organism, especially in organs most susceptible to its modulating action on membrane phospholipids esterified fatty acids, such as the liver. Ketogenic diets, widely used to treat various disorders, often use long-chain triacylglycerols, however, medium chain triglycerides are considered an alternative source, since they are digested, absorbed, oxidized and removed from circulation more rapidly than long chain triacylglycerols. In addition, it lead to the formation of ketone bodies quickly, without the influence of hormonal or nutritional factors. To contribute to the discussion, this paper presents a review chapter, entitled Mitochondrial dysfunction in nonalcoholic fatty liver disease, which gathers information on oxidative stress and consequent mitochondrial dysfunction in the context of liver disease. It also presents an original article, entitled Fatty acid profile in liver tissue of rats subjected to ketogenic diet rich in trienantin, which discusses the effects of a ketogenic diet based on trienantin, a homogeneous triacylglycerol composed of enantic (heptanoic) acid, or based on soybean oil on this profile. The livers of animals fed the ketogenic diet showed a significant reduction in the concentration of monounsaturated fatty acids, 9-hexadecenoic and 9-octadecenoic compared to control group, suggesting a potential reduction in the activity of the enzyme Stearoil-CoA Desaturase 1. Additionally, the group fed trienantin showed increased levels of octadecanoic acid compared to control group, which corroborated the hypothesis of decreased enzyme activity in this group. Thus trienantin in ketogenic proportions exerted modulatory effects similar to soybean oil, widely used conventional oil, despite the differences in fatty acid composition between these two fat sources. The study showed that rats fed a ketogenic diet experienced modulation of fatty acid profile of hepatic lipids and that in the conditions set forth herein, this modulation was due especially to the nature of the ketogenic diet itself and not to the type of oil used in their preparation. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Os lipídios dietéticos determinam a incorporação e a composição em ácidos graxos do organismo, especialmente em órgãos mais susceptíveis a sua ação moduladora sobre os ácidos graxos esterificados a fosfolipídios de membrana, como, por exemplo, o fígado. Dietas cetogênicas, amplamente utilizadas para o tratamento de diversas desordens, normalmente utilizam triacilgliceróis de cadeia longa, porém, triacilgliceróis de cadeia média são considerados uma fonte alternativa, visto que são digeridos, absorvidos, oxidados e removidos da circulação mais rapidamente que os de cadeia longa. Além disso, levam à formação de corpos cetônicos de forma acelerada, não sofrendo a influência de fatores hormonais ou nutricionais. Visando contribuir com a discussão, a presente dissertação apresenta um capítulo de revisão, intitulado Disfunção mitocondrial na doença do fígado gorduroso não alcoólica, que reúne informações sobre o estresse oxidativo e a conseqüente disfunção mitocondrial, no contexto desta doença hepática. E, um artigo original, intitulado Perfil em ácidos graxos do tecido hepático de ratos submetidos à dieta cetogênica rica em trienantina, que discute os efeitos de dieta cetogênica à base de trienantina, um triacilglicerol homogêneo do ácido enântico (heptanóico), ou de óleo de soja, sobre tal perfil. Os fígados dos animais alimentados com as dietas cetogênicas apresentaram redução significativa na concentração de ácidos graxos monoinsaturados, 9-hexadecenóico e 9-octadecenóico, quando comparados ao grupo Controle, sugerindo uma possível redução na atividade da enzima Estearoil-CoA Dessaturase 1. Adicionalmente, o grupo alimentado com trienantina apresentou aumento dos níveis de ácido octadecanóico, quando comparado ao grupo Controle. Assim, a trienantina, em proporções cetogênicas, exerceu efeito modulador similar ao do óleo de soja, um óleo convencional largamente utilizado, apesar das diferenças na composição em ácidos graxos entre essas duas fontes lipídicas. Neste estudo constatou-se que os ratos alimentados com dieta cetogênica sofreram modulação do perfil em ácidos graxos dos lipídios hepáticos e que, nas condições aqui estabelecidas, tal modulação se deveu especialmente à natureza cetogênica da dieta per si e não ao tipo de óleo empregado em sua elaboração.

Many diets of different compositions are available for use in animal experiments and have been used as standard, but they may induce adverse metabolic effects, compromising the comparison between the results of several studies. The literature records many reports of changes related to the use of these diets, however it lacks studies that compare metabolic effects of consumption of the different standard diets in animal experiments. Accordingly, the objective of this dissertation was to evaluate the metabolic effects of the consumption of diets considered standard widely used in animal research, being presented in the form of two articles: a review of the literature that gathers evidence yet little discussed by the scientific community on the feeding of laboratory animals; the second article refers to an experimental study with rats newly weaned, who received two types of diets: a commercial cereal-based, Nuvilab®, and another purified proposal by the American Institute of Nutrition, the AIN-93. Under the experimental conditions established, coefficients of protein and feeding efficiency presented significantly higher in group AIN-93 than in group Nuvilab®. The AIN-93 showed significantly higher lipid and protein digestibility than Nuvilab®. The different diets did not cause weight difference evolution of animals and histological analysis to the optical microscope of the kidneys, heart, spleen, stomach and small intestine showed no changes in the structures of these bodies, despite the different treatments. Animals fed the AIN-93 diet, regardless of age, had hepatic steatosis in frequency significantly higher than the animals that received the commercial Nuvilab®. The different diets did not cause influence on the absolute and relative weights of organs of animals, except for the absolute weight of the liver among younger animals and relative weight of the intestine among older animals. There was no influence of different diets on biochemical parameters evaluated, and the differences detected possibly resulting from the interaction between age and length of exposure of animals to diets. The markers of damage of kidney and liver function were similar and serum creatinine varied according to age. It was shown that both diets, AIN-93 and Nuvilab ®, are able to promote the growth of rats for a period of study considered subchronic. However, the occurrence of hepatic steatosis in animals fed the AIN-93 diet in pellets, reinforces the importance of tracking the standard protocols of experimentation and is indicative of nutritional inadequacies by imposing the need for further investigations to clarify which components or characteristics of this diet, widely used in animal experiments, may have contributed to this result. For instance, it is suggested that diets in the form of flour are used preferably those pellets, particularly on protocols to investigate metabolic effects. Fundação de Amparo a Pesquisa do Estado de Alagoas Várias dietas de diferentes composições estão disponíveis para o uso em experimentação animal e têm sido utilizadas como padrão, mas podem induzir efeitos metabólicos distintos, comprometendo a comparação entre os resultados dos diversos estudos. A literatura registra inúmeros relatos de alterações relacionadas ao uso dessas dietas, porém, há carência de estudos que comparem os efeitos metabólicos do consumo das diferentes dietas padrão utilizadas em experimentos animais. Assim sendo, o objetivo da presente dissertação foi avaliar as repercussões metabólicas do consumo de dietas consideradas padrão, amplamente utilizadas na pesquisa animal, sendo apresentada na forma de dois artigos: uma revisão da literatura, que reúne evidências ainda pouco debatidas pela comunidade científica relativas à alimentação de animais de laboratório, e um segundo artigo, que se refere a um estudo experimental com ratos Wistar recémdesmamados, que receberam dois tipos de dieta: uma comercial à base de cereais, Nuvilab®, e outra purificada proposta pelo American Institute of Nutrition, a AIN-93. Nas condições experimentais estabelecidas, coeficientes de eficiências protéica e alimentar apresentaram-se significativamente maiores no grupo AIN-93 que no grupo Nuvilab®. A AIN-93 apresentou digestibilidades lipídica e protéica significativamente maiores que a Nuvilab®. As diferentes dietas não causaram diferença na evolução ponderal dos animais e a análise histológica ao microscópio óptico dos rins, coração, baço, estômago e intestinos não evidenciou alterações nas estruturas desses órgãos, apesar dos diferentes tratamentos. Os animais alimentados com a dieta AIN-93, independente da idade, apresentaram esteatose hepática em uma frequência significativamente maior que os animais que receberam a comercial Nuvilab®. As diferentes dietas não exerceram influência sobre os pesos absoluto e relativo dos órgãos dos animais, com exceção do peso absoluto do fígado, entre os animais mais jovens, e do peso relativo do intestino, entre os animais mais velhos. Não se observou influência das diferentes dietas sobre os parâmetros bioquímicos avaliados, sendo as diferenças detectadas possivelmente resultantes da interação entre a idade e o tempo de exposição dos animais às dietas. Os marcadores de lesão e função hepática e renal foram similares e a creatinina sérica variou em função da idade. Demonstrou-se que ambas as dietas, AIN-93 e Nuvilab®, são capazes de promover o crescimento de ratos Wistar por um período de estudo considerado subcrônico. Porém, a ocorrência de esteatose hepática nos animais alimentados com a dieta AIN-93 peletizada, reforça a importância de monitoramento de protocolos padrão de experimentação e é indicativa de inadequações nutricionais, impondo a necessidade de investigações adicionais para esclarecer que componentes ou características dessa dieta, amplamente utilizada em experimentação animal, podem ter contribuído para tal resultado. Por hora, sugere-se que a dieta AIN-93 seja oferecida preferencialmente na forma de farinha, particularmente em protocolos que investiguem efeitos metabólicos.
