The distribution and potential bioaccumulation of dietary and waterborne cadmium and lead in tissues of sea bream (Sparus aurata), a major aquaculture species, was studied in relation to three different fish farming systems. Metallothionein levels in fish tissues were also evaluated. Results demonstrate that metal concentrations in various tissues significantly vary among fish culture systems. Different tissues show different capacity for accumulating heavy metals. The content of both cadmium and lead is not strictly correlated with that of metallothionein. Indeed, the marked accumulation of both metals in liver, as well as the high lead content found in gills and kidney, are not accompanied by a concomitant accumulation of metallothioneins in these tissues. No correlation is present between heavy metals and metallothionein content in muscle tissue. The results also demonstrate that cadmium accumulates mainly via dietary food, whereas lead accumulation is not of food origin. Noteworthy is that the concentration of the two metals found in muscle in all instances is lower than the limits established by European Union legislation for fish destined for human consumption.

DOSI CITOTOSSICHE DI MANGANESE ED EFFETTI SULLO SVILUPPO EMBRIONALE. E’ un elemento in traccia appartenente alla categoria dei nutrienti o metalli essenziali; utilizzato ed accumulato da tutte le forme di vita, è coinvolto nel normale funzionamento di meccanismi cellulari come la replicazione, la mineralizzazione e la protezione cellulare. D’altro canto, se la sua presenza è fondamentale per la vita, l’esposizione di cellule/organismi ad elevate concentrazioni di Mn causa tossicità. Il manganese rappresenta oggi, un “nuovo” importante fattore di contaminazione ambientale, conseguenza dell’intensa attività antropica dell’ultimo secolo. Il trattamento di embrioni di riccio di mare della specie Paracentrotus lividus con dosi citotossiche di Mn influisce sullo sviluppo embrionale producendo un alternativo morfotipo di sviluppo, attivando le principali MAPK (ERK e p38) coinvolte sia nella regolazione dello sviluppo che nello stress, provocando una modulazione nei livelli di sintesi delle principali proteine da stress HSC70 e HSC60. Nello specifico, troviamo che dosi crescenti di Mn (da 7,7 a 61,6 mg/L) provocano ritardi e malformazioni nello sviluppo embrionale secondo un andamento dose-effetto, chiaramente caratterizzato dall’aumento in frequenza del numero di embrioni con totale assenza o ridotto allungamento dello scheletro di CaCO3 (spicole). Il processo di spicologenesi sembra comunque ripristinarsi quando il Mn viene allontanato dal mezzo di coltura, per i diversi tempi di trattamento/ sviluppo, anche se dopo 40 ore di trattamento gli embrioni non riescono a recuperare il corretto disegno della struttura scheletrica. Mediante l’uso di marcatori territorio-specifici per i tre foglietti embrionali già differenziati in ecto-, meso- ed endoderma (anticorpi monoclonali Ecto V, UH2-95, 1D5) è stato osservato che tutti gli antigeni specifici per i vari territori sono presenti, sia negli embrioni controllo che negli embrioni trattati (61,6 mg Mn/L), in una posizione spaziale e per un tempo di sviluppo appropriato. Invece, le ibridazioni in situ per i trascritti codificanti per msp130, sm30 ed sm50 (geni espressi esclusivamente nelle cellule deputate alla secrezione delle spicole, PMC), hanno messo in evidenza, per i trattati, una forte inibizione nei livelli di espressione di sm30, l’unico dei tre geni che risponde a segnali mediati dalla matrice extracellulare. Inoltre, ERK e p38 rimangono nella forma fosforilata per tutto lo sviluppo degli embrioni trattati: questo risultato potrebbe dipendere da una quantità di calcio fisiologico non sufficiente a regolare gli eventi di fosfo-/defosforilazione delle MAPK, dovuto all’accumulo di Mn negli embrioni (AAS analisi). Per concludere l’accumulo di Mn negli embrioni di P. lividus stimola una duplice soluzione adattativa: attivazione di una risposta da stress e regolazione di pathways alternativi per consentire il proseguo dello sviluppo. http://www.unisi.it/eventi/gei/programma.pdf

Apoptosis is a genetic program of cell death that eliminates superfluous or compromised cells during development and adult life of many organisms. In sea urchin embryos, apoptosis is not only a physiological event during larval metamorphosis, but also a process induced by cadmium accumulation or other stressor like TPA (12-O-tetradecanoylphorbol-13-acetate) followed by an increase of temperature to 31°C. Apoptosis is a highly conserved process usually operated by a proteolytic cascade that involves caspase activation by two different pathways: extrinsic and intrinsic. The first one involves membrane death receptors, while the second involves mitochondria. In this work we analyzed the possible involvement of extrinsic and intrinsic apoptotic pathways in physiological and stressful conditions in Paracentrotus lividus embryos. By fluorescent TUNEL assays we demonstrate that apoptosis is part of cadmium and TPA+31°C stress response. We find that Cd and TPA+31°C treatments induce apoptosis through caspase-3 activation, while caspase-7 is the main effector of physiological apoptosis. Caspase-10 is active only in physiological apoptosis, while caspase-8 is mainly involved in stress-induced apoptosis. In addition, we did not find any involvement of mitochondria. Moreover we observed, in Cd-treated embryos, a Reactive Oxygen Species (ROS) increase, that could be related to the induction of apoptosis.

The exposure to environmental toxicants such cadmium (Cd) is an important research area in wildlife protection. In this study, the effect of Cd oral administration on the ovarian structure and function and on reproductive performance of the Italian wall lizard Podarcis sicula was studied. In vivo, adult female lizards were randomly assigned to three groups. Cd was given with food in single dose and in multiple doses 3 days/week for 4 weeks at dose of 1.0 μg/g body weight. Following euthanasia, the ovaries were removed and analyzed for morpho-functional changes. Results demonstrated that Cd increases prefollicular germ cells number; the evaluation of the number of follicles detects significantly higher number of atretic growing follicles, whereas primary follicles remain unchanged with respect to controls. After Cd treatments, follicles are deformed by the presence of large protrusions and a general dysregulation in the follicle organization is observed. The zona pellucida is also affected. Cd causes alteration in sugar metabolism and in metallothionein gene expression. Finally, Cd administration significantly reduces clutch size and dramatically increases embryo mortality. In conclusion, data here described show that Cd induces morpho-functional alterations in lizard follicles and indicates that these are responsible for a significant impairment of oogenesis. The effects of the dose are time independent, persisting essentially unchanged regardless of single or multiple administration, so it can be concluded that even occasional, sublethal Cd contamination may significantly impair reproductive performance in these animals.

Organisms living in the Southern Ocean are exposed to strong environmental constraints, especially temperature. The Perciform suborder Notothenioidei, today largely endemic to the Antarctic, has developed numerous cold-adapted characters. The most striking peculiarity of this group of fish is the drastic reduction of hemoglobin content in their blood. This condition gives rise to a low metabolic demand of iron. Recently, we have undertaken a study to add new insights on iron metabolism in hemoglobinless fish. By taking advantage to our previous studies on transferrins from Antarctic Notothenioids, in this article we compared the evolutionary properties of these sequences to those from other fish groups and mammals. Our findings based on analysis of dN/dS ratio and on the results of the relative rate ratio test suggest that the transferrin gene has undergone positive selection in fish but not in mammals. The results of type I functional divergence provide statistical evidence for shifted evolutionary rate after speciation. Finally, when superimposed onto the three-dimensional structure of transferrin, the sites identified as responsible of the divergence in Notothenioids appear to cluster preferentially at non-iron binding locations, occupying surface locations that might affect the rate of iron binding and/or release in a cold environment.

The aim of this study was to verify if the freshwater safety values established from the European Community (Council Directive 1998) and the Italian Ministry of Health (2001) for cadmium (44.5 nM/L in drinking water and 178 nM/L in sewage waters) were safe for amphibians, since at these same concentrations cadmium induced endocrine disruption in the newt Triturus carnifex. Adult male specimens of T. carnifex were exposed daily to cadmium (44.5 nM/L and 178 nM/L as CdCl2, nominal concentrations), respectively, during 3- and 9-months; at the same time, control newts were exposed to tap water only. The accumulation of cadmium in the skin, liver and kidney, the levels of metallothioneins in the skin and the liver, the expression of metallothionein mRNA in the liver, as well as the presence of histological alterations and of apoptosis in the target organs were evaluated. The 9-months exposure induced cadmium accumulation in all the tissues examined; moreover, histological changes were observed in all the tissues examined, irrespective of the dose or the time of exposure. Apoptosis was only detected in the kidney, whereas metallothioneins and metallothionein mRNA did not increase. This study demonstrates that the existing chronic water quality criterion established for cadmium induces in the newt T. carnifex cadmium accumulation and histological alterations in the target organs examined. Together with our previous results, showing that, at these same concentrations, cadmium induced endocrine disruption, the present results suggest that the existing chronic water quality criterion for cadmium appears to be not protective of amphibians.

The biological effect of seasonality on cadmium, lead and metallothionein contents was assessed in mussels Mytilus galloprovincialis from natural banks located along the coastline of the Gulf of Naples (Campania, Italy). Heavy metals and metallothionein concentrations were measured in digestive and reproductive glands. The results showed a clear correlation between metallothionein content and the reproductive gland status determined during the seasons; on the contrary, no correlation was found between metallothionein and metal contents. Data allow us to hypothesize that metallothionein functions go beyond metal detoxification, thus opening new scenarios for these proteins in invertebrates. The effect of seasons on metals concentration in mussel tissues showed similar seasonal patterns between the sites, regardless of their anthropogenic impacts. Cadmium content was not strictly related to seasonal periods, whereas lead content was significantly lower in summer. The results also indicate that the metal contents in mussels from the Gulf of Naples do not represent a risk to human health, even in the period of their maximum accumulation, and that the relaying of mussels before marketing could improve the animal stress conditions, but having a slight effect on metal excretion.

Ferritin is a major intracellular iron storage protein in higher vertebrates and plays an important role in iron metabolism. This study reports the identification from the Antarctic icefish Chionodraco rastrospinosus of a complete mRNA sequence and four partial mRNA sequences, all encoding the ferritin M subunit and sharing a clear homology with the ferritin M-chain of other fish species. The open reading frame of the complete ferritin M transcript is of 528 base pairs and encodes a protein of 176 amino acids that retains the residues involved in the ferroxidase diiron center and in the ferrihydrite nucleation center. Despite the absence of hemoglobin and of any appreciable amount of iron in the icefish blood, RT-PCR analysis shows that H and M ferritin subunits are expressed both in blood and in other tissues, such as spleen, head kidney, liver and kidney. Phylogenetic analysis shows that the H and M subunits form two well separated clusters. Basal to these two clusters emerges a heterogeneous cluster, formed by two Danio rerio M, a Salmo salar M and an Orechromis niloticus H isoforms; these forms maybe represent the heritage of ancestral forms from which arose the two major H and M subunits of the fishes.

In the marine environment increasing concentrations of bio-available compounds often result from anthropogenic activities. Among metal ions, manganese represents a new emergent factor in environmental contamination. Here, we studied the effects of manganese on Paracentrotus lividus sea urchin embryos using biological and biochemical approaches for the analysis of impact on development, tissue accumulation and stress markers. Embryos were continuously exposed from fertilization to manganese at concentrations ranging from 1.0 to 61.6 mg l(-1), monitored for developmental abnormalities at 48 h after fertilization, and used for atomic spectrometric analysis at various times from 6 to 72 h. We found that concentration- and time-dependent increases in morphological abnormalities were directly correlated to manganese accumulation, with major defects in skeleton formation at 48 h. Concurrently, we found an upregulation of the hsc70 and hsc60 stress proteins detected by immunoblotting, whereas no induction of apoptosis or ROS production was observed by TUNEL and live tests, respectively. Taken together, our findings demonstrate that the observed manganese embryo-toxicity is related to both its intracellular accumulation and misregulated homeostasis, and confirm the importance of stress proteins as protective agents in the acquisition of tolerance and resistance to apoptosis.

The possible role of metallothionein (MT) in metal homeostasis has been investigated in growing oocytes and eggs of the lizard Podarcis sicula. Chromatographic analysis does not reveal the presence of MT in both ovary and eggs, the only metal-binding proteins detected being represented by high molecular mass components. De novo synthesis of MT could be observed in the ovary of cadmium-treated lizards. A cDNA encoding MT was obtained from the liver of P. sicula by RT-PCR followed by a RACE strategy, using primers designed on consensus motifs of vertebrate MT. In spite of the lack of MT in the ovary of untreated animals, Northern blot analysis demonstrates that the maternal untranslated MT transcript is expressed constitutively in the ovary in all the periods of the ovarian cycle. MT mRNA content increases during the oocyte growth, reaching the highest level in ovulated eggs, concomitantly with the accumulation of zinc and copper. Our findings suggest that maternal MT mRNA accumulates in the egg and is translated sometime during development to cope with the future needs of the growing embryo. The appearance of MT after cadmium treatment suggests that the block that makes the oocytarian MT mRNA untranslatable is removed by the metal.
