Background: Disinfection byproducts (DBPs) as ovarian toxicants have been documented in toxicological studies. However, no human studies have explored the effects of exposure to DBPs on diminished ovarian reserve (DOR)., Objective: To assess whether urinary biomarkers of exposure to drinking-water DBPs were associated with DOR risk., Methods: A total of 311 women undergoing assisted reproductive technology were diagnosed with DOR in the Tongji Reproductive and Environmental (TREE) cohort from December 2018 to August 2021. The cases were matched to the controls with normal ovarian reserve function by age in a ratio of 1:1. Urinary trichloroacetic acid (TCAA) and dichloroacetic acid (DCAA) were quantified as biomarkers of drinking-water DBP exposures. The conditional logistic regression and restricted cubic spline (RCS) were used to explore urinary biomarkers of drinking-water DBP exposures in associations with the risk of DOR., Results: Elevated urinary DCAA levels were associated with higher DOR risk [adjusted odds ratio (OR) = 1.87; 95 % confidence interval (CI): 1.16, 3.03 for the highest vs. lowest quartiles; P for trend = 0.016]. The association was confirmed in the RCS model, with a linear dose-response curve (P for overall association = 0.029 and P for non-linear association = 0.708). The subgroup analysis by age and body mass index (BMI) showed that urinary DCAA in association with DOR risk was observed among women ≥35 years old and leaner women (BMI < 24 kg/m 2 ), but the group differences were not statistically significant. Standardised tests are often used to determine the ecotoxicity of chemicals and focus mainly on one or a few generic endpoints (e.g. mortality, growth), but information on the sub-cellular processes leading to these effects remain usually partial or missing. Flow cytometry (FCM) can be a practical tool to study the physiological responses of individual cells (such as microalgae) exposed to a stress via the use of fluorochromes and their morphology and natural autofluorescence. This work aimed to assess the effects of five chlorine-based disinfection by-products (DBPs) taken individually on growth and sub-cellular endpoints of the green microalgae Raphidocelis subcapitata. These five DBPs, characteristic of a chlorinated effluent, are the following monochloroacetic acid (MCAA), dichloroacetic acid (DCAA), trichloroacetic acid (TCAA), bromochloroacetic acid (BCAA) and 1,1-dichloropropan-2-one (1,1-DCP). Results showed that 1,1-DCP had the strongest effect on growth inhibition (EC 50 = 1.8 mg.L -1 ), followed by MCAA, TCAA, BCAA and DCAA (EC 50 of 10.1, 15.7, 27.3 and 64.5 mg.L -1 respectively). Neutral lipid content, reactive oxygen species (ROS) formation, red autofluorescence, green autofluorescence, size and intracellular complexity were significantly affected by the exposure to the five DBPs. Only mitochondrial membrane potential did not show any variation. Important cellular damages (>10%) were observed for only two of the chemicals (BCAA and 1,1-DCP) and were probably due to ROS formation. The most sensitive and informative sub-lethal parameter studied was metabolic activity (esterase activity), for which three types of response were observed. Combining all this information, an adverse outcome pathways framework was proposed to explain the effect of the targeted chemicals on R. subcapitata. Raphidophytes are flagellate unicellular algae that causes algal blooms in drinking water sources. In Japan, it was recently reported that the concentration of trichloroacetic acid (TCAA), a major chlorinated disinfection byproduct (DBP), increased dramatically in drinking water when the source water contained raphidophytes. Additionally, raphidophytes produced haloacetic acid (HAA) precursors, especially TCAA precursors, in high concentrations. However, their properties are still unknown, and thus, well-designed countermeasures against DBP formation have not yet been established. Therefore, in this study, the HAA precursors originated from raphidophytes in natural water collected from the algal blooms in Muro Dam (Nara Prefecture, Japan) and Gonyostomum semen (G. semen), a raphidophyte species, cultivated in the laboratory, were characterized to provide the information for establishing suitable treatment strategies. Using several high-performance liquid chromatography columns, solid-phase extraction cartridges, and ultrafiltration devices, and the spectral profiles, we discovered that the HAA precursors are highly hydrophilic and high-molecular-weight compounds with acidic and phenolic functional groups. Further characterization of the high-molecular-weight fraction (> 3 kDa) from the G. semen culture showed that the HAA precursors had a molecular weight of ~10-60 kDa, and that they were not protein molecules despite containing a large amount of nitrogen atoms. Furthermore, the TCAAFP of the fraction (310 ± 25 μg/mg C) were as high as phenol, known as a reactive TCAA model precursor. Water toxicity detection, as a valuable supplement to conventional water quality measurement, is an important method for evaluating water environmental quality standards. However, the toxicity of composite pollutants is more complicated due to their mixture effects. This study developed a novel, rapid and interference-resistant detection method for water toxicity based on an electrochemical biosensor using peak current from nitrite oxidation as a signal. Toxicants could weaken the characteristic peak current of nitrite to indicate the magnitude of toxicity. The proof-of-concept study was first conducted using a synthetic water sample containing trichloroacetic acid (TCAA), and then the results were compared with those of the traditional toxicity colorimetric method (CCK-8 kit) and laser confocal microscopy (CLSM). The accuracy of the biosensor was further verified with water samples containing individual pollutants such as Cd 2+ (50-150 μg/L), Cr 6+ (20-80 μg/L) mixture, triclosan (TCS; 0.1-1.0 μg/L) and TCAA (10-80 μg/L), or a mixture of the above. The viability of the sensor was further validated with the actual water sample from the Tuojiang River. The results demonstrated that although the concentration of a single conventional pollutant in water did not exceed the discharge standard for surface water, the comprehensive toxicity of natural water should not be ignored. In this study, we developed and validated a simple, fast and sensitive LC-MS/MS method for the measurement of tetrodotoxin (TTX) in human plasma. Three HILIC-type solid phase extraction (SPE) carriers (PSA, silica, Siphila i HILIX) with different stationary phase functional groups were compared. The Siphila i HILIX SPE plate containing multi-carboxyl groups was finally selected due to obviously better extraction recovery of TTX (about 80% of recovery from plasma samples) than the other two and no significant matrix effects were observed, which was speculated to have mixed-mode synergistic effects of hydrophilic interaction and ion exchange. 100μL plasma sample was precipitated rapidly with acetonitrile containing 1% trichloroacetic acid, and filtrates were loaded onto Siphila i HILIX 96 well SPE plate. After washed with 95% acetonitrile, TTX was eluted with 200μL of 50% acetonitrile containing 1% trichloroacetic acid. 2μL of elution solution was directly injected into LC-MS/MS and the total run time on a BEH amide column was 4.5 min. The method avoids the evaporation and ultrafiltration processes which is simple and timesaving (<30 min). TTX and internal standard (arginine- 15 N 4 ) were monitored in positive mode using m/z 320.3→162.2 (quantification transition for TTX), 320.3→284.1 (confirmation transition for TTX) and 179.2→63.0 (transition for IS), respectively. The method was linear in the range of 0.1-20 ng/mL for TTX with the low limit of quantification (S/N > 10) of 0.1 ng/mL; the intra- and inter-assay accuracies were in the range of 98.5%-99.8% (relative standard deviations, RSDs ≤ 5.92%) and 98.8-99.5% (RSDs ≤ 6.23%), respectively. Biases of spiking analysis were ranged from -7.00% to 7.43% for healthy human plasma samples (RSDs ≤ 8.83%) and from -5.00% to 3.93% for hemolytic, high triglyceride, high cholesterol and high bilirubin plasma samples (RSDs ≤ 6.40%), which proved the good anti-interference property of the method. Discriminative and sensitive detection of environmentally important and health-related trichloroacetic acid (TCA) suffers from various problems such as bulky instruments and time-consuming operation as well as complex sample processing. Herein, we present a rapid, sensitive, and specific method for the detection of gaseous TCA using a fluorescent single-molecule array. An o -carborane-based benzothiazole derivative (CB-BT-OCH 3 ) with specific fluorescence properties was specifically designed and utilized to fabricate a film-based single-molecule array. It was revealed that the fluorescent film is photochemically stable and extremely sensitive to TCA vapor, depicting an observable fluorescence color change from green to blue. The experimental detection limit is 0.2 ppm, which is lower than the safety limit (1 ppm) required by the threshold limit values and biological exposure indices. In addition, the film could show detectable intensity change within 0.2 s. On the basis of multiple signal responses, a conceptual two-channel-based fluorescent TCA sensor was developed. Importantly, the proposed conceptual sensor paves a new route to the development of specific fluorescent film-based sensor arrays with a single fluorophore as sensing units.

It is crucial to explore the source, formation process and interdependence of disinfection byproducts (DBPs) to reduce their risk on public health. In this investigation, a source water was chlorinated to evaluate the initial formation rates and the maximum yields of trichloromethane (TCM), dichloroacetic acid (DCAA), and trichloroacetic acid (TCAA) based on a hyperbola model. The results showed that TCM achieved the highest initial formation rate and maximum theoretical concentration compared with DCAA and TCAA. The TCM yield can be used to forecast the yields of DCAA and TCAA throughout the whole reaction process, and the yields of chloral hydrate (CH), dichloroacetonitrile (DCAN) and 1,1,1-trichloropropanone (1,1,1-TCP) within the initial reaction stage. Besides, the raw water, settled water and filtered water collected from a drinking water treatment plant were divided into five fractions, respectively, by ultrafiltration membranes to evaluate their DBP formation after chlorination. Compared with the medium molecular weight species, high and low molecular weight organic matters exhibited relatively high specific regulated and unregulated DBP yields (expressed as μg/mg C), respectively. Humic acid-like compositions predominantly contributed to regulated DBP yields, while soluble microbial by-product-like compounds preferentially generated DCAN. The correlation study revealed that the TCM could also serve as an indicator for the measured DBPs from chlorination of sample fractions with different molecular weight. In this paper cobalt oxide (Co 3 O 4 ) nanoparticles were mixed with polyacrylonitrile to prepare Co 3 O 4 doped carbon nanofiber (CNF) composite by electrospinning and carbonization, which was further used to modify on carbon ionic liquid electrode (CILE). Hemoglobin (Hb) was immobilized on Co 3 O 4 -CNF/CILE surface with Nafion acted as the protective film to fabricate an electrochemical biosensor (Nafion/Hb/Co 3 O 4 -CNF/CILE). Electrochemical behavior of Hb on the electrode was investigated with a pair of quasi-reversible redox peak appeared on cyclic voltammogram and electrochemical parameters were calculated. Moreover, this biosensor had good analytical capabilities for electrocatalytic reduction of different substrates including trichloroacetic acid, potassium bromate and sodium nitrite with wider detection range from 40.0 to 260.0 mmol L -1 , 0.1 to 48.0 mmol L -1 and 1.0 to 12.0 mmol L -1  by cyclic voltammetry, respectively. Rapid and accurate determination of disinfection byproducts (DBPs) has become an emerging need for environmental monitoring and has yet to be realized in electrochemical sensors with metal organic framework (MOF)-based materials. In this study, a highly sensitive electrochemical sensor for trichloroacetic acid (TCAA) detection based on iron(II) phthalocyanine (PcFe) and a Zn-based metal organic framework (ZIF-8) composite is fabricated. As an electrode material, ZIF-8 possesses a large surface area and porous structure, which exhibits high absorbability; meanwhile, PcFe (II), as the sensing element, undergoes a reduction process from PcFe (II) to PcFe (I) during the sensing process. In the presence of TCAA, PcFe (I) is reoxidized by TCAA, which shifts the reaction equilibrium and accelerates the electron transfer on the electrode interface. By analyzing the reduction current of PcFe (II), the quantitative detection of TCAA is realized. The sensor shows a limit of detection (LOD) of 1.89 nM, which is superior to other reported TCAA sensors, as well as a high sensitivity (826 μΑ/μM). Moreover, the good selectivity and stability of this sensing platform demonstrate its capability and promise in determination of trace DBPs. The reported sensor provides a new strategy for electrochemical detection of DBPs and could expand the applications of MOFs in emerging technologies for monitoring contaminants.

People are exposed to chlorinated by-products (CBPs) through the consumption of cold tap water (CTW) (ingestion, inhalation, dermal contact) but also through the use of hot tap water (HTW) in such activities as showering and bathing (inhalation, dermal contact). This study focuses on the impact of residential water heating on CBP levels in tap water. Trihalomethane (THM) and haloacetic acid (HAA) levels were measured in the CTW and HTW of 50 residences located in two distribution systems supplied by chlorinated surface water during summer and winter. Results show important differences between CBP levels measured in cold and hot tap water. However, the magnitude of changes differs according to the specific species of THMs and HAAs, the season, the distribution system and the location within the same distribution system. Residential water heating led to an increase in average THM levels for the two distribution systems studied, which tended to be greater in winter. Residential water heating affected the two main HAA species found in the area studied (dichloroacetic (DCAA) and trichloroacetic (TCAA) acids) differently. In fact, the average DCAA levels increased due to water residential heating while a small change in average levels was observed for TCAA. However, the water heating impact on HAAs (in terms of importance and sometimes of tendency (increase vs. decrease)) may also differed between residences. The influence of seasons on the change in the average DCAA and TCAA levels (in μg/L) from residential water heating was not statistically significant except for TCAA levels in one distribution system. Results show the importance of considering site-specific characteristics of CTW (CBP level, temperature, residual chlorine, etc.) to estimate the levels of CBPs in HTW in CBP exposure assessment studies (and not to generalize for an entire population). We are reporting a novel biosensing platform based on a hybrid nanomaterial that combines the advantages of Nafion-coated multiwalled carbon nanotubes (MWCNTs) and mesoporous silica MCM41 nanoparticles functionalized with hemoglobin (Hb). MWCNTs-MCM41-Hb hybrid bioconjugate was characterized by scanning electron microscopy (SEM), UV-vis spectroscopy and electrochemical techniques after deposition at glassy carbon electrodes (GCE). The combination of the high surface area, biocompatibility and protein loading capacity of MCM41 nanoparticles and the high surface area and catalytic properties of MWCNTs allowed the direct electron transfer (DET) between Hb and the electrode surface. The electron transfer rate constant (k) and the surface coverage of electroactive Hb (Γ Hb ) were 5.2 s -1 and 4.7 × 10 -10 mol cm -2 , respectively. The GCE modified with the nanostructured architecture (GCE/MWCNTs-MCM41-Hb) was successfully used as a third-generation biosensor for the highly sensitive and selective quantification of nitrite (NO 2 - ) and trichloroacetic acid (TCA) by taking advantage of the excellent biocatalytic activity of Hb and the efficient direct charge transfer of the heme group., (Copyright © 2018 Elsevier B.V. Objective: The aim of this study was to examine associations between craniofacial birth defects (CFDs) and disinfection by-product (DBP) exposures, including the sum of four trihalomethanes (THM4) and five haloacetic acids (HAA5) (ie, DBP9)., Methods: We calculated first trimester adjusted odds ratios (aORs) for different DBPs in a matched case-control study of 366 CFD cases in Massachusetts towns with complete 1999 to 2004 THM and HAA data., Results: We detected elevated aORs for cleft palate with DBP9 (highest quintile aOR = 3.52; 95% CI: 1.07, 11.60), HAA5, trichloroacetic acid (TCAA), and dichloroacetic acid. We detected elevated aORs for eye defects with TCAA and chloroform., Conclusion: This is the first epidemiological study of DBPs to examine eye and ear defects, as well as HAAs and CFDs. The associations for cleft palate and eye defects highlight the importance of examining specific defects and DBPs beyond THM4.

Background: Evaluation of interindividual variability is a challenging step in risk assessment. For most environmental pollutants, including perchloroethylene (PERC), experimental data are lacking, resulting in default assumptions being used to account for variability in toxicokinetics and toxicodynamics., Objective: We quantitatively examined the relationship between PERC toxicokinetics and toxicodynamics at the population level to test whether individuals with increased oxidative metabolism are be more sensitive to hepatotoxicity following PERC exposure., Methods: Male mice from 45 strains of the Collaborative Cross (CC) were orally administered a single dose of PERC (1,000 mg/kg) or vehicle (Alkamuls-EL620) and euthanized at various time points (n = 1/strain/time). Concentration–time profiles were generated for PERC and its primary oxidative metabolite trichloroacetate (TCA) in multiple tissues. Toxicodynamic phenotyping was also performed., Results: Significant variability among strains was observed in toxicokinetics of PERC and TCA in every tissue examined. Based on area under the curve (AUC), the range of liver TCA levels spanned nearly an order of magnitude (~8-fold). Expression of liver cytochrome P4502E1 did not correlate with TCA levels. Toxicodynamic phenotyping revealed an effect of PERC on bodyweight loss, induction of peroxisome proliferator activated receptor-alpha (PPARα)-regulated genes, and dysregulation of hepatic lipid homeostasis. Clustering was observed among a ) liver levels of PERC, TCA, and triglycerides; b ) TCA levels in liver and kidney; and c ) TCA levels in serum, brain, fat, and lung., Conclusions: Using the CC mouse population model, we have demonstrated a complex and highly variable relationship between PERC and TCA toxicokinetics and toxicodynamics at the population level. https://doi.org/10.1289/EHP788.

Water disinfection plays a crucial role in water safety but it is also a matter of concern as the use of disinfectants promotes the formation of disinfection by-products (DBPs). Haloacetic acids (HAAs) are one of the major classes of DBPs since they are frequently found in treated water, are ubiquitous, pervasive and have high water solubility, so a great concern emerged about their formation, occurrence and toxicity. Exposure to HAAs is influenced by consumption patterns and diet of individuals thus their bioavailability is an important parameter to the overall toxicity. In the current study the bioacessibility of the most representative HAAs (chloroacetic acid - MCAA, bromoacetic acid - MBAA, dichloroacetic acid - DCAA, dibromoacetic acid - DBAA, and trichloroacetic acid - TCAA) after simulated in vitro digestion (SIVD) in tap water and transport across Caco-2 monolayers was evaluated. Compounds were monitored in 8 points throughout the digestion phases by an optimized LC-MS/MS methodology. MCAA and MBAA were not bioaccessible after SIVD whereas DCAA, DBAA and TCAA are highly bioaccessible (85 ± 4%, 97 ± 4% and 106 ± 7% respectively). Concerning transport assays, DCAA and DBAA were highly permeable throughout the Caco-2 monolayer (apparent permeability and calculated fraction absorbed of 13.62 × 10(-6) cm/s and 90% for DCAA; and 8.82 × 10(-6) cm/s and 84% for DBAA), whereas TCAA showed no relevant permeability. In this paper a three-dimensional (3D) reduced graphene oxide (RGO) and gold (Au) composite was synthesized by electrodeposition and used for the electrode modification with carbon ionic liquid electrode (CILE) as the substrate electrode. Myoglobin (Mb) was further immobilized on the surface of 3D RGO-Au/CILE to obtain an electrochemical sensing platform. Direct electrochemistry of Mb on the modified electrode was investigated with a pair of well-defined redox waves appeared on cyclic voltammogram, indicating the realization of direct electron transfer of Mb with the modified electrode. The results can be ascribed to the presence of highly conductive 3D RGO-Au composite on the electrode surface that accelerate the electron transfer rate between the electroactive center of Mb and the electrode. The Mb modified electrode showed excellent electrocatalytic activity to the reduction of trichloroacetic acid in the concentration range from 0.2 to 36.0 mmol/L with the detection limit of 0.06 mmol/L (3σ)., (Copyright © 2015 Elsevier B.V. By using the hydrothermal method, carbon microspheres (CMS) were fabricated and used for electrode modification. The characteristics of CMS were investigated using various techniques. The biocompatible sensing platform was built by immobilizing hemoglobin (Hb) on the micrometer-sized CMS-modified electrode with a layer of chitosan membrane. On the cyclic voltammogram, a couple of quasi-reversible cathodic and anodic peaks appeared, showing that direct electrochemistry of Hb with the working electrode was achieved. The catalytic reduction peak currents of the bioelectrode to trichloroacetic acid was established in the linear range of 2.0~70.0 mmol·L(-1) accompanied by a detection limit of 0.30 mmol·L(-1) (3σ). The modified electrode displayed favorable sensitivity, good reproducibility and stability, which suggests that CMS is promising for fabricating third-generation bioelectrochemical sensors.

Exposure to the ubiquitous environmental contaminant trichloroethylene (TCE) is associated with cancer and non-cancer toxicity in both humans and rodents. Peroxisome proliferator-activated receptor-alpha (PPARα) is thought to be playing a role in liver toxicity in rodents through activation of the receptor by the TCE metabolite trichloroacetic acid (TCA). However, most studies using genetically altered mice have not assessed the potential for PPARα to alter TCE toxicokinetics, which may lead to differences in TCA internal doses and hence confound inferences as to the role of PPARα in TCE toxicity. To address this gap, male and female wild type (129S1/SvImJ), Pparα-null, and humanized PPARα (hPPARα) mice were exposed intragastrically to 400 mg/kg TCE in single-dose (2, 5 and 12 h) and repeat-dose (5 days/week, 4 weeks) studies. Interestingly, following either a single- or repeat-dose exposure to TCE, levels of TCA in liver and kidney were lower in Pparα-null and hPPARα mice as compared with those in wild type mice. Levels of trichloroethanol (TCOH) were similar in all strains. TCE-exposed male mice consistently had higher levels of TCA and TCOH in all tissues compared with females. Additionally, in both single- and repeat-dose studies, a similar degree of induction of PPARα-responsive genes was observed in liver and kidney of hPPARα and wild type mice, despite the difference in hepatic and renal TCA levels. Additional sex- and strain-dependent effects were observed in the liver, including hepatocyte proliferation and oxidative stress, which were not dependent on TCA or TCOH levels. These data demonstrate that PPARα status affects the levels of the putative PPARα agonist TCA following TCE exposure. Therefore, interpretations of studies using Pparα-null and hPPARα mice need to consider the potential contribution of genotype-dependent toxicokinetics to observed differences in toxicity, rather than attributing such differences only to receptor-mediated toxicodynamic effects., (© The Author 2015. Water used in a food factory is frequently disinfected with chlorine, which originates disinfection by-products: haloacetic acids (HAAs) make up the second most prevalent class of these products. In this paper we propose the first static HS-GC-MS method developed for direct HAA determination in beverages; the method has higher sensitivity, simplicity and reliability than the only alternative available in the literature. From 150 beverages analysed, it is possible to conclude that at least 2 HAAs (dichloro- and trichloroacetic acids, DCAA and TCAA) are always present in beverages prepared with treated water, which remains constant for 2 or 3 months in the beverages. Moreover, beverages of 100% fruit juices and soft drinks prepared with mineral water (free of HAAs) do not contain any HAA at significant values. A universal application of compound-specific isotope analysis of chlorine was thus far limited by the availability of suitable analysis techniques. In this study, gas chromatography in combination with a high-temperature conversion interface (GC-HTC), converting organic chlorine in the presence of H2 to gaseous HCl, was coupled to a dual-detection system, combining an ion trap mass spectrometer (MS) and isotope-ratio mass spectrometer (IRMS). The combination of the MS/IRMS detection enabled a detailed characterization, optimization, and online monitoring of the high-temperature conversion process via ion trap MS as well as a simultaneous chlorine isotope analysis by the IRMS. Using GC-HTC-MS/IRMS, chlorine isotope analysis at optimized conversion conditions resulted in very accurate isotope values (δ(37)Cl(SMOC)) for measured reference material with known isotope composition, including chlorinated ethylene, chloromethane, hexachlorocyclohexane, and trichloroacetic acids methyl ester. Respective detection limits were determined to be <15 nmol Cl on column with achieved precision of <0.3‰.

A meta-analysis was conducted to inform the epistemology, or theory of knowledge, of contaminants of emerging concern (CECs). The CEC terminology acknowledges the existence of harmful environmental agents whose identities, occurrences, hazards, and effects are not sufficiently understood. Here, data on publishing activity were analyzed for 12 CECs, revealing a common pattern of emergence, suitable for identifying past years of peak concern and forecasting future ones: dichlorodiphenyltrichloroethane (DDT; 1972, 2008), trichloroacetic acid (TCAA; 1972, 2009), nitrosodimethylamine (1984), methyl tert-butyl ether (2001), trichloroethylene (2005), perchlorate (2006), 1,4-dioxane (2009), prions (2009), triclocarban (2010), triclosan (2012), nanomaterials (by 2016), and microplastics (2022 ± 4). CECs were found to emerge from obscurity to the height of concern in 14.1 ± 3.6 years, and subside to a new baseline level of concern in 14.5 ± 4.5 years. CECs can emerge more than once (e.g., TCAA, DDT) and the multifactorial process of emergence may be driven by inception of novel scientific methods (e.g., ion chromatography, mass spectrometry and nanometrology), scientific paradigm shifts (discovery of infectious proteins), and the development, marketing and mass consumption of novel products (antimicrobial personal care products, microplastics and nanomaterials). Dissolved organic matter (DOM) represents the major pool of organic precursors for harmful disinfection byproducts, such as haloacetic acids (HAAs), formed during drinking water chlorination, but much of it remains molecularly uncharacterized. Knowledge of model precursors is thus a prerequisite for understanding the more complex whole water DOM. The utility of HAA formation potential data from model DOM precursors, however, is limited due to the lack of comparability to water samples. In this study, the formation kinetics of dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA), the two predominant HAA species, were delineated upon chlorination of seventeen model DOM precursors and sixty-eight inland lake water samples collected from the Upper Midwest region of the United States. Of particular interest was the finding that the DCAA and TCAA formation rate constants could be grouped into four statistically distinct clusters reflecting the core structural features of model DOM precursors (i.e., non-β-diketone aliphatics, β-diketone aliphatics, non-β-diketone phenolics, and β-diketone phenolics). A comparative approach built upon hierarchical cluster analysis was developed to gain further insight into the chlorine reactivity patterns of HAA precursors in inland lake waters as defined by the relative proximity to four model precursor clusters. This work highlights the potential for implementing an integrated kinetic-clustering approach to constrain the chlorine reactivity of DOM in source waters.

In this study, we propose a simple strategy based on flow injection and field-amplified sample-stacking CE-ESI-MS/MS to analyze haloacetic acids (HAAs) in tap water. Tap water was passed through a desalination cartridge before field-amplified sample-stacking CE-ESI-MS/MS analysis to reduce sample salinity. With this treatment, the signals of the HAAs increased 300- to 1400-fold. The LODs for tap water analysis were in the range of 10 to 100 ng/L, except for the LOD of monochloroacetic acid (1 μg/L in selected-ion monitoring mode detection). The proposed method is fast, convenient, and sensitive enough to perform on-line analysis of five HAAs in the tap water of Taipei City. Four HAAs, including trichloroacetic acid, dichloroacetic acid, dibromoacetic acid, and monobromoacetic acid, were detected at concentrations of approximately 1.74, 1.15, 0.16, and 0.15 ppb, respectively., (© 2013 WILEY-VCH Verlag GmbH & Co. This paper demonstrates that disinfection byproducts (DBP) concentration profiles in heated water were quite different from the DBP concentrations in the cold tap water. Chloroform concentrations in the heated water remained constant or even decreased slightly with increasing distribution system water age. The amount of dichloroacetic acid (DCAA) was much higher in the heated water than in the cold water; however, the maximum levels in heated water with different distribution system water ages did not differ substantially. The levels of trichloroacetic acid (TCAA) in the heated water were similar to the TCAA levels in the tap water, and a slight reduction was observed after the tap water was heated for 24 h. Regardless of water age, significant reductions of nonregulated DBPs were observed after the tap water was heated for 24 h. For tap water with lower water ages, there were significant increases in dichloroacetonitrile (DCAN), chloropicrin (CP), and 1,1-dichloropropane (1,1-DCP) after a short period of heating. Heating of the tap water with low pH led to a more significant increase of chloroform and a more significant short-term increase of DCAN. High pH accelerated the loss of the nonregulated DBPs in the heated water. The results indicated that as the chlorine doses increased, levels of chloroform and DCAA in the heated water increased significantly. However, for TCAA, the thermally induced increase in concentration was only notable for the chlorinated water with very high chlorine dose. Finally, heating may lead to higher DBP concentrations in chlorinated water with lower distribution system temperatures.

Chlorine has been utilized in the early stages of water treatment processes as disinfectant. Disinfection for drinking water reduces the risk of pathogenic infection but may pose a chemical threat to human health due to disinfection residues and their by-products (DBP) when the organic and inorganic precursors are present in water. In the last two decades, many modeling attempts have been made to predict the occurrence of DBP in drinking water. Models have been developed based on data generated in laboratory-scale and field-scale investigations. The objective of this paper is to develop a predictive model for DBP formation in the Alexandria governorate located at the northern west of Egypt based on field-scale investigations as well as laboratory-controlled experimentations. The present study showed that the correlation coefficient between trihalomethanes (THM) predicted and THM measured was R (2)=0.88 and the minimum deviation percentage between THM predicted and THM measured was 0.8 %, the maximum deviation percentage was 89.3 %, and the average deviation was 17.8 %, while the correlation coefficient between dichloroacetic acid (DCAA) predicted and DCAA measured was R (2)=0.98 and the minimum deviation percentage between DCAA predicted and DCAA measured was 1.3 %, the maximum deviation percentage was 47.2 %, and the average deviation was 16.6 %. In addition, the correlation coefficient between trichloroacetic acid (TCAA) predicted and TCAA measured was R (2)=0.98 and the minimum deviation percentage between TCAA predicted and TCAA measured was 4.9 %, the maximum deviation percentage was 43.0 %, and the average deviation was 16.0 %.

Haemin-functionalised magnetic iron(II, III) oxide (Fe3O4) nanoparticles (Fe3O4/haemin) were synthesised by changing the acidity of a solution of the two compounds. The nanoparticles were characterised by transmission electron microscopy, UV-vis absorption spectroscopy, X-ray diffraction, Fourier transform infrared spectroscopy, measurement of magnetisation, and electrochemical techniques. The properties of both haemin and Fe3O4 were retained. Thus, Fe3O4/haemin nanoparticles exhibited pronounced electrocatalytic activity towards trichloroacetic acid (TCA) like haemin itself. Interestingly, electrocatalytic activity towards TCA was affected by detection temperature, which was controlled via electrically heated carbon paste electrodes. The maximal catalytic current was 5.8 times higher at 60°C than at room temperature (25°C). A graphene (GR) and multi-walled carbon nanotubes (MWCNT) hybrid was prepared and modified on a 1-hexylpyridinium hexafluorophosphate based carbon ionic liquid electrode (CILE). Hemoglobin (Hb) was immobilized on GR-MWCNT/CILE surface with Nafion as the film forming material and the modified electrode was denoted as Nafion/Hb-GR-MWCNT/CILE. Spectroscopic results revealed that Hb molecules retained its native structure in the GR-MWCNT hybird. Electrochemical behaviors of Hb were carefully investigated by cyclic voltammetry with a pair of well-defined redox peaks obtained, which indicated that direct electron transfer of Hb was realized in the hybrid modified electrode. The result could be attributed to the synergistic effects of GR-MWCNT hybrid with enlarged surface area and improved conductivity through the formation of a three-dimensional network. Electrochemical parameters of the immobilized Hb on the electrode surface were further calculated with the results of the electron transfer number (n) as 1.03, the charge transfer coefficient (a) as 0.58 and the electron-transfer rate constant (ks) as 0.97 s(-1). The Hb modified electrode showed good electrocatalytic ability toward the reduction of different substrates such as trichloroacetic acid in the concentration range from 0.05 to 38.0 mmol L(-1) with a detection limit of 0.0153 mmol L(-1) (3σ), H2O2 in the concentration range from 0.1 to 516.0 mmol L(-1) with a detection limit of 34.9 nmol/L (3σ) and NaNO2 in the concentration range from 0.5 to 650.0 mmol L(-1) with a detection limit of 0.282 μmol L(-1) (3σ). A highly sensitive electrochemical sensor for the detection of trichloroacetic acid (TCA) is developed by subsequent immobilization of phthalocyanine (Pc) and Fe(II) onto multiwalled carbon nanotubes (MWCNTs) modified glassy carbon (GC) electrode. The GC/MWCNTs/Pc/Fe(II) electrode showed a pair of well-defined and nearly reversible redox couple correspondent to (Fe(III)Pc/Fe(II)Pc) with surface-confined characteristics. The surface coverage (Γ) and heterogeneous electron transfer rate constant (ks) of immobilized Fe(II)-Pc were calculated as 1.26×10(-10) mol cm(-2) and 28.13 s(-1), respectively. Excellent electrocatalytic activity of the proposed GC/MWCNTs/Pc/Fe(II) system toward TCA reduction has been indicated and the three consequent irreversible peaks for electroreduction of CCl3COOH to CH3COOH have been clearly seen. The observed chronoamperometric currents are linearly increased with the concentration of TCA at concentration range up to 20mM. Detection limit and sensitivity of the modified electrode were 2.0 μM and 0.10 μA μM(-1) cm(-2), respectively. The applicability of the sensor for TCA detection in real samples was tested. Disinfection byproducts were measured in the finished drinking water from ten water treatment plants in three Chinese cities - Guangzhou, Foshan and Zhuhai. A total of 155 water samples were collected in 2011 and 2012. The median (range) of trihalomethane (THM) and haloacetic acid (HAA) levels were 17.7 (0.7-62.7) μg/L and 8.6 (0.3-81.3) μg/L, respectively. Chloroform, dichloroacetic acid and trichloroacetic acid were the dominant species observed in Guangzhou and Foshan water, while brominated THMs predominated in water from Zhuhai. Haloacetonitriles, haloketones, chloral hydrate and trichloronitromethane were usually detected at levels ranging from unquantifiable (<0.2μg/L) to 12.2μg/L (choral hydrate). THMs and HAAs showed clear seasonal variations with the total concentrations higher in winter than in summer. Correlations among DBP levels varied, with the strongest linear correlation observed between chloroform and chloral hydrate levels (R(2)=0.77). The risk of cancer from ingestion, inhalation and dermal contact exposure to THMs was estimated. An innovative haloacetic acid (HAA) removal process was developed. The process consisted of a zero-valent iron (Fe(0)) column followed by a biologically active carbon (BAC) column that were efficient in degrading tri- and di-HAAs, and mono- and di-HAAs, respectively. The merit of the process was demonstrated by its performance in removing trichloroacetic acid (TCAA). An empty bed contact time of 10 min achieved nearly complete removal of 1.2 μM TCAA and its subsequent products, dichloroacetic acid (DCAA) and monochloroacetic acid (MCAA). HAA removal was a result of chemical dehalogenation and biodegradation rather than physical adsorption. Preliminary kinetic analyses were conducted and the pseudo-first-order rate constants were estimated at ambient conditions for Fe(0) reduction of TCAA and biodegradation of DCAA and MCAA by BAC. A solid phase microextraction method was used for the analysis of nine haloacetic acids (HAAs) in water and air (aerosols) from indoor swimming pools (ISPs). The analysis is characterized by derivatization of HAAs to their methyl-esters with dimethyl sulphate, headspace solid phase microextraction (HS-SPME) with a Carboxen-polydimethylsiloxane (CAR-PDMS) fiber and gas chromatography - electron capture detector (GC/ECD). High correlation coefficients were obtained for esters mixture calibration lines and detection limits were found to be at the low ppb level. Repeatability was assessed and coefficients of variation varied from 10 to 20%. Reproducibility was also evaluated and coefficients of variation from 15 to 25% were obtained. Analytical results from four Portuguese ISPs showed that the mean concentration of total HAAs (THAAs) in water ranged from 10 ± 2 to 183 ± 28 μg/L in which 55 ± 20% corresponded to trichloroacetic and dichloroacetic acids (TCAA and DCAA). THAAs highest concentrations were directly related to higher ISPs' water organic matter content. In the lack of European specific regulation for water from ISPs and taking into consideration that ingestion is a form of exposure, THAAs concentration values were compared with drinking water maximum contamination level (MCL) of 60 μg/L proposed by the US EPA for the sum of five HAAs. In 35% of water sampling campaigns the sum of MBAA (monobromoacetic acid), MCAA (monochloroacetic acid), DCAA and TCAA exceeded that MCL value. The concentrations obtained for THAAs in the ISPs' atmosphere ranged from 5 ± 1 to 64 ± 10 μg/m(3) (T = 28°C at 5 cm above the water surface) and were proportional to the aerosols' quantity, which was deeply related to indoor air ventilation system.

Advanced treatment technologies that control multiple contaminants are beneficial to drinking water treatment. This research applied UV/H(2)O(2) for the simultaneous degradation of geosmin, 2-methylisoborneol, four trihalomethanes and six haloacetic acids. Experiments were conducted in de-ionized water at 24 ± 1.0 °C with ng/L amounts of odorants and μg/L amounts of disinfection byproducts. UV was applied with and without 6 mg/L H(2)O(2.) The results demonstrated that brominated trihalomethanes and brominated haloacetic acids were degraded to a greater extent than geosmin and 2-methylisoborneol. Tribromomethane and dibromochloromethane were degraded by 99% and 80% respectively at the UV dose of 1200 mJ/cm(2) with 6 mg/L H(2)O(2), whereas 90% of the geosmin and 60% of the 2-methylisoborneol were removed. Tribromoacetic acid and dibromoacetic acid were degraded by 99% and 80% respectively under the same conditions. Concentrations of trichloromethane and chlorinated haloacetic acids were not substantially reduced under these conditions and were not effectively removed at doses designed to remove geosmin and 2-methylisoborneol. Brominated compounds were degraded primarily by direct photolysis and cleavage of the C-Br bond with pseudo first order rate constants ranging from 10(-3) to 10(-2) s(-1). Geosmin and 2-methylisoborneol were primarily degraded by reaction with hydroxyl radical with direct photolysis as a minor factor. Perchlorinated disinfection byproducts were degraded by reaction with hydroxyl radicals. Eight typical drinking water supplies in China were selected in this study. Both source and tap water were used to investigate the occurrence of chlorinated disinfection byproducts (DBPs), and seasonal variation in the concentrations of trihalomethanes (THMs) of seven water sources was compared. The results showed that the pollution level for source water in China, as shown by DBP formation potential, was low. The most encountered DBPs were chloroform, dichloroacetic acid, trichloroacetic acid, and chlorodibromoacetic acid. The concentration of every THMs and haloacetic acid (HAA) compound was under the limit of standards for drinking water quality. The highest total THMs concentrations were detected in spring.

Pollution with organochlorines has received major attention due to various environmental effects, but it is now increasingly recognized, that they also take part in biogeochemical cycles and that natural background concentrations exist for several chlorinated compounds. We here report the natural occurrence and cycling of organic compounds with a trichloromethyl moiety in common. The study areas are temperate coniferous forests. Trichloromethyl compounds can be found in all compartments of the forests (groundwater, soil, vegetation and throughfall), but not all compounds in all compartments. The atmospheric input of trichloromethyl compounds is found to be minor, with significant contributions for trichloroacetic acid (TCAA), only. In top soil, where the formation of the compounds is expected to occur, there is a clear positive relationship between chloroform and trichloroacetyl containing compounds. Other positive relations occur, which in combination with chlorination experiments performed in the laboratory, point to the fact that all the trichloromethyl compounds may be formed concurrently in the soil, and their subsequent fates then differ due to different physical, chemical and biological properties. TCAA cannot be detected in soil and groundwater, but sorption and mineralization experiments performed in the laboratory in combination with analyses of vegetation, show that TCAA is probably formed in the top soil and then partly taken up by the vegetation and partly mineralized in the soil. Objective: The purpose of this study is to investigate the changes of concentration of trihalomethanes (THMs: chloroform, bromodichloromethane, chlorodibromomethane, bromoform) and haloacetic acids (HAAs: monochloroacetic acid, monobromoacetic acid, dichloroacetic acid, dibromoacetic acid, trichloroacetic acid, bromochloroacetic acid) in water samples with holding time and their stability in extract., Method: Two water samples from pipe network were collected and added 10 microg/L of THMs mixed standards and 10 microg/L of HAAs mixed standards respectively. The concentrations of THMs and HAAs were measured by U.S. EPA methods 551.1 and 552.3 on the day after sampling, 3 days, 7 days and 14 days, and compared the changes with holding time. The extract with THMs or HAAs mixed standards in the preserving of the day, 7 days, 14 days, 21 days and 30 days were measured by the concentration of THMs and HAAs, compared the changes of targets concentration with holding time., Results: Impact of sample holding time is an important factor in the concentration of the targets which significantly decreased with the extension of the time. The concentration of targets in extract except drichloroactic acid, keep stable in the periods of measurement., Conclusion: Sample holding times have a greater impact on the determination of THMs and HAAs, therefore, we suggest that samples be measured or finished pretreatment on the day when sample was collected. In addition to drichloroacetic acid, the concentration of other targets in the extract can be stable for more than 30 days.

Trichloroacetic acid (TCAA) is a pollutant with several sources and is also formed naturally in soil. We show that almost all investigated environmental compartments (soil, soil water, groundwater, spruce needles and throughfall, but not rain) contain compounds, which make false positives in the thermal decarboxylation method often used for determination of TCAA in environmental samples. The compounds are dominating quantitatively over TCAA in soil, soil water and groundwater, while TCAA is dominating in needle and throughfall samples. The compounds behave differently from TCAA with regard to the velocity and the pH-dependence of the chloroform release. We did not manage to reveal the whole chemical structure of the compounds, but a trichloroacetyl group seems to be the only plausible structure giving rise to CHCl(3) both upon heating and under alkaline conditions. Besides the trichloroacetyl group, the compounds did in general contain a carboxylic acid group, although in needle and throughfall samples, trichloroacetyl compounds with a neutral charge at pH 7.5 seemed to co-exist with the carboxylic acids. Trichloroacetyl groups in humic substances and possibly other macromolecular structures contribute to the major portion of the total trichloroacetyl-CHCl(3) in topsoil, but smaller molecules with less UV-VIS absorption seem to constitute the major part of trichloroacetyl-CHCl(3) in soil water and groundwater. The trichloroacetyl containing compounds are most likely naturally occurring compounds formed in the natural chlorination processes in soil, but additional studies are needed to substantiate this hypothesis.

A stable composite film composed of 1-butyl-3-methyl-imidazolium tetrafluoroborate, chitosan, zirconia nanoparticles and the hemoglobin (Hb) was cast on the surface of an ionic liquid of an N-butylpyridinium hexafluorophosphate modified carbon paste electrode to fabricate a modified electrode. Scanning electron microscopy images of the modified electrode indicated that there was a dendritic structure on the modified electrode, and UV-Vis and FT-IR spectra showed that Hb in the composite film retained its native structure. Voltammetric experiments at the modified electrode showed that, in a pH 7.0 phosphate buffer solution, a pair of well-defined and quasi-reversible redox voltammetric peaks was obtained with the formal potential located at -0.216 V. The electrochemical parameters of Hb in the composite film were further carefully calculated: the electron-transfer rate constant was 0.52 s(-1) and the charge transfer coefficient was 0.34. The catalytic reduction peak current had a linear relationship with the concentration of trichloroacetic acid (TCA) in the range from 0.2 to 10.3 mmol/L with a detection limit of 66.7 µmol/L. Therefore, the composite film as a novel matrix opened up a possibility for further study on the design of enzymatic biosensors with potential applications.

This study was designed to analyse the reliability of using urinary and blood trichloroacetic acid (TCAA) as a biomarker of exposure. A total of 46 healthy women consumed supplied TCAA-containing tap water for 15 days and provided urine and blood samples for TCAA measurements. The findings revealed that the reliability of measurements was excellent by using measures of TCAA ingestion, blood concentration and urinary excretion (intraclass correlation coefficients (ICC) > 0.75, p < 0.001). Volume of tap water consumption (ICC = 0.69) and creatinine-adjusted urinary concentration (ICC = 0.72) were less reliable. This indicated that the intraindividual variability was small and the interindividual reliability was high by using these measures in this cohort study. Laboratory variability did not significantly contribute to total variance (ICC > 0.95, p < 0.001). Other possible sources of variation such as bathing, showering, dishwashing and physical activities were unlikely to contribute significantly to total variance. For sampling strategies, 1-day blood sampling and 2-day urine sampling are sufficient to achieve reliability for an epidemiological study if a quasi-steady-state TCAA level in the body is reached. The results suggest that TCAA ingestion, TCAA loading in blood and urinary TCAA excretion are reliable measures for use as biomarkers in epidemiological studies.

The direct electrochemistry of myoglobin (Mb) entrapped in the Nafion film on a multi-walled carbon nanotubes (MWCNTs) modified carbon ionic liquid electrode (CILE) had been investigated in this paper. By using a hydrophilic ionic liquid of 1-ethyl-3-methylimidazolium tetrafluoroborate (EMIMBF(4)) as the modifier, a high-performance basal electrode was fabricated and further modified by MWCNTs, Mb and Nafion by a step-by-step casting method. Spectroscopic results indicated that the Mb molecule on the surface of MWCNTs/CILE retained its native structure. Cyclic voltammetric results showed that a pair of well-defined quasi-reversible redox peaks appeared in the pH 7.0 phosphate buffer solution (PBS), which was attributed to the direct electron transfer of Mb heme Fe(III)/Fe(II) redox couples with the modified electrode. The Nafion/Mb/MWCNTs/CILE gave excellent electrocatalytic activity towards different substrates including trichloroacetic acid (TCA), hydrogen peroxide (H(2)O(2)) and sodium nitrite (NaNO(2)).

A conductive biocomposite film (MWCNTs-NF-Hb) containing multi-walled carbon nanotubes (MWCNTs) incorporated with entrapped haemoglobin (Hb) in nafion (NF) has been synthesized on glassy carbon electrode (GCE), gold (Au), indium tin oxide (ITO) and screen printed carbon electrode (SPCE) separately by potentiostatic methods. The presence of both MWCNTs and NF in the biocomposite film enhances the surface coverage concentration (Gamma), and increases the electron transfer rate constant (K(s)) to 132%. The biocomposite film exhibits a promising enhanced electrocatalytic activity towards the reduction of O(2), H(2)O(2) and CCl(3)COOH. The cyclic voltammetry has been used for the measurement of electrocatalysis results of analytes by means of biocomposite film-modified GCEs. The MWCNTs-NF-Hb-modified GCEs' sensitivity values are higher than the values obtained for other film modified GCEs. The surface morphology of the biocomposite films which have been deposited on ITO has been studied using scanning electron microscopy and atomic force microscopy. The studies have revealed that there was an incorporation of NF and immobilization of Hb on MWCNTs. Finally, the flow injection analysis has been used for the amperometric studies of analytes at MWCNTs-Hb and MWCNTs-NF-Hb film modified SPCEs. The amperometric study results have shown higher slope values for MWCNTs-NF-Hb biocomposite film.

Haloacetic acids (HAAs) are a class of byproducts resulting from the reaction of chlorinated disinfectants with natural organic matter. These chemicals have been found in animal studies to possibly influence hepatic, reproductive, and developmental functions, and they may be mutagenic and carcinogenic. Because HAAs are hydrophilic and strongly acidic, it is a challenge to measure them at low levels. In this study, nine traditional HAAs and monoiodoacetic acid, an emerging disinfection byproduct, are analyzed in water directly. HAAs were separated on a BetaMax Acid column or a HILIC UPLC column, and they were detected by negative electrospray ionization-tandem mass spectrometry. Although the on-column limits of detection of HAAs were lower when using an HILIC UPLC column (0.08-2.73 microg/L) than when using a BetaMax Acid column (0.18 to 71.5 microg/L), to use an HILIC UPLC column, it was required to dissolve water samples in 90% acetonitrile before injection and result in sample dilution. BetaMax Acid column was found to be more suitable for the analysis of HAAs in drinking water because there was no need of sample preparation. Major species of HAAs, such as dichloroacetic acid and trichloroacetic acid, and other primary species (e.g., dibromoacetic acid, bromochloroacetic acid and bromodichloroacetic acid) can be detected using the BetaMax Acid column at concentrations higher than 1-3 microg/L.

The interaction between erythrosine (ET) and tetracaine hydrochloride (TA) was studied by resonance Rayleigh scattering (RRS), frequency doubling scattering (FDS) and second-order scattering (SOS) combining with absorption spectrum. In a weak acidic medium of Britton-Robinson (BR) buffer solution of pH 4.5, erythrosine reacted with tetracaine hydrochloride to form 1:1 ion-association complex. As a result, the new spectra of RRS, SOS and FDS appeared and their intensities enhanced greatly. The maximum peaks of RRS, SOS and FDS were at 342 nm, 680 nm and 380 nm, respectively. The intensities of the three scattering were directly proportional to the concentration of TA in the range of 0.008-4.2 microg mL(-1) for RRS, 0.027-4.2 microg mL(-1) for SOS and 0.041-4.2 microg mL(-1) for FDS. The methods had very high sensitivities and good selectivities, and the detection limits were 0.003 microg mL(-1) for RRS, 0.008 microg mL(-1) for SOS and 0.012 microg mL(-1) for FDS, respectively. Therefore, a new method was developed to determinate trace amounts of TA. The recovery for the determination of TA in blood serum and urine samples was between 97.0% and 103.8%. In this study, mean polarizability was calculated by AM1 quantum chemistry method. In addition, the reasons for the enhancement of scattering spectra and the energy transfer between absorption, fluorescence and RRS were discussed.

Detection of the abnormal or the pathogenic form of prion protein (PrP(Sc)) by Western blot (WB) is challenging, especially, for samples derived from cell cultures that contain low levels of PrP(Sc). A variety of PrP(Sc) concentration methods have been reported with various PrP(Sc) recovery efficiencies. Ultracentrifugation is one of the methods used frequently to enrich the pathogenic form of PrP(Sc) prior to WB analyses. The resulting PrP(Sc) pellet is extremely insoluble and often requires sonication to be dissolved, potentially generating aerosols. We modified the common protein-precipitating protocol using trichloroacetic acid to concentrate PrP(Sc) by slow-speed centrifugation, followed by solubilization of the pellets with 6 mol/L urea prior to sodium dodecyl sulphate -- polyacrylamide gel electrophoresis and WB analyses. Comparative studies suggest this simple trichloroacetic acid protocol was more effective in enriching PrP(Sc) presented in cell cultures and brain homogenates than other reported protein-precipitating methods. Furthermore, incorporation of the urea treatment step to dissolve the precipitated PrP(Sc) pellets helped to reduce the infectivity of PrP(Sc).

Aim and Scope: This paper is a companion to the recent review paper by Laturnus et al. (2005) on TCA in soils, presenting a complementary review of knowledge gaps in the sources and fate of trichloroacetic acid (TCA) in plants., Main Features: The review considers the various sources of TCA precursors, including the question of how much atmospheric TCA comes from naturally-produced precursors, and addresses the implications of climate change on atmospheric TCA formation. Models of the conversion of precursors to TCA in the atmosphere are critically compared with field measurements of concentrations, deposition and budgets; data on the quantitative relationships between gas-phase TCA, particulate TCA, and TCA dissolved in rain and clouds are reviewed. Methods for quantifying TCA are summarised, along with a description of what the different techniques measure, and how results can be compared. A distinction is made between 'extractable' TCA and 'total' TCA in vegetation. Evidence for the various pathways by which TCA enters plants is given, including the in situ production of TCA in leaves. This leads to a better understanding of how plant tissue concentrations depend on uptake, production and removal rates. Finally, knowledge of the toxic effects of TCA on plants and TCA metabolism in plant tissues is summarised., Results and Discussion: The discussion highlights knowledge gaps, and is intended to aid the reader in interpreting previously published results through identifying where different ways of expressing data have been used, and the consequent conclusions that can be drawn. CONCLUSION AND FURTHER RESEARCH DIRECTIONS: Recommendations are given for future research directions--in identifying precursor sources, quantifying heterogeneous atmospheric processes, recognising and quantifying uptake pathways, and elucidating the biochemical mechanisms involved in sequestering and degrading TCA inside leaves.

Radioisotopes carbon 14 and chlorine 36 were used to elucidate the environmental role of trichloroacetic acid (TCA) formerly taken to be a herbicide and a secondary air pollutant with phytotoxic effects. However, use of 14C-labeling posed again known analytical problems, especially in TCA extraction from the sample matrix. Therefore--after evaluation of available methods--a new procedure using decarboxylation of [1,2-14C]TCA combined with extraction of the resultant 14C-chloroform with a non-polar solvent and its subsequent radiometric measurement was developed. The method solves previous difficulties and permits an easy determination of amounts between 0.4 and 20 kBq (10 - 500 ng g(-1)) of carrier-less [1,2-14C]TCA in samples from environmental investigations. The procedure is, however, not suitable for direct [36Cl]TCA determination in chlorination studies with 36Cl. Because TCA might be microbially degraded in soil during extraction and sample storage and its extraction from soil or needles is never complete, the decarboxylation method--i.e. 2 h TCA decomposition to chloroform and CO2 in aqueous solution or suspension in closed vial at 90 degrees C and pH 4.6 with subsequent CHCl3 extraction-is recommended here, estimated V < 7%. Moreover, the influence of pH and temperature on the decarboxylation of TCA in aqueous solution was studied in a broad range and its environmental relevance is shown in the case of TCA decarboxylation in spruce needles which takes place also at ambient temperatures and might amount more than 10-20% after a growing season. A study of TCA distribution in spruce needles after below-ground uptake shows the highest uptake rate into current needles which have, however, a lower TCA content than older needle-year classes, TCA biodegradation in forest soil leads predominatingly to CO2.

A simultaneous conductometric titration method for determination of mixtures of acetic acid, monochloroacetic acid and trichloroacetic acid based on the multivariate calibration partial least squares is proposed. It is possible to obtain an adjustable model to relate squared concentration values of the mixtures used in the calibration range by conductance. The effect of orthogonal signal correction (OSC) as a preprocessing technique used to remove the information unrelated to the target variables is studied. The calibration model was build using conductometric titrations data of 16 mixtures of three acids. The concentration matrix was designed by a orthogonal design. The root mean squares error of prediction (RMSEP) for acetic acid, monochloroacetic acid and trichloroacetic acid with and without OSC were 0.08, 0.30 and 0.08, and 0.15, 0.40 and 0.18, respectively. The results obtained by OSC-PLS are better than the PLS and this indicate the successful application of the OSC filter as a good preprocessing method in multivariate calibration methods. The proposed procedure allows the simultaneous determination of these acids, in the synthetic mixtures.

Haloacetic acids (HAAs) are a family of compounds whose environmental concentrations have been extensively studied, primarily in Europe. Depending on the compound, their sources are believed to be both natural and anthropogenic. To better understand possible sources and contribute to the knowledge of the global distribution of these compounds, especially between the Northern and Southern Hemispheres, samples of precipitation, soils, and conifer needles were collected from Canada, Malawi, Chile, and the U.K. Precipitation samples exhibited highest HAA concentrations in collections from Canada, and lowest in those from Malawi. Malawi samples contained measurable levels of monobromoacetic acid (MBA) (56 ng/ L) unlike those from most other locations (< 9 ng/L). Soil HAA concentration levels were highest in the U.K. (e.g., 7.3 ng/g average TCA) and lowest in Malawi (0.8 ng/g average TCA), with Chile having higher levels (4.8 ng/g average TCA) than Canada (3 ng/g average TCA). Malawi soils contained small amounts of MBA (2 ng/g), in common with the two most southern of the 11 Chilean sites. Analysis of soil cores (10-cm depth sliced at 1 cm) from sites in Malawi and Chile showed that trichloroacetic acid (TCA) generally declined with depth while mono- and dichloroacetic acid (MCA and DCA) showed no trend. MCA, DCA, and TCA concentrations in archived U.K. soil samples increased by factors of 2, 4, and 5-fold over 75 years while TFA showed no consistent trend. Monochloroacetic acid (MCA) was detected in pine needles collected from Malawi. U.K. needle samples had the highest concentrations of all chloroacetic acids (CAAs): MCA, 2-18 ng/g; dichloroacetic acid (DCA), 2-38 ng/g; and trichloroacetic acid (TCA), 28-190 ng/g. Conifer needles from Canada and Chile contained CAAs at levels ranging from < 2 to 16 ng/g wet wt. Trifluoroacetic acid concentrations generally declined with increasing elevation in the samples from the Rocky Mountains in western Canada. The results indicate that concentrations of HAAs are greatest in the industrialized Northern Hemisphere but there are significant amounts of these compounds in the less industrialized Southern Hemisphere.

Trichloroethylene (TCE) and some of its metabolites are potentially carcinogenic compounds that the general population is commonly exposed to in drinking water. Concentrations of TCE, dichloroacetic acid (DCA) and trichloroacetic acid (TCA) given to laboratory animals in cancer bioassays are high, whereas drinking water levels of the compounds are very low. It is not clear whether the trace amounts of TCE, DCA and TCA in drinking water pose a cancer risk to humans. The accuracy of pharmacokinetic studies relies on the analytical method from which blood and tissue concentration data are obtained. Models that extrapolate cancer risks of TCE and its metabolites from laboratory animals to humans, in turn, rely on the results of pharmacokinetic studies. Therefore, it is essential to have reliable analytical methods for the analysis of TCE and its metabolites. This paper reviews the methods currently in the literature for the analysis of TCE, DCA, TCA and, to a lesser extent, chloral hydrate (CH). Additional aspects of analytical methods such as method validation, species preservation and future directions in the analysis of TCE and its metabolites are also discussed., (Copyright (c) 2005 John Wiley & Sons, Ltd.)

A method using liquid chromatography with tandem mass spectrometry was developed for the determination of 11 commonly used aminoglycoside antibiotics in meat. The proposed method is sufficiently sensitive (detection limits of 15 to 40 ppb for the various antibiotics) and highly selective. It is suitable for the quantitation and confirmation of aminoglycosides in a variety of matrixes (pork muscle, fish, and veal liver). Any multiresidue method for aminoglycosides must take into account their high affinity toward sample proteins and the significantly different pK values of the various analytes. The developed method uses a low-pH extraction with trichloracetic acid to ensure complete extraction of the analytes from the matrix. An anion-exchange step is used to remove the acid from the centrifuged extract. Aminoglycosides in this solution of low ionic strength can be quantitatively retained and afterwards eluted from a weak cation-exchanger solid-phase extraction (SPE) cartridge. The highly selective SPE steps produce clean extracts, which minimize possible suppression of the mass spectrometer signal.

The aim of the present study was to investigate the hypothesis that liver tissue repair induced by exposure to chloroform (CHCl(3))+trichloroethylene binary mixture (BM) is dose-dependent similar to that elicited by exposure to these compounds individually. Male Sprague-Dawley rats (250-300 g) received three dose combinations of binary mixture (74+250, 185+500 and 370+1250 mg CHCl(3)+trichloroethylene/kg, intraperitoneally) in corn oil (maximum of 0.5 ml/kg). Liver injury was assessed by plasma alanine amino transaminase (ALT) activity and histopathology by haematoxylin & eosin. Liver tissue repair was measured by (3)H-thymidine incorporation into hepatonuclear DNA. Blood and liver levels of both the parent compounds and two major metabolites of trichloroethylene (trichloroacetic acid and trichloroethanol) were quantified by gas chromatography. The blood and liver CHCl(3) levels after the administration of binary mixture were similar compared to the administration of CHCl(3) alone. The blood and liver trichloroethylene levels after the binary mixture were significantly lower compared to trichloroethylene alone due to higher elimination in presence of CHCl(3), resulting in decreased production of metabolites. The antagonistic toxicokinetics resulted in lower liver injury than the summation of injury caused by the individual components at all three dose levels. On the other hand, tissue repair elicited by the binary mixture was dose-dependent. The interactive toxicity of this binary mixture of CHCl(3) and trichloroethylene led to subadditive initial liver injury because of a combined effect of higher elimination of TCE and mitigated progression of liver injury was prevented by timely dose-dependent stimulation of compensatory tissue repair. Even though the doses of the toxicants employed in this study are much higher than found in the environment, the results suggest that a mixture of these two compounds at environmental levels is unlikely to cause any exaggerated interactive acute liver toxicity of any biological significance.
