An invasive population of the coconut rhinoceros beetle (Oryctes rhinoceros; CRB) was discovered in Honiara, Guadalcanal, Solomon Islands in 2015. The beetle has caused severe damage to coconut palms in the outbreak area and its continued spread threatens the food security and livelihood of thousands of smallholder farmers in the region. Spread and abundance of the beetle were monitored using bucket traps baited with the aggregation pheromone, ethyl-4 methyloctanoate. Beetles were collected from traps approximately bi-weekly for two periods; one during 2017–18 and the other during 2019–2020. Trap catches showed that CRB was present throughout the whole survey region with significantly higher numbers of female CRB trapped than males. Results indicate a significant 1.5-fold increase in CRB trap catch numbers from 2017–2018 to 2019–2020 despite control efforts. The number of CRB adults trapped also varied between sites and months during both time periods but with no clear patterns. Removal of breeding sites along with strong local quarantine should remain the top priority of the local government to contain CRB expansion within Solomon Islands and beyond.

Actualmente, el abuso y mal uso de los productos quimicos ha causado una serie de problemas como contaminacion (suelo y agua) y resistencia de las plagas a los ingredientes activos. El cambio climatico y otras transformaciones por las que atraviesa la agri-cultura, generaron hace decadas la aparicion de la industria de insumos biologicos, que son productos amigables con el medio ambiente y la salud humana. Estos productos tienen caracteristicas y modos de accion que ayudan al control de plagas con menos riesgos que el uso de pesticidas. Esta revision recopila la importancia, el uso y las aplicaciones de productos biologicos basados en hongos beneficos en America Latina, destacando el proyecto "Biocontrol para Sistemas Agricolas Sustentables" ejecutado en Ecuador, financiado por MFAT Nueva Zelandia, el cual ha demostrado la eficiencia del uso de microorganismos en la agricultura a traves de los resultados de las investigaciones llevadas a cabo durante la ejecucion de este Proyecto. Ademas, se mencionan aspectos relacio-nados con el control de calidad de productos biologicos y las regulaciones para su registro. EnglishAgriculture is going through a global crisis, productivity and sustainability are two factors, which farmers are challenging to produce more food, preserve the environment and the health of living beings. Developed and devel-oping countries need to adopt new productive alternatives, which improve the quality of food, preserving the envi-ronment and the ecosystems. Actually, the abuse and misuse of chemical products has caused a series of problems such as contamination (soil and water) and pest resistance to active ingredients. Climate change and other transfor-mations that agriculture is going through, generated decades ago the emergence of the industry of biological inputs, which are friendly products with the environment and human health. These products have characteristics and modes of action that help pest control with fewer risks than the use of pesticides. This review compiles the importance, use and applications of biological products based on beneficial fungi in Latin America, highlighting the project "Bio-control for Sustainable Farming Systems" executed in Ecuador, funded by MFAT New Zeland, which has shown the efficiency of the use of microorganisms in the agriculture through the research results obtained during the execution of this Project. In addition, aspects related to quality control of bioproducts and regulations for their registration are also mentioned.

Beauveria pseudobassiana formed three-dimensional aggregates of cells (CAs) in liquid culture. CAs were formed mainly by blastospores and conidia, distinct from microsclerotia formed through adhesion of hyphae. The formation, germination and sporulation of CAs were studied, as well as the pathogenicity of conidia produced from them against adults of black beetle. After 4 days of culture, CAs were formed, becoming compact and melanised after 10 days of incubation. Electron microscopy showed three-dimensional CAs averaging 431.65 µm in length with irregular shapes and rough surfaces, where cells were trapped within an extracellular matrix. CAs germinated after 2 days of incubation on agar-plates producing hyphae and forming phialides and conidia after 4 days. Produced conidia caused 45% mortality of black beetle adults. CAs germination and sporulation on soil were directly correlated with soil moisture, reaching 80% and 100% germination on the surface of soil with 17% and 30% moisture, respectively. CAs maintained 100% germination after 2 years of storage under refrigeration. These CAs could have a similar function as microsclerotia in nature, acting as resistant structures able to protect internal cells and their ability to sporulate producing infective conidia, suggesting their potential to be used as bioinsecticides to control soil-dwelling insects.

El cultivo de mora de castilla (Rubus glaucus) es de mucha importancia para los agricultores ya que deben implementar prácticas culturales que permitan reducir el uso de pesticidas sintéticos. El objetivo de este estudio fue evaluar el efecto de diferentes alternativas tecnológicas para el control de Botrytis sp. en el cultivo de mora de castilla. Los tratamientos fueron 1: Testigo absoluto (silwet + agua), 2: Fungicida tradicional sintético (Carbendazim), 3: Extracto de mirtáceas (Iridium®), 4: Metabolitos bacterianos (Balus®) y 5: Trichoderma spp. (Tricomix®). Las aplicaciones se realizaron cada 15 días para el producto sintético y cada 8 días los biológicos, orgánicos y el testigo. Se utilizó un diseño de bloques completamente al azar (DBCA) con 3 repeticiones y un total de 10 unidades experimentales. Las variables estudiadas del fruto fueron: incidencia de Botrytis sp. (%), rendimiento (g/parcela), peso (g), diámetro polar y ecuatorial (mm), firmeza (N), sólidos solubles totales (Brix), acidez (% ácido cítrico) y pH (log H+). Se realizó un análisis de varianza y prueba de Tukey al 5 %. A partir de un análisis de incidencia semanal, se identificó que Carbendazim, Trichoderma spp. y extracto mirtáceas tuvieron un control en la incidencia de Botrytis sp. en el fruto de mora de castilla.

Mora de Castilla (Rubus glaucus, Benth), also called blackberry, is grown mainly in Colombia and Ecuador. This fruit crop is of economic importance for farmers in the Province of Tungurahua (Ecuador). In this field research, the effect of Trichoderma asperellum (1.53 × 109 CFU/g) inoculation was assessed on crop yield and fruit weight at three production sites (Huachi Grande, Pillaro and Tisaleo in Ecuador). Factorial experiments were conducted consisting of treatments of 0.18 g of a commercial product per plant and a second uninoculated treatment). The product was applied eight times per month and the weekly fruit harvest recorded in each of the eight applications. The inoculation of T. asperellum had a significant effect on crop yield and fruit weight: yield was greatest in Tisaleo (5350 g/plant) with T. asperellum treatment, which was increased by 17% over the uninoculated witness (4447 g/plant). Fruit weight in Pillaro was improved from 5.36 g/fruit for the uninoculated control to 6.04 g/fruit (12.6%). Additionally, the organic matter (OM) content was correlated with the fungal population in the soil. Tisaleo had the highest OM (4%) and the highest yield in this study. In conclusion, the inoculation of T. asperellum in the soil positively affected crop productivity.

Avocado is a fruit crop of high economic importance for Ecuador, both locally and for exportation. Fuerte and Hass varieties are cultivated, both of which are grafted onto rootstocks from the cultivar Criollo. The growth of rootstocks in the nursery is a critical stage in the production of avocado plants, and application of beneficial microorganisms could be used to improve plant nutrition and growth of the rootstocks. In this research, the effect of inoculation of Trichoderma harzianum or Glomus iranicum var. tenuihypharum on avocado rootstock seedlings (cultivar Criollo) was evaluated. Application of T. harzianum significantly increased the absorption of N5+ and Mg2+ in the roots; while in the aerial parts (leaves and stem), it increased the absorption of N5+, Ca2+, Mg2+, Mn2+ and Cu2+. On the other hand, G. iranicum var. tenuihypharum significantly increased the absorption of Ca2+ and Fe3+ in the root; but no major effect was observed in the aerial part. However, a positive tendency toward an increase in the amount of P5+ was observed in the root; while in the aerial part this trend was observed for N3- and Ca2+ absorption. Neither of the two microorganisms influenced the absorption of S4+, an element that was present at stable levels in the whole plant. In conclusion, the applied microorganisms produced an increase in absorption of several macro and micronutrient in Criollo avocado rootstock seedlings.

The coconut rhinoceros beetle (CRB: Oryctes rhinoceros Linnaeus) is one of the most damaging pests to coconut and oil palms in Asia and the Pacific Islands. Adults bore into the crown and damage developing fronds, which affects tree development and yield. The insect is native to South and Southeast Asia and was inadvertently introduced into the Pacific in 1909. It has since spread to several Pacific island nations and territories, causing significant economic impact on these important coconut and palm-growing regions. In the 1950s and 1960s, an international biological control effort was initiated to search for and release natural enemy species. Release of the Oryctes rhinoceros nudivirus Huger (OrNV) and the species complex of Metarhizium Sorokin (Hypocreales: Clavicipitaceae) was successful in controlling CRB in its invaded range. Recently a new biotype of the beetle, known as CRB-G, has spread into the Pacific Islands causing unprecedented levels of damage due to the failure of previously successful biological control agents (BCAs) to suppress this biotype. The re-emergence of CRB as a serious pest warrants a rigorous re-evaluation of potential BCAs and a new search for effective natural enemies if necessary. In this article, we review literature on CRB to 1) analyze past introductions of BCAs and their effectiveness; 2) identify potentially important natural enemies and their geographical origins; and 3) assess possible approaches for utilization of BCAs against the new wave of CRB invasion. Research gaps and directions deserving future attention are highlighted and a strategy for renovation of biological controls for CRB suggested.

El género Trichoderma y en menor grado Purpureocillium, han sido estudiados ampliamente por sus propiedades como microorganismos benéficos y su eficacia en el control de enfermedades. Sin embargo, la eficacia biológica, no es el único parámetro que se considera al momento de formular un microorganismo; la viabilidad, la pureza y la concentración en la formulación, son características importantes para asegurar el éxito en campo. El propósito de este estudio fue probar distintos tipos de formulaciones (gránulos, polvos, líquidos) registrar su comportamiento en el tiempo, utilizando las variables viabilidad, concentración y pureza para seleccionar las mejores formulaciones en función de estos parámetros. Como resultado se obtuvo que los bioformulados a base de T. asperellum presentaron mayor estabilidad durante el almacenamiento, siendo más estables las formulaciones sólidas secas en comparación con los productos líquidos. En los formulados para P. lilacinum, se observó que la presentación de polvos mojables mostró la concentración y la estabilidad más alta en el tiempo.

Some species in the genus Pyronota are known to be serious pests of pasture. The establishment of new dairy pastures on recently flipped soils on Cape Foulwind, Buller provided a unique opportunity to investigate the flight behaviour and biology of Pyronota festiva and P. setosa from 2008—2012. Flight behaviour was monitored by fixed-vane interruption traps and by field observations. Adults of both species flew through late spring/early summer (Nov—Dec) of each year and seasonal flight timing was consistent between years. Pyronota festiva flight timing and duration were similar to those previously reported in the literature and were slightly female dominated. Pyronota setosa flights started one to two weeks in advance of P. festiva and were dominated by males (>95%). Female P. setosa were rarely observed flying and were rarely caught in sweep-net sampling. Pyronota setosa beetles were never observed swarming or congregating and feeding on the foliage of mānuka. These differences in flight behaviour may help to explain the field observations that P. festiva can rapidly invade new pastures while P. setosa populations appear to spread more slowly.

Root feeding larvae of Phyllophaga vetula causes severe damage in maize agroecosystems and their conventional management with chemical pesticides adversely affects the environment. Plant beneficial rhizosphere microorganisms such as mycorrhzial and entomopathogenic fungi (EPF) offer an alternative sustainable pest management strategy, but their successful integration in agroecosystems requires profound knowledge about their ecology. Here we explored interactions between a commercial isolate of the EPF Beauveria bassiana and indigenous maize mycorrhizas and their single and combined effects against root herbivory from the insect larvae P. vetula. Maize plants were grown in a greenhouse for seven weeks in pots with P limited soil with all possible combinations of the three main factors including native arbuscular mycorrhizal fungi (AMF) (without and with), EPF (without and with) and P. vetula (without and with). Root herbivory by P. vetula reduced plant growth and nutrition, and in addition reduced AMF root colonization. In contrast, AMF improved plant growth and P nutrition, but also reduced the population density of B. bassiana . Inoculation with B. bassiana increased root dry weight and counteracted a decrease in shoot N content from P. vetula root herbivory, despite that only 5% of P. vetula larvae were infected with B. bassiana . Dual inoculation with AMF and B. bassiana increased plant growth and N shoot content allowing the plants to compensate for the damage caused by P. vetula herbivory. In conclusion our results show that combination of native AMF populations and the EPF B. bassiana induce tolerance in maize from root herbivory by P. vetula in terms of plant growth performance and nutrition.

The 2019 Inter-governmental Panel (IPCC) Report on Climate Change and Land highlighted the urgency and scale of the environmental impact from human-induced landscape change. Palm oil has historically had a particularly negative reputation for driving deforestation, biodiversity loss, greenhouse gas emissions, social exploitation and damaging health. In the eyes of many in the West, it is regarded as the world’s most hated crop. However, palm is highly productive compared with other crops and produces 40% of the worlds edible oil from only 5% of vegetable oil producing land and 0.4% of agricultural land in total. It has the potential to meet future demand for oil with minimum additional environmental and climate impact compared with other sources of vegetable oil. The related high value density has the potential to move millions of vulnerable smallholder farmers out of poverty. Given the conclusions of the IPCC Climate and Land Report, it is therefore important to re-examine the crop’s reputation in light of the accumulated evidence and to properly understand the full impacts across the environmental, health, social and economic factors. We present a comprehensive review of the benefits and risks of the crop across these dimensions and provide a new synthesis. We conclude that while oil palm has had a significant negative impact on habitat and biodiversity, it plays a minor role compared with poaching, illegal logging and threats from climate change. There are important opportunities for the industry to reverse this damage. Its reputation for negative health impacts are not backed up by the scientific evidence and indeed there may be health benefits from substituting some oils in the diet with oil palm. Positive social and economic impacts are most obvious in areas where proper market-led economies are in place, but there can be significant negative social impacts in less developed areas. We conclude that much of the reputation of palm oil is not based on a balanced interpretation of the scientific evidence. Provided future development is zero deforestation, does not occur on peat, uses methane capture technology at the mills, empowers indigenous smallholders and supports the regeneration of secondary forest, we conclude that oil palm can be the most environmentally, socially and economically sustainable means to meet future demand for vegetable oil. Indeed, with pro-active collaboration with relevant non-government organisations, oil palm can be part of the solution to reversing the degradation of tropical forest biomes

Some entomopathogenic fungi, such as Metarhizium spp. and Beauveria spp., closely interact with plants by colonizing the rhizosphere or growing endophytically inside root tissues. These characteristics increase the potential for endophytic entomopathogenic fungi to persist in the root system when used as biological control agents. The mechanism by which endophytic entomopathogenic fungi are capable to colonize the internal plant tissues is also influenced by plants and can be regarded as an adaptive protection acquired by plants against herbivorous insects. The mutual benefits obtained by plants and fungi through this peculiar endophytic association supports the bodyguard hypothesis. The objectives of this study were to evaluate the ability of different isolates of Metarhizium spp. and a single isolate of Beauveria bassiana, coated onto seeds, to colonize the rhizosphere and root tissues of maize (Zea mays) and to assess whether the presence of an insect pest and fungal root pathogen influenced this interaction. All the entomopathogenic fungal isolates tested were able to colonize the rhizosphere to some extent, but one isolate of M. anisopliae and the one of M. robertsii performed best. Overall, the presence of Costelytra giveni (Scarabaeidae) larvae significantly decreased rhizosphere colonization by the entomopathogens, while the phytopathogenic fungus Fusarium graminearum (Nectriaceae: Hypocreales) did not. Similarly, the presence of C. giveni decreased endophytic growth in root tissues by around 75% when considering all entomopathogens together. In contrast, a 112% increase in overall root endophytic colonization occurred in the presence of F. graminearum. Salicylic acid (SA) and jasmonic acid (JA) content in maize plants changed depending on the isolate of M. anisopliae incorporated through seed coating and root herbivory by larval presence. In the absence of C. giveni, plants treated with M. anisopliae A1080 had higher SA and JA root content than control plants. Conversely, when larvae were present, maize plants treated with M. anisopliae F672 had increased levels of both phytohormones, indicating a potentially primed state. The observed decrease in root colonization by some entomopathogenic fungal isolates in the presence of larvae of C. giveni and the increase in endophytic colonization in the presence of F. graminearum are in accordance with the bodyguard hypothesis. Additionally, the changes observed in the SA and JA content in Metarhizium treated plants corresponded to plant responses related to systemic acquired resistance and to induced systemic resistance. Our results demonstrate that biotic soil factors can shape the outcome of endophytic entomopathogenic fungi-plant interaction, as well as affecting the plant physiology.

The coconut rhinoceros beetle (CRB; Oryctes rhinoceros) is a major pest of coconut and oil palm, but the discovery and release of Oryctes rhinoceros nudivirus (OrNV) in the 1960s and 70s suppressed the pest such that no new invasions of uninfested islands by CRB were reported for over 30years after implementation of the biocontrol programme. Surprisingly, a highly damaging outbreak was reported from Guam (2007), which could not be controlled by OrNV. Subsequently, new invasions have been reported from Port Moresby, Papua New Guinea (2009); O'ahu, Hawai'i (2013); and Honiara, Solomon Islands (2015). We have found that all of these outbreaks have been caused by a previously unrecognized haplotype, CRB-G, which appears to be tolerant to OrNV. PCR analysis shows that OrNV is generally present at high incidence in established populations of CRB, but is generally absent from the invasive CRB-G populations. CRB-G from Guam was not susceptible to OrNV infection by oral delivery, but injection of the virus did cause mortality. Further genetic analysis shows that CRB populations can be divided into a number of clades that coincide with the endemic and invasive history of the beetle. Analysis suggests that CRB-G originated in Asia, though the precise location remains to be discovered.

Antigua is a small island at the northern end of the Lesser Antilles chain. Its geology is largely limited to the Upper Oligocene and it records with unusual fidelity the transition from island arc volcanism to quiescence and limestone deposition. Despite the apparent limitation of a rock record confined to a short stratigraphical interval, Antigua has a geodiversity that ranges from thick andesites through silicified logs and freshwater snails in cherts to limestones that include both shallow and deeper water facies, and fine karstic features.

Summary Coconut rhinoceros beetle (CRB; Oryctes rhinoceros [L.]; Coleoptera: Scarabaeidae) grubs feed only on decaying vegetation and usually are found in dead standing coconut trees and decaying coconut debris on the ground. However, in Guam, a significant proportion of the CRB population develops in detritus caught within the crowns of live coconut palms. This habitat extension may be due to almost total absence of predation by insectivorous birds and mammals as a result of heavy predation by the brown tree snake, Boiga irregularis (Bechsterin) (Squamata: Colubridae).

The bacterium Yersinia entomophaga was isolated from larvae of the New Zealand grass grub, Costelytra zealandica (Coleoptera: Scarabaeidae), found in soil. Following ingestion of a lethal dose of bacteria, larvae of C. zealandica reduced feeding activity and movement. After approximately 4 h infected larvae convulsed and regurgitated dark digestive fluid and expelled frass pellets leaving the midgut empty and the larva amber in appearance. In the initial stages of infection, ingested bacteria were mostly contained within the peritrophic membrane and expelled with the gut fluid or transferred into the hind gut. While few Y. entomophaga were associated with the midgut epithelial cells, by 24 h cells were swelling and bursting with vesicles being expelled into the midgut lumen. By 48 h, bacteria had entered the haemocoel and the midgut cells had further deteriorated. After 72 h, the cellular remnants were totally detached from the basal membrane the infected insects were filled with bacteria and moribund or dead with septicaemia. Mortality was directly proportional to dose and time after infection. By applying a range of doses, the LD50 was determined as 2.9 × 104 Y. entomophaga per C. zealandica larva, with an LT50 of 2.94 days for doses of >1 × 105 per larva. Ingestion of low doses of bacteria did not inhibit feeding activity but led more slowly to death. By time of death, Y. entomophaga had multiplied, approximately 500 fold, in the cadavers of the infected larvae.

Biocontrol of the whitefly Trialeurodes vaporariorum (Westwood) (Hemiptera: Aleyrodidae) using entomopathogenic fungi has been a difficult challenge under greenhouse conditions. In order to select fungal isolates adapted to high temperature and extremely low moisture nine isolates of Lecanicillium lecanii (Zimmerman) Zare & W. Gams, L. attenuatum Zare & W. Gams and L. longisporum (Petch) Zare & W. Gams (Hypocreales: Clavicipitaceae) were evaluated. In vitro assays were performed to determine colony radial growth, conidial production and conidial germination in three water activity media (aw = 0.97, 0.98 and 1.00) at 28 and 32 °C. Virulence of Lecanicillium spp. isolates was evaluated against third instar T. vaporariorum on tomato plants at 23 °C. Colony radial growth, conidial production and germination decreased with the reduction in water activity, while 32 °C was extremely detrimental for all fungal isolates. However, some isolates were able to grow and produce conidia at low water activity and high temperature. Additionally, mortality above 60 % was recorded for one of these isolates. Practical implementation of biocontrol of T. vaporariorum under greenhouse production systems should consider the selection of those Lecanicillium isolates that show tolerance to the adverse environmental conditions in greenhouses.

Carriacou is one of the small islands in the Grenadine chain in the southern Lesser Antilles. It preserves two Miocene successions, that on the south coast shallowing upwards and separated by a probable fault from the extensively exposed turbidite sequence, called the Grand Bay Formation, on the east coast. These formations show a range of features beautifully exposed in coastal sections, including unconformities, turbidites and a starfish bed.

Different stages of volcanic island evolution are evident in the Cenozoic Lesser Antilles arc developed near the eastern edge of the Caribbean Plate. The early stage of seamount evolution is represented by the Kick-em-Jenny submarine volcano near the southern end of the arc. The volcanically active islands known as the Volcanic Caribbees are composed of a series of composite volcanoes and are examples of an emergent volcanic island stage. Those islands that were formerly active, known as the Limestone Caribbees, provide evidence that an erosional to submergent stage occurred in the northern part of the island arc during the Oligocene and Miocene epochs. Growth of the islands and underlying crust is dependent upon the magma production rate along the arc, which has for the last 100 ka ranged from less than 1 km 3 to 40 km 3 . These differences in magma production and volcanic rock composition along the arc are attributed to the obliquity of plate convergence and the plate convergence rates.

In the last few years, there have been numerous developments in the commercialisation of bionematicides. Here we briefly review the commercial status of four key active ingredients backed by large international companies and which are, or are likely to become widely available soon. We also identify research priorities. While scientists have intensively researched Pasteuria spp. and Purpureocillium lilacinus as potential nematicides, little research has been done on Bacillus firmus and Myrothecium verrucaria fermentation products. Conversely, a survey of the trade press suggests that these latter two agents, particularly B. firmus, are the most widely used. There is a clear need to further understand the ecology and mode of action of B. firmus when used as a bionematicide. Furthermore, while all active ingredients have proven efficacy in laboratory and/or small plot trials, there is a paucity of independent data showing product efficacy in target markets.

Outbreaks of insect pests can cause major losses in pasture productivity, but farmers are often poorly equipped to evaluate risk factors, predict pest impacts and determine appropriate control actions. Decision tools are described for pasture pest management using three case studies. For grass grub, "at risk" paddocks are identified based on factors such as time since sowing, soil type and larval damage the previous autumn/ winter. Targeted populations measurements can then be taken and a decision to control grass grub made using this objective measurement and/or an assessment of the pasture value, termed the insurance approach. For manuka beetle, pesticide application decisions are based on damage scores of individual paddocks and the correlation between these scores and larval densities. With porina, information on moth flight times, larval populations and pasture damage are used to make key control decisions. For all pests, decisions to control should be made in conjunction with other farm systems information, including the costs of lost forage production and long-term impacts on pasture persistence. Dr. Rer. Nat. Alois M. Huger had a long and distinguished career at the Institut fur biologische Schadlingsbekampfung (Institute for Biological Pest Control) Darmstadt, Germany, where he became one of the world's leading insect pathologists. He applied his experience and understanding of insect tissues and specialist skills in microscopy to diagnosis and elucidation of the pathology of insect diseases. During his career he discovered the Oryctes nudivirus and Bacillus thuringiensis subsp. tenebrionis, and was a member of teams unravelling amber disease in scarab beetles and the role of bacteria in parthenogenesis of parasitoids. He researched and described the life cycles of microsporidian and rickettsial pathogens of insects and was the first to describe the ultrastructure of a microsporidian spore. Dr. Huger carried out diagnosis of diseased insects over a period of 55years and has left us with many publications and a database containing thousands of records for ongoing investigation. Working with multiple pathogens in different systems, Dr. Huger obtained an overview second to none in the complexities of insect/pathogen relationships and has been at the forefront of making these discoveries benefit humanity. He is a worthy recipient of the Founders' Lecture Honoree Award, the highest honour of the Society for Invertebrate Pathology.

Protein degradation is a critical component of cellular maintenance. The intracellular translocation and targeting of the Ubiquitin Proteasome System (UPS) differentially coordinates a protein’s half-life and thereby its function. Nucleus Accumbens 1 (NAC1), a member of the Pox virus and Zinc finger/Bric-a-brac Tramtrack Broad complex (POZ/BTB) family of proteins, participates in the coordinated proteolysis of synaptic proteins by mediating recruitment of the UPS to dendritic spines. Here we report a novel interaction between NAC1 and TAR DNA-binding protein 43 (TDP-43), a protein identified as the primary component of ubiquitinated protein aggregates found in patients with Amyotrophic Lateral Sclerosis (ALS). In vitro translated full-length TDP-43 associated with both the POZ/BTB domain and the non-POZ/BTB domain of NAC1 in GST pulldown assays. Other POZ/BTB proteins (including zinc finger POZ/BTB proteins and atypical POZ/BTB proteins) showed weak interactions with TDP-43. In addition, NAC1 and TDP-43 were present in the same immunocomplexes in different regions of mouse brain and spinal cord. In primary spinal cord cultures, TDP-43 expression was mainly nuclear, whereas NAC1 was both nuclear and cytoplasmic. In order to mimic ALS-like toxicity in the spinal cord culture system, we elevated extracellular glutamate levels resulting in the selective loss of motor neurons. Using this model, it was found that glutamate toxicity elicited a dose-dependent translocation of TDP-43 out of the nucleus of cholinergic neurons and increased the co-localization of NAC1 and TDP-43. These findings suggest that NAC1 may function to link TDP-43 to the proteasome; thereby, facilitating the post-translational modifications of TDP-43 that lead to the development of ALS.

Biopesticides based on living microbes and their bioactive compounds have been researched and promoted as replacements for synthetic pesticides for many years. However, lack of efficacy, inconsistent field performance and high cost have generally relegated them to niche products. Recently, technological advances and major changes in the external environment have positively altered the outlook for biopesticides. Significant increases in market penetration have been made, but biopesticides still only make up a small percentage of pest control products. Progress in the areas of activity spectra, delivery options, persistence of effect and implementation have contributed to the increasing use of biopesticides, but technologies that are truly transformational and result in significant uptake are still lacking.

Pasture pests are often held in check by natural enemies but we have observed that severe pest attack over a wide area can occur after large scale land use change. Data were reviewed from current projects and databases for pest density and damage records covering the past 30 years. The focus was on areas where large scale land change has been implemented, including new irrigation schemes, pasture development from tussock or scrub, and land moulding for drainage via "flipping" or "hump and hollow". In these situations, pest outbreaks reached unprecedented levels, e.g. 2200 grass grub larvae/m2 in the Amuri irrigation scheme, 770 porina larvae/m2 on the East Otago Plateau and 3500 manuka beetle larvae/ m2 at Cape Foulwind on the West Coast. With major land use change, a new environment is created where pest species are provided abundant resources and the initial invaders multiply rapidly, free from the pressure of natural enemies. Monitoring systems, to provide early warning of pest attack, and remediation strategies can be used to overcome damage by these pests. Through transposon mutagenesis and DNA sequence analysis, the main disease determinant of the entomopathogenic bacterium Yersinia entomophaga MH96 was localized to an ∼32-kb pathogenicity island (PAI) designated PAI Ye96 . Residing within PAI Ye96 are seven open reading frames that encode an insecticidal toxin complex (TC), comprising not only the readily recognized toxin complex A (TCA), TCB, and TCC components but also two chitinase proteins that form a composite TC molecule. The central TC gene-associated region (∼19 kb) of PAI Ye96 was deleted from the Y. entomophaga MH96 genome, and a subsequent bioassay of the ΔTC derivative toward Costelytra zealandica larvae showed it to be innocuous. Virulence of the ΔTC mutant strain could be restored by the introduction of a clone containing the entire PAI Ye96 TC gene region. As much as 0.5 mg of the TC is released per 100 ml of Luria-Bertani broth at 25°C, while at 30 or 37°C, no TC could be detected in the culture supernatant. Filter-sterilized culture supernatants derived from Y. entomophaga MH96, but not from the ΔTC strain grown at temperatures of 25°C or less, were able to cause mortality. The 50% lethal doses (LD 50 s) of the TC toward diamondback moth Plutella xylostella and C. zealandica larvae were defined as 30 ng and 50 ng, respectively, at 5 days after ingestion. Histological analysis of the effect of the TC toward P. xylostella larva showed that within 48 h after ingestion of the TC, there was a general dissolution of the larval midgut.

New Zealand pastures are host to a range of native and exotic invertebrates. Many of these are pests that feed on the sown plant species, often causing plant death and deterioration in the productivity and persistence of the sward. While most research has focused on pest biology and control, studies of plant productivity show dramatic acute (short-medium term) and chronic (longterm) effects. For example, grass grub has been reported to cause 50% losses to ryegrass swards and Argentine stem weevil can cause a 20% loss of productivity, but the long term effects through loss of the sown species and changes in composition can be even more severe. Insect damage can be exacerbated by other biotic (e.g. weed invasion, grazing animals, plant pathogens) or abiotic factors (e.g. climate, soil fertility) to further reduce plant persistence. Some biotic and abiotic factors may interact with insect pests to have positive effects on pasture persistence, e.g. biocontrol agents that reduce pest density or addition of fertiliser to enable plants to recover from insect damage. For many pasture pests, knowledge of their ecology is based on research from over 20 years ago that, while still valid, needs to be updated to account for modern intensive pasture management practices, such as intensive grazing with a lack of reseeding, and new cultivars. To improve pasture persistence, integrated research programmes are needed that examine the relative importance of all factors contributing to plant survival and develop strategies to mitigate their effects. The island of Tobago is only about 40 km in length and to the geologist is a demonstration of the old saying that good things come in small packages. Situated in the southeast corner of the Caribbean Plate, it is the eastern extremity of the Caribbean Mountain System of northern South America, and is divided into three geologic provinces, one metamorphic, one igneous and one sedimentary.

Manuka beetles (Pyronota sp.; Scarabaeidae) are serious and persistent pests of dairy pastures on Cape Foulwind, Westport. When a selection of scarab-active fungal isolates were tested against 3 rd instar larvae of two Pyronota species, a locally sourced Beauveria brongniartii (F636) isolate consistently achieved the fastest and highest levels of larval mortality. Topical application of F636 caused an average of 80% larval mortality 6 weeks post-treatment. Mortality was shown to be dose rate dependent for both Pyronota species. When treatments were applied by incorporating rice grains colonised by the fungus into soil, simulating field application, F636 again produced the fastest and highest levels of larval mortalities, averaging 70% 6 weeks post-application. Mortalities of both Pyronota species reached 100% after 8 weeks when the assay medium was a grey sand based soil (ex Cape Foulwind). Isolate F636 shows promise as a biological control agent for this pest and field trials have been carried out in the autumn of 2010.

Cellulose and hemicelluloses are the most prevalent sources of carbon in nature. Currently many approaches employ micro-organisms and their enzyme products to degrade plant feedstocks for production of bioenergy. Scarab larvae are one such model. Theyconsumecellulosesfromavarietyofsourcesincludingplantroots,soilorganicmatter and decaying wood, and are able to extract nutrients and energy from these sources. In this paper, we review the physicochemical properties of the scarab larval gut, the diversity and digestive role that microflora play in the scarab gut and discuss the potential for applying these digestive processes in bioreactors for improving bio-fuel production. Scarab larvae are characterised by their highly alkaline midgut which is dominated by serine proteinase enzymes, and a modified hindgut which harbors the majority of the intestinal microbiota under anaerobic conditions. Evidence suggests that digestion of recalcitrant organic matter in scarab larvae likely results from a combination of endogenous gut proteinases and cellulolyticenzymesproducedbysymbioticmicro-organisms.Mostoftheeasilydigestible proteins are mobilized and absorbed in the midgut by endogenous proteinases. The hindgut contentsofscarablarvaearecharacterizedbyhighconcentrationsofvolatilefattyacids,the presence of fermenting bacteria, and typical anaerobic activities, such as methanogenesis. The hindgut typically contains a wide diversity of micro-organisms, some of which appear to be obligate symbionts with cellulolytic potential. As a result, the scarab larval gut can be regarded as a small bioreactor resembling the rumen of sheep or cattle, where solid food particles composed of cellulose, hemicellulose, pectin and polysaccharides are degraded through enzymatic and fermentation processes. Together these observations suggestscarablarvaehavepotentialtoassistthebio-fuelindustrybyprovidingnewsources of (hemi)cellulolytic bacteria and bacterial (hemi)cellulolytic enzymes.

Incoloy 903 overlays have been used to provide hydrogen environment embrittlement (HEE) resistance to welds in nickel alloy 718 structures. This is problematic because application of the required overlays has a history of high rejection and rework due to interpass microfissuring. Kovar has been identified as a potential hydrogen resistant replacement for Incoloy 903. A weldability study was initiated to compare the hot crack (microfissure) resistance of the two alloys to determine if substitution of Kovar for Incoloy 903 has the potential to improve the fabricability of HEE overlays. Varestraint testing indicates that Kovar has much higher crack initiation strains for both HAZ and weld metal cracking. Crack initiation strains were approximately 2% for Kovar while Incoloy 903 crack initiation strains were only 0.25% . Maximum crack lengths (MCL) observed on Kovar Varestraint tests were 0.12mm and 0.58mm for base and weld metal respectively, while 903 MCLs were 0.56mm and 2.3mm. Gleeble hot ductility testing indicates that Kovar has a nil ductility range of 7 degrees C while Incoloy 903 has a range of approximately 45 degrees C. The larger range observed for 903 is an indication of its greater crack susceptibility. Fabricability was correlated to material microstructure using optical microscopy, scanning electron microscopy and microprobe analysis.

Dead larvae and pupae of the sheep blowfly, Lucilia sericata, were collected from field soil and subjected to pathological examination. A haemocoel sample of each cadaver was screened for pathogens by growth on specific media, and dominant isolates were characterised with biochemical and morphological tests. Thirty‐eight bacterial and 15 fungal isolates were isolated and bioassayed against L. sericata larvae in soil. Nearly all larvae challenged with microbes survived to emergence of the adult fly, but two Tolypocladium cylindrosporum isolates caused a 100% reduction in emergence. A selected isolate was further tested against larvae of L. sericata, L. cuprina, Calliphora stygia and Chry‐somya rufifacies, and there was strong suppression of adult emergence (0–20% emergence) compared to untreated controls (93–100%).

Amber disease of the New Zealand grass grub Costelytra zealandica (Coleoptera: Scarabaeidae) is caused by ingestion of pADAP plasmid carrying isolates of Serratia entomophila or Serratia proteamaculans (Enterobacteriaceae) and causes infected larvae to cease feeding and clear their midgut to a pale amber colour where midgut serine protease activities are virtually eliminated. Using bacterial strains and mutants expressing combinations of the anti-feeding (afp) and gut clearance (sep) gene clusters from pADAP, we manipulated the disease phenotype and demonstrated directly the relationship between gene clusters, phenotype and loss of enzyme activity. Treatment with afp-expressing strains caused cessation of feeding without gut clearance where midgut protease activity was maintained at levels similar to that of healthy larvae. Treatment with strains expressing sep-genes caused gut clearance followed by a virtual elimination of trypsin and chymotrypsin titre in the midgut indicating both the loss of pre-existing enzyme from the lumen and a failure to replenish enzyme levels in this region by secretion from the epithelium. Monitoring of enzymatic activity through the alimentary tract during expression of disease showed that loss of serine protease activity in the midgut was matched by a surge of protease activity in the hindgut and frass pellets, indicating a flushing and elimination of the midgut contents. The blocking of enzyme secretion through amber disease appears to be selective as leucine aminopeptidase and alpha-amylase were still detected in the midgut of diseased larvae.

Rapid elimination of midgut luminal proteinase activity and gut clearance are the two major symptoms of amber disease in Costelytra zealandica larvae because of the three-subunit protein toxin complex produced in Serratia entomophila and Serratia proteamaculans. Quantitative PCR analysis of mRNA from the major serine proteinase gene families showed that loss of proteinase activity did not result from transcriptional downregulation. Unexpectedly, protein levels and rates of protein synthesis increased, rather than decreased, in the midgut of diseased insects. Proteomic analysis of midgut tissues showed marked differences between healthy and diseased midguts. Large increases in soluble forms of both actin and tubulin were identified from 2D-gels, together with concurrent decreases in the levels of polymeric actin-associated proteins: actin depolymerizing factor and cyclophilin. These results suggest that the Serratia toxin acts to cause degradation of the cytoskeletal network and prevent secretion of midgut gut digestive proteinases as both the actin cytoskeleton and microtubules are involved in exocytosis. Proteinases synthesized in the diseased midgut must be rapidly degraded because they do not accumulate in an inactive form.

The bacterium Serratia entomophila (Enterobacteriaceae) has been developed as a commercially available biopesticide for control of the pasture pest Costelytra zealandica. The influence of culture medium composition, dissolved oxygen (DO) concentration and harvesting time were investigated in order to optimise the production of S. entomophila. In batch fermentations, highest yields were achieved using sucrose (40 g L−1) as the carbon source, followed closely by fructose and molasses. The effect of yeast extract (YE), marmite and bakery yeast as cell growth enhancers was also examined in both batch and fed-batch mode. Culture medium containing 20 g L−1 of YE (fed-batch) produced the highest cell density. No significant effect on cell yield was detected when cultures were supplemented with bakery yeast or marmite. The Middle Miocene Grand Bay Formation is exposed on the eastern half of the island of Carriacou, Grenada Grenadines. The formation was deposited in water depths of greater than 150 m and is mainly composed of a sequence of bioclastic and volcanogenic turbidites. The dominant rock type in the Grand Bay Formation is fine- to coarse-grained sandstones. The volcaniclastic sandstones are poorly sorted and immature, and contain volcanic clasts, clinopyroxene, amphibole, plagioclase feldspar and opaque crystals in an argillaceous matrix. Within the Grand Bay Formation are beds of subaequeous lapilli and ash tuff representing pyroclastic fall deposits that have a mineralogy that is similar to the heavy minerals in the turbidites. The mineral assemblage of the lapilli tuffs suggests that these eruptions were basaltic andesite or andesite in composition.
