ROADMAP is a multi-actor project, including multiple institutions and organisations as project partners from different countries. In each country, partners formed working groups so called “Living Labs” composed of stakeholders from different groups, such as vets, farmers, farmer institutions, pharmaceutical companies. There are 12 Living Labs (LL) in total and each LL works differently, focusing on four levels, 1) farming systems, 2) sector, 3) regulatory, and 4) societal – but no LL works on all four levels. To describe the outcomes of the living labs, technical leaflets have been developed for all living labs. The pressure to increasingly optimize the breeding of livestock monogastric animals resulted in antimicrobials often being misused in an attempt to improve growth performance and counteract diseases in these animals, leading to an increase in the problem of antibiotic resistance. To tackle this problem, the use of probiotics, also known as direct in-feed microbials (DFM), seems to be one of the most promising strategies. Among probiotics, the interest in Bacillus strains has been intensively increased in recent decades in pigs and poultry. The aim of the present review was to evaluate the effectiveness of Bacillus strains as probiotics and as a potential strategy for reducing the misuse of antibiotics in monogastric animals. Thus, the potential modes of action, and the effects on the performance and health of pigs (weaning pigs, lactation and gestation sows) and broilers are discussed. These searches yielded 131 articles (published before January 2021). The present review showed that Bacillus strains could favor growth in terms of the average daily gain (ADG) of post-weaning piglets and broilers, and reduce the incidence of post-weaning diarrhea in pigs by 30% and mortality in broilers by 6–8%. The benefits of Bacillus strains on these parameters showed results comparable to the benefit obtained by the use of antibiotics. Furthermore, the use of Bacillus strains gives promising results in enhancing the local adaptative immune response and in reducing the oxidative stress of broilers. Fewer data were available regarding the effect on sows. Discordant effects have been reported regarding the effect on body weight (BW) and feed intake while a number of studies have supported the hypothesis that feeding probiotics to sows could benefit their reproductive performance, namely the BW and ADG of the litters. Taken all the above-mentioned facts together, this review confirmed the effectiveness of Bacillus strains as probiotics in young pigs and broilers, favoring their health and contributing to a reduction in the misuse of direct in-feed antibiotics. The continuous development and research regarding probiotics will support a decrease in the misuse of antibiotics in livestock production in order to endorse a more sustainable rearing system in the near future.

Hemicelluloses (HC) are polysaccharides constituents of the cell walls of plants. They are fermented in the gut to produce volatile fatty acids (VFA). The present study investigated the effects of decreasing HC level in a sow’s lactation diet on sow performances, offspring development and milk composition. From 110 days (d) of gestation until weaning (26 ± 0.4 d post-farrowing), 40 Swiss Large White sows were assigned to one of the four dietary treatments: (1) T13 (HC: 127 g/kg), (2) T11 (HC: 114 g/kg), (3) T9 (HC: 94 g/kg) and (4) T8 (HC: 80 g/kg). Milk was collected at 3 and 17d of lactation. At birth, piglets were divided into two groups according to their birthweight (BtW): normal (N-BtW; BtW >1.20 kg) or low (L-BtW; BtW ≤1.20 kg). Decreased HC levels in the maternal diet linearly increased (p ≤ .05) the body weight of L-BtW piglets at two weeks post-weaning and linearly decreased (p ≤ .05) diarrhoea incidence and duration in this category. The concentrations of copper, threonine and VFA, as well as the proportion of butyrate, in milk linearly increased (p ≤ .05), whereas lactose content linearly decreased (p ≤ .05) with decreased HC in the maternal diet. The present study provides evidence that decreasing HC level in a sow’s lactation diet can positively affect the composition and VFA profile of milk and ultimately favour the growth and health of L-BtW piglets.HighlightsThe results of this study showed that decreasing the level of hemicelluloses in a sow’s lactation diet modified milk composition and had positive effects on the post-weaning performance of low birthweight piglets.This study highlighted the role of dietary fibres in the maternal diet to alleviate body weight variations at two weeks post-weaning.Nevertheless, before advising pig producers, further research should elucidate the optimal level of hemicelluloses for lactating sows. Pig livestock was influenced by several global concerns that imposed a re-thinking of the farming system, which included the reduction in chemical dependency and the development of antimicrobial alternatives. Post-weaning diarrhea and enterotoxaemia caused by Escherichia coli, are serious threats that are responsible for the economic losses related to mortality, morbidity and stunted growth in weaning piglets. The aim of the study was to set up experimental conditions to simulate the simultaneous outbreak of post-weaning diarrhea and enterotoxaemia in weaned piglets, through verocytotoxic O138 Escherichia coli challenge, with a multidisciplinary approach. Eighteen piglets susceptible to F18 VTEC infection were selected by polymerase chain reaction for polymorphism on the fucosyltransferase 1 gene and randomly divided in two experimental groups, non-infected controls (C, n = 6) and infected ones (I, n = 12) and housed into individual pens at the same environmental conditions for 29 days. At day 20, I pigs were orally inoculated with Escherichia coli O138 and fed a high protein ration for 3 days. Zootechnical, clinical, microbiological, histological and immunological parameters were evaluated along the follow up (3 and 9 days). Experimental infection, confirmed by bacteria faecal shedding of the I group, significantly affected the clinical status. The I group showed significantly higher total scores, corresponding to medians of the sum of daily scores from days 1 to 3 (Σ3) and 1 to 9 (Σ9) post infection, epiphora, vitality, hair irregularity, oedema and depression. Histological examination showed evident inflammatory infiltrate of lymphocytes, and follicular hyperplasia in I pigs, in the same group, the immunohistochemical and immunological assays revealed an increase in IgG in the intestinal crypts and CD3-positive T cells in intestinal epithelium. The experimental Escherichia coli infection in controlled conditions is crucial for both the evaluation of innovative compounds and the elucidation of the mechanisms associated with the persistence of antibacterial resistant strains. In conclusion, the adopted infection model, carried out on receptor-mediated susceptible piglets, allowed us to identify a discriminative panel of clinical symptoms related to Escherichia coli O138 infection, and could be used to assess the protective effect of antibiotic alternatives.
