Based on a sample of 406 seven-year-old twins, this study examined whether exposure to friends’ social or physical aggression, respectively, moderates the effect of heritability on children’s own social and physical aggression. Univariate analyses showed that children’s own social and physical aggression were significantly explained by genetic factors, whereas friends’ social and physical aggression represented “true” environmental factors that were unrelated to children’s genetic dispositions. Multivariate analyses further suggested a possible gene–environment interaction in the link between friends’ and children’s physical aggression but not in the link between friends’ and children’s social aggression. Instead, friends’ social aggression was directly related to children’s social aggression, in addition to genetic effects on this behavior. BACKGROUND: The psychomotor stimulant theory of addiction posits that sensitivity to the positively rewarding properties of alcohol puts certain individuals at higher risk for alcohol abuse. A valid and reliable index of overactivation in the reward system has been a heightened baseline heart rate (HR) increase on the ascending limb of the blood alcohol curve. The main goal of this study was to investigate the relationship between this HR response and a questionnaire measuring sensitivity to reward and sensitivity to punishment. Additional goals included looking at (1). the association between a high HR response and various personality traits (hopelessness/introversion, anxiety sensitivity, impulsivity, and sensation-seeking) and (2). the relationship between these personality traits and stimulant use. METHODS: A total of 18 low- and 19 high-HR responders completed the Sensitivity to Punishment and Sensitivity to Reward Questionnaire (SPSRQ), the Substance Use Risk Profile Scale (SURPS), and a modified version of the Addiction Severity Index. RESULTS: High-HR responders obtained significantly higher scores than low-HR responders on the sensitivity to reward scale of the SPSRQ, as well as increased sensation-seeking scores on the SURPS. High-HR responders were not at significantly higher risk of having used stimulants, but stimulant use was associated with higher impulsivity scores on the SURPS. CONCLUSIONS: Novelty/sensation-seeking is among the personality traits that have been linked to heavy alcohol use. Bivalve culture in Canada increased by 25% from 2000 to 2016. In Prince Edward Island (PEI), bivalves are cultivated in bays and estuaries and there is limited space for further aquaculture expansion. Thus, there is merit in developing a numerical model determining the abundance of bivalve populations in relation to their food availability in order to assess the carrying capacity of shellfish growing areas. This modelling will take into account the different bivalve species present in the bay, as the cultivated Mytilus edulis and Crassostrea virginica and wild, M. edulis, C. virginica, Mya arenaria and Mercenaria mercenaria. As a first step toward a modelling goal, this study compared the physiological differences of the 6 bivalve groups. Three physiological parameters were measured: clearance rate, oxygen consumption and assimilation to determine the energy budget or scope for growth (SFG). These measurements were carried out on individuals contained in metabolic chamber at summer and autumn temperatures (20 and 8 °C, respectively). Our results show that M. edulis is best adapted to these temperature in PEI as it maintains high SFG at both temperatures. For C. virginica and M. arenaria, high physiological parameters under summer conditions were observed, followed by a decrease in autumn. For M. mercenaria rates were low at both temperatures indicating a persistently low growth potential. These results demonstrate the adaptive physiological capacity of each species and provide insight into the underlying reasons some species such as C. virginica and M. mercenaria are at their northern distribution limit in the Gulf of St. Lawrence. Finally, no differences in the SFG between cultivated and wild bivalves have been observed. These results are discussed within the context of estimating the impact of each bivalve group in bays environment from PEI and particularly on food availability.

Post-settlement dispersal potential of four commercial bivalve species (Mytilus edulis, Pecten maximus, Venus verrucosa and Ruditapes philippinarum) were studied through the assessment of recruits' sinking velocities by using a sinking velocity tube of five meters height. In parallel, dynamics of shear stress were monitored for five months on a tidal habitat characterized by the presence and the dispersal of the four species. By coupling both datasets we propose first theoretical estimates of temporal windows of secondary migrations. These experiments revealed interspecific differences in migration potential relate to shell shapes and behaviour, especially to secretion of byssal threads. The sensitivity to passive and active post-settlement migrations seems to rely on the synchronisation between the arrival on the sediment, the tidal regime (spring tide, neap tide), but also the rate of growth of the recruits. The present study confirms that patterns of secondary migrations of bivalve recruits result from a close physical-biological coupling involving benthic boundary layer (BBL) hydrodynamics and shell morphology as well as eco-ethological responses to environmental conditions but clearly modulated by the growth dynamics until a threshold size when drifting is no longer possible.

Oligotrophic conditions may impose a nutritional challenge for the larval and early post-larval development of bivalves during the search for a suitable benthic habitat. Here we investigated what settlement cues might be important for mytilid populations in southeastern Brazil. Our results point to a trophic trigger mediating larval settlement that may include an effect of saturated fatty acids, probably linked to organic detritus and bacterial production deriving from terrestrial inputs. The prevalence of drifting in this population suggests it may be a strategy for individuals to delay final settlement until encountering favorable trophic environmental conditions.

In this study, we investigated the food sources of eastern oysters Crassostrea virginica cultivated in Atlantic Canada. Stable isotopes (13C and 15N) and fatty acid biomarkers were used to identify these sources under in situ conditions for suspended (∼0.5 m below surface) and bottom (∼2 m) culture stocks. It was found that particulate organic matter represented the main food source, with major contributions from live phytoplankton. Higher lipid contents were detected in the digestive glands of suspended oysters compared to bottom oysters (p 2 μm, 1.05 ± 0.15 μg l−1, mean ± SEM). To determine whether the small size PPP was captured and assimilated by C. virginica, feeding trials were conducted in the laboratory using three PPP/NPP diets (20%, 50%, and 80% PPP), consisting of isotopically-labelled (δ13C) PPP cells (Nannochloropsis oculata) and non-labelled NPP cells (Tisochrysis lutea). An isotopically-labelled fatty acids analysis indicated PPP assimilation in various tissues (digestive gland, gills, mantle, and abductor muscle), including from oysters fed the reduced (20%) PPP diet. Isotopic enrichment (13C) in the FA 22:2 (non-methylene-interrupted or NMI) showed that precursors of NMIs utilized PPP carbon in its biosynthesis process. In conclusion, C. virginica assimilated primarily particulate organic matter (POM), including PPP, which dominated the phytoplankton community in near surface waters. C. virginica can exploit PPP carbon during fatty acid production and further biosynthesis.

Abstract Background Human glutamic acid decarboxylase 65 (hGAD65) is a key autoantigen in type 1 diabetes, having much potential as an important marker for the prediction and diagnosis of type 1 diabetes, and for the development of novel antigen-specific therapies for the treatment of type 1 diabetes. However, recombinant production of hGAD65 using conventional bacterial or mammalian cell culture-based expression systems or nuclear transformed plants is limited by low yield and low efficiency. Chloroplast transformation of the unicellular eukaryotic alga Chlamydomonas reinhardtii may offer a potential solution. Results A DNA cassette encoding full-length hGAD65, under the control of the C. reinhardtii chloroplast rbcL promoter and 5'- and 3'-UTRs, was constructed and introduced into the chloroplast genome of C. reinhardtii by particle bombardment. Integration of hGAD65 DNA into the algal chloroplast genome was confirmed by PCR. Transcriptional expression of hGAD65 was demonstrated by RT-PCR. Immunoblotting verified the expression and accumulation of the recombinant protein. The antigenicity of algal-derived hGAD65 was demonstrated with its immunoreactivity to diabetic sera by ELISA and by its ability to induce proliferation of spleen cells from NOD mice. Recombinant hGAD65 accumulated in transgenic algae, accounts for approximately 0.25–0.3% of its total soluble protein. Conclusion Our results demonstrate the potential value of C. reinhardtii chloroplasts as a novel platform for rapid mass production of immunologically active hGAD65. This demonstration opens the future possibility for using algal chloroplasts as novel bioreactors for the production of many other biologically active mammalian therapeutic proteins.

Abstract Background The maize INDETERMINATE1 gene, ID1, is a key regulator of the transition to flowering and the founding member of a transcription factor gene family that encodes a protein with a distinct arrangement of zinc finger motifs. The zinc fingers and surrounding sequence make up the signature ID domain (IDD), which appears to be found in all higher plant genomes. The presence of zinc finger domains and previous biochemical studies showing that ID1 binds to DNA suggests that members of this gene family are involved in transcriptional regulation. Results Comparison of IDD genes identified in Arabidopsis and rice genomes, and all IDD genes discovered in maize EST and genomic databases, suggest that ID1 is a unique member of this gene family. High levels of sequence similarity amongst all IDD genes from maize, rice and Arabidopsis suggest that they are derived from a common ancestor. Several unique features of ID1 suggest that it is a divergent member of the maize IDD family. Although no clear ID1 ortholog was identified in the Arabidopsis genome, highly similar genes that encode proteins with identity extending beyond the ID domain were isolated from rice and sorghum. Phylogenetic comparisons show that these putative orthologs, along with maize ID1, form a group separate from other IDD genes. In contrast to ID1 mRNA, which is detected exclusively in immature leaves, several maize IDD genes showed a broad range of expression in various tissues. Further, Western analysis with an antibody that cross-reacts with ID1 protein and potential orthologs from rice and sorghum shows that all three proteins are detected in immature leaves only. Conclusion Comparative genomic analysis shows that the IDD zinc finger family is highly conserved among both monocots and dicots. The leaf-specific ID1 expression pattern distinguishes it from other maize IDD genes examined. A similar leaf-specific localization pattern was observed for the putative ID1 protein orthologs from rice and sorghum. These similarities between ID1 and closely related genes in other grasses point to possible similarities in function.

OBJECTIVES: Physical aggression in children is a major public health problem. Not only is childhood physical aggression a precursor of the physical and mental health problems that will be visited on victims, but also aggressive children themselves are at higher risk of alcohol and drug abuse, accidents, violent crimes, depression, suicide attempts, spouse abuse, and neglectful and abusive parenting. Furthermore, violence commonly results in serious injuries to the perpetrators themselves. Although it is unusual for young children to harm seriously the targets of their physical aggression, studies of physical aggression during infancy indicate that by 17 months of age, the large majority of children are physically aggressive toward siblings, peers, and adults. This study aimed, first, to identify the trajectories of physical aggression during early childhood and, second, to identify antecedents of high levels of physical aggression early in life. Such antecedents could help to understand better the developmental origins of violence later in life and to identify targets for preventive interventions. METHODS: A random population sample of 572 families with a 5-month-old newborn was recruited. Assessments of physical aggression frequency were obtained from mothers at 17, 30, and 42 months after birth. Using a semiparametric, mixture model, distinct clusters of physical aggression trajectories were identified. Multivariate logit regression analysis was then used to identify which family and child characteristics, before 5 months of age, predict individuals on a high-level physical aggression trajectory from 17 to 42 months after birth. RESULTS: Three trajectories of physical aggression were identified. The first was composed of children who displayed little or no physical aggression. These individuals were estimated to account for approximately 28% of the sample. The largest group, estimated at approximately 58% of the sample, followed a rising trajectory of modest aggression. Finally, a group, estimated to comprise approximately 14% of the sample, followed a rising trajectory of high physical aggression. Best predictors before or at birth of the high physical aggression trajectory group, controlling for the levels of the other risk factors, were having young siblings (odds ratio [OR]: 4.00; confidence interval [CI]: 2.2-7.4), mothers with high levels of antisocial behavior before the end of high school (OR: 3.1; CI: 1.1-8.6), mothers who started having children early (OR: 3.1; CI: 1.4-6.8), families with low income (OR: 2.6; CI: 1.3-5.2), and mothers who smoked during pregnancy (OR: 2.2; CI: 1.1-4.1). Best predictors at 5 months of age were mothers' coercive parenting behavior (OR: 2.3; CI: 1.1-4.7) and family dysfunction (OR: 2.2; CI: 1.2-4.1). The OR for a high-aggression trajectory was 10.9 for children whose mother reported both high levels of antisocial behavior and early childbearing. CONCLUSIONS: Most children have initiated the use of physical aggression during infancy, and most will learn to use alternatives in the following years before they enter primary school. Humans seem to learn to regulate the use of physical aggression during the preschool years. Those who do not, seem to be at highest risk of serious violent behavior during adolescence and adulthood. Results from the present study indicate that children who are at highest risk of not learning to regulate physical aggression in early childhood have mothers with a history of antisocial behavior during their school years, mothers who start childbearing early and who smoke during pregnancy, and parents who have low income and have serious problems living together. All of these variables are relatively easy to measure during pregnancy. Preventive interventions should target families with high-risk profiles on these variables. Experiments with such programs have shown long-term impacts on child abuse and child antisocial behavior. However, these impacts were not observed in families with physical violence. The problem may be that the prevention programs that ion programs that were provided did not specifically target the parents' control over their physical aggression and their skills in teaching their infant not to be physically aggressive. Most intervention programs to prevent youth physical aggression have targeted school-age children. If children normally learn not to be physically aggressive during the preschool years, then one would expect that interventions that target infants who are at high risk of chronic physical aggression would have more of an impact than interventions 5 to 10 years later, when physical aggression has become a way of life. Frequent use of physical aggression by humans appears to reach its peak between 2 and 3 years of age. In the following years most children learn alternatives to physical aggression. Approximately 4% of children have high levels of physical aggression from early childhood to late adolescence. These children can be considered to show chronic physical aggression. They are at high risk of causing injuries to others and to themselves. They are also at high risk of many other co-morbid mental health conditions, school failure, substance abuse, depression, unemployment, spouse abuse, child abuse, and suicide. There is some evidence that, because of their risky style of behavior, they are also at high risk of many other medical conditions such as cardiovascular problems, cancer, and brain damage. The aim of this exploratory study was to identify developmental patterns of adolescent concurrent alcohol, cannabis, and other illicit drug use and their preadolescent individual, familial, and social risk factors in a population-representative cohort from the province of Quebec, Canada ( N = 1,593; 48.4% male). Age 12-17 years self-reports of alcohol, cannabis, and other illicit drug use were collected. Latent growth modeling was used to analyze developmental patterns of single- or polysubstance use (SU/PSU), and multinomial regression examined their association with risk factors assessed at age 10-12 years. Five developmental patterns were revealed, including nonusers (12.8% sample) and four classes reflecting different levels of SU/PSU (5.8%-37.5%), varying in severity based on onset, frequency, and type of substances used. Boys and girls were similarly represented throughout SU/PSU patterns. In comparisons with nonusers, several preadolescent risk factors were associated with increasing severity of SU/PSU. Possibly indexing fearlessness/disinhibition, low internalizing symptoms were common to all adolescent users. An earlier onset of substance use and increasing use of substances throughout adolescence were linked with having deviant peers for all user classes but later-onset users. Preadolescents manifesting externalizing problems and exposed to family adversity in addition to the above risk factors showed the earliest onset and most frequent adolescent SU/PSU, especially those also exposed to less appropriate parenting. Consistent with the developmental model of substance use, the nature, number, and severity of preadolescent risk factors distinguished between the type and severity of SU/PSU patterns in adolescence and call for a consistent strategy of universal, selective, and indicated preventive interventions. Knowledge is lacking on whether adolescents' motivations for social media use predict internalizing symptoms, and few studies have considered the moderating role of extraversion. In 2017 (T1) and 2018 (T2), 197 adolescents (49.70% girls, M age  = 13.15, SD = 0.36) from a large metropolitan area participated in this study. Adolescents reported on their social media use motivations and extraversion (at T1) and depressive and anxiety symptoms (T1 and T2). Latent class analysis revealed four motivation profiles at T1: (1) entertainment and fun (30.97%); (2) avoidance and escapism (14.21%); (3) meeting new people, feel involved (16.75%); (4) boredom (38.07%). Social motivations (profile 3) predicted elevated internalizing symptoms at T2. Avoidance and boredom motivations (profile 2 and 4) predicted internalizing symptoms for adolescents with low extraversion. Early risk factors for gambling participation (GP) and substance use (SU) in adolescents have usually been studied separately, although these disorders were integrated into the same clinical category over a decade ago. This exploratory study aimed to investigate the early individual, parental, familial and social risk factors associated with developmental patterns of adolescent GP and SU in a population-representative cohort (N = 1594, 51.2% boys). Using a person-centered strategy and multiple assessments from age 12 to 17, six developmental patterns describing joint GP and SU courses were revealed. Non-substance users/non-gamblers served as the reference class in an integrated longitudinal-multivariate analysis framework examining 15 distinct risk factors. Results showed that a core of risk factors were common to all trajectory-classes of substance users with or without GP. For a similar level of SU, most of the risk factors associated with non-gambling users also affected their gambling peers. However, additional risk factors were specifically related to GP. Thus, substance users who also gamble were affected by a greater number of risk factors than non-gambling substance users. Findings are consistent with a developmental syndrome of addiction, which posits a shared etiology between different expressions of addiction as well as differences in risk factors that lead to distinct trajectories of addictive behaviors. They highlight the importance of considering both GP and SU for a comprehensive assessment of adolescents' level of risk with regard to addictive behaviors., (© 2024. The present study investigated whether distinct developmental patterns of externalizing behaviors (EBs: hyperactivity-impulsivity, noncompliance, physical aggression) based on parent reports were repeatedly and differentially associated with separate dimensions of internalizing problems such as general anxiety, separation anxiety, and depressive symptoms across the early, middle, and late preschool years in a population birth cohort ( N = 2,057, 50.7% boys). Six high trajectory classes obtained by latent growth modeling were used as longitudinal indicators of single EB and co-occurrent EBs. Children following low or moderate trajectories for all EBs served as the reference class. Results revealed that children in trajectory classes reflecting high levels of co-occurring EBs showed higher levels of general anxiety, separation anxiety, and depressive symptoms across the preschool years. In contrast, children in trajectory classes reflecting single EB manifested higher levels of some, but not all, dimensions of internalizing problems. In addition, their scores varied from one period to another. No sex differences were observed in the above associations. These results underline the need for comprehensive assessments across distinct types of EBs and internalizing problems to better reflect the characteristics that distinguish individual children. Finally, results suggest that children showing early co-occurrent EBs and internalizing symptoms may be an important group to target for in-depth assessment and possibly preventive intervention. The developmental association between disruptive behaviors (DBs: hyperactivity-impulsivity, non-compliance, physical aggression) and internalizing problems in early childhood is not well understood and has generated competing hypotheses and mixed results. Using a person-centered strategy, the present study aimed to examine concurrent trajectories of DBs and trajectories of internalizing problems from age 1.5 to 5 years in a population-representative sample (N = 2057; 50.7% boys). Six trajectories of DBs and three trajectories of internalizing problems, based on parent reports and obtained via latent growth modeling across five periods of assessment, were used as longitudinal indicators of each type of behaviors. Children following low or moderate trajectories served as the reference class. Compared to children in the reference class, those in trajectory classes characterized by high levels of co-occurring DBs (OR = 6.60) and, to a lesser extent, those in single high DB classes (OR = 2.78) were more likely to follow a high trajectory of internalizing problems simultaneously. These results support a multiple problem hypothesis regarding the association between DBs and internalizing problems, consistent with a developmental perspective that includes a general factor underpinning different psychopathologies. These findings highlight the importance of considering the co-occurrence between DBs and internalizing problems when studying either construct in children.
