Juniperus sabina essential oil was evaluated for its fetotoxic potential on mice. Pregnant dams were injected s.c. (15-45 or 135 mg essential oil/kg body weight) on days 6 to 15 of gestation. They were killed and the uterine contents were examined on day 19 of pregnancy. The fetuses were removed for examination. The dams of the two higher treated groups showed a significant weight loss as compared to controls. An hepatotoxicity was observed among females that resorbed their whole litter, thus indicating a greater susceptibility towards Juniperus sabina essential oil during pregnancy. The essential oil induced, in the three treated groups, an embryotoxicity as manifested by a statistically significant increase in the number of affected litters; but no fetotoxicity.

Quaternary alkaloid extracts of Ptelea trifoliata showed potentially interesting antimicrobial activity. Chromatographic examination showed the presence of six components. Of these, choline and O-4-methyl ptelefolinium were known compounds. Rational structures are proposed for the new alkaloids, O-4-methylhydroxyluninium, neohydroxylunine and pteleatinium salts. Pteleatinium salt is responsible for the antimicrobial activity of the extracts of the plant.

Further investigation of a CH2Cl2 fraction prepared from the South African tree Combretum caffrum for substances inhibitory to the murine P-388 lymphocytic leukemia (PS system) cell line has led to the isolation of two new bibenzyls, designated combretastatins B-3 and B-4, accompanied by the previously known bibenzyls 7, 8, and 9. The structure of each substance was ascertained by results of mass and nmr spectral analyses and confirmed by crystal structure determination (for 7) or synthesis. Combretastatins B-3 and B-4 gave PS ED50 values of 0.4 and 1.7 micrograms/ml, respectively, and bibenzyls 7, 8, and 9 were comparably cell growth inhibitory against the PS cell line with ED50 results of 1.7, 2.5, and 0.25 ug/ml, respectively. All the bibenzyls caused leukemia cells to accumulate in mitosis at cytotoxic drug concentrations; however, a wide range of in vitro activity against the protein tubulin (the major component of the mitotic spindle) was observed.

Bark of Tecoma arequipensis yielded the major alkaloidal component (-)-delta-N-normethylskytanthine whose structure was proven by X-ray diffraction analysis of the N-thiourea derivative. The stereochemistry of 1 at all four centers was enantiomeric with that of skytanthine previously reported from Tecoma stans and Skytanthus acutus. Gc-ms of the crude base fraction indicated the presence of several related alkaloids as very minor constituents.

From the weakly antibacterial non-quaternary alkaloidal fractions from Ptelea trifoliata L. (Rutaceae), ten tertiary quinol-2-one and quinol-4-one alkaloids were isolated and identified. In addition, beta-sitosterol, beta-sitosteryl-beta-D-glucoside and bergapten were isolated. None of these compounds possessed perceptible antimicrobial activity. The weak antimicrobial activity of the neutral and alkaloidal fractions was traced to small amounts of pteleatinium chloride which had not been completely separated by bulk processes. Alkaloids previously known to be present in P. trifoliata which were found in this study ptelefoline methyl ether, pteleine and skimmianine. Alkaloids previously known but new to this plant were lunidoine and isomaculasidine. Alkaloids newly found in nature were neohydroxylunine, hydroxylunidonine, 6-methoxylunidoine, 6-methoxylunineand 6-methoxy-hydroxylunidine. The structure of the latter three bases is proposed tentatively.
