In a world where climate change is real and its consequences are unprecedented, understanding of the plant adaptive capacity and native stress-responsive machinery is crucial. In recent years, universal stress proteins (USPs) have received much attention in the field of plant science due to their stress-specific transcriptional regulation. This study focuses on the extensive characterization of the USP gene family members in the monocot crop rice (Oryza sativa L. var. japonica). Here, we report a total of 44 USP genes in the rice genome. In silico characterization of these genes showed that domain architecture played a major role in the functional diversification of the USP gene family which holds for all plant USPs. On top of that, a higher conservation of OsUSP members has been exhibited with a monocot genome (Zea mays L.) as compared to a dicot genome (Arabidopsis thaliana L.). Expression profiling of the identified genes led to the discovery of multiple OsUSP genes that showed pronounced transcript alteration under various abiotic stress conditions, indicating their potential role as multi-functional stress-specific modules. Furthermore, expression validation of OsUSP genes using qRT-PCR provided a strong evidence for the utility OsUSP genes in building multi-stress tolerant plants. Altogether, this study provides leads to suitable USP candidates that could be targeted for plant breeding and genetic engineering experiments to develop stress resilient crop species.

Abstract Background Grape leaves provide the biochemical substrates for berry development. Thus, understanding the regulation of grapevine leaf metabolism can aid in discerning processes fundamental to fruit development and berry quality. Here, the temporal alterations in leaf metabolism in Merlot grapevine grown under sufficient irrigation and water deficit were monitored from veraison until harvest. Results The vines mediated water stress gradually and involving multiple strategies: osmotic adjustment, transcript-metabolite alteration and leaf shedding. Initially stomatal conductance and leaf water potential showed a steep decrease together with the induction of stress related metabolism, e.g. up-regulation of proline and GABA metabolism and stress related sugars, and the down-regulation of developmental processes. Later, progressive soil drying was associated with an incremental contribution of Ca2+ and sucrose to the osmotic adjustment concomitant with the initiation of leaf shedding. Last, towards harvest under progressive stress conditions following leaf shedding, incremental changes in leaf water potential were measured, while the magnitude of perturbation in leaf metabolism lessened. Conclusions The data present evidence that over time grapevine acclimation to water stress diversifies in temporal responses encompassing the alteration of central metabolism and gene expression, osmotic adjustments and reduction in leaf area. Together these processes mitigate leaf water stress and aid in maintaining the berry-ripening program.

Glyoxalase is an evolutionary highly conserved pathway present in all organisms. Conventional glyoxalase pathway has two enzymes, glyoxalase I (GLYI) and glyoxalase II (GLYII) that act sequentially to detoxify a highly cytotoxic compound methylglyoxal (MG) to D-lactate with the help of reduced glutathione. Recently, proteins with DJ-1/PfpI domain have been reported to perform the same conversion in a single step without the help of any cofactor and thus termed as “unique glyoxalase III” enzyme. Genome-wide analysis of glyoxalase genes have been previously conducted in Arabidopsis, rice and Soybean plants, but no such study was performed for one of the agricultural important model legume species, Medicago truncatula. A comprehensive genome-wide analysis of Medicago identified a total of putative 29 GLYI, 14 GLYII genes, and 5 glyoxalase III (DJ-1) genes. All these identified genes and their corresponding proteins were analyzed in detail including their chromosomal distribution, gene duplication, phylogenetic relationship, and the presence of conserved domain(s). Expression of all these genes was analyzed in different tissues as well as under two devastating abiotic stresses- salinity and drought using publicly available transcript data. This study revealed that MtGLYI-4, MtGLYII-6, and MtDJ-1A are the constitutive members with a high level of expression at all 17 analyzed tissues; while MtGLYI-1, MtGLYI-11, MtGLYI-5, MtGLYI-7, and MtGLYII-13 showed tissue-specific expression. Moreover, most of the genes displayed similar pattern of expression in response to both salinity and drought stress, irrespective of stress duration and tissue type. MtGLYI-8, MtGLYI-11, MtGLYI-6, MtGLYI-16, MtGLYI-21, and MtGLYII-9 showed up-regulation, while MtGLYI-17 and MtGLYI-7/9 showed down-regulation in response to both stresses. Interestingly, MtGLYI-14/15 showed completely opposite pattern of expression in these two stresses. This study provides an initial basis about the physiological significance of glyoxalase genes in plant development and stress response of Medicago that could be explored further.

Abstract: Perennial ryegrass (Lolium perenne L.) is a popular turfgrass species. To understand the molecular mechanisms of salinity tolerance, a suppression subtractive cDNA library was constructed for a salinity-tolerant ryegrass accession, with NaCl-treated (255mM) plants as the tester. Differentially expressed cDNA fragments were cloned and screened. BLAST search revealed that 268 clones exhibited significant homologies to known genes. These genes could be categorized into 11 different functional groups, including metabolism, energy transfer, detoxification, compatible solute, cellular transport, transcription, signal transduction, etc. The salinity-regulated expression of selected genes was confirmed by RT-PCR analysis. The results suggested that these putatively salinity up-regulated genes may play a vital role in the salinity tolerance of perennial ryegrass. They can be used as candidate genes for creating stress-tolerant grasses and for understanding molecular mechanisms of plant adaptation to salinity stress. [Copyright &y& Elsevier]

An alloplasmic cytoplasmic male sterile (CMS) line of Brassica juncea was developed by repeated backcrossing of the sexual hybrid ( Diplotaxis erucoides × Brassica rapa) with B. juncea cv. ‘Pusa Bold’. The CMS line is comparable to the B. juncea nuclear donor line for all morphological traits except male fertility. Southern and cleaved amplified polymorphic sequence (CAPS) analyses showed that the CMS line carries organellar genomes from the wild species that differ from the organellar genomes of B. juncea. Northern analysis with three mitochondrial gene probes revealed altered atp α transcription pattern associated with male sterility. The restorer of CMS ( Moricandia arvensis) B. juncea was found to restore male fertility to the new CMS system. The mode of fertility restoration was found to be monogenic and gametophytic. The study identified CAPS markers for the psbB gene that could be useful in quick identification of the erucoides CMS line. Broad similarities and important differences observed at the molecular level between M. arvensis and D. erucoides CMS systems of B. juncea are discussed. [ABSTRACT FROM AUTHOR]
