Abstract

Construction of the JT-60SA tokamak was completed on schedule in March 2020. Manufacture and assembly of all the main tokamak components satisfied technical requirements, including dimensional accuracy and functional performances. Development of the plasma heating systems and diagnostics have also progressed, including the demonstration of the favourable electron cyclotron range of frequency (ECRF) transmission at multiple frequencies and the achievement of long sustainment of a high-energy intense negative ion beam. Development of all the tokamak operation control systems has been completed, together with an improved plasma equilibrium control scheme suitable for superconducting tokamaks including ITER. For preparation of the tokamak operation, plasma discharge scenarios have been established using this advanced equilibrium controller. Individual commissioning of the cryogenic system and the power supply system confirmed that these systems satisfy design requirements including operational schemes contributing directly to ITER, such as active control of heat load fluctuation of the cryoplant, which is essential for dynamic operation in superconducting tokamaks. The integrated commissioning (IC) is started by vacuum pumping of the vacuum vessel and cryostat, and then moved to cool-down of the tokamak and coil excitation tests. Transition to the super-conducting state was confirmed for all the TF, EF and CS coils. The TF coil current successfully reached 25.7 kA, which is the nominal operating current of the TF coil. For this nominal toroidal field of 2.25 T, ECRF was applied and an ECRF plasma was created. The IC was, however, suspended by an incident of over current of one of the superconducting equilibrium field coil and He leakage caused by insufficient voltage holding capability at a terminal joint of the coil. The unique importance of JT-60SA for H-mode and high-β steady-state plasma research has been confirmed using advanced integrated modellings. These experiences of assembly, IC and plasma operation of JT-60SA contribute to ITER risk mitigation and efficient implementation of ITER operation.

Abstract

It has been recently demonstrated that NKG2D is an activating costimulatory receptor on natural killer (NK) cells, natural killer T (NKT) cells, activated [CD8.sup.+] T cells, and [gamma][delta] T cells, which respond to cellular stress, such as inflammation, transformation, and infection. Here we show that intestinal inflammation in colitic SCID mice induced by adoptive transfer of [CD4.sup.+][CD45RB.sup.high] T cells is characterized by significant increase of [CD4.sup.+][NKG2D.sup.+] T cells and constitutive expression of NKG2D ligands, such as H60, Mult-1, and Rae-1, by lamina propria [CD11c.sup.+] dendritic cells. Furthermore, treatment with nondepleting and neutralizing anti-NKG2D MAb after transfer of [CD4.sup.+][CD45RB.sup.high] T cells into SCID mice significantly suppressed wasting disease with colitis, abrogated leukocyte infiltration, and reduced production of IFN-[gamma] by lamina propria [CD4.sup.+] T cells. These findings demonstrate that NKG2D signaling pathway is critically involved in [CD4.sup.+] T cell-mediated disease progression and suggest a new therapeutic target for inflammatory bowel diseases. Abstract

The ITER Remote Experimentation Centre (REC) project (one of the three sub-projects of the International Fusion Energy Research Centre (IFERC)) is progressing under the agreement between the Government of Japan and the European Atomic Energy Community for the joint implementation of the Broader Approach (BA) activities in the field of fusion energy research. The objectives of the REC activity are to identify the functions and solve the technical issues for the construction of the REC for ITER at Rokkasho, and to develop the remote experiment system and verify the functions required for remote experimentation by using the Satellite Tokamak (JT-60SA) facilities to facilitate the future exploitation of ITER and JT-60SA. The functions of REC will be tested, and the total system will be demonstrated using JT-60SA and existing facilities in the EU, such as JET and WEST. The hardware of the REC has been prepared in Rokkasho Japan, which has the remote experiment room with a large video wall to show the plasma and operation status, IT equipment and a storage system by the reuse of the Helios supercomputer tape library. A broadband network infrastructure of 10Gbps has been installed connected to SINET5. Using this network system, fast data transfer from ITER to REC was examined in 2016, and the transfer of the data volumes expected for the initial ITER experiments has been demonstrated. A secure remote experimentation system has been developed, using JT-60SA, that has functions for preparing and setting of shot parameters, viewing the status of control data, streaming of the plasma status, data-exchange function of shot events, and monitoring of the facility operation. Remote data analysis techniques, data visualisation software, a documentation management and experiment planning system and numerical simulation codes for the preparation and performance estimation of discharges have also been developed.

Abstract

FTY720, a sphingosine-derived immunomodulator, causes immunosuppression via enhancement of lymphocyte sequestration into secondary lymphoid organs, thereby preventing their antigen-activated T cell egress to sites of inflammation. FTY720 is highly effective in inhibiting autoimmunity in various animal models. However, there is little known about how FTY720 controls the migration property of memory T cells. Here, we demonstrated that FTY720 pre%nts the development of colitis induced by the adoptive transfer of lamina propria (LP) colitogenic effector memory CD[4.sup.+] T cells ([T.sub.EM] cells; CD45R[B.sup.low]WCD[44.sup.high]CD62[L.sup.-]) into severe combined immunodeficiency (SCID) mice and suppresses interferon-% interleukin-2, and tumor necrosis factor-a production by LP CD[4.sup.+] T cells. The numbers of spleen, peripheral blood, mesentefic lymph node, and LP CD[4.sup.+] T cells in FTY720-treated mice were significantly reduced compared with those in control mice. Notably, LP CD[4.sup.+] [T.sub.EM] cells as well as splenic CD[4.sup.+]CD45R[B.sup.high] T cells expressed several spingosine-l-phosphate receptors that are targets for FRY720. Furthermore, FTY720 also prevented the development of colitis induced by the adoptive transfer of splenic CD[4.sup.+]CD45R[B.sup.high] T cells into SCID mice. Naturally arising [CD4.sup.+][CD25.sup.+] regulatory T ([T.sub.R]) cells have been shown to prevent and cure murine T cell-mediated colitis. However, their exact mechanism of controlling colitogenic memory [CD4.sup.+] T cells in in vivo systems excluding the initial process of naive T cell activation and differentiation has not been examined to date. Using the colitogenic effector memory ([T.sub.EM]) [CD4.sup.+] cell-mediated colitis model induced by adoptive transfer of colitogenic [CD4.sup.+][CD44.sup.high][CD62L.sup.-] lamina propria (LP) T cells obtained from colitic [CD4.sup.+] [CD45RB.sup.high] T cell-transferred mice, we have shown in the present study that [CD4.sup.+][CD25.sup.+] TR cells are able not only to suppress the development of colitis, Th1 cytokine production, and the expansion of colitogenic LP [CD4.sup.+] [T.sup.EM] cells but also to expand these cells by themselves extensively in vivo. An in vitro coculture assay revealed that [CD4.sup.+][CD25.sup.+] TR cells proliferated in the presence of IL-2-producing colitogenic LP [CD4.sup.+] [T.sub.EM] cells at the early time point (48 h after culture), followed by the acquisition of suppressive activity at the late time point (96 h after culture). Abstract

Fas/Fas ligand (FasL) interaction has been implicated in the pathogenesis of various diseases. To clarify the involvement of Fas/ FasL in the pathogenesis of intestinal inflammation, we investigated the preventive and therapeutic effects of neutralizing anti-FasL monoclonal antibody (MAb) on the development of chronic colitis induced by adaptive transfer of CD[4.sup.+] CD45R[B.sup.high] T cells to SCID mice. Administration of anti-FasL MAb from 1 day after T cell transfer (prevention study) resulted in a significant improvement of clinical manifestations such as wasting and diarrhea. However, histological examination showed that mucosal inflammation in the colon, such as infiltration oft cells and macrophages, was not improved by the anti-FasL MAb treatment. In vitro studies showed that anti-FasL MAb did not inhibit IFN-[gamma] production by anti-CD3/CD28-stimulated lamina propria CD[4.sup.+] T cells but suppressed TNF-[alpha] and IL-1[beta] production by lamina propria mononuclear cells. Therapeutic administration of anti-FasL MAb from 3 wk after T cell transfer also improved ongoing wasting disease but not intestinal inflammation. These results suggest that the Fas/FasL interaction plays a critical role in regulating systemic wasting disease but not local intestinal inflammation. Abstract

Interaction of OX40 (CD134) on T cells with its ligand (OX40L) on antigen-presenting cells has been implicated in pathogenic T cell activation. This study was performed to explore the involvement of OX40/OX40L in the development of T cell-mediated chronic colitis. We evaluated both the preventive and therapeutic effects of neutralizing anti-OX40L MAb on the development of chronic colitis in SCID mice induced by adoptive transfer of CD[4.sup.+]CD45R[B.sup.high] T cells as an animal model of Crohn's disease. We also assessed the combination of anti-OX40L and anti-TNF-[alpha] MAbs to improve the therapeutic effect. Administration of anti-OX40L MAb markedly ameliorated the clinical and histopathological disease in preventive and therapeutic protocols. In vivo treatment with anti-OX40L MAb decreased CD[4.sup.+] T cell infiltration in the colon and suppressed IFN-[gamma], IL-2, and TNF-[alpha] production by lamina propria CD[4. sup.+] T cells. The combination with anti-TNF-[alpha] MAb further improved the therapeutic effect by abolishing IFN-[gamma], IL-2, and TNF-[alpha] production by lamina propria CD[4.sup.+] T cells. Our present results suggested a pivotal role of OX40/OX40L in the pathogenesis of T cell-mediated chronic colitis. The OX40L blockade, especially in combination with the TNF-[alpha] blockade, may be a promising strategy for therapeutic intervention of Crohn's disease. Abstract

The ITER Remote Experimentation Centre (REC) in Rokkasho is a project implemented within the Broader Approach (BA) agreement at IFERC. The long-term objective of the REC is to allow researchers to take part in the experimentation on ITER from a remote location. On a shorter time scale, before ITER will be operated, the REC facility will be used to test technologies for remote participation in collaboration with existing European tokamaks and with JT-60SA, whose first operations are envisaged in 2020. Other than setting up and equipping the REC control room, during the first phase of REC (2013-2017) the project scope included activities aimed at developing and evaluating software tools for fast data transfer, remote participation, data analysis, and plasma simulation. These activities have been carried out by an Extended-Integrated Project Team (E-IPT). Indeed, due to the characteristics of the remote experiment, the collaborations with experts in other institutes providing experimental data, network infrastructure, data transfer protocol, and experiences on intercontinental data transfer and data acquisition were essential for the success of the REC activities. As a consequence, members from different Japanese and European institutions were invited to join in E-IPT; among the various contributors, there are members of the ITER project and of the Satellite Tokamak Programme (STP), members of the NII and the NIFS in Japan, and members of JET and WEST in Europe. In this paper, we report on the results of the REC verification tests that have been carried out in 2017. These tests were mainly aimed at assessing the functionalities of the REC control room (i.e., the configurability of the room layout, the capabilities of the video wall, etc.), as well as the functionalities of the software tools for remote participation that have been developed during the first BA period. A report on the preliminary remote participation tests carried out in collaboration with the JET tokamak will be also given, together with a description of the tests with both JET and WEST that have been planned for 2018.

Abstract

The project of the ITER Remote Experimentation Centre (REC) (one of the three sub-projects of the International Fusion Energy Research Centre (IFERC)) is progressing under the agreement between the Government of Japan and the European Atomic Energy Community for the joint implementation of the Broader Approach (BA) activities in the field of fusion energy research. The objectives of the REC activity are to identify the functions and solve the technical issues for the construction of the REC for ITER at Rokkasho, and to develop the remote experiment system and verify the functions required for the remote experiment by using the Satellite Tokamak (JT-60SA) facilities in order to facilitate the future exploitation of the future experiments of ITER and JT-60SA [1, 2]. In the end, the functions of REC will be tested, and the total system is demonstrated by using JT-60SA and existing facilities in the EU, such as JET and WEST. The hardware of the REC has been prepared in Rokkasho Japan, which has the remote experiment room with a large video wall to show the plasma and operation status, IT equipment and the storage system by the reuse of the supercomputer of Helios. The REC facility will be widely used for the remote collaboration. Also, the broadband network infrastructure of 10Gbps has been produced. Using this network system, the fast data transfer from ITER to REC has been examined in 2016, and the transfer of the full data in the initial ITER experiment has been demonstrated [3]. Secure remote experiment system has been developed, using JT-60SA, that has functions for preparing and setting of shot parameters, viewing the status of control data, streaming of the plasma status, data-exchange function of shot events, and monitoring of the facility operation. The software of the remote data analysis has been progressed significantly. The unified software infrastructure to access data stored both at on- and remote- sites and the documentation management system are also being developed. Numerical simulation for preparation and performance estimation of the shots by the appropriate implementation of the simulation code, such as the integrated simulation code, and the analysis codes for the plasma shots.
