Gravity-driven microfluidics, which utilizes gravity force to drive liquid flow, offers portability and multi-condition setting flexibility because they do not require pumps or connection tubes to drive the flow. However, because the flow rate decreases with time in gravity-driven microfluidics, it is not suitable for stem cell experiments, which require long-term (at least a day) stability. In this study, gravity-driven microfluidics and a slow-tilting table were developed to culture cells under constant unidirectional perfusion. The microfluidic device was placed on a slow-tilting table, which tilts unidirectionally at a rate of approximately 7° per day to compensate for the reduction in the flow rate. Computational simulations showed that the pulsation of the flow arising from the stepwise movement of the table was less than 0.2%, and the flow was laminar. Hydrophilization of the tanks increased the flow rate, which is consistent with the theoretical values. We showed that vitronectin is better than laminin 511 fragments as a coating material for adhering human induced pluripotent stem cells on a microchamber made of polydimethylsiloxane, and succeeded in culturing the cells for 3 days. It is believed that the system offers easy-to-use cell culture tools, such as conventional multiwell culture vessels, and enables the control of the cell microenvironment.

Here we report the perfusion culture of a multi-layered tissue composed of HepG2 cells (a human hepatoma line) in a pressure-driven microphysiological system (PD-MPS), which we developed previously as a multi-throughput perfusion culture platform. This paper reports perfusion culture of human umbilical vein endothelial cells (HUVECs) on a microporous membrane in a pressure-driven microphysiological system (PD-MPS), which we developed previously as a multi-throughput perfusion culture platform. We designed fluidic culture unit with microporous membrane to culture HUVECs under fluidic shear stress and constructed a perfusion culture model in the PD-MPS platform. Four fluidic culture units were arranged in the microplate-sized device, which enables four-throughput assay for characterization of HUVECs under flow. Medium flow was generated above and below the membrane by sequential pneumatic pressure to apply physiological shear stress to HUVECs. The microphysiological system (MPS) is a promising tool for predicting drug disposition in humans, although limited information is available on the quantitative assessment of sequential drug metabolism in MPS and its extrapolation to humans. In the present study, we first constructed a mechanism-based pharmacokinetic model for triazolam (TRZ) and its metabolites in the entero-hepatic two-organ MPS, composed of intestinal Caco-2 and hepatic HepaRG cells, and attempted to extrapolate the kinetic information obtained with the MPS to the plasma concentration profiles in humans. In the two-organ MPS and HepaRG single culture systems, TRZ was found to be metabolized into α- and 4-hydroxytriazolam and their respective glucuronides. All these metabolites were almost completely reduced in the presence of a CYP3A inhibitor, itraconazole, confirming sequential phase I and II metabolism. Both pharmacokinetic model-dependent and -independent analyses were performed, providing consistent results regarding the metabolic activity of TRZ: clearance of glucuronidation metabolites in the two-organ MPS was higher than that in the single culture system. The plasma concentration profile of TRZ and its two hydroxy metabolites in humans was quantitatively simulated based on the pharmacokinetic model, by incorporating several scaling factors representing quantitative gaps between the MPS and humans. Thus, the present study provided the first quantitative extrapolation of sequential drug metabolism in humans by combining MPS and pharmacokinetic modeling.

The use of organ-on-a-chip (OOC) devices is a promising alternative to existing cell-based assays and animal testing in drug discovery. A rapid prototyping method with polydimethylsiloxane (PDMS) is widely used for developing OOC devices. However, because PDMS tends to absorb small hydrophobic molecules, the loss of test compounds in cell-based assays and increases in background fluorescence during observation often lead to biased results in cell-based assays. To address this issue, we have fabricated a glass-based OOC device and characterized the medium flow and molecular absorption properties in comparison with PDMS-based devices. Consequently, we revealed that the glass device generated a stable medium flow, restricted the absorption of small hydrophobic molecules, and showed enhanced cell adhesiveness. This glass device is expected to be applicable to precise cell-based assays to evaluate small hydrophobic molecules, for which PDMS devices cannot be applied because of their absorption of small hydrophobic molecules.

This study aims to investigate bacteriorhodopsin (bR) molecules reconstituted in lipid bilayers composed of di(nonafluorotetradecanoyl)-phosphatidylcholine (F4-DMPC), a partially fluorinated analogue of dimyristoyl-phosphatidylcholine (DMPC) to clarify the effects of partially fluorinated hydrophobic chains of lipids on protein's stability. Calorimetry measurements showed that the chain-melting transition of F4-DMPC/bR systems occurs at 3.5 °C, whereas visible circular dichroism (CD) and X-ray diffraction measurements showed that a two-dimensional (2D) hexagonal lattice formed by bR trimers in F4-DMPC bilayers remains intact even above 30 °C, similar to bR in a native purple membrane. Complete dissociation of the trimers into the monomers detected by visible CD almost coincides with the complete melting of 2D lattice observed by X-ray diffraction, in which both take place at around 65 °C (10 °C lower than that for bR in a native purple membrane). However, it is extremely high in comparison with the bR reconstituted in DMPC bilayers in which the dissociation of bR trimer in DMPC bilayers occurs near the chain-melting transition temperature of DMPC bilayers at approximately 18 °C. In order to explore the rationale behind the difference in stability, a further investigation of the detailed structural features of pure F4-DMPC bilayers was performed by analyzing the lamellar diffraction data using simple electron density models. The results suggested that the perfluoroalkyl groups do not exhibit any conformation change even if the chain-melting transition occurs, which is likely to contribute to the stability of the 2D hexagonal lattice formed by the bR trimers.

Perfluoroalkyl (Rf) chains have a specific helical conformation due to the steric repulsion between the adjacent CF2 units. Although Rf chains have no chiral center, two chiral structures, i.e., the right-handed (R) and left-handed (L) helices, are available as the most stable conformations, which are atropisomers to each other. According to the stratified dipole array (SDA) theory, the helical structure about the chain axis plays a key role in the spontaneous molecular aggregation of Rf chains in a two-dimensional manner, and the Rf chains having the same chirality tend to be aggregated spontaneously to generate molecular domains. This implies that an Rf compound in a solid state should be a mixture of the R and L domains, and each domain should exhibit distinguishable optical activity. Fluorous properties represented by water-and-oil repellency are perfluoroalkyl (Rf) compound-specific characteristics, which are widely used for surface coating of glass, electronic devices and textiles for preventing water and grease fouling. According to the stratified dipole-arrays (SDA) theory, the minimum Rf length of (CF₂)₇ is theoretically necessary for realizing fluorous properties. Unfortunately, however, production of compounds involving this chemical unit is strictly banned because of concerns of environmental pollution, which is a big dilemma. Here, we show that the fluorous properties can be realized by self-assembled monolayer (SAM) even with a short Rf-containing compound, since the SAM technique makes the best use of the self-aggregation property of the Rf groups, and it readily makes the molecules immobile.

Examining intestine–liver interactions is important for achieving the desired physiological drug absorption and metabolism response in in vitro drug tests. Multi-organ microphysiological systems (MPSs) constitute promising tools for evaluating inter-organ interactions in vitro. For coculture on MPSs, normal cells are challenging to use because they require complex maintenance and careful handling. Herein, we demonstrated the potential of coculturing normal cells on MPSs in the evaluation of intestine–liver interactions. To this end, we cocultured human-induced pluripotent stem cell-derived intestinal cells and fresh human hepatocytes which were isolated from PXB mice with medium circulation in a pneumatic-pressure-driven MPS with pipette-friendly liquid-handling options. The cytochrome activity, albumin production, and liver-specific gene expressions in human hepatocytes freshly isolated from a PXB mouse were significantly upregulated via coculture with hiPS-intestinal cells. Our normal cell coculture shows the effects of the interactions between the intestine and liver that may occur in vivo. This study is the first to demonstrate the coculturing of hiPS-intestinal cells and fresh human hepatocytes on an MPS for examining pure inter-organ interactions. Normal-cell coculture using the multi-organ MPS could be pursued to explore unknown physiological mechanisms of inter-organ interactions in vitro and investigate the physiological response of new drugs.

Cell culture under medium flow has been shown to favor human brain microvascular endothelial cells function and maturation. Here a three-dimensional in vitro model of the human brain microvasculature, comprising brain microvascular endothelial cells but also astrocytes, pericytes and a collagen type I microfiber – fibrin based matrix, was cultured under continuous medium flow in a pressure driven microphysiological system (10 kPa, in 60–30 s cycles). The cells self-organized in micro-vessels perpendicular to the shear flow. Comparison with static culture showed that the resulting interstitial flow enhanced a more defined micro-vasculature network, with slightly more numerous lumens, and a higher expression of transporters, carriers and tight junction genes and proteins, essential to the blood-brain barrier functions.

Inside living organisms, concentration gradients dynamically change over time as biological processes progress. Therefore, methods to construct dynamic microscale concentration gradients in a spatially controlled manner are needed to provide more realistic research environments. Here, we report a novel method for the construction of dynamic microscale concentration gradients in a stepwise manner around cells in micropatterned hydrogel. In our method, cells are encapsulated in a photodegradable hydrogel formed inside a microfluidic perfusion culture device, and perfusion microchannels are then fabricated in the hydrogel by micropatterned photodegradation. The cells in the micropatterned hydrogel can then be cultured by perfusing culture medium through the fabricated microchannels. By using this method, we demonstrate the simultaneous construction of two dynamic concentration gradients, which allowed us to expose the cells encapsulated in the hydrogel to a dynamic microenvironment.

The first microfluidic microphysiological systems (MPS) entered the academic scene more than 15 years ago and were considered an enabling technology to human (patho)biology in vitro and, therefore, provide alternative approaches to laboratory animals in pharmaceutical drug development and academic research. Nowadays, the field generates more than a thousand scientific publications per year. Despite the MPS hype in academia and by platform providers, which says this technology is about to reshape the entire in vitro culture landscape in basic and applied research, MPS approaches have neither been widely adopted by the pharmaceutical industry yet nor reached regulated drug authorization processes at all. Here, 46 leading experts from all stakeholders - academia, MPS supplier industry, pharmaceutical and consumer products industries, and leading regulatory agencies - worldwide have analyzed existing challenges and hurdles along the MPS-based assay life cycle in a second workshop of this kind in June 2019. They identified that the level of qualification of MPS-based assays for a given context of use and a communication gap between stakeholders are the major challenges for industrial adoption by end-users. Finally, a regulatory acceptance dilemma exists against that background. This t4 report elaborates on these findings in detail and summarizes solutions how to overcome the roadblocks. It provides recommendations and a roadmap towards regulatory accepted MPS-based models and assays for patients' benefit and further laboratory animal reduction in drug development. Finally, experts highlighted the potential of MPS-based human disease models to feedback into laboratory animal replacement in basic life science research.

Spatial pattern formation is a critical step in embryogenesis. Bone morphogenetic protein 4 (BMP4) and its inhibitors are major factors for the formation of spatial patterns during embryogenesis. However, spatial patterning of the human embryo is unclear because of ethical issues and isotropic culture environments resulting from conventional culture dishes. Here, we utilized human pluripotent stem cells (hiPSCs) and a simple anisotropic (unidirectional perfusion) culture chamber, which creates unidirectional conditions, to measure the cell community effect. The influence of cell density on BMP4-induced differentiation was explored during static culture using a conventional culture dish. Immunostaining of the early differentiation marker SSEA-1 and the mesendoderm marker BRACHYURY revealed that high cell density suppressed differentiation, with small clusters of differentiated and undifferentiated cells formed. Addition of five-fold higher concentration of BMP4 showed similar results, suggesting that suppression was not caused by depletion of BMP4 but rather by high cell density. Quantitative RT-PCR array analysis showed that BMP4 induced multi-lineage differentiation, which was also suppressed under high-density conditions. We fabricated an elongated perfusion culture chamber, in which proteins were transported unidirectionally, and hiPSCs were cultured with BMP4. At low density, the expression was the same throughout the chamber. However, at high density, SSEA-1 and BRACHYURY were expressed only in upstream cells, suggesting that some autocrine/paracrine factors inhibited the action of BMP4 in downstream cells to form the spatial pattern. Human iPSCs cultured in a perfusion culture chamber might be useful for studying in vitro macroscopic pattern formation in human embryogenesis.

We report a sharp photoinduced aqueous dissolution of the copolymer through phase transition based on the photochemical reaction of o-nitrobenzaldehyde (NBA) and the principle of polymer effect. We synthesized the copolymers having poly( N-isopropylacrylamide) main chain and NBA side chain at 4, 7, and 10 mol % functionalizations and analyzed their photoresponsive characteristics. Light with 365 nm wavelength converted NBA groups at copolymer side chains to carboxylic acid efficiently at the rate of 7.3 cm2/J, and in the case of 10 mol % functionalization, the irradiation dosage no more than 56 mJ/cm2 induced sharp aqueous dissolution of the copolymer thin layer in pH 7.4 at 25 °C. As example applications, we demonstrated on-demand release of polyethylene beads and fluorescent-labeled albumins, which had been immobilized on a substrate surface via the copolymers, by the precisely controlled light irradiation using a microprojection system. Also, we examined application of the copolymers to the selective recovery of living cells from culture substrate under microscopic observation. As a result, mild light irradiation at room temperature triggered immediate detachment of the cultured adherent cells only in the irradiated areas without critical influence on their viability.

In vertebrate development, bone morphogenetic protein (BMP) 4 and its inhibitor, Noggin, are important factors determining differentiation patterns along the body axis. In a previous study on the effects of BMP4 on cell differentiation using a perfusion culture chamber and human induced pluripotent stem (iPS) cells, we found that cells located upstream of the chamber differentiated, whereas those located downstream remained undifferentiated. Noggin specifically binds to BMP4 and prevents it from binding to BMP receptors. Therefore, cell differentiation pattern may be attributed to the spatial pattern of secreted Noggin to inhibit differentiation. In this study, we simulated concentrations of BMP4, Noggin, and BMP4-Noggin complex as a reaction-diffusion model, under three experimental conditions. We considered four boundary conditions for differentiation and Noggin secretion. The optimal simulation conditions that mimic cell differentiation were determined by comparing the experimental and simulation results. The simulation demonstrated that the two boundary conditions where cells secreted Noggin before differentiation marker expression, matched the experimental results of our previous work and references. Thus, the spatial pattern of cell differentiation can be attributed to the distribution of Noggin secreted before differentiation marker expression and to the reaction-diffusion of BMP4 and Noggin.

The CF2 symmetric stretching vibration (νs(CF2)) band of a perfluoroalkyl (Rf) group in an infrared (IR) spectrum exhibits a unique character, i.e., an apparent high wavenumber shift with increasing the chain length, which is an opposite character to that of the CH stretching vibration band of a normal alkyl chain. To reveal the mechanism of the unusual IR band shift, two vibrational characters of an Rf chain are focused: (1) a helical conformation of an Rf chain, (2) the carbon (C) atoms having a smaller mass than the fluorine (F) atom dominantly vibrate as a coupled oscillator leaving F atoms stay relatively unmoved. These indicate that a ‘coupled oscillation of the skeletal C atoms’ of an Rf chain should be investigated considering the helical structure. In the present study, therefore, the coupled oscillation of the Rf chain dependent on the chain length is investigated by Raman spectroscopy, which is suitable for investigating a skeletal vibration. The Raman active νs(CF2) band is found to be split into two bands, and the splitting is readily explained by considering the helical structure and length with respect to group theory, and the unusual IR peak shift is concluded to be explained by the helical length.

A novel cell processing tool to cut and section a cultured cell monolayer was implemented by the micro-projection of grid pattern onto a substrate functionalized with photo-acid-generating polymer. In adequate conditions of irradiation dosage and line width of grid pattern, Madin-Darby canine kidney cell monolayer was cut into cell clumps with similar size corresponding to the projected grid pattern. The sectioned monolayers were collected as cell clumps with the similar size from the substrate maintaining their viability enough to outgrow after subculture to the other dish. Also the influence of cutout size on the growth rate of collected cell clumps after subculture was also investigated. Further, passage of human induced pluripotent stem cells with size uniformity of the colonies was achieved by using this sectioning method without critical influence on their viability and undifferentiated state.

MPS (microphysiological system) is in-vitro cell-culture environment, which is precisely maintained in a micro space manufactured using MEMS (micro electro mechanical systems) technology, to derive in-vivo like functions from human cells. From the viewpoint of pharmacokinetics, it can be considered as a wet PBPK/PD simulator consisting of the micro organs (cell culture units) connected each other with a circulating medium, which mimic the organs responsible for the drug's ADME (absorption-distribution-metabolism-elimination). In this review, we identify two types of the cell culture units consisting of the MPS for pharmacokinetics, and overview the characteristics of each type. Then, we discuss about the technical requirements needed for the cell culture unit from the point of view of both cell-culture environmental design and cell function, and introduce the world-wide current situation of the commercialization of the MPS.

Photoresponsive actuators composed of a hydrogel functionalized with spiropyran are described. The hydrogel exhibits drastic shrinking in rapid response to blue light irradiation in acidic aqueous systems, and is examined for the application of several photoresponsive actuator systems. Rod-like hydrogel bends drastically after 1 s light irradiation, and microrelief is formed instantly on the hydrogel sheet by the micropatterned light irradiation. Based on these characteristics of the hydrogel, a photo-controllable microfluidic system is constructed with a hydrogel sheet, and the microchannels with arbitrary width, height and pathway are formed instantly by the micropatterned light irradiation. Also independent and parallel control of microvalve array by local light irradiation is demonstrated for a similar microfluidic system combined with fixed microchannel.

To examine the applicability of fluorinated membrane-forming phospholipids to reconstitution matrices for functional membrane proteins, the membrane properties of a synthetic ether-type phosphatidylcholine (PC) bearing partially fluorinated C18-monoacetylenic (9-octadecynyl) chains, DF8CCH8PC, were compared with those of its non-fluorinated counterpart, DH8CCH8PC. Light-harvesting complex 2 (LH2) and the light-harvesting 1‒reaction center core complex (LH1-RC) isolated from purple photosynthetic bacteria were employed as probe membrane proteins to evaluate the extent to which their reconstitution into DF8CCH8PC membranes could proceed. DF8CCH8PC formed more expanded and more stable fluid monolayers than DH8CCH8PC at the air-water interface at 25 °C; the former PC molecule occupied an area of ca. 0.70 nm2 at a collapse pressure, πc, of 52 mN/m, while the latter occupied an area of ca. 0.55 nm2 at a πc of 45 mN/m. In contrast, the molecular motion detected using fluorescent probes was much more restricted in DF8CCH8PC bilayers than in DH8CCH8PC ones. Although the reconstitution efficiencies of both LH2 and LH1-RC into DF8CCH8PC bilayers were lower than those into DH8CCH8PC bilayers, the membrane proteins incorporated into DF8CCH8PC bilayers showed increased thermostability. The increased thermostability of these proteins in fluorinated PC membranes might be due to the restricted molecular motion in the hydrophobic chains. The results of this study suggest that partially fluorinated PCs can be useful materials for the construction of lipid‒functional membrane protein assemblies including large membrane protein complexes, such as LH1-RC, for biotechnological applications.

The development of biologically relevant three-dimensional (3D) tissue constructs is essential for the alternative methods of organ transplantation in regenerative medicine, as well as the development of improved drug discovery assays. Recent technological advances in hydrogel microfabrication, such as micromolding, 3D bioprinting, photolithography, and stereolithography, have led to the production of 3D tissue constructs that exhibit biological functions with precise 3D microstructures. Furthermore, microfluidics technology has enabled the development of the perfusion culture of 3D tissue constructs with vascular networks. In this review, we present these hydrogel microfabrication technologies for the in vitro reconstruction and cultivation of 3D tissues. Additionally, we discuss current challenges and future perspectives of 3D tissue engineering., Highlights • The overview of recent technological advances in hydrogel microfabrication. • Hydrogel microfabrication technologies in 3D tissue engineering. • The usability of microfluidics combined with hydrogel microfabrication.

Dynamically controlled construction of microstructures was demonstrated using a spirobenzopyran-containing poly(N-isopropylacrylamide). Under patterned light irradiation, the polymer accumulated at the irradiated area from a uniformly dissolved state and formed micro-patterned structures based on phase transition. Furthermore the microstructures were collapsed due to redissolution of the precipitated polymer when light irradiation was stopped.

Here, we report a pneumatic pressure-driven microfluidic device capable of multi-throughput medium circulation culture. The circulation culture system has the following advantages for application in drug discovery: (i) simultaneous operation of multiple circulation units, (ii) use of a small amount of circulating medium (3.5 mL), (iii) pipette-friendly liquid handling, and (iv) a detachable interface with pneumatic pressure lines via sterile air-vent filters. The microfluidic device contains three independent circulation culture units, in which human umbilical vein endothelial cells (HUVECs) were cultured under physiological shear stress induced by circulation of the medium. Circulation of the medium in the three culture units was generated by programmed sequentially applied pressure from two pressure-control lines. HUVECs cultured in the microfluidic device were aligned under a one-way circulating flow with a shear stress of 10 dyn cm(-2); they exhibited a randomly ordered alignment under no shear stress and under reciprocating flow with a shear stress of 10 dyn cm(-2). We also observed 2.8- to 4.9-fold increases in expression of the mRNAs of endothelial nitric oxide synthase and thrombomodulin under one-way circulating flow with a shear stress of 10 dyn cm(-2) compared with conditions of no shear stress or reciprocating flow.

Polymer bonding is widely employed in thermal welding, ultrasonic welding, laser welding, and surface-activated bonding. To adapt bonding properties to the smaller microstructure and complexity of a microfluidic chip, the bonding process should produce low polymer structure deformation and strong bonding strength. To fabricate a microfluidic chip without an adhesive layer, a new precision polymer bonding process that employs a nanostructure and two different layers of the same polymer material with different glass transition temperatures (Tg ) is devised. Low-temperature bonding (less than 20 °C) was achieved. A deformation ratio of 1.1% was obtained, which is 10 times smaller than that for the typical thermal-bonding process. Thus, a precision bonding technique for a low-deformation microstructured chip was successfully developed.

Interfacial properties and membrane protein solubilization activity of a series of partially fluorinated octyl-phosphocholine (PC) surfactants were investigated from the viewpoint of the fluorination degree of the hydrophobic chain. The critical micelle concentration (CMC), surface tension lowering activity, molecular occupied area at the CMC and free energy changes of micellization as well as adsorption to the air-water interface for each PC surfactant were estimated from surface tension measurements at 25 °C. The PCs with higher degree of fluorination exhibited low CMC and high surface activity, while the single trifluoromethyl group at the end of the chain appeared to enhance the hydrophilicity of the surfactant molecule. Under conditions where conventional short-chain surfactants, n-octyl-β-D-glucoside, Triton X-100 and dioctanoylphosphatidylcholine significantly solubilize purple membranes (PM), none of the fluorinated-PCs solubilized PM. This suggests that fluorinated-PCs are low-invasive enough to maintain the structure of lipids/protein assemblies like PM.

Photolithographic fabrication of unique microstructures composed of flexible hydrogel sheets is proposed and demonstrated by using photo-acid-generating poly(methyl methacrylate). Crosslinking of a hydroxyl-rich polymer and lifting off of the crosslinked polymer layer from the substrate are controlled respectively in an area-selective manner upon micropatterned light irradiation, and various pocket-like microstructures are fabricated resultantly.

Conventional cell handling and sorting methods require manual labor, which decreases both cell quality and quantity. To purify adherent cultured cells, cell purification technologies that are high throughput without dissociation and can be utilized in an on-demand manner are expected. Here, we developed a Laser-induced, Light-responsive-polymer-Activated, Cell Killing (LiLACK) system that enables high-speed and on-demand adherent cell sectioning and purification. This system employs a visible laser beam, which does not kill cells directly, but induces local heat production through the trans-cis-trans photo-isomerization of azobenzene moieties. Using this system in each passage for sectioning, human induced pluripotent stem cells (hiPSCs) maintained their pluripotency and self-renewal during long-term culture. Furthermore, combined with deep machine-learning analysis on fluorescent and phase contrast images, a label-free and automatic cell processing system has been developed by eliminating unwanted spontaneously differentiated cells in undifferentiated hiPSC culture conditions., Yohei Hayashi et al. present a method for high-speed adherent cell sectioning and purification, along with a label-free and automatic cell processing system. They show that this method is able to section human induced pluripotent stem cells without losing pluripotency and viability.

Summary Blue light-responsive photoacid generator (PAG) was pendant to poly(N-isopropylacrylamide) gel which bound on a glass plate. The gel was compressed in thickness direction by natural drying and then was irradiated with blue light. Reswelling of the gel in water was suppressed at irradiated area with a dose-dependent effect. The results were probably due to formation of additional crosslinkng of polymer network by blue light-irradiation at dry state. By using the PAG-bearing gel, a microfluidic device was fabricated in which microchannel was formed by light irradiation and subsequent water filling.

This paper reports a multi-throughput multi-organ-on-a-chip system formed on a pneumatic pressure-driven medium circulation platform with a microplate-sized format as a novel type of microphysiological system. The pneumatic pressure-driven platform enabled parallelized multi-organ experiments (i.e. simultaneous operation of multiple multi-organ culture units) and pipette-friendly liquid handling for various conventional cell culture experiments, including cell seeding, medium change, live/dead staining, cell growth analysis, gene expression analysis of collected cells, and liquid chromatography–mass spectrometry analysis of chemical compounds in the culture medium. An eight-throughput two-organ system and a four-throughput four-organ system were constructed on a common platform, with different microfluidic plates. The two-organ system, composed of liver and cancer models, was used to demonstrate the effect of an anticancer prodrug, capecitabine (CAP), whose metabolite 5-fluorouracil (5-FU) after metabolism by HepaRG hepatic cells inhibited the proliferation of HCT-116 cancer cells. The four-organ system, composed of intestine, liver, cancer, and connective tissue models, was used to demonstrate evaluation of the effects of 5-FU and two prodrugs of 5-FU (CAP and tegafur) on multiple organ models, including cancer and connective tissue.

Microphysiological systems (MPS) are currently attracting a lot of interest from pharmaceutical companies worldwide. In the United States and European Union, several large government projects related to MPS have been initiated, and, in Japan, pharmaceutical companies interested in MPS are watching the recent trends and developments in the field. In July 2017, the Japan Agency for Medical Research and Development initiated a research program to develop chip-based MPS. In this review, we examine the technical aspects of commercializing chip-based MPS.

The photo- and thermoresponse of poly(N-isopropylacrylamide) (pNIPAAm) functionalized with spiropyran chromophore is examined with respect to the influence of molecular recognition by cyclodextrin (CD). Characterization in aqueous solutions of spiropyran-functionalized poly(N,N-dimethylacrylamide) under coexistence of α-, β-, or γ-CD reveals that β-CD selectively includes the ring-closing isomer of the chromophore, which is dominant under light irradiation, while no inclusion is observed for the protonated ring-opening isomer, which is dominant in the dark before irradiation. As a result, it is shown that the selective inclusion of the chromophore at a polymer side chain is switched by light irradiation. Further, drastic photoresponsive dehydration of spiropyran-functionalized pNIPAAm is inhibited only by β-CD out of three examined CDs, demonstrating that the molecular recognition regulates the dehydration of the whole polymer triggered by the photoswitching of the chromophore introduced at only 1 mol% functionalization.

Understanding tumor heterogeneity is an urgent and unmet need in cancer research. In this study, we used a morphology-based optical cell separation process to classify a heterogeneous cancer cell population into characteristic subpopulations. To classify the cell subpopulations, we assessed their morphology in hydrogel, a three-dimensional culture environment that induces morphological changes according to the characteristics of the cells (i.e., growth, migration, and invasion). We encapsulated the murine breast cancer cell line 4T1E, as a heterogeneous population that includes highly metastatic cells, in click-crosslinkable and photodegradable gelatin hydrogels, which we developed previously. We observed morphological changes within 3 days of encapsulating the cells in the hydrogel. We separated the 4T1E cell population into colony- and granular-type cells by optical separation, in which local UV-induced degradation of the photodegradable hydrogel around the target cells enabled us to collect those cells. The obtained colony- and granular-type cells were evaluated in vitro by using a spheroid assay and in vivo by means of a tumor growth and metastasis assay. The spheroid assay showed that the colony-type cells formed compact spheroids in 2 days, whereas the granular-type cells did not form spheroids. The tumor growth assay in mice revealed that the granular-type cells exhibited lower tumor growth and a different metastasis behavior compared with the colony-type cells. These results suggest that morphology-based optical cell separation is a useful technique to classify a heterogeneous cancer cell population according to its cellular characteristics.

The effect of the fluorination degree of hydrophobic chains on the monolayer behavior of unsaturated diacylphosphatidylcholines (PCs) was examined by employing a series of PCs bearing partially fluorinated 9-octadecynoyl (stearoloyl) groups (DFnStPCs, n: the number of fluorinated carbon atoms in a stearoloyl group; n = 1, 2, 4, 8), including their hydrophobic parts – partially fluorinated stearolic acids (FnStAs) – at the air–water interface. π–A isotherm measurements and Brewster angle microscope observations revealed: (i) all DFnStPCs including FnStAs form monolayers of liquid character at 25 °C; (ii) they form more expanded monolayers than their non-fluorinated counterparts, distearoloyl-PC (DStPC) and stearolic acid, while the monolayer stability increases with n; (iii) compared with DStPC and DF8StPC, DFnStPCs (n = 1, 2, 4) in the low-π region tend to show a weakening in their self-aggregation property and an increase in the work required for monolayer compression; (iv) although DF8StPC forms the most expanded monolayer, the behavior of DF8StPC resembles that of DStPC rather than that of DFnStPCs (n = 1, 2, 4). The monolayer behavior of DFnStPCs (n = 1, 2, 4) is explained by postulating a flatly-lying conformation of hydrophobic chains, in which three polar parts (ester group, triple bond, CF2 CH2 linkage) in chains are immersed in the subphase at large areas. DStPC and DF8StPC lacking a CF2 CH2 linkage, however, do not likely adopt such a conformation.

We developed a microfluidic perfusion cell culture chip that provides three different shear stress strengths and a large cell culture area for the analysis of vascular endothelial functions. The microfluidic network was composed of shallow flow-control channels of three different depths and deep cell culture channels. The flow-control channels with high fluidic resistances created shear stress strengths ranging from 1.0 to 10.0 dyn/cm(2) in the cell culture channels. The large surface area of the culture channels enabled cultivation of a large number (approximately 6.0 × 10(5)) of cells. We cultured human umbilical vein endothelial cells (HUVECs) and evaluated the changes in cellular morphology and gene expression in response to applied shear stress. The HUVECs were aligned in the direction of flow when exposed to a shear stress of 10.0 dyn/cm(2). Compared with conditions of no shear stress, endothelial nitric oxide synthase mRNA expression increased by 50% and thrombomodulin mRNA expression increased by 8-fold under a shear stress of 9.5 dyn/cm(2).

Perfluoroalkyl compounds are known to exhibit a hydrophobic character on the surface of the material, although the CF bond has a large dipole, which should make the molecular surface polar and hydrophilic. This inconsistency has long been a chemical matter to be solved. Herein, a stratified dipole-arrays model is proposed: the molecular polar surface can be fully hidden by forming a two-dimensional aggregate of perfluoroalkyl (Rf) groups; this aggregate is spontaneously induced by dipole–dipole interaction arrays owing to the helical structure of the Rf group. In this model, a ‘short’ Rf group should play the role of a single Rf group with a hydrophilic character, whereas a ‘long’ Rf group should spontaneously form a hexagonal aggregate. To examine this model, Rf-containing myristic acids with various Rf lengths have been synthesized and their aggregation properties are analyzed by using the Langmuir monolayer technique aided by precise IR spectroscopic analysis.

Novel hydrogels that shrink in response to light irradiation and temperature change have been developed. Since any technical application of polymer gels critically depends on the response rate, therefore porous hydrogels have been prepared in order to accelerate the response reaction. Poly( N -isopropylacrylamide) gels were functionalized with photo-responsive spirobenzopyran and solvent/nonsolvent mixtures were used as swelling agent to introduce network heterogeneities during the gelation process. Both photo- and thermo-responsive properties of gels have been investigated and compared. The time dependence of bending deformation due to the asymmetric light irradiation as well as kinetics of shrinking has been measured. It was found that the fastest photo- and thermo-responsive shrinking was observed for loosely crosslinked porous hydrogels. Furthermore, it was confirmed that the highly cross-linked cylindrical gel rod with macroporous structure showed rapid bending deformation due to the asymmetric light irradiation.
