Abstract Background Bacteria of the candidate phyla radiation (CPR), constituting about 25% of the bacterial biodiversity, are characterized by small cell size and patchy genomes without complete key metabolic pathways, suggesting a symbiotic lifestyle. Gracilibacteria (BD1-5), which are part of the CPR branch, possess alternate coded genomes and have not yet been cultivated. The lifestyle of Gracilibacteria, their temporal dynamics, and activity in natural ecosystems, particularly in groundwater, has remained largely unexplored. Here, we aimed to investigate Gracilibacteria activity in situ and to discern their lifestyle based on expressed genes, using the metaproteogenome of Gracilibacteria as a function of time in the cold-water geyser Wallender Born in the Volcanic Eifel region in Germany. Results We coupled genome-resolved metagenomics and metaproteomics to investigate a cold-water geyser microbial community enriched in Gracilibacteria across a 12-day time-series. Groundwater was collected and sequentially filtered to fraction CPR and other bacteria. Based on 725 Gbps of metagenomic data, 1129 different ribosomal protein S3 marker genes, and 751 high-quality genomes (123 population genomes after dereplication), we identified dominant bacteria belonging to Gallionellales and Gracilibacteria along with keystone microbes, which were low in genomic abundance but substantially contributing to proteomic abundance. Seven high-quality Gracilibacteria genomes showed typical limitations, such as limited amino acid or nucleotide synthesis, in their central metabolism but no co-occurrence with potential hosts. The genomes of these Gracilibacteria were encoded for a high number of proteins involved in cell to cell interaction, supporting the previously surmised host-dependent lifestyle, e.g., type IV and type II secretion system subunits, transporters, and features related to cell motility, which were also detected on protein level. Conclusions We here identified microbial keystone taxa in a high-CO2 aquifer, and revealed microbial dynamics of Gracilibacteria. Although Gracilibacteria in this ecosystem did not appear to target specific organisms in this ecosystem due to lack of co-occurrence despite enrichment on 0.2-µm filter fraction, we provide proteomic evidence for the complex machinery behind the host-dependent lifestyle of groundwater Gracilibacteria. ABSTRACT: Sustainable development principles have become a common consensus and a challenge to humanity, shifting the societal focus towards greener alternatives. The chemical sector, in particular analytical chemistry tends to attain a leading role in this initiative as it strives to incorporate green chemistry principles when designing and proposing new chemical solutions. Sample preparation is considered a critical stage in an analytical procedure regarding waste generation and resource consumption. The main goal of this work was to evaluate the sample preparation greenness of selected analytical standard procedures. Accordingly, three reference methods from the United States Environmental Protection Agency (EPA methods 523, 528, and 610) and one method from the German Institute for Standardization (DIN 38047–37), which use classical solid-phase and liquid-liquid extraction as the core of their sample preparation procedures, were evaluated via AGREEprep, a green metric tool designed specifically for evaluation of sample preparation greenness. In addition, the methods were compared with twenty novel analytical alternatives reported in the literature, including a variety of solid-phase and liquid-phase microextraction techniques. In all cases, the miniaturized sample preparation strategies showed superior greenness over the standard methods, while providing similar or better analytical performance. The main evidenced shortcomings of the standard methods were the large sample volume required for the extraction and the use of large volumes of organic solvents. The overall results confirmed that the shift toward green approaches for sample preparation integrating sustainable extraction protocols and greener methodologies has the potential to improve the environmental performance of the analytical method.

Abstract While macroinvertebrate dispersal operates at the individual level, predictions of their dispersal capabilities often rely on indirect proxies rather than direct measurements. To gain insight into the dispersal of individual specimens, it is crucial to mark (label) and capture individuals. Isotopic enrichment with 15N is a non‐invasive method with the potential of labelling large quantities of macroinvertebrates. While the analysis of 15N is widely utilised in food web studies, knowledge on the specific utility of isotopic enrichment with 15N for mass labelling of macroinvertebrate individuals across different taxa and feeding types is limited. Previous studies have focused on single species and feeding types, leaving gaps in our understanding of the broader applicability of this method. Therefore, this study aimed to test and compare isotopic mass enrichment across several macroinvertebrate taxa and feeding types. We released 15NH4Cl at five stream reaches in North‐Rhine Westphalia, Germany, and successfully enriched 12 distinct macroinvertebrate taxa (Crustacea and Insecta). Significant enrichment was achieved in active and passive filter feeders, grazers, shredders and predators, and predominantly showed positive correlations with the enrichment of the taxa's main food sources phytobenthos and particulate organic matter. Enrichment levels rose rapidly and peaked at distances between 50 m and 300 m downstream of the isotopic inlet; significant enrichment occurred up to 2000 m downstream of the isotopic inlet in all feeding types. Macroinvertebrate density estimates on the stream bottom averaged to a total of approximately 3.4 million labelled individuals of the 12 investigated taxa, thus showing the high potential of isotopic (15N) enrichment as a non‐invasive method applicable for mass labelling across different macroinvertebrate feeding types. Hence, isotopic enrichment can greatly assist the analysis of macroinvertebrate dispersal through mark‐and‐recapture experiments, as it allows to measure the movement at the level of individual specimens.

The rising discharge of anthropogenic chemicals into aquatic environments poses a significant threat, necessitating effective monitoring strategies. This study introduces an innovative approach to effect-directed analysis (EDA) by coupling liquid chromatography (LC) with high-performance thin-layer chromatography (HPTLC), utilizing a modified MALDI spotter. The objective is to optimize fractionation parameters for sample application and assess the method's viability in identifying acetylcholinesterase (AChE) inhibitors, specifically malathion, parathion, and chlorfenvinphos.The optimization process involves controlling sample volume, spot shape, and spot distances on HPTLC plates. Successful application is demonstrated by a miniaturized LC system coupled to the HPTLC plate via spotter, allowing effective separation and identification of AChE inhibitors. The study further explores the method's application to water samples from a river with predominantly agricultural drainage area.Spiked samples reveal distinct active spots, identified through extraction and subsequent high-resolution mass spectrometry (HRMS) measurements. However, results indicate the absence of AChE inhibitors in non-spiked water samples, affirming the efficacy of the EU ban on most organophosphate pesticides. The usefulness of HPTLC in separation of coeluting substances from HPLC is emphasized, demonstrating its suitability for the effect-directed analysis of complex samples.This work shows the integration of HPTLC with liquid chromatography for EDA, offering a powerful tool for identifying and monitoring AChE inhibitors in water samples. The approach addresses limitations in current monitoring strategies and provides insights into the presence and impact of chemicals in aquatic ecosystems. The study contributes to ongoing efforts to enhance water quality monitoring, aligning with the principles of the EU Water Framework Directive.

The investigation of atmospheric microplastic pollution is a rising topic within microplastic research. However, sampling strategies concerning atmospheric microplastics are not standardized yet and strongly depend on the scientific objectives investigated. A few research groups are currently focused on determining atmospherically deposited microplastics in moss, as it can be a biomonitoring system for other atmospheric pollutants such as heavy metals, nitrogen, and persistent organic components. In this context, and for atmospheric microplastics in general, almost all examinations focus on determining microplastic numbers and shapes, while polymer masses are barely investigated. However, particle- and mass-based information are needed to assess the fate of atmospherically transported microplastics in the environment. For this purpose, this study shows the development and evaluation of a new sample preparation method for determining microplastics in moss for both analytical approaches using thermal extraction desorption-gas chromatography-mass spectrometry (TED-GC-MS) and Raman microspectroscopy (µRaman). Further, this newly developed microplastics/moss separation method (µPEEL) was compared to oxidative digestion using Fenton's reagent, usually used as a sample preparation method for organic-rich samples and moss. The method comparison was performed concerning green analytical chemistry (GAC) and its respective method functionality. The greenness was assessed using the software tool AGREEprep, which covers ten comprehensive aspects of GAC. Both methods’ functionality was assessed regarding observable matrix interferences and the data quality achieved. It is demonstrated that µPEEL benefits GAC and method functionality since harmful chemicals can be avoided, matrix interferences can be minimized, and the separation quality is increased.

Waterpipe tobacco wastes (WTWs) may contain considerable levels of hazardous contaminants such as BTEX (benzene, toluene, ethylbenzene, o-xylene, and m/p-xylene). However, no research has been carried out on BTEX levels in WTWs and the release of these pollutants into the water environment. This research examined the levels of BTEX in WTWs of flavored/local tobacco and also the release rate of these toxins into three kinds of water, including seawater (SW), tap water (TW), and distilled water (DW) with different leaching times (15, 30 min, 1.2, 4, 8 h, and 1, 2, and 4 days). The mean contents of BTEX in WTW samples of Al-Mahmoud, Al-Fakher, Mazaya, Al-Ayan brands, and local tobacco samples were 17.0 ± 4.14, 19.1 ± 4.65, 19.6 ± 4.19, 18.8 ± 4.14, and 3.16 ± 0.63 μg/kg, respectively. Abstract Background Stone wool is an inorganic mineral insulation material increasingly used to reduce the climate impact of buildings. The acute and chronic ecotoxicological potential of stone wool eluates have been studied in a battery of standardized laboratory ecotoxicological tests. The experiments were conducted with stone wool test materials in fibrous and milled form, with and without the presence of organic binder. For the preparation of eluates, the OECD protocol on the transformation/dissolution of metals and metal compounds was applied. The resulting eluates were used in acute tests, i.e., bioluminescence test with Aliivibrio fischeri (DIN EN ISO 11348-1:2009), algae growth test with Desmodesmus subspicatus (OECD No. 201) and immobilization test with Daphnia magna (OECD No. 202), as well as chronic tests, i.e., the Daphnia magna reproduction test (OECD No. 211) and the nematode growth and reproduction test with Caenorhabditis elegans (ISO 10872:2010). Results While no acute or chronic ecotoxicological effects of the eluates were observed for fibrous stone wool material, the milled test materials showed some chronic effects on aquatic invertebrates. Depending on the test materials and concentrations of milled stone wool used in the eluate preparation, these chronic effects included significant stimulation or inhibition of daphnid reproduction and nematode growth. The chemical analysis conducted in parallel to the ecotoxicological assessment indicated no leaching of organic substances from the applied binder or mineral oils and no formation of nanoparticles by the milling of stone wool. Furthermore, ICP-MS and ICP-OES analysis of eighteen elements revealed that only aluminum and nickel could be quantified in the eluates, at concentrations of approximately 750 µg/L and 7 µg/L, respectively. Conclusions Based on the present ecotoxicological assessment, eluates from stone wool fibers cannot be considered as chemically hazardous to the aquatic environment. However, additional investigations of the ecotoxicological potential of the milled material and the environmental exposure of stone wool products are necessary for a complete evaluation of potentially negative effects of stone wool insulation materials.

Abstract Stable isotope analysis of individual compounds is emerging as a powerful tool to study nutrient origin and conversion in host-parasite systems. We measured the carbon isotope composition of amino acids and glucose in the cestode Schistocephalus solidus and in liver and muscle tissues of its second intermediate host, the three-spined stickleback (Gasterosteus aculeatus), over the course of 90 days in a controlled infection experiment. Similar linear regressions of δ13C values over time and low trophic fractionation of essential amino acids indicate that the parasite assimilates nutrients from sources closely connected to the liver metabolism of its host. Biosynthesis of glucose in the parasite might occur from the glucogenic precursors alanine, asparagine and glutamine and with an isotope fractionation of − 2 to – 3 ‰ from enzymatic reactions, while trophic fractionation of glycine, serine and threonine could be interpreted as extensive nutrient conversion to fuel parasitic growth through one-carbon metabolism. Trophic fractionation of amino acids between sticklebacks and their diets was slightly increased in infected compared to uninfected individuals, which could be caused by increased (immune-) metabolic activities due to parasitic infection. Our results show that compound-specific stable isotope analysis has unique opportunities to study host and parasite physiology.

The choice of the extraction material is a critical step in solid-phase microextraction (SPME), especially the extraction of analytes which strongly differ in their polarity is challenging. This study is intended to give first insights into the headspace extraction of analytes with large differences in polarity by using a novel hydrophilic-lipophilic-balanced (HLB) SPME material with different particle sizes (5 µm, 5 + 30 µm, and 30 µm) in classical SPME, as well as SPME arrow format. The obtained results were compared to conventional and already established divinylbenzene carbon wide-range polydimethylsiloxane (DVB-CWR-PDMS) and PDMS SPME arrow fibers. The chosen model analytes toluene, indole, phenol, anisole, o-xylene, naphthalene, 2-heptanone, n-dodecane, and lindane are assigned to different substance classes with Kaw values ranging from -5.0–2.5 and Kow values from 1.5 to 7.5. The highest amount of the analytes was extracted by the HLB 5 + 30 µm arrow and the HLB 5 µm arrow, whereas the HLB SPME with a six times smaller phase volume compared to an SPME arrow, performed almost as well as the DVB-CWR-PDMS arrow. The 8-point calibration with the 5 + 30 µm HLB SPME arrow in the range of 5–70 µg L−1 of the analytes showed good linearity with R2 values ranging from 0.9765 to 0.9982, MDLs from 0.9 to 6.2 µg L−1 and RSDs from 2 to 31 %. For all of the observed analytes, the HLB material performed better than the conventional extraction materials and has great potential to replace the conventional extraction materials in many applications.

Aromatic amines (AAs) in urine typically require complex, labor-intensive and time-consuming sample preparation procedures before they can be analyzed. Usually it consists of hydrolysis, extraction and, especially when analyzed with GC, derivatization. Traditionally, these steps are done manually, significantly contributing to the total analysis time and providing opportunities for human errors. Automation presents several advantages, such as minimized human intervention and errors, and an overall greener analytical procedure.In this study, the automation of the AAs sample preparation procedure for urine samples was investigated. Problems encountered during the automation and adjustments made to the original protocol are discussed in detail. Some examples include volume limitations or needle penetration depth adjustments. Taking advantage of the automation, several steps of the sample preparation procedure could be further optimized, such as reaction/extraction times or some of the reagents which were not optimal for the automated set-up.The automated procedure presented here enables a user-friendly and green approach for the analysis of AA in urine, which could be used to gain a better understanding between smoking, AA concentrations in urine, and the risk of developing smoking related diseases.

Abstract Interpretation of stable isotope data is of upmost importance in ecology to build sound models for the study of animal diets, migration patterns and physiology. However, our understanding of stable isotope fractionation and incorporation into consumer tissues is still limited. We therefore measured the δ13C values of individual amino acids over time from muscle and liver tissue of three-spined sticklebacks (Gasterosteus aculeatus) on a high protein diet. The δ13C values of amino acids in the liver quickly responded to small shifts of under ± 2.0‰ in dietary stable isotope compositions on 30-day intervals. We found on average no trophic fractionation in pooled essential (muscle, liver) and non-essential (muscle) amino acids. Negative Δδ13C values of − 0.7 ± 1.3‰ were observed for pooled non-essential (liver) amino acids and might indicate biosynthesis from small amounts of dietary lipids. Trophic fractionation of individual amino acids is reported and discussed, including unusual Δδ13C values of over + 4.9 ± 1.4‰ for histidine. Arginine and lysine showed the lowest trophic fractionation on individual sampling days and might be useful proxies for dietary sources on short time scales. We suggest further investigations using isotopically enriched materials to facilitate the correct interpretation of ecological field data.

Abstract Coffee is typically brewed by extracting roasted and milled beans with hot water, but alternative methods such as cold brewing became increasingly popular over the past years. Cold-brewed coffee is attributed to health benefits, fewer acids, and bitter substances. But the preparation of cold brew typically needs several hours or even days. To create a cold-brew coffee within a few minutes, we present an approach in which an ultrashort-pulsed laser system is applied at the brewing entity without heating the powder suspension in water, efficiently extracting caffeine and aromatic substances from the powder. Already 3 min irradiation at room temperature leads to a caffeine concentration of 25 mg caffeine per 100 ml, comparable to the concentrations achieved by traditional hot brewing methods but comes without heating the suspension. Furthermore, the liquid phase’s alkaloid content, analyzed by reversed-phase liquid chromatography coupled to high-resolution mass spectrometry, is dominated by caffeine and trigonelline and is comparable to traditional cold-brewed coffee rather than hot-brewed coffee. Furthermore, analyzing the head-space of the prepared coffee variants, using in-tube extraction dynamic head-space followed by gas chromatography coupled to mass spectrometry, gives evidence that the lack of heating leads to the preservation of more (semi-)volatile substances like pyridine, which provide cold-brew coffee its unique taste. This pioneering study may give the impetus to investigate further the possibility of cold-brewing coffee, accelerated by more than one order of magnitude, using ultrafast laser systems.

Microplastics analysis in solid environmental samples like sediments and soils requires complex preparation steps to eliminate interfering matrix components. This is often combined with the use of highly concentrated, expensive, and environmentally harmful chemicals. These sample preparation methods can be very time-consuming. Hence, with regard to green analytical chemistry, reduction of emissions and hazards, these sample preparation methods should be modified or avoided, and more environmentally friendly methods should be developed. The characterization and evaluation of the optimized hydrophobicity-water/air-based enrichment cell for microplastics (µSEP) are presented in this study. The separation mechanism of µSEP is based on the hydrophobic adhesion of microplastic on finely dispersed upstreaming air bubbles, serving as a sustainable and practical microplastic sample preparation method. The separation mechanism requires only water and air to considerably reduce the matrix for microplastic analysis. Using soil samples as an example, the applicability of the µSEP sample preparation method for the analysis of microplastics by Raman microspectroscopy (µRaman) and thermal extraction-desorption gas chromatography-mass spectrometry (TED-GC–MS) is demonstrated. Within a sample preparation time of 60 min, for polyethylene terephthalate (PET), an average recovery of 77% ± 11%, for polystyrene (PS), 42% ± 11%, and for polypropylene (PP) 65% ± 12% was determined. Furthermore, this study evaluates µSEP and other sample preparation methods regarding their ecotoxicological impact based on the recently released software tool AGREEprep. This tool considers ten individual criteria to cover influencing factors of green analytical chemistry.

Abstract Intraspecific diet specialization, usually driven by resource availability, competition and predation, is common in natural populations. However, the role of parasites on diet specialization of their hosts has rarely been studied. Eye flukes can impair vision ability of their hosts and have been associated with alterations of fish feeding behavior. Here it was assessed whether European perch (Perca fluviatilis) alter their diet composition as a consequence of infection with eye flukes. Young-of-the-year (YOY) perch from temperate Lake Müggelsee (Berlin, Germany) were sampled in two years, eye flukes counted and fish diet was evaluated using both stomach content and stable isotope analyses. Perch diet was dominated by zooplankton and benthic macroinvertebrates. Both methods indicated that with increasing eye fluke infection intensity fish had a more selective diet, feeding mainly on the benthic macroinvertebrate Dikerogammarus villosus, while less intensively infected fish appeared to be generalist feeders showing no preference for any particular prey type. Our results show that infection with eye flukes can indirectly affect interaction of the host with lower trophic levels by altering the diet composition and highlight the underestimated role of parasites in food web studies.

Honey is the oldest and nowadays widely used natural sweetener for food worldwide. Its composition is associated with its botanical and geographical origin and honey is often mislabeled and has a high potential for food fraud. Thus, quick easy and sensitive analyses are required. For the first time, we developed and applied an automated, fast, sensitive and robust, in-tube extraction dynamic headspace in-tube extraction-dynamic headspace (ITEX-DHS) method for a variety of Honey containing VOCs in connection with GC–MS. Another advantage of ITEX is, that it is a green analytical solventless method. The method provides very low method detection limits (MDL) from 0.8 to 47 ng g−1 for VOCs in honey samples as well as very good repeatabilities with averages below 9 % RSD. Recoveries are between 83 and 100 %. Only octanal possess a repeatability 13 % and a recovery of 62 % due to its high polarity. 38 honey samples were measured after method validation. Four acacia honeys (A), six forest honeys (F) and 22 blossom honeys (B). The type of six honeys was not known (U) but could be predicted with the help of a linear discriminant analysis (LDA). The LDA was carried out with the three groups (A, B, F) leading to a proportion of correct predictions of 90.6 %. With the help of a scatterplot, two of the unknown samples were classified as forest honeys and four of them as blossom honeys.

Stable isotope analysis offers a unique tool for comparing trophic interactions and food web architecture in ecosystems based on analysis of stable isotope ratios of carbon (13C/12C) and nitrogen (15N/14N) in organisms. Clarias gariepinus were collected from six sites along the Vaal River, South Africa and were assessed for ectoparasites and endoparasites. Lamproglena clariae (Copepoda), Tetracampos ciliotheca and Proteocephalus glanduligerus (Cestoda), and larval Contracaecum sp. (Nematoda) were collected from the gills, intestine and mesenteries, respectively. Signatures of δ13C and δ15N were analysed in host muscle tissue and parasites using bulk stable isotope analysis. Variable stable isotope enrichment between parasites and host were observed; L. clariae and the host shared similar δ15N signatures and endoparasites being depleted in δ13C and δ15N relative to the host. Differences in stable isotope enrichment between parasites could be related to the feeding strategy of each parasite species collected. Geographic and spatial differences in enrichment of stable isotopes observed in hosts were mirrored by parasites. As parasites rely on a single host for meeting their nutritional demands, stable isotope variability in parasites relates to the dietary differences of host organisms and therefore variations in baseline stable isotope signatures of food items consumed by hosts.

Safeguarding water quality is resource-intensive and costly. Especially in cases of accidents or disasters, compact devices that allow quick assessment of the current situation are lacking. The objective of this work is the development of a portable measuring device for on-site detection of pollutants in water based on nano-high performance liquid chromatography (nano-HPLC) and electrospray ionization (ESI) ion mobility spectrometry (IMS). Integrated online enrichment by means of solid phase extraction (SPE) further improves sensitivity. In this work, an SPE cartridge exchange unit is presented, which was developed by additive manufacturing in a cost-effective and resource-efficient way. Prerequisites are an easy adaptation to commercially available SPE cartridges and pressure stability of up to 50 bar. In addition, a compact ESI-IMS with 75 mm drift tube length and a resolving power of R = 100 is presented that enables ionization, separation based on ion mobility and detection of analytes at a flow rate of 0.6 – 1.8 µL/min from the liquid phase. This approach also allows miniaturization of the overall system leading to a reduction in energy requirements and the amount of solvents used. For future environmentally benign systems, complete elimination of toxic solvents would be ideal. Therefore, acetonitrile and the nontoxic ethanol are compared as organic mobile phase in terms of elution and ionization efficiency. For characterization, a test mixture containing relevant target analytes, such as chlortoluron, isoproturon, pyrimethanil, mepanipyrim, cyprodinil and carbamazepine, is analyzed. In addition, the analytical greenness metric approach tool is used to evaluate the overall system in terms of its green credentials.It was shown that ethanol can be well used as an organic solvent for the mobile phase and even exhibits better ionization than acetonitrile for the selected model analytes. Furthermore, a relatively high orthogonality of O=0.53 and an effective 2D peak capacity of 2Dneff=174 are reached. Due to the overall miniaturization and splitless coupling, the aims of green chemistry can be met, and ideally a value of 0.92 can be achieved using the AGREE tool for the presented system.

With the increasing demand for ultrapure water in the pharmaceutical and semiconductor industry, the need for precise measuring instruments for those applications is also growing. One critical parameter of water quality is the amount of total organic carbon (TOC). This work presents a system that uses the advantage of the increased oxidation power achieved with UV/O3 advanced oxidation process (AOP) for TOC measurement in combination with a significant miniaturization compared to the state of the art. The miniaturization is achieved by using polymer-electrolyte membrane (PEM) electrolysis cells for ozone generation in combination with UV-LEDs for irradiation of the measuring solution, as both components are significantly smaller than standard equipment. Conductivity measurement after oxidation is the measuring principle and measurements were carried out in the TOC range between 10 and 1000 ppb TOC. The suitability of the system for TOC measurement is demonstrated using the oxidation by ozonation combined with UV irradiation of defined concentrations of isopropyl alcohol (IPA).

The analysis of environmental microplastic particles using FTIR microscopy is a challenging task, due to the very high number of individual particles within a single sample. Therefore, automatable, fast and robust approaches are highly requested. Micro particles were commonly enriched on filters, and sub- or the whole filter area was investigated, which took more than 20h and produced millions of data, which had to be evaluated. This paper presents a new approach of such filter area analysis using an intelligent algorithm to measure only those spots on a filter that would produce evaluable FTIR data. Empty spaces or IR absorbers like carbon black particles were not measured which successfully reduced the total analysis time from 50h to 7h. The analysis of microplastics in sediments, soils or beach samples is commonly paired with a separation step to enrich microplastics or to remove non-plastics, respectively. Those steps are often very time consuming and are performed in presence of high concentrated solvents. The latter are also suspected to corrode or decompose the analyte particles, which hamper further identification processes. This paper describes a new fast and effective microplastics separation apparatus for analytical issues that was based on hydrophobic adhesion of microplastics and fine air bubbles. The presented prototype could successfully enrich over 90 %wt of 30ppmw microplastics in 200 g sand in 20 min. Additionally, it could be demonstrated that the new separation technique was very suitable for further microplastics identification by FTIR microscopy. In this context, a sample with different polymers and matrix components was analyzed and the results were presented within this article. • Microplastics were enriched selectively by hydrophobic adhesion. • No additional chemicals except water and air were used. • Separation took only 20 min and 90 %wtof microplastics were recovered.

The combination of a representative microplastic sampling method and a fast-quantitative analysis using Pyrolysis-GC–MS (Py-GC–MS) for investigation of the microplastic load and mass balances is presented in this work. A representative microplastic filtration requires a method allowing quick extraction of the sample. The developed steel based cascadic microplastic filtration uses steel basket filters with mesh sizes of 100 μm, 50 μm and 10 μm and a mean recovery of 86 % without cross contamination was achieved. Thermoanalytical methods have the advantage of minimal sample preparation with short analysis times. The presented platinum filament-based Py-GC–MS method requires little sample preparation and quantification limits for polystyrene (PS) and polyethylene (PE) were 0.03 μg and 1 μg absolute, respectively. The relative standard deviation of the analytical method is 11 %. The combined method allows representative sampling and analysis of MP from water bodies and waste water treatment plants within 48 h. • Presentation of a validated steel based cascadic microplastic filtration plant. • Fast and reproduceable Py-GC–MS analysis method for microplastic. • Py-GC–MS allows microplastic analysis with little sample preparation.
