Abstract

Insects are potential sources of industrial proteolytic enzymes that could lead to new enzymatic applications. Study and characterization of intestinal proteolytic activity in insects can lead to understanding physiological processes, development, and ecological aspects such as effects of diet on their digestive physiology. This work presents partial characterization of intestinal proteolytic activity in adult Chromacris colorata (Serville) collected in a natural ecosystem. This is the first report of trypsin-type serine protease and papain-type cysteine protease activity statistically similar in males and females (13,100 ± 2,995.4321 and 14,495.83 UAP ± 4,153.5011, respectively). Proteolytic activity was sensitive to chemical inhibitors phenylmethylsulfonyl fluoride (PMSF), and N-(trans-epoxysuccinyl)-L-leucine 4-guanidinobutylamide (E-64), reducing total activity about 30% (F = 8.743; df = 2,45; p = 0.0006199). Proteolytic activity was sensitive to the Ca+2 cation, indicating this cation might not be a required co-factor for functioning of dominant enzymes (F = 12.338; df = 2,24; p = 0.0002064). This first report on intestinal proteolytic activity of C. colorata showed differential response to cations and inhibitors, which contributes to knowledge of the digestive system of C. colorata and emphasizes need for further study to characterize physiology and biochemistry of the digestive system of this insect to better understand its biology and possible applications. Los insectos son fuentes potenciales de enzimas proteolíticas para la industria que podrían conducir a nuevas aplicaciones enzimáticas. El estudio y a caracterización de la actividad proteolítica intestinal en insectos puede conducir a un entendimiento de procesos fisiológicos, desarrollo y aspectos ecológicos tales como los efectos de la dieta en su fisiología digestiva. Este trabajo presenta una caracterización parcial de la actividad proteolítica intestinal de Chromacris colorata (Serville) colectada en un ecosistema natural. Este es el primer reporte de actividad de serin proteasas tipo tripsina y cisteín proteasas tipo papaína similares estadísticamente en machos y hembras (13,100 ± 2,995.4321 y 14,495.83 UAP ± 4,153.5011, respectivamente). La actividad proteolítica fue sensible a los inhibidores químicos fenilmetilsulfonil fluoruro (PMSF) y al N-(trans-expoxysuccinil)-L-leucina 4-guanidinbutilamida (E-64), reduciendo la actividad total cerca de un 30% (F = 8.743; gl = 2,45; p = 0.0006199). The native ant Solenopsis geminata (Fabricius, 1804) is a continuous pest of manzano pepper plants (Capsicum pubescens Ruiz and Pav.) cultivated in the northern highlands of Puebla, Mexico. This is the first report of S. geminata substantially affecting planations of manzano pepper in disturbed mesophyll rainforest of Mexico. Abstract

The Central American locust (CAL), Schistocerca piceifrons piceifrons (Walker) (Acrididae: Cyrtacanthacridinae), is a harmful agricultural pest from Mexico to Costa Rica. This study assessed the nutritional contents of the locust. Enzymatic digestibility of hydrosoluble proteins and the antioxidant capacities of the derived peptides were analyzed. Locust flour was used to make corn tortillas to analyze the water retention and physical properties of the tortillas. Protein content was 72.4%, and protein extract yielded 4.77 ± 0.095 mg per gram of dry weight. Trypsin provided the strongest protein digestion at 1 and 3 hours, compared to pepsin. Regarding peptide scavenging ability of free radicals, digestion with trypsin allowed detection of 0.068 and 0.055 mM TE/mg of protein, at digestive times of 1 and 3 hours, respectively. Overall, results were not statistically different. Corn tortillas displayed changes in color and consistency related to quantity of locust flour. When more locust flour was used, more water was lost during the cooking process. The research documents the potential of Central American locust as a protein source and brings the possibility of using this undervalued natural resource. Abstract

It has been reported that stingless bees of the genus Trigona are regular visitors to the Jatropha curcas bioenergetic plant, physic nut. These bees use their mandibular system to extract cellulose and resin from the stem, materials necessary for the construction of nests. The objective of this work was to evaluate the prevalence of begomovirus (BV) in populations of Trigonas visitors of J. curcas in Soconusco, Chiapas, Mexico. Eleven individuals of the genus Trigona were collected in five populations, individuals were grouped as T. fuscipennisand T. fulviventris, using taxonomic clues. Using the PA/PB primers, 79% and 69% of T. fuscipennis and T. fulviventris, respectively, had BVs. The results suggest that bees of the genus Trigona could be transmitters of BV, a potential risk in the crop J. curcas. [ABSTRACT FROM AUTHOR]

Abstract

Lepidopterism caused by caterpillar contact is considered a public health problem around the world. The local and systemic responses of this pathology include short- and long-term inflammatory events. Although the proteolytic activity of the venoms from caterpillars is strongly associated with an inflammatory response in humans and murine models, fast and acute symptoms such as a burning sensation, itching, and pain should be related to the presence of low-weight hydrophilic molecules which easily influence cell metabolism. This investigation reports on the 1H-Nuclear Magnetic Resonance (NMR) profiling of the venom from the larva of Hylesia continua, a caterpillar linked to frequent cases of lepidopterism in the northern highlands of Puebla, Mexico. According to one-dimensional (1D) and two-dimensional (2D) NMR data, the venom of H. continua contained 19 compounds with proven pain-inducing activity (i.e., acetic acid, lactic acid, formic acid, succinic acid, 2-hydroxyglutaric acid, ethanol, and glutamate), inflammatory activity (i.e., cadaverine, putrescine, and acetoin), as well as natural immunosuppressive activity (i.e., O-phosphocholine and urocanic acid). The levels of the 19 compounds were calculated using quantitative-NMR (qNMR) and extensively discussed on the basis of their toxic properties which partially explain typical symptoms of lepidopterism caused by the larvae of H. continua. To the best of our knowledge, this is the first investigation reporting a complex mixture of small molecules with inflammatory properties dissolved in the venom of a lepidopteran larva. Abstract

Purslane (Portulaca oleraceaL.) is a species recognized for its high levels of bioactive compounds, among which the compounds with antioxidant properties stand out. The objective of the work was to determine the variation of phenolic metabolites and their antiradical capabilities inwild accessions of P. oleracea. The research was conducted with samples from five municipalities in Tamaulipas, Mexico collected in October 2018. Thecontents of total phenolic compounds (CFT) and free radical scavenging capacity against ABTS (2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulfonic)acid) and DPPH (2,2-diphenyl-1-picrylhydracyl)were determined. Extractions were made from vegetative parts using three solvents: acetone, water and ethanol. The accession of the localities of Abasolo and Padilla were the ones that had the highest amount of CFT with 5.8938±0.03 and 5.3742±0.11 mEAGg-1PS, respectively, using water in the extraction. Regarding the free radical scavengingcapacity against ABTS, the accession of Abasolo and Jiménez recorded the highest values, with 3.27±0.06 and 3.2226±0.04 mM ETg-1PS. Regarding thelevel against DPPH radicals, the accession of Abasolo was the highest with 2.0204±0.05 mM ETg-1PS,using water in the extraction. Water was the best solvent for the extraction of the determined contents. Heterogeneity was observed in the composition and levels of the parameters evaluated among the accessions. Wild P. oleraceaaccessions represent reservoirs of phenolic compounds and free radical scavenging capacity, including cultivated and wild varieties, La verdolaga (Portulaca oleracea L.) es una especie reconocida por sus altos niveles de compuestos bioactivos, entre los que destacan los compuestos antioxidantes y su capacidad colectora de radicales libres. El objetivo del trabajo fue determinar la variación de metabolitos fenólicos y sus capacidades antirradicalarias en accesiones silvestres de P. oleracea. La investigación se realizó con muestras provenientes de cinco municipios de Tamaulipas, México recolectadas en octubre 2018. Se determinaron los contenidos de compuestos fenólicos totales (CFT) y capacidad colectora de radicales libres contra ABTS (ácido 2,2'-azino-bis- (3-etilbenzotiazolina-6-sulfónico)) y DPPH (2,2-difenil-1-picrilhidracilo). Las extracciones se realizaron de partes vegetativas usando tres solventes: acetona, agua y etanol. La accesión de las localidades de Abasolo y Padilla fueron la que presentaron mayor cantidad de CFT con 5.8938±0.03 y 5.3742±0.11 mEAG/g PS, respectivamente, usando agua en la extracción y la accesión de Padilla fue la de menor contenido con 1.2409±0.45 mEAG/g PS usando etanol para la extracción. Respecto a la capacidad colectora de radicales libres contra ABTS y DPPH la accesión de Abasolo registró los valores más altos, con 3.2700±0.06 y 2.0204±0.05 mM ET/g PS, respectivamente, usando agua en la extracción. El agua fue el mejor solvente para la extracción de todos los contenidos determinados. Se observó una heterogeneidad en la composición y niveles de los parámetros evaluados entre las accesiones. Las accesiones silvestres de P. oleracea representan reservorios importantes de compuestos fenólicos y capacidad colectora de radicales libres, los cuales deben caracterizarse a fondo para apoyar el aprovechamiento de la especie, incluyendo las variedades cultivadas y silvestres.

Abstract

Trichoderma species is a heterotrophic saprobion organism, which competes with other microorganisms, preventing them from developing. This fungus has mechanisms of action, among which are: production of antibiotics and metabolites, and hyperparasitism, which also promotes the induction of systemic resistance in plants. In this study, five native Trichoderma strains which were collected from agricultural land in Tamaulipas, Mexico were evaluated. The objective of this research was to identify the secondary metabolites produced by native strains of Trichoderma spp. under in vitro conditions and evaluation of the effect of these compounds on the growth of the pathogen Fusarium oxysporum and the germination of tomato seeds (Solanum esculentum). The production of Trichoderma's metabolites was performed by culturing in 150 ml of potato broth supplemented with sucrose (5%) and yeast extract (5%). Inoculation was performed by the addition of a 4 mm disk of active mycelium and allowed to grow at 25C ± 2°C and 12 h light/dark on a rotary shaker at 150 rpm for three weeks. Subsequently, the extraction of metabolites was performed using liquid: liquid phase which consisted of the filtrate and methylene chloride in a ratio of 1:3, and then the organic phase was recovered. The organic fraction was evaporated at 40°C with the aid of a rotary evaporator (Buchi®); the sample was recovered in 1 ml of methylene chloride. Analysis by GC-MS indicates that Trichoderma isolates produced 41 secondary metabolites of volatile and semi-volatile molecules and was observed, such that the number of compounds varies from the species analyzed and the collection site. The evaluation of the antagonistic activity of the extracts of Trichoderma spp. on F. oxysporum showed no significant differences between treatments (P=0.05); however, it was observed that pigmentation decreased in the mycelium of the pathogen. Regarding the evaluation of the effect of the extracts on the seeds of tomato, it was observed that the compounds of Trichoderma asperellum (TV1) induced the germination and the development of the seedling. This species grows mainly at low altitudes and its upper limit reaches 1000m., it prefers temperatures above 18.3O C and its production and harvest may ocurr at two seasons, one during the beginning of the summer and the main one ocurring in the middle of the autumn in northeast México. It is estimated that the main production occurs in the state of Tamaulipas with 84 tons per year and is harvested from 23 municipalities. Concentration of capseicins and dihydrocapsaicine from wild populations may vary considerably from one location to another in the municipalities, ecotypes and diverse climatic conditions from this wide geographic zone. The high demands of wild chili as well as the variability on concentration of capseicins in fruits are considered as some of the main reasons why intensive cultivation of this species should be carried out in Northeastern Mexico.
