Abstract

The continuous monitoring of meteor and fireball activity provides helpful information about meteoroid streams, but also about the mechanisms that deliver these particles of interplanetary matter to the Earth [1, 2, 3]. Besides, by means of meteor spectroscopy we can obtain an insight into the chemical nature of meteoroids ablating in the atmosphere. Very bright fireball events are particularly interesting, as under appropriate circumstances they may give rise to meteorite falls. With this aim, the SPanish Meteor Network (SPMN) is performing a continuous meteor monitoring and spectroscopic campaign from 25 observing stations located in Spain. These monitor the night sky over the Iberian Peninsula and neighbouring areas. In this work we present the analysis of a mag. -9 sporadic fireball recorded over the Doñana Natural Park on Jan. 25, 2012.

8. The Kamayoq in Peru: farmer-to-farmer extension and experimentation

Abstract

One of the most effective ways to address farmers’ needs is through a farmer-to-farmer extension approach that also encourages farmer experimentation. This is clear from Practical Action’s work in Peru. The Kamayoq are farmers selected by their communities, who receive specific training and then return to their villages to train neighbouring farmers. They work with other farmers to develop solutions to local agricultural and veterinary problems, generally following a Participatory Technology Development approach. Positive results also include an increase in self-confidence among the Kamayoq and those working with them, something which further encourages local experimentation

Abstract

Applications of 5 × 10-4 M Zn(II) and 3 × 10-5M Cd(II) depressed but did not block root growth in Allium cepa L. Both heavy metals were indirect clastogens at these moderate concentrations, for they induced the appearance of chromosomal bridges, but no breaks were detected in the first ana-telophases. On the other hand, Zn and Cd respectively potentiated six and eleven times the direct clastogenic effect of the ß-irradiation produced by [3H]thymidine incorporation into DNA. The chromosomal breaks at mitosis were more abundant when [3H] was incorporated into early replicating DNA. On the other hand, when the incorporation took place in the last half of interphase, the subsequent G2 as well as the prophase were extended (17-fold for Zn and 27-fold for Cd), and the Sexcess lesions were repaired before anaphase. The whole cycle experienced 1.5 and 2.0-fold increases in the presence of Zn and Cd in relation to that in control meristems (15 h). [ABSTRACT FROM AUTHOR]

21. Nuclear ploidy is contingent on the microtubular cycle responsible for plant cytokinesis

Abstract

Summary. Division of the plant cell relies on the preprophase band of microtubules (PPB)-phragmoplast system. Cells of onion ( Allium cepa L.) root meristems were rendered binucleate by preventing the consolidation of cell plate formation in telophase with 5 mM caffeine. These binucleates developed either a single PPB around one of their two nuclei or two PPBs, one per nucleus, in the prophase of the ensuing mitosis. Prophase cells developing one single PPB were shorter in length (42.3 ± 4.1 µm) than those developing 2 PPBs (49.8 ± 4.1 µm), and interphase duration was inversely related to cell length. Cells whose length was less than or equal to 42 µm, i.e., which had not even reached the mean size of the small binucleates in prophase, were followed throughout mitosis. In metaphase, they always assembled two mitotic spindles (one per nucleus). However, the cells that had assembled a single PPB also developed a single phragmoplast in telophase, leading to polyploidization. As these meristematic cells were not wide enough to accommodate the midzones of both mitotic spindles in any single plane transversal to the cell elongation axis, the spindles tilted until their midzones formed a continuum where the single common phragmoplast assembled. Its position was thereby uncoupled from that of the preceding PPB. Subsequently, the chromosomes from two different half-spindles were included, by a common nuclear envelope, in a single tetraploid nucleus. Finally, the cytokinetic plate segregated the two tetraploid nuclei formed at each side of the phragmoplast into two independent sister cells.

22. HSP90 and checkpoint-dependent lengthening of the G 2 phase observed in plant cells under hypoxia and cold

Abstract

Summary. Proliferating cells of Allium cepa L. roots became adapted to hypoxia (5% oxygen) and cold (10°C) by acquiring new steady-state kinetics of growth. The cell cycle time increased from the 17.6 h in control meristems up to 29.7 and 69.0 h under hypoxia and cold conditions, respectively. Acclimation of the proliferating cells was stress specific. No acclimation took place after 24 h of heat treatment (40°C). Under cold treatment, all cycle phases enlarged uniformly. However, under hypoxia, while the G 1 and S cycle phases roughly doubled in their timing, the expected checkpoint-dependent lengthening of G 2 did not take place. This failure in lengthening G 2 in response to hypoxia correlated with a failure in the overinduction of a single peptide with a molecular mass of about 134 kDa which is among those recognised by an HSP90 antibody. Moreover, the presence of this large peptide of the HSP90 family proved to be a marker for cell proliferation. It was always absent from the contiguous differentiated cells of the root. Lastly, the mitochondrial chaperonin recognized by an HSP60 antibody in these roots not involved in photosynthesis was always higher in the proliferating than in the nonproliferating cells.

24. Default cycle phases determined after modifying discrete DNA sequences in plant cells

Abstract

After bromosubstituting DNA sequences replicated in the first, second, or third part of the S phase, in Allium cepa L. meristematic cells. Radiation at 313 nm wavelength under anoxia allowed us to adscribe different sequences to both the positive and negative regulation of some cycle phase transitions. The present report shows that the radiation forced cells in late G1 phase to advance into S, while those in G2 remained in G2 and cells in prophase returned to G2 when both sets of sequences involved in the positive and negative controls were bromosubstituted. In this way, not only G2 but also the S phase behaved as cycle phases, where cells accumulated by default when signals of different sign functionally cancelled out. The treatment did not halt the rates of replication or transcription of plant bromosubstituted DNA. The irradiation under hypoxia apparently prevents the binding of regulatory proteins to Br-DNA.

25. Stringency at four regions of the plant cell cycle where proteins regulating its progression are synthesized*

Abstract

Different responses were detected when inhibiting protein synthesis (by 1μg ml-1 anisomycin) for similar times (3 h) in each of the four regions of the plant cell cycle where synthesis of specific proteins seems to control further cycle progression. Synchronous cells, labelled as binucleate by 1-h, 5 mM-caffeine treatment, were used for this study in Allium cepa L. root meristems proliferating under steady-state kinetics. In the experimental conditions used, G1 lasts 8 h in the synchronous control populations. G1 houses two such regions. The first region is located in early G1. Inhibition of protein synthesis in this region produced 8·2 h of delay in reaching the S period. Moreover, the treatment halved the prolUerative fraction of the population, behaving as a restriction area where cells should be rendered competent to go on cycling by newly synthesized proteins. The second region, located in late G1 (before the 6th hour of the cycle), produced, when inhibiting protein synthesis in the population, a delay of 2·4h (shorter than the treatment) in reaching the S period and no decrease of its proliferative fraction. A similar inhibition of protein synthesis in the third region mapped in early G2 (before the 20th hour of the cell cycle) produced 4·4h of delay in reaching mitosis and a 20 % decrease in the proliferative fraction. Finally, there is a fourth region in early prophase (before the 25th hour of the cell cycle) where all the cells seemed to abandon the cycle during inhibition of protein synthesis.

26. Cell proliferation in Allium cepa L. meristems under 8-hydroxyquinoline, a chelating agent that affects DNA and RNA polymerases

Abstract

8-Hydroxyquinoline (HQ) chelates Mg2+ and Mn2+ and, secondarily, affects the activities of DNA and RNA polymerases. The in vivo effect of HQ has been estimated in Allium cepa L. meristems growing under new growth kinetics in the presence of this agent. HQ (at both 5 × 10−5 M and 10−4M) depressed incorporation of [3H]uridine much more effectively than that of [3H]-thymidine. Cycle kinetics in meristems behaved as if they were independent of the rates of synthesis or accumulation of RNA since, under HQ, cycle time was only moderately modified and the new cycle kinetics achieved could be explained by the new rates of [3H] thymidine incorporation. Lengthened S periods were partially compensated for by shortened G2 phases, suggesting that, in these cells, both the growth cycle and its coupling with the DNA-division cycle were not disturbed by a decreased amount of RNA. Finally, the nucleolar cycle during mitosis, but not the interphase nucleolus, was modified under the new rates of RNA synthesis.

27. Nucleolar development in the interphase of the cell cycle

Abstract

Stereology of nucleoli at 3 different points of the cell cycle, in the middle of the G1 S and G2 of interphase, was accomplished in a naturally synchronous cell population rendered binucleate and thus ‘labelled’ and made identifiable by 1 h caffeine treatment in root meri-stems of Allium cepa L. Consistent structural changes were found so that nucleolar parameters by themselves can locate a cell at any interphase period. The growth of the nucleolus in the course of interphase takes place exclusively in its granular portion. The growth rate of this last component was found to be greater in the first half of interphase (5·4μm3h−1) than in the second (2·9 μm3 h”−1), as calculated for a nucleus with only one fused nucleolus. The changes in volume of the nucleolar components during interphase do not parallel the gene dosage. Nucleolar surface at each point seems to be the factor which determines the growth rate in the preceding interval, since these rates are inverted for fused and unfused nucleoli. The nucleolar volume occupied by lacunae is minimal in the S-period and shows an enormous increase by the middle of G2. We think our data may be the structural basis on which a model for nucleolar functioning in proliferating cells could be built.

28. Dominance of a NOR (nucleolar organizer region) over its allele and over its sister NOR after asymmetric 5-azacytidine substitution in plant chromosomes

Abstract

Dominance of a 5-azacytidine-substituted nucleolar organizer region (AZA NOR) over another with normal DNA when confined to a common nucleus or to different nuclei in a common cell was studied in Allium cepa L. root meristems. AZA administration took place for one S period. The assessment of NOR dominance was done either in early Gi (nucleologen-esis) or later in interphase (mature nucleoli). At the mitosis immediately preceding the second interphase after AZA, the segregated AZA and normal chromatids of each chromosome of the pair of homologues were confined to a single binucleate cell by inhibiting the formation of the cell plate. Anaphase segregation of the homologous AZA NORs was seen to occur at random. The AZA NORs completed nucleologenesis after a shorter time than normal NORs, independently of the intranuclear or intracellular presence of other NORs. After nucleologenesis was over in the AZA NORs, a few of the normal NORs, when in a common nucleus, remained permanently inefficient in developing a full-size mature nucleolus or any nucleolus at all (partial or total dominance, respectively). Dominance of an AZA NOR over its sister or allelic NOR with normal DNA led to the new nucleolar patterns of asymmetry and inverse symmetry, respectively, making their appearance in these binucleate cells.

29. Timing of Nucleolar Activity in Meristems

Abstract

The nucleoli in 4 regions of the root meristem of Zea mays have been examined and the time spent in each of the phases of the nucleolar cycle has been calculated from estimates of the rates of mitosis and the fraction of cycling cells in each region. The nucleolar cycles have been fitted to the mitotic cycles of cycling cells in the same regions. The timing of activity of the r-RNA genes differs in relation to the chromosomal cycles of condensation and movement in the different regions of the meristem. In the stelar regions disorganization of the nucleolus starts in the middle of prophase and reorganization starts towards the end of telophase and is completed in the following interphase. In the quiescent centre of the meristem the phases of both cycles are lengthened except for telophase and reorganization of the nucleolus does not start until after the end of telophase. The acceleration of telophase in the quiescent centre is discussed in relation to inhibition of protein synthesis. In the adjacent cap initials, which have the shortest mitotic cycle, the nucleolus starts to disorganize before the onset of prophase and reorganization is completed halfway through telophase. This fits with the fact that DNA synthesis is advanced into the telophase of the previous cycle in these cells.
