The aerobic hyperthermophile "Fervidibacter sacchari" catabolizes diverse polysaccharides and is the only cultivated member of the class "Fervidibacteria" within the phylum Armatimonadota. It encodes 117 putative glycoside hydrolases (GHs), including two from GH family 50 (GH50). In this study, we expressed, purified, and functionally characterized one of these GH50 enzymes, Fsa16295Glu. We show that Fsa16295Glu is a β-1,3-endoglucanase with optimal activity on carboxymethyl curdlan (CM-curdlan) and only weak agarase activity, despite most GH50 enzymes being described as β-agarases. The purified enzyme has a wide temperature range of 4-95°C (optimal 80°C), making it the first characterized hyperthermophilic representative of GH50. The enzyme is also active at a broad pH range of at least 5.5-11 (optimal 6.5-10). Fsa16295Glu possesses a relatively high kcat/KM of 1.82 × 107 s-1 M-1 with CM-curdlan and degrades CM-curdlan nearly completely to sugar monomers, indicating preferential hydrolysis of glucans containing β-1,3 linkages. Finally, a phylogenetic analysis of Fsa16295Glu and all other GH50 enzymes revealed that Fsa16295Glu is distant from other characterized enzymes but phylogenetically related to enzymes from thermophilic archaea that were likely acquired horizontally from "Fervidibacteria." Given its functional and phylogenetic novelty, we propose that Fsa16295Glu represents a new enzyme subfamily, GH50_3.

Abstract Few aerobic hyperthermophilic microorganisms degrade polysaccharides. Here, we describe the genome-enabled enrichment and optical tweezer-based isolation of an aerobic polysaccharide-degrading hyperthermophile, Fervidibacter sacchari, previously ascribed to candidate phylum Fervidibacteria. F. sacchari uses polysaccharides and monosaccharides for growth at 65–87.5 °C and expresses 191 carbohydrate-active enzymes (CAZymes) according to RNA-Seq and proteomics, including 31 with unusual glycoside hydrolase domains (GH109, GH177, GH179). Fluorescence in-situ hybridization and nanoscale secondary ion mass spectrometry confirmed rapid assimilation of 13C-starch in spring sediments. Purified GHs were optimally active at 80–100 °C on ten different polysaccharides. Finally, we propose reassigning Fervidibacteria as a class within phylum Armatimonadota, along with 18 other species, and show that a high number and diversity of CAZymes is a hallmark of the phylum, in both aerobic and anaerobic lineages. Our study establishes Fervidibacteria as hyperthermophilic polysaccharide degraders in terrestrial geothermal springs and suggests a broad role for Armatimonadota in polysaccharide catabolism.

Psoriatic disease (PD) and non-alcoholic fatty liver disease (NAFLD) potentially share disease pathways given the numerous inflammatory pathways involved in both diseases and a higher prevalence of NAFLD in PD patients.  Metabolic syndrome and obesity are a key link between the two diseases, but even when controlling for this, associations between both diseases are still seen. Therapeutics that impact metabolic or inflammatory pathways may be impactful in both PD and NAFLD. In this review, we describe common inflammatory pathways contributing to both PD and NAFLD and critically review the potential impact of treatments for and on both diseases.

Myeloid-derived suppressor cells (MDSCs) are innate immune cells that acquire the capacity to suppress adaptive immune responses during cancer. It remains elusive how MDSCs differ from their normal myeloid counterparts, which limits our ability to specifically detect and therapeutically target MDSCs during cancer. Here, we sought to determine the molecular features of breast cancer-associated MDSCs using the widely studied mouse model based on the mouse mammary tumor virus (MMTV) promoter-driven expression of the polyomavirus middle T oncoprotein (MMTV-PyMT). To identify MDSCs in an unbiased manner, we used single-cell RNA sequencing to compare MDSC-containing splenic myeloid cells from breast tumor-bearing mice with wild-type controls. Our computational analysis of 14,646 single-cell transcriptomes revealed that MDSCs emerge through an aberrant neutrophil maturation trajectory in the spleen that confers them an immunosuppressive cell state. We establish the MDSC-specific gene signature and identify CD84 as a surface marker for improved detection and enrichment of MDSCs in breast cancers.

PurposeLocally advanced pancreatic cancer (LAPC) treatment has varying practice patterns with poor outcomes. We investigated treatment using single-agent chemotherapy and multiagent chemotherapy (MAC) with or without radiation therapy (RT) at high-volume facilities (HVFs) and academic centers (ACs).Methods and materialsThe National Cancer Database was used to obtain data on 10,139 patients with LAPC. HVF was defined as the top 5% of facilities per number of patients treated at each facility. Univariate and multivariable (MVA) analysis Cox regressions were performed to identify the impact of HVF, AC, MAC, and RT on overall survival (OS).ResultsThe median age of patients was 66 years (range, 22-90); 50.1% were male and 49.9% female. Of the patients, 46.1% received MAC, 53.8% received single-agent chemotherapy, 45.7% received RT, 54.3% did not receive RT, and 5% underwent surgical resection. The median follow-up was 48.8 months. Background and purposeAdaptive radiation planning for pancreatic adenocarcinoma (PA) relies on accurate treatment response assessment, while traditional response evaluation criteria inefficiently characterize tumors with complex morphological features or intrinsically low metabolism. To better assess treatment response of PA, we quantify and compare regional morphological and metabolic features of the 3D pre- and post-radiation therapy (RT) tumor models.Materials and methodsThirty-one PA patients with pre and post-RT Positron emission tomography/computed tomography (PET/CT) scans were evaluated. 3D meshes of pre- and post-RT tumors were generated and registered to establish vertex-wise correspondence. To assess tumor response, Mahalanobis distances (M dist∣Fusion) between pre- and post-RT tumor surfaces with anatomic and metabolic fused vectors were calculated for each patient. M dist∣Fusion was evaluated by overall survival (OS) prediction and survival risk classification. As a comparison, the same analyses were conducted on traditional imaging/physiological predictors, and distances measurements based on metabolic and morphological features only.ResultsAmong all the imaging/physiological parameters, M dist∣Fusion was shown to be the best predictor of OS (HR = 0.52, p = 0.008), while other parameters failed to reach significance. Moreover, M dist∣Fusion outperformed traditional morphologic and metabolic measurements in patient risk stratification, either alone (HR = 11.51, p < 0.001) or combined with age (HR = 9.04, p < 0.001).ConclusionsWe introduced a PET/CT-based novel morphologic and metabolic pipeline for response evaluation in locally advanced PA. The fused M dist∣Fusion outperformed traditional morphologic, metabolic, and physiological measurements in OS prediction and risk stratification. The novel fusion model may serve as a new imaging-marker to more accurately characterize the heterogeneous tumor RT response.

Background: Alcohol use after liver transplant (LT) is associated with higher rates of graft loss and increased mortality; however, there are limited data evaluating predictors of return to alcohol use using biochemical markers like phosphatidylethanol (PEth). Methods: This multicenter retrospective cohort study evaluated psychosocial predictors of return to alcohol use using PEth testing in patients transplanted for alcohol‐associated liver disease (ALD). The study included 223 patients at three centers who had received a LT for ALD and had at least one PEth measurement post‐LT. Results: The rate of return to alcohol use was 6.9 cases per 100 person‐years (26 patients total) over a median 555 days of follow‐up after transplant. Younger age (HR 0.96; 95% CI 0.92–0.99, p = 0.02), mental health comorbidities (HR 2.83; 95% CI 1.25–6.39, p = 0.01), and non‐Hispanic White race (HR 3.79; 95% CI 1.42–10.15, p = 0.01) were associated with return to alcohol use post‐LT. There was no difference between post‐LT return to alcohol use rates or short‐term survival among patients with less than 6 months of sobriety prior to listing compared with those with more than 6 months. Patients with sustained alcohol use post‐LT had increased odds of history of illicit substance use (OR 5.20; 95% CI 1.01–26.83, p = 0.04) but no significant difference in time from the last drink to listing (OR 1.03; 95% CI 0.18–5.80, p = 0.97). Conclusions: These findings emphasize the importance of mental health comorbidities rather than period of sobriety in predicting post‐LT return to alcohol use. Furthermore, the higher risk of return to alcohol use in non‐Hispanic White patients suggests a potential disparity with referral and selection of higher risk White patients. [ABSTRACT FROM AUTHOR]

Background & Objective: Breast cancer is the most common cancer among women worldwide. Cell-cycle is controlled by regulatory proteins called cyclins and Cyclin-Dependent Kinases. Aberrant activity of cyclins plays an important role in cancer development and progression. The Cyclin E gene, called CCNE1 or pCCNE1, is one of the four cyclins groups of the cell cycle, reported in a variety of cancers as well as breast cancer, and is a notice target in cancer research. In this regard, Cyclin E gene amplification in breast cancer patients was evaluated and investigated and its capacity as a molecular biomarker was assessed. Materials & Methods: Tumor tissue samples as well as adjacent healthy tissues, were excised and collected from breast cancer patients undergoing surgery in Al-Zahra hospital. Genomic DNA was extracted using phenol-chloroform method. The extracted products concentration was normalized and applied in Real-time PCR technique. Amplification of aforementioned gene was determined by calculating Cts using 2-∆∆Ct method and was evaluated by Wilcoxon statistical test. Result: The Cyclin E gene amplification in breast cancer tissue was significantly higher than healthy tissue (p = 0.002). Furthermore, the amplification of Cyclin E gene was investigated as a breast cancer biomarker by the ROC curve statistical analysis. Conclusion: The results revealed, the area under the ROC curve (AUC) was close in 70 percent. As a result, the Cyclin E gene amplification in neoplasm dramatically increases and can be used as a biomarker for the diagnosis and prognosis of breast cancer.

At the dawn of the twentieth century, droves of former Ottoman subjects including Armenians and Syrians began to set foot in the United States searching for better opportunities. Many faced American white supremacist xenophobia and fell victim to racial discrimination. Various Ottoman diasporic communities responded to this harassment by expressing an increasing investment in the question of American whiteness and vigorously yearning to move beyond its fringes. Their voices, however, remain considerably muted; their stories are largely excluded from most American immigration narratives and conventional area-studies histories. This study endeavors to help reverse this scholarly tradition by examining the mindset of Ottoman Armenian expatriates as articulated in the editorials of Asbarēz, an Armenian-language weekly published in Fresno, California starting in 1908. As this micro-study shows, the migrants used the European racialist knowledge imported from the Ottoman empire to lay claim to whiteness and achieve integration in the US but also to affect change at home.

Phytaspases differ from other members of the plant subtilisin-like protease family by having rare aspartate cleavage specificity and unusual localization dynamics. Phytaspases are secreted from healthy plant cells but are re-internalized upon perception of death-inducing stresses. Although proteolytic activity is required for the secretion of plant subtilases, its requirement for the retrograde transportation of phytaspases is currently unknown. To address this issue, we employed an approach to complement in trans the externalization of a prodomain-less form of Nicotiana tabacum phytaspase (NtPhyt) with the free prodomain in Nicotiana benthamiana leaf cells. Using this approach, the generation of the proteolytically active NtPhyt and its transport to the extracellular space at a level comparable to that of the native NtPhyt (synthesized as a canonical prodomain-containing precursor protein) were achieved. The application of this methodology to NtPhyt with a mutated catalytic Ser537 residue resulted in the secretion of the inactive, although processed (prodomain-free), protein as well. Notably, the externalized NtPhyt Ser537Ala mutant was still capable of retrograde transportation into plant cells upon the induction of oxidative stress. Candida dubliniensis is an uncommon source of ocular infections and has only been reported in a single previous case of keratitis. This report documents the course of a combined Candida dubliniensis and Candida albicans keratitis following a chemical injury. Antifungal sensitivities of the two different Candida species are also demonstrated.
