This paper presents an analysis of the socio-scientific dimensions which 46 11th graders, enrolled in the subject scientific culture, consider to justify their COVID-19 vaccination decision and how they managed to combine and integrate these dimensions before and after working on a sequence of activities aimed at learning about argumentation in science. Although almost all the students intended to be vaccinated against COVID-19, the results show that before the sequence, they used one or two dimensions to justify their decision, while after it, some combined up to five dimensions. The ethical and social-cultural dimensions were two of the most frequently used. The scientific dimension was infrequent in the initial argument but was, however, the most used dimension in the final argument, after the intervention. Working on socioscientific issues encourages the development of argumentation skills, and, in this specific case, health education skills as well.

Abstract Background Critical limb-threatening ischemia (CLTI) constitutes the most severe manifestation of peripheral artery disease, usually induced by atherosclerosis. CLTI patients suffer from high risk of amputation of the lower extremities and elevated mortality rates, while they have low options for surgical revascularization due to associated comorbidities. Alternatively, cell-based therapeutic strategies represent an effective and safe approach to promote revascularization. However, the variability seen in several factors such as cell combinations or doses applied, have limited their success in clinical trials, being necessary to reach a consensus regarding the optimal “cellular-cocktail” prior further application into the clinic. To achieve so, it is essential to understand the mechanisms by which these cells exert their regenerative properties. Herein, we have evaluated, for the first time, the regenerative and vasculogenic potential of a combination of endothelial colony forming cells (ECFCs) and mesenchymal stem cells (MSCs) isolated from adipose-tissue (AT), compared with ECFCs from umbilical cord blood (CB-ECFCs) and AT-MSCs, in a murine model of CLTI. Methods Balb-c nude mice (n:32) were distributed in four different groups (n:8/group): control shams, and ischemic mice (after femoral ligation) that received 50 µl of physiological serum alone or a cellular combination of AT-MSCs with either CB-ECFCs or AT-ECFCs. Follow-up of blood flow reperfusion and ischemic symptoms was carried out for 21 days, when mice were sacrificed to evaluate vascular density formation. Moreover, the long-term molecular changes in response to CLTI and both cell combinations were analyzed in a proteomic quantitative approach. Results AT-MSCs with either AT- or CB-ECFCs, promoted a significant recovery of blood flow in CLTI mice 21 days post-ischemia. Besides, they modulated the inflammatory and necrotic related processes, although the CB group presented the slowest ischemic progression along the assay. Moreover, many proteins involved in the repairing mechanisms promoted by cell treatments were identified. Conclusions The combination of AT-MSCs with AT-ECFCs or with CB-ECFCs promoted similar revascularization in CLTI mice, by restoring blood flow levels, together with the modulation of the inflammatory and necrotic processes, and reduction of muscle damage. The protein changes identified are representative of the molecular mechanisms involved in ECFCs and MSCs-induced revascularization (immune response, vascular repair, muscle regeneration, etc.).

IntroductionSolid organ transplant (SOT) recipients display weak seroconversion and neutralizing antibody (NAb) responses after severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccination and remain at risk of severe coronavirus disease 2019 (COVID-19). While B-cell memory is the hallmark of serological immunity, its role in driving successful vaccine responses and providing immune protection in SOT patients remains unclear.MethodsWe investigated the function and interplay of SARS-CoV-2-specific memory B cells (mBc), different cytokineproducing T cells, and cross-reactive NAb in driving seroconversion and protection against COVID-19 in two cohorts. First, we studied a large cohort of 148 SOT recipients and 32 immunocompetent individuals who underwent several vaccinations. Subsequently, we assessed 25 SOT patients participating in a randomized controlled trial to compare two different immunosuppressive strategies for allowing successful seroconversion and memory-cell responses after booster vaccination.ResultsWe corroborate previous findings that B- and T-cell memory responses are weaker and more delayed in SOT patients than in immunocompetent (IC) individuals; however, within the SOT cohort, we found that these responses are relatively stronger and more robust in patients not receiving mycophenolate mofetil (MMF)-based therapies. Anti- spike IgG titers strongly correlated with RBD-specific IgG-producing mBc, with both displaying broad viral cross reactivity. Prebooster SARS-CoV-2-specific mBc and IL-2- producing T cells accurately predicted Nab seroconversion (AUC, 0.828) and protection against severe COVID-19. While switching unresponsive SOT patients from calcineurin inhibitors (CNI)/MMF to a low-exposure CNI/mTOR-i regimen favored wider SARS-CoV-2-specific immune responses after a fourth booster vaccination, preformed RBD-specific mBc predicted NAb seroconversion.DiscussionOur study adds new insights into the pathobiology of immune memory and highlights the pivotal role of SARS-CoV-2-specific mBc in promoting immune protection inSOT patients.

Urban habitats represent new opportunities to study natural processes such as ecological succession. Our work focused for the first time on the changes in ecosystem services occurring in the ecological succession from annual to perennial grasslands in Mediterranean peri-urban greenspaces. Student’s t test was used to analyse the influence of the grassland type on plant and soil features, and showed that soil under perennial grassland had a significantly higher C content and arylamidase and arylsulfatase activity than annual grasslands. In contrast, annual grasslands showed a significantly higher plant-species richness, and their soils had higher bulk density and phenoloxidase and arylamidase activity compared to perennial grasslands. Neither the labile nor recalcitrant fractions of the organic matter showed any significant difference between communities. When all the soil factors were included together, Redundancy Analysis revealed a significant gradient in soil phenoloxidase activity and organic matter distinguishing perennial from annual grasslands. We conclude that when annual grasslands give way to perennial grasslands through natural succession in Mediterranean greenspaces, certain ecosystem services such as soil carbon storage and water regulation improve, while biodiversity maintenance declines. Thus, Mediterranean urban greenspaces where natural succession occurs should be handled in such a way as to preserve both type of habitats in order to improve a wider range of ecosystem services.

Esta investigación no experimental, estructurada y de recolección transversal busca brindar un panorama acerca de la enseñanza de conceptos y normas jurídicas que inciden en la profesión bibliotecaria, tomando como ejemplo a la carrera de Bibliotecología y Ciencia de la Información de la Universidad de Buenos Aires. El enfoque de la investigación es mixto: se combinaron los métodos cuantitativo (encuestas a graduados) y cualitativo (entrevistas con distintos profesionales). Contiene un breve repaso de las leyes implicadas y se brindan consejos para su inclusión en los planes de estudio de la carrera. La mayoría de los bibliotecarios graduados no cuentan con conocimientos suficientes de las leyes relacionadas a su profesión. ¿Cómo puede un bibliotecario fomentar el libre acceso a la información y defender las excepciones para bibliotecas sin fundamentos sólidos del tema y herramientas para lograrlo? = This non-experimental, structured, cross-sectional research seeks to provide an overview of the teaching of laws and regulations that affect the library profession, taking as an example the library science career at the Universidad de Buenos Aires. The research approach is mixed: quantitative (graduate surveys) and qualitative (interviews with different professionals) methods were combined. It contains a brief review of the laws involved and provides advice for their inclusion in the degree study plans. Most graduate librarians do not have sufficient knowledge of the laws related to their profession. How can a librarian promote free access to information and defend exceptions for libraries without solid rationale for the topic and tools to achieve it?

Cell division is one of the most relevant processes in Biology, for instance, to understand the reproduction of species or the development of tumours. Learning this content requires not addressing it out of context but integrating it into real-life problems that demand students apply their knowledge about cell division. In this study, the authors examine how ten small groups of 10th grade students apply their mitosis models to solve a problem about cancer treatment; and how they progress in their application in terms of students' discursive moves. Throughout their discussions, learners must explain what happens to a treated tumour cell which cannot finish its division. To do so, they should consider understudied ideas such as the prometaphase stage or the importance of bipolar spindle formation. Data collected are the groups' audio recordings and their written reports. The results show that all groups handle notions, such as the need for centrosomes' duplication or microtubules-chromosomes' attachment for mitosis to be complete, throughout the activity. Although all groups integrate two or three ideas at a time into their discourse, none of them manage to combine and contextualise all the expected ones. Implications for Biology education are discussed.

IntroductionPeripheral blood (PB) molecular patterns characterizing the different effector immune pathways driving distinct kidney rejection types remain to be fully elucidated. We hypothesized that transcriptome analysis using RNA sequencing (RNAseq) in samples of kidney transplant patients would enable the identification of unique protein-coding and noncoding genes that may be able to segregate different rejection phenotypes.MethodsWe evaluated 37 biopsy-paired PB samples from the discovery cohort, with stable (STA), antibody-mediated rejection (AMR), and T cell-mediated rejection (TCMR) by RNAseq. Advanced machine learning tools were used to perform 3-way differential gene expression analysis to identify gene signatures associated with rejection. We then performed functional in silico analysis and validation by Fluidigm (San Francisco, CA) in 62 samples from 2 independent kidney transplant cohorts.ResultsWe found 102 genes (63 coding genes and 39 noncoding genes) associated with AMR (54 upregulated), TCMR (23 upregulated), and STA (25 upregulated) perfectly clustered with each rejection phenotype and highly correlated with main histologic lesions (ρ = 0.91). For the genes associated with AMR, we found enrichment in regulation of endoplasmic reticulum stress, adaptive immunity, and Ig class-switching. In the validation, we found that the SIGLEC17P pseudogene and 9 SIGLEC17P-related coding genes were highly expressed among AMR but not in TCMR and STA samples.ConclusionsThis analysis identifies a critical gene signature in PB in kidney transplant patients undergoing AMR, sufficient to differentiate them from patients with TCMR and immunologically quiescent kidney allografts. Our findings provide the basis for new studies dissecting the role of noncoding genes in the pathophysiology of kidney allograft rejection and their potential value as noninvasive biomarkers of the rejection process.

La publicación de los Objetivos de Desarrollo Sostenible ha supuesto un cambio de rumbo en la Educación Ambiental, principalmente porque plantean los problemas ambientales desde distintos ámbitos (social, económico, cultural o religioso), es decir, desde una perspectiva integral y multidimensional. Por ello, cabría esperar una transformación en la investigación educativa y la práctica de aula sobre Educación Ambiental. Este estudio pretende conocer qué y cómo se ha abordado la Educación Ambiental en la investigación educativa nacional e internacional de alto impacto desde la publicación de los Objetivos de Desarrollo Sostenible (2015-2021). Para ello, se realiza una revisión de 715 artículos en revistas del ámbito de la Didáctica de las Ciencias Experimentales y la Justicia Social. Dichos artículos se han categorizado en función de los términos para la identificación de la muestra y la tipología de estudio. Los resultados muestran en primer lugar, un predominio del término Educación Ambiental en las publicaciones, frente a otros que explicitan, en mayor medida, la visión multidisciplinar de esta, como Justicia Ambiental. En segundo, destaca la alta presencia de estudios de intervenciones frente a otro tipo de trabajos (percepciones, reflexión o revisión bibliográfica) entre las publicaciones analizadas. Con los resultados obtenidos se podría considerar, por un lado, que la investigación educativa en Educación Ambiental y su dimensión social continúan distanciadas; y por otro, que dicha investigación está migrando desde un enfoque teórico a uno de aplicación.

The most relevant features of FLIPEC (Free fLow Iterative Plasma Equilibrium Code) are presented. This new code iteratively calculates free-boundary, axisymmetric ideal MHD equilibria with arbitrary poloidal and toroidal plasma flows. FLIPEC is a mature code that has emerged from a complete overhaul of a previous version (F-Torija Daza 2022 et al Nucl. Fusion 62 126044). It uses a (inverse) curvilinear coordinate representation for the Grad–Shafranov–Bernoulli equation system, which allows FLIPEC to extend its free-boundary capabilities to arbitrary plasma shapes and removes many limitations with regards to the distance between plasma and external coils. Run-time stabilization of vertical modes has also been implemented by means of artificial feedback coils. Finally, active targeting schemes have also been included. These capabilities are illustrated on two very different cases: the ITER tokamak baseline configuration and a NSTX spherical tokamak equilibrium.

As a result of the integration of the university system in the European Higher Education Area in 2010, competences, methodology and evaluation have attracted the attention of researchers in the field of university teaching, while the contents have been relegated to a background. The present research aims to find out whether the contents that are taught respond to the changing demands of society or, otherwise, remain static and may become obsolete. The compulsory subject "Facilities Project" of the Degree in Fundamentals of Architecture at the Polytechnic University of Madrid is chosen as a case study. For this, an open survey is carried out to the students who take it in two successive years. As it is an open survey, students are offered a greater opportunity to express freely, both their answers and the reasons they have forgiving them. It is detected that they positively value much of the content and that they also value, to a lesser extent, other content negatively. Different evaluations are collected for each of the topics that make up the subject and also the reasons that, together with the suggestions, allow measures to be taken to improve the content offered. It is concluded that the contact with the opinion of the students allows obtaining a valuable point of view when keeping the contents updated.

Abstract Background Emotional and stress-related disorders show high incidence, prevalence, morbidity, and comorbidity rates in Mexico. In recent decades, research findings indicate that cognitive behavioral interventions, from a disorder-specific perspective, are the effective front-line treatment for anxiety and depression care. However, these treatments are not often used. Reasons include limited access and low availability to effective interventions and comorbidity between mental disorders. Emotional deregulation of negative affectivity has been found to be a mediating factor in addressing emotional disorders from a transdiagnostic perspective, aimed at two or more specific disorders. In addition, technological advancement has created alternatives for psychological assistance, highlighting the possibilities offered by technologies since Internet-supported intervention programs have been empirically tested for effectiveness, efficiency and efficacy and can be key to ensuring access to those who are inaccessible. The aim of the study is to evaluate the efficacy, moderators of clinical change and acceptability of a transdiagnostic guided Internet-delivered intervention versus a transdiagnostic self-guided Internet-delivered intervention for emotional, trauma and stress-related disorders, and waiting list in community sample. Methods A three-armed, parallel group, superiority randomized controlled clinical trial with repeated measurements at four times: pretest, posttest, follow-up at 3, 6 and 12 months. Outcomes assessor, participant, care provider and investigator will be blinded. Participants aged 18 to 70 years will be randomly allocated 1:1:1 to one of three study arms: a) Transdiagnostic guided internet-delivered intervention with synchronous assistance, b) Transdiagnostic self-guided internet-delivered intervention, c) Waiting list group. Based on sample size estimation, a minimum of 207 participants (69 in each intervention group) will be included. Discussion The study could contribute to improving the efficacy of transdiagnostic internet-delivered interventions to promote the dissemination of evidence-based treatments and eventually, to decrease the high prevalence of emotional and trauma-related disorders in the Mexican population. Los espacios urbanos han cobrado una dimensión significativamente simbólica en las formas literarias contemporáneas, en donde la ciudad interactúa, afecta y modifica el devenir metafísico urbano. La reflexión que gira en torno a la ciudad como espacio narrativo no se limita únicamente a un inventario de lo espacial o referencial, sino que entiende a la ciudad como una elaboración sensible de la sociedad contemporánea. Las tensiones y resonancias de la vida en la ciudad construyen los imaginarios sobre los que se fundamenta la experiencia urbana. En El asesinato de Paulina Lee (2016) del escritor regiomontano Hugo Valdés, el espacio urbano de Monterrey, Nuevo León, desempeña un rol protagónico. En este ensayo se analizará la cartografía urbana como retrato de una ciudad que se adentró en la modernidad industrial del siglo XX y cuya expansión acelerada dio lugar a un territorio fragmentado por la desigualdad económica en donde predominaron la marginalidad, la pobreza y la violencia.
