6. Sensory and memory stimulation as a means to care for individuals with dementia in long-term care facilities

Michael Mileski, Joseph Baar Topinka, Matthew Brooks, Corie Lonidier, Kelly Linker, Kelsey Vander Veen School of Health Administration, Texas State University, San Marcos, TX, USA Objective: The primary objective of this study was to identify and further examine the facilitators and barriers of utilizing sensory and memory stimulation as a means to care for individuals with dementia who live in long-term care settings. Materials and methods: The authors conducted a literature review of 30 academic articles found using the databases such as CINAHL, PubMed, and Academic Search Ultimate from the past 15 years. Facilitator and barrier themes were found within each article and analyzed for their relevance to sensory and memory stimulation therapies and their effects on individuals with dementia. Results: The most common facilitator was improved communication. The top three barriers were access, staff training, and mixed results. Discussion: Reminiscence therapy appears to provide a person-centered method of care for those who otherwise have problems communicating. These implementations will be more effective if they have the support of staff and management. Conclusion: The authors conclude that sensory and memory stimulation therapies have the potential to help improve many dementia-specific issues for individuals living in long-term care settings. 8. Characterization of microsatellite loci in Kearney's bluestar (Amsonia keameyana) and cross-amplification in other Amsonia species

Abstract

Kearney's bluestar (Amsonia keameyana) is a highly endangered herbaceous perennial in the family Apocynaceae. The species is found only in the Baboquivari Mountains of southern Arizona. We report the isolation and development of 12 microsatellite loci for Kearney's bluestar. Numbers of alleles ranged from two to four and observed heterozygosities ranged from 0.20 to 0.80 in the nine loci found to be polymorphic in the test population. All loci were also tested for cross-amplification in five other Amsonia species representing two sub-genera from the southwestern United States. Some loci that were not polymorphic in the Kearney's bluestar were polymorphic in other species.

Abstract

Olfactory function, orchestrated by the cells of the olfactory mucosa at the rooftop of the nasal cavity, is disturbed early in the pathogenesis of Alzheimer's disease (AD). Biometals including zinc and calcium are known to be important for sense of smell and to be altered in the brains of AD patients. Little is known about elemental homeostasis in the AD patient olfactory mucosa. Here we aimed to assess whether the disease-related alterations to biometal homeostasis observed in the brain are also reflected in the olfactory mucosa. We applied RNA sequencing to discover gene expression changes related to metals in olfactory mucosal cells of cognitively healthy controls, individuals with mild cognitive impairment and AD patients, and performed analysis of the elemental content to determine metal levels. Results demonstrate that the levels of zinc, calcium and sodium are increased in the AD olfactory mucosa concomitantly with alterations to 17 genes related to metal-ion binding or metal-related function of the protein product. A significant elevation in alpha-2-macroglobulin, a known metal-binding biomarker correlated with brain disease burden, was observed on the gene and protein levels in the olfactory mucosa cells of AD patients. These data demonstrate that the olfactory mucosa cells derived from AD patients recapitulate certain impairments of biometal homeostasis observed in the brains of patients.

Abstract

Copper oxide nanoparticles (CuO NPs) are used in numerous applications including antimicrobial agents, semiconductors, chemical sensors, catalysts and others. However, their high toxic potential and growing industrial production increases the concern for health and environmental risks. The present study aimed to investigate the toxicity of pristine versus surface-modified CuO NPs in relation to their stability/dissolution in cell culture medium, cellular uptake, cytotoxicity and ROS production in mouse macrophages RAW264.7. Our results showed that NPs modified with different coating agents [citrate (CIT), ascorbate (ASC), polyethyleneimine (PEI) and polyvinylpyrollidone (PVP)] affected the dissolution of the NPs in water but not in cell culture medium. Further, in vitro testing demonstrated that PEI-coated NPs were most cytotoxic while ASC-coated material exerted the lowest cytotoxicity. The anionic coatings CIT and ASC also provided protection against CuO NPs-induced oxidative stress and decreased levels of ROS production. Cellular internalization assessed by TEM was detected in cells exposed to CIT, ASC, PVP-coated and pristine NPs but not PEI-CuO NPs. Intracellular Cu content in cells tended to decrease with time, resulting in reduced cell/total Cu ratios, with the highest reduction noted for cells exposed to uncoated NPs. Overall, the toxicity of the surface-modified CuO NPs could be explained by synergistic interactions between the NPs, their dissolution, and the toxicity of the coating agents.

38. Characterization of marine macroalgae by fluorescence signatures

Abstract

The feasibility of distinguishing macroalgal classes by their fluorescence signatures was investigated using narrow-waveband light to excite groups of accessory pigments in brown, red, and green macroalgae and measuring fluorescence emission at 685 nm. Results obtained on 20 marine macroalgae field-collected samples showed that fluorescence excitation signatures were relatively uniform within phylogenetic classes but were substantially different for different classes. It is suggested that it may be possible to characterize the type and the abundance of subtidal macroalgae from low-flying aircraft using existing laser-induced fluorescence methodology.
