Dendrobium loddigesii is a precious traditional Chinese medicine with high medicinal and ornamental value. However, the characterization of its mitochondrial genome is still pending. Here, we assembled the complete mitochondrial genome of D. loddigesii and discovered that its genome possessed a complex multi-chromosome structure. The mitogenome of D. loddigesii consisted of 17 circular subgenomes, ranging in size from 16,323 bp to 56,781 bp. The total length of the mitogenome was 513,356 bp, with a GC content of 43.41%. The mitogenome contained 70 genes, comprising 36 protein-coding genes (PCGs), 31 tRNA genes, and 3 rRNA genes. Furthermore, we detected 403 repeat sequences as well as identified 482 RNA-editing sites and 8154 codons across all PCGs. Following the sequence similarity analysis, 27 fragments exhibiting homology to both the mitogenome and chloroplast genome were discovered, accounting for 9.86% mitogenome of D. loddigesii. Synteny analysis revealed numerous sequence rearrangements in D. loddigesii and the mitogenomes of related species. Phylogenetic analysis strongly supported that D. loddigesii and D. Amplum formed a single clade with 100% bootstrap support. Micro-pits are widely used in the aerospace and tribology sectors on cylindrical surfaces and electrochemical micromachining which are of great significance for the high material removal rate, absence of tool wear, and mechanical stress, while facing significant challenges such as stray corrosion and low machining efficiency. Aiming at the above problems, this paper proposes a comprehensive method called radial ultrasonic rolling electrochemical micromachining (RUREMM) in which an ultrasonic field has been added onto the cylindrical surface. First, a theoretical model was created to gain the rules of the formation and collapse of bubbles in the liquid medium. Second, to analyze the optimal size of the cathode electrode, the COMSOL5.2 simulation software was proposed to research the influence of the electric field on the different dimensions, and the influences of different parameters in RUREMM on material depth/diameter ratio and roughness are explored through processing experiments. Research results found that the cavitation bubble undergoes expansion, compression, collapse and oscillation, where the max deviation is less than 12.5%. The optimized size was chosen as 200 × 200 μm2 and an electrode spacing of 800 μm through a series of electric field model simulation analyses. Relevant experiments show that the minimum pits with a width of 212.4 μm, a depth of 21.8 μm, and a surface roughness (Ra) of 0.253 μm were formed due to the optimized parameters. The accurate and timely detection of bacteria is critically important for human health as it helps to determine the original source of bacterial infections and prevent disease spread. Herein, gold nanoparticles (AuNPs) were synthesized using polyoxometalates (POMs) as the stabilizing agent. Since AuNPs have glucose oxidase (GOx)-like activity and POMs possess peroxidase (HRP)-like activity, the as-prepared Au@POM nanoparticles have double enzyme-like activities and facilitate cascade reaction. As known, glucose is required as an energy resource during bacterial metabolism, the concentration of glucose decreases with the increase of bacteria content in a system with bacteria and glucose. Therefore, when we use Au@POM nanozymes to trigger the cascade catalysis of glucose and 3,3′,5,5′-tetramethylbenzidine (TMB), the concentration of glucose and bacteria can be sensitively detected using the absorbance intensity at 652 nm in the visible spectrum. As demonstration, S. aureus and E. coli were used as model bacteria. The experimental results show that the present method has a good linear relationship in the bacterial concentration range of 1 to 7.5 × 107 colony-forming units (CFU) mL−1 with a detection limit of 5 CFU mL−1. To achieve high precision, stability, and good surface quality when producing micro-dimple arrays on cylindrical surfaces, we propose a new processing method known as radial ultrasonic rolling electrochemical micromachining (RUREMM) in this study. This method is based on the electrochemical micromachining (EMM) and ultrasonic machining principle. The relevant simulation model was created, and ANSYS researched the flow field characteristics of the electrolyte between the array electrodes and the workpiece. Micro-dimple arrays were created on a SS304 cylindrical surface with the consideration of the effects of the machining parameters, including ultrasonic amplitude and applied pulse voltage. Compared with the EMM, the average width of the micro-dimples is reduced by 24.5%, the aspect ratio of the dimple is increased by 108.0%, and the surface roughness of micro-dimples is decreased by 59.7%. In addition, the localization and the surface quality of micro-dimples by RUREMM can be improved when using appropriate machining parameters. [ABSTRACT FROM AUTHOR]

An AS-EMM method was presented to process the microstructures with different array shapes on planes and cylindrical metal surfaces. Comparisons were carried out between AS-EMM and EMM by analyzing the machining accuracy, surface quality and friction properties of the micro-texture. The results show that the eroded dimple aspect ratios of planes and cylinder pairs are increased by 45.6% and 25.8% respectively which may improve the dissolution localization and the machining precision by AS-EMM. Besides, the friction-wear test results express the 13.6% and 16.2% reduction respectively in average coefficient of friction values on different pairs by AS-EMM. [ABSTRACT FROM AUTHOR]

Summary: The genus Rhizobium usually has a multipartite genome architecture with a chromosome and several plasmids, making these bacteria a perfect candidate for plasmid biology studies. As there are no universally shared genes among typical plasmids, network analyses can complement traditional phylogenetics in a broad‐scale study of plasmid evolution. Here, we present an exhaustive analysis of 216 plasmids from 49 complete genomes of Rhizobium by constructing a bipartite network that consists of two classes of nodes, the plasmids and homologous protein families that connect them. Dissection of the network using a hierarchical clustering strategy reveals extensive variety, with 34 homologous plasmid clusters. Four large clusters including one cluster of symbiotic plasmids and two clusters of chromids carrying some truly essential genes are widely distributed among Rhizobium. In contrast, the other clusters are quite small and rare. Symbiotic clusters and rare accessory clusters are exogenetic and do not appear to have co‐evolved with the common accessory clusters; the latter ones have a large coding potential and functional complementarity for different lifestyles in Rhizobium. The bipartite network also provides preliminary evidence of Rhizobium plasmid variation and formation including genetic exchange, plasmid fusion and fission, exogenetic plasmid transfer, host plant selection, and environmental adaptation.
