Since sweetness is one of the most important qualities of many fruits, and since sugars are translocated from leaves to fruits, the present study investigates photosynthetic activity, activity of sugar metabolizing enzymes, sugar content in leaves and fruits and endogenous levels of hydrogen peroxide in leaves of melon plants treated with various dilutions of hydrogen peroxide, a nonspecific signaling molecule in abiotic stress. For this purpose, 4-month-old melon plants were treated with various concentrations (50mM) of hydrogen peroxide by applying 300 mL per day to the soil of potted plants. The treatments resulted in increased fructose, glucose, sucrose and starch in the leaves and fruits. The most effective concentration of hydrogen peroxide was 20mM. During the day, soluble sugars in leaves were highest at 12:00 h and starch at 15:00 h. Furthermore, the peroxide treatment increased the photosynthetic activity and the activities of chloroplastic and cytosolic fructose-1,6-bisphosphatase, sucrose phosphate synthase and invertases. Thus, our data show that exogenous hydrogen peroxide, applied to the soil, can increase the soluble sugar content of melon fruits.

The DnaK/Hsp70 family is a molecular chaperone that binds non-native states of other proteins, and concerns to various physiological processes in the bacterial, plant and animal cells. Previously, we showed that overexpression of DnaK from a halotolerant cyanobacterium Aphanothece halophytica (ApDnaK) enhances tolerance to abiotic stresses such as high salinity and high temperature in tobacco plants. Here, we tested the transformation of poplar (Populus alba) with ApDnaK for enhancing the growth of transformed poplar plants. Under control growth conditions, transgenic poplar plants exhibited similar growth rates with the wild-type plants during young seedlings under low light intensity, whereas they showed faster growth, larger plant size, and higher cellulose contents when poplar plants were grown under high light intensity. Transgenic young poplar plants exhibited more rapid recovery from the stresses of high salinity, drought, and low temperature compared with those of the wild type plants when poplar plants were grown under low light intensity. These results suggest that ApDnaK could be useful to enhance the growth rate as well as to increase the stress tolerance.

• Background and Aims Glycinebetaine (GB), a quaternary ammonium compound, is a very effective compatible solute. In higher plants, GB is synthesized from choline (Cho) via betaine aldehyde (BA). The first and second steps in the biosynthesis of GB are catalysed by choline monooxygenase (CMO) and by betaine aldehyde dehydrogenase (BADH), respectively. Rice (Oryza sativa), which has two genes for BADH, does not accumulate GB because it lacks a functional gene for CMO. Rice plants accumulate GB in the presence of exogenously applied BA, which leads to the development of a significant tolerance to salt, cold and heat stress. The goal in this study was to evaluate and to discuss the effects of endogenously accumulated GB in rice. • Methods Transgenic rice plants that overexpressed a gene for CMO from spinach (Spinacia oleracea) were produced by Agrobacterium-mediated transformation. After Southern and western blotting analysis, GB in rice leaves was quantified by 1H-NMR spectroscopy and the tolerance of GB-accumulating plants to abiotic stress was investigated. • Key Results Transgenic plants that had a single copy of the transgene and expressed spinach CMO accumulated GB at the level of 0·29–0·43 μmol g−1 d. wt and had enhanced tolerance to salt stress and temperature stress in the seedling stage. • Conclusions In the CMO-expressing rice plants, the localization of spinach CMO and of endogenous BADHs might be different and/or the catalytic activity of spinach CMO in rice plants might be lower than it is in spinach. These possibilities might explain the low levels of GB in the transgenic rice plants. It was concluded that CMO-expressing rice plants were not effective for accumulation of GB and improvement of productivity.

We have found that hydrogen peroxide at low levels can induce synthesis of osmoprotectants, e.g. glycinebetaine in barley, and proline and sucrose in rice. Then we have succeeded in enhancing salt-stress tolerance by hydrogen peroxide treatment in rice, which is known to be one of the most salt-sensitive crops. Furthermore we have found that hydrogen peroxide treatment induces a higher sugar content in tomato and melon fruits. Hydrogen peroxide was effective in accelerating germination of a turf grass (Kentucky bluegrass) under cool conditions. Thus we conclude that application of hydrogen peroxide can be a useful technique for agriculture.

To uncover the adaptation mechanisms of photosystems for halotolerance, changes in stoichiometry and activity of photosystems in response to changes of salinities were examined in a halotolerant cyanobacterium, Aphanothece halophytica. Photosynthetic O2 evolution was high even at high salinities. O2 evolution activity increased with increasing external concentration of NaCl, reached a maximum at 1.5 M NaCl, and then decreased. Similar salt dependence was observed for photosystem II activity. On the other hand, photosystem I activity increased concomitantly with increase in salinity. Photoacoustic measurements indicated that appreciable energy storage by photosystem I mediated cyclic electron flow at high salinities. Significant electron donation to photosystem I reaction centres through NAD(P)H-dehydrogenase complexes was observed in high salt media. The contents of cytochrome b6/f and photosystem II were almost constant under various salinity conditions, whereas the levels of chlorophyll α, photosystem I, soluble cytochrome c-553, and NAD(P)H-dehydrogenase increased in the cells grown with high salinities. These results indicate that salt specifically induces an increase of protein levels involving cyclic electron flow around photosystem I that may entail an important role for adaptation of Aphanothece halophytica cells to high salinities.
