6. Escherichia coli removal in down-flow hanging sponge reactors: insights from laboratory reactor studies.

Abstract

Down-flow hanging sponge (DHS) reactors, employed in domestic wastewater treatment, have demonstrated efficacy in eliminating Escherichia coli and other potentially pathogenic bacteria. The aim of this study was to elucidate the mechanism of removal of E. coli by employing a cube-shaped polyurethane sponge carrier within a compact hanging reactor. An E. coli removal experiment was conducted on this prepared sponge. Escherichia. coli level was found to decrease by more than 2 logs after passing through five nutrient-restricted DHS sponges. Conversely, a newly introduced sponge did not exhibit a comparable reduction in E. coli level. Furthermore, under conditions of optimal nutritional status, the reduction in E. coli level was limited to 0.5 logs, underscoring the crucial role of nutrient restriction in achieving effective elimination. Analysis of the sponge-associated bacterial community revealed the presence of a type VI secretion system (T6SS), a competitive mechanism observed in bacteria. Abstract

The aim of this thesis is to investigate how Stevens creates a new Romanticism. It argues that Stevens demonstrates a double view of Romanticism as having positive and negative aspects and it relates discussion of this double view to the development of his poetry and theories of poetry. Stevens shares with the Romantics the belief that through the power of imagination the problem of dualism - especially the split between art and existential reality - can be solved. Prom Stevens's perspective, thinking about what should be respected and what should be corrected in Romanticism provides grounds for the creation of his own new Romanticism. In chapters one and two, by examining the conflict between imagination and reality in the works of Coleridge, Wordsworth, Shelley and Keats, I explore the intertextual relations between Stevens and the Romantics from a perspective informed by the implications of Stevens's work and thought. In chapters three and four, focusing on Stevens's treatment of the relation between imagination and reality, I examine the nuanced differences between his work and that of the Romantics. Chapter five provides a prologue to 'Notes toward a Supreme Fiction', the culmination of Stevens's concern with imagination and reality. In the final chapter I examine how Stevens's new Romanticism, especially its emphasis on the imagination's activity, is concretised in 'Notes toward a Supreme Fiction'. I also explore how the later development of his sense of reality affects his poetic creativity. By examining the influence of the Romantics on Stevens and his response to them, the nature of his poetry can be more accurately understood. Throughout the thesis, I engage, as appropriate, with the work of many critics who have written on Stevens. It is my hope that my own approach gives a folly considered and detailed account of a topic often addressed more briefly by other commentators.

Abstract

Rhodococcus qingshengii CS98 is a bacterium isolated from soil in Japan that shows strong cesium-accumulating ability. The genome sequence of R. qingshengii CS98 was 6,726,107 bp in total length, G+C average content of 62.4% and comprised a circular chromosome (6,240,414 bp) and a linear plasmid (485,693 bp). The genome was predicted a total of 6,255 protein-coding sequences (CDSs), 59 tRNAs and 5 rRNA operons (5S/16S/23S). The complete genome sequence of R. qingshengii CS98 represents a valuable resource for future comparative genomic analysis and will help elucidate the genetic basis of bioremediation features.

Abstract

Two cesium-accumulating soil bacteria, Rhodococcus sp. strain CS402 and R. erythropolis CS98, accumulate rich concentrations of cesium on incubation with 0.5 mM of cesium for 24 hours. The optimum pH value for cesium intake was 8.5. Potassium and rubidium suppress cesium deposition in both the strains, indicating the presence of a potassium transport channel for cesium accumulation.

Abstract

Two strains of bacteria, CS98 and CS402 of the genus Rhodococcus, with the ability to accumulate cesium, were isolated and characterized. The strains had a high-level ability to accumulate cesium, however, the accumulatedcesium was released after 24 and 48 hours for CS98 and CS402, respectively. CS98 and CS402 were able to concentrate cesium with maximum concentration factors of 3.5 x 10(super 4) and 3.6 x 10(super 3), respectively. It is expected that the two strains can be used for the removal and monitoring of radioactive cesium in natural environments.

Abstract

Bacterial production has been often estimated from DNA synthesis rates by using tritium-labeled thymidine. Some bacteria species cannot incorporate extracellular thymidine into their DNA, suggesting their biomass production might be overlooked when using the conventional method. In the present study, to evaluate appropriateness of deoxyribonucleosides for evaluating bacterial production of natural bacterial communities from the viewpoint of DNA synthesis, incorporation rates of four deoxyribonucleosides (thymidine, deoxyadenosine, deoxyguanosine and deoxycytidine) labeled by nitrogen stable isotope (15N) into bacterial DNA were examined in both ocean (Sagami Bay) and freshwater (Lake Kasumigaura) ecosystems in July 2015 and January 2016. In most stations in Sagami Bay and Lake Kasumigaura, we found that incorporation rates of deoxyguanosine were the highest among those of the four deoxyribonucleosides, and the incorporation rate of deoxyguanosine was approximately 2.5 times higher than that of thymidine. Whereas, incorporation rates of deoxyadenosine and deoxycytidine were 0.9 and 0.2 times higher than that of thymidine. These results clearly suggest that the numbers of bacterial species which can incorporate exogenous deoxyguanosine into their DNA are relatively greater as compared to the other deoxyribonucleosides, and measurement of bacterial production using deoxyguanosine more likely reflects larger numbers of bacterial species productions. Abstract

We observed phosphorus (P) compound groups in sediment in Lake Kasumigaura in winter (February and March) and summer (July and August) to identify how their composition differs between the seasons. The concentration of soluble unreactive P in sediment pore water (SUP) was significantly higher in winter than in summer, whereas the concentration of soluble reactive P (SRP) was significantly higher in summer than in winter. In summer, when the concentration of SRP was high, the concentration of orthophosphate was greatest (~80 %) among the P compound groups. The concentration of orthophosphate diesters had a significant negative correlation with SUP concentration and was significantly lower in winter (high SUP concentration) than in summer. Such relative abundance of P compound groups in sediment could have resulted from adsorption of orthophosphate in summer and degradation of orthophosphate diesters in winter; these seasonal processes could contribute significantly to the changes in the concentrations of SRP and SUP, possibly influencing the P concentrations in the water column in Lake Kasumigaura. Abstract

This study proposed a non-radioactive quantitative measurement of bacterial production using stable isotope nitrogen-15-labeled deoxyadenosine ([N]-2′-deoxyadenosine; N-dA) by liquid chromatography mass spectrometry (LC-MS). The method of preparing N-dA described in this study included incubation of seawater or lake water with N-dA for 5-24 h, filtration onto a membrane filter, DNA extraction, enzymatic hydrolysis of DNA to nucleosides and quantification of N-dA by LC-MS. In the DNA extraction, the silica beads method was examined first, but a large amount of salts and enzymatic inhibitors used in the method caused failure of subsequent procedures, such as enzymatic hydrolysis and LC-MS analysis. On the other hand, the magnetic beads method showed much better results for the extraction. The incorporation rate of N-dA was significantly positively correlated to that of tritium-labeled thymidine (H-TdR) in samples of coastal seawaters and lake waters. The average N-dA: H-TdR incorporation ratio for the seawater sample was 0.55 with 2.5 and 97.5 % confidence intervals of 0.51 and 0.58, respectively; the average ratio for the lake water sample was 0.28 with 2.5 and 97.5 % confidence intervals of 0.23 and 0.34, respectively. The results suggest that the N-dA method can be applied to the measurement of bacterial production in aquatic ecosystems, and that this method can accurately predict the DNA synthesis rates measured by the conventional method.
