BACKGROUND: Beside others, neuroinhibitory and sedative effects of CBD were documented. AIM AND METHODS: The aim of the study was to assess the dose-related effects of CBD premedication on the course of isoflurane anesthesia. Wistar rats were pretreated with different doses of CBD 1 h before isoflurane anesthesia. In the pretreatment, animals were given CBD at doses of 100, 20, 10, or 2 mg kg(−1). Before the fifth (control) anesthesia, the animals were given only mid-chain triglyceride oil, which served as a solvent in the CBD formulation. The induction time was determined, and on awakening, the time to appearance of the flexion reflex and the recovery from anesthesia were determined. RESULTS: Statistical analysis showed a significantly shorter induction time if animals were pretreated with 20 mg kg(−1) CBD. In addition, pretreatment with 100 mg kg(−1) CBD resulted in a prolonged induction time, while on awakening, delayed appearance of reflexes and prolonged recovery from anesthesia compared to pretreatment with 20 mg kg(−1) CBD were observed. CONCLUSIONS: The results indicate that the influence of CBD on the course of isoflurane anesthesia depends on the dose and can reduce the induction time. Although this study was performed in laboratory rats, in clinical practice, these data should be considered when CBD-treated patients undergo isoflurane anesthesia.

Comprehensive comparisons of similar lipid based drug delivery systems produced by different technologies are scarce. Spray drying and fluid bed layering technologies were compared with respect to the process and product characteristics of otherwise similar simvastatin loaded dry emulsion systems. Fluid bed layering provided higher process yield (83.3 % vs 71.5 %), encapsulation efficiency (80.0 % vs 68.4 %), relative one month product stability (93.8 % vs 85.5 %), larger and more circular particles (336 µm vs 56 µm) and lower median oil droplet size after product reconstitution in water (2.85 µm vs 4.27 µm), compared to spray drying. However, spray dried products exhibited higher drug content (22.2 mg/g vs 9.34 mg/g). An in-vivo pharmacokinetic study in rats was performed and a pharmacokinetic model was developed in order to compare the optimised simvastatin loaded dry emulsion systems, a simvastatin glyceride mimetic loaded in the dry emulsion and a simvastatin loaded SMEDDS with a reference physical mixture. Of the formulation tested, fluid bed layered pellets excelled and provided a 115 % relative increase in bioavailability. Among the two technologies, fluid bed layering provided dry emulsion products with higher relative bioavailability and better product characteristics for further processing into final dosage forms. In the last 60 years, milk, meat, and egg worldwide consumption per capita increased 2-, 3-, and 5-fold, respectively. In the same period, several studies have reported a drop of semen quality and an increased incidence of cryptorchidism, hypospadias, and some hormone-related cancers (testicular, prostate, breast, endometrial cancer) in humans. A possible reason for these disruptions is increased exposure to exogenous hormones in food of animal origin. Hormones in food are considered to affect the endocrine system and cell signaling and thus disrupt homeostasis in the consumers. Since food safety assurance is a part of the veterinary service, the risk assessment of hormones in food as potential disruptive compounds is a significant challenge. Numerous cohort and epidemiologic studies and meta-analyses have been performed with respect to find an association between the consumption of food of animal origin and particular health disorders. Additionally, several studies in animal models have been performed to explain the mechanisms of disruptive effects of exogenous hormones. Since contradictory results have been reported, there is no general conclusion about the disruptive effects of exogenous hormones. Further experiments on animals in which long-term treatment with exogenous hormones is performed and further comprehensive endocrinological, toxicological, and human epidemiological studies are needed to confirm or deny the role of exogenous hormones in human health disorders. Moreover, long-term exposure to a combination of several exogenous compounds, such as environmental pollutants and dietary hormones and their additive effects, are also not well known and should be a topic for further studies.

Extracts of sweet chestnut (Castanea sativa Mill.) which contain hydrolysable tannins are used to prevent diarrhoea in farm animals due to the astringent effect of tannins. The aim of this study was to establish whether long-term treatment with the chestnut extract as a feed additive affects the acetylcholine (ACh)-triggered contractions of isolated rat ileum. Wistar rats of both sexes were divided into 4 male and 4 female groups with 6 animals in each group. The first, second and third groups of both sexes received feed with 3, 15 and 30 g of chestnut extract per kg, respectively. The fourth group of both sexes received feed without the added chestnut extract and served as the control group. After the 90-day trial, the animals were euthanized, and ilea were isolated and placed in an organ bath filled with Tyrode’s solution. Ilea were connected to an isometric transducer. Ileal contractions were triggered by 0.005, 0.01, 0.05, 0.1, 0.5, 1.0, 5.0 and 10.0 mg of ACh, and contractions were recorded. The groups exhibited dose responsiveness to the ACh treatment steps; however, no significant differences in the contraction intensity were observed among the groups at the same ACh dose level. These results suggest that long-term treatment with chestnut extract as an astringent does not influence the intensity of ileal contractions triggered by ACh. Thus, no adverse effect on intestinal motility can be expected during a long-term use of chestnut extract as a feed additive in livestock production; however, further studies in target animal species are needed to confirm the findings.

Possible effects of xenoestrogens on human health, in particular on male reproductive health, have attracted considerable attention in recent years. Cow's milk was suggested in numerous publications as one of possible sources of xenoestrogens that could affect human health. Although milk has undoubtedly many beneficial health effects and could even have a role in reducing incidence of some cancers, concerns were raised about presumably high levels of estrogens in cow's milk. In intensive farming, concentrations of estrogens in milk are higher due to long milking periods that today extend long into the pregnancy, when concentrations of estrogens in the cow's body rise. Numerous studies examined potential effects of milk on reproductive health and endocrine-related cancers in both experimental studies with laboratory animals, and in human epidemiological studies. In the present review article, we compiled a review of recently published literature about the content of estrogens in cow's milk and potential health effects, in particular on reproductive system, in humans. Although results of published studies are not unequivocal, it seems that there is stronger evidence suggesting that amounts of estrogens in cow's milk are too low to cause health effects in humans.

Maternal stress could reprogram the developing fetal nervous system. A common target of maternal glucocorticoids is fetal neuro-endocrine axis. In the present study, pregnant mice were exposed to stress by injection and their male offspring were tested for sexual and aggressive behaviors in adult life. Three groups of pregnant mice were exposed to stress by sham syringe injection. The first group was injected on days 13, 14, and 15 p.c., the second group was injected on days 17 and 18 p.c., and the third group was injected daily from days 13 to 18 p.c. while control mice were not injected. Male offspring that were exposed to stress on days 13-18 p.c. and 17-18 p.c. were less aggressive and had lower blood testosterone levels in comparison to the control group. In male sexual behavior, there were no statistically significant differences between the groups. Body weight differed significantly with groups injected on days 13-18 p.c. and 13-15 p.c. having significantly higher body weight in adult life than the other two groups. After behavioral testing, brains were processed for immunohistochemical staining with antibodies against vasopressin (AVP) and calbindin (CALB). The expression of AVP and CALB in the lateral septum and in the preoptic area, respectively, did not differ between groups, suggesting that these two masculinization markers were not affected by prenatal stress. Present study therefore shows that even presumably mild and short prenatal stress weakens aggressive behavior of adult male mice, possibly due to reduced testosterone levels in blood.

Welfare of dairy cows can be assessed using welfare assessment protocols consisting of resource, management and animal-based measures. Welfare Quality® Assessment Protocol is one of the best-known protocols, which depends almost entirely on animal-based measures. To gain more objective and rapid welfare assessment, new techniques have been developed to measure welfare of animals, such as hair cortisol concentration. As cortisol is released in response to stress, it has long been used as a biomarker of stress in animals. While the precise mechanism of cortisol incorporation into hair is unknown, hair cortisol concentration seems to be a marker of long-term systemic cortisol concentration. Hair cortisol is, therefore, a potential marker of chronic stress and is not likely to be affected by acute stress. Studies on cattle show connections between hair cortisol concentration and factors such as pregnancy, parity, diseases, ectoparasites, body condition score, environmental changes, stocking density and milk yield. Hair cortisol concentration appears to be affected by time of sampling, cow age and breed, UV radiation, season, body region of sampled hair and hair colour. Its concentration also depends on sampling and analytical methods. Hair cortisol is a promising non-invasive tool to evaluate welfare of dairy cows, however, more research is needed to determine the extent of effects on its concentration and the appropriate method of sampling and analysis. Correlations between Welfare Quality® Assessment Protocol scores and pooled hair cortisol concentrations have not yet been found, and more research is needed with larger sample size, a standardized protocol of hair sampling, processing and analysis. With proper attention to detail, hair cortisol levels in pooled hair samples might come to be used as a reliable indicator of dairy animal welfare.

Background Early maternal separation may have long-lasting physiological effects on different organ systems. Although long-lasting effects of early maternal separation are mostly studied in connection with the development and function of the central nervous system hypothalamic–pituitary–adrenal axis, animal health, behaviour and productivity, there is a lack of information about its impacts on the reproductive system. In the dairy industry, calves are often separated from mothers 1 or 2 days after birth and are not nursed. In the present retrospective study based on data from an artificial insemination (AI) centre, we compared semen parameters and fertility in bulls that were separated from their mothers one day after birth with the semen parameters of bulls that remained with their mothers and were nursed for approximately 2 months. Semen parameters were followed in 3 consecutive years in 52 maternally separated and 22 nursed bulls. Results Ejaculate volume and total sperm count in ejaculate were significantly higher in nursed bulls in comparison to maternally separated bulls at the age 25–36 and 37–48 months, but interestingly, not at the age 12–24 months, during the first year in the AI centre. Non-return rates did not differ between separated and nursed bulls. Conclusion The results suggest that early maternal separation causes long-lasting effects on the functioning of the male reproductive system, evident by reduced production of semen in adult bulls. The data suggest that with a standard of 20 million sperms per straw of frozen semen, 27–78 fewer straws can be obtained from one ejaculate of maternally separated bulls in comparison to the nursed bulls.

Cows are often milked until 60 d before their next expected calving. Milk from cows in the third trimester of pregnancy contains up to 20 times more estrogens than milk from nonpregnant cows. The aim of this study was to evaluate whether exposure to known doses of estrogens from bovine milk could affect blood hormone levels in mice and influence their reproductive organs. This study was performed with 30 intact male and 30 ovariectomized female mice. Mice of each sex were randomly divided into 3 experimental groups, each with 6 animals of each sex, and a control group with 12 animals of each sex. The first experimental group received 4mL of milk each day from a pregnant cow with natural estrone (E1) and 17β-estradiol (E2) in concentrations 0.093 and 0.065ng/mL, respectively. The second experimental group received 4mL of the same milk each day, with an added 10ng/mL of both E1 and E2. The third experimental group received 4mL of the same milk each day, with an added 100ng/mL of both E1 and E2. The control group received no milk. After 8 d of treatment, mice were euthanized, blood was collected, and the uteruses, testes, and seminal vesicles were weighed. The results of our study demonstrated that consumption of native milk from a pregnant cow did not affect plasma E1 and E2 levels in either sex; uterine weight in females; or testosterone levels and testes and seminal vesicle weights in males. Similarly, we found no changes in the group that received the milk with an added 10ng/mL of E1 and E2. We did observe elevated plasma estrogens in both sexes, increased uterus weight in females, and decreased plasma testosterone levels in males from the group that received milk with an added 100ng/mL of E1 and E2. However, concentrations in the third group exceeded the physiological concentration of milk estrogens by 1,000 times, so it would be extremely unlikely to find such concentrations in native cow milk.

The aim of our study was to establish whether heat treatment and souring of milk affect its estrone (E1) and 17β-estradiol (E2) concentrations. Milk samples were collected from 10 Holstein cows in late pregnancy. Concentrations of E1 and E2 were measured in milk samples that were previously heated to 70 and 95°C for 5 min. Additionally, E1 and E2 concentrations were determined in the same milk samples after 2 d of spontaneous souring at room temperature, and these samples were compared with E1 and E2 levels in raw, unprocessed milk. Concentrations of both hormones were determined by commercial ELISA kits. Concentrations of E1 in unprocessed and processed milk (milk heated to 70 and 95°C and soured milk) were (mean ± SE) 47.25 ± 4.16, 44.84 ± 3.47, 41.00 ± 4.55, and 44.92 ± 3.91 pg/mL, respectively. Concentrations of E2 in the same milk samples were 36.11 ± 10.01, 32.46 ± 9.88, 31.78 ± 9.56, and 31.43 ± 8.00 pg/mL, respectively. Concentrations of E1 and E2 in heat-treated milk did not differ significantly from those in unprocessed milk. Similarly, E1 and E2 concentrations in soured milk did not differ significantly from those in unprocessed milk samples. These results indicate that E1 and E2 are stable in milk and that milk processing (heating and souring) does not influence their degradation. Therefore, E1 and E2 concentrations are expected to be similar between commercial full-fat milk and the raw milk from which it was produced.

International audience; AbstractResidues of acaricide coumaphos were assessed in honey, bee brood, and beeswax during a 2-year field experiment. Honey, bee brood, and beeswax samples were collected before and after routine use of coumaphos in the treatment of bee colonies against varroosis in two consecutive years. Determination of coumaphos in honey and bee brood was based on RP-HPLC with UV detection after a liquid-liquid extraction with hexane or ethyl acetate. Coumaphos in beeswax was identified and quantified by GC/MS. Results indicate the undetectable presence of coumaphos in honey. In bee brood, coumaphos was observed after the treatment. In beeswax, the accumulation of coumaphos was determined not only in hives where it was used but also in hives nearby in which coumaphos was not used. Results indicate the accumulation of coumaphos in bee brood and beeswax. Due to the coumaphos accumulation this drug should be used only in strongly affected bee colonies.

Besides genetic factors, the season of semen collection could have an important effect on ejaculate characteristics, although results from previously published studies are somewhat variable. To determine seasonal effects on semen characteristics, we have analyzed 71,983 ejaculates collected from bulls of four different breeds over a 31-year period. Ejaculate volume, semen concentration, and total sperm output were analyzed with the respect to season and age of bull. Results revealed that semen concentration did not vary significantly during seasons, and ejaculate volume and total sperm output are influenced by season in all breeds. The highest ejaculate volume and total number of sperm in ejaculates were observed during the summer, followed by spring, autumn, and winter. Results suggest that the gradual increase in the day length in the spring is most likely responsible for the highest sperm output during the summer months, suggesting that seasonal effects are also present in cattle, which is not normally considered a seasonal species.
