This cruise was the second of in total four seasonal cruises with RV Kronprins Haakon in 2019/20 focusing on biology in the project Arven etter Nansen (AeN). This seasonal cruise was named Q4 (Q4= 4th quarter of the year) investigating in total 17 stations of the established AeN transect along 34 E in the Northern Barents Sea and adjacent Arctic Basin from 76 to 82°N (see Fig. 1 below). The cruise addressed objectives of the research foci in RF1 on Physical drivers, RF2 on Human drivers, RF3 on the living Barents Sea and RA-C Technology and method development, and collected a multitude of data along the Nansen Legacy transect which was ice covered except the southernmost station P1. In addition to in situ sampling, on board experiments were conducted to quantify biological processes, rates and interactions that will also be important feeds into modeling work and projections in RF4 The future Barents Sea. The cruise took a variety of continuous ship measurements (Weather station, EK80, EM203, ADCP, thermosalinograph, pCO2 underway) as well as station measurements such as CTD with water samples, biological sampling of the benthos (box corer, benthic trawl), water column (multinet, MIK net, macrozooplankton trawl and many other smaller nets) and sea ice (snow, ice cores, water just underneath sea ice). In addition, experimental work (respiration, grazing and egg production) was conducted in the ship’s laboratories. The chemistry team onboard measured oxygen, nutrients and pH from standard depths on most CTD stations and sea ice samples. The cruise started in Longyearbyen and ended in Tromsø (28.11.-17.12.2019). The sampling began at the deep (>3000 m) northernmost station of the transect, Stn. P7, and continued along the southward transect until station P1, in open water and Atlantic dominated water masses. During the expedition the Barents Sea was characterized by a relatively large sea ice cover with consolidated sea ice all the way from P7 to P2. The Polar Front was located just north of P1. All process stations were sampled (P7-P1) as well as two ice stations: one close to P7 ad one close to P5. At the southernmost station P1, stormy weather challenged sampling, but most tasks were in the end accomplished except of deploying the box corer, sediment trap and the AUV. These operations were considered too challenging due to strong drift and ship movement, and it was not safe to conduct small boat operations. Challenges with the box corer was also experienced at the deep station P7 due to technical issues. In the end, most work was accomplished despite challenging weather, sea ice conditions and some technical issues making this cruise successful in gaining new important knowledge about the Northern Barents Sea in the polar night season which is extremely poorly studied. The overall high biological activity and biomass at this time of the year, November-December, was surprising for most of us.

(IF 3.25 [2018]; Q1); International audience; Measurement of suspended particulate matter concentration (SPMC) spanning large time and geographical scales have become a matter of growing importance in recent decades. At many places worldwide, complex observation platforms have been installed to capture temporal and spatial variability over scales ranging from cm (turbulent regimes) to whole basins. Long-term in situ measurements of SPMC involve one or more optical and acoustical sensors and, as the ground truth reference, gravimetric measurements of filtered water samples. The estimation of SPMC from optical and acoustical proxies generally results from the combination of a number of independent calibration measurements, as well as regression or inverse models. Direct or indirect measurements of SPMC are inherently associated with a number of uncertainties along the whole operation chain, the autonomous field deployment, to the analyses necessary for converting the observed proxy values of optical and acoustical signals to SPMC. Controlling uncertainties will become an important issue when the observational input comprises systems of sensors spanning large spatial and temporal scales. This will be especially relevant for detecting trends in the data with unambiguous statistical significance, separating anthropogenic impact from natural variations, or evaluating numerical models over a broad ensemble of different conditions using validated field data.The aim of the study is to present and discuss the benefits and limitations of using optical and acoustical backscatter sensors to acquire long-term observations of SPMC. Additionally, this study will formulate recommendations on how to best acquire quality-assured SPMC data sets, based on the challenges and uncertainties associated with those long-term observations. The main sources of error as well as the means to quantify and reduce the uncertainties associated with SPMC measurements are also illustrated.

We investigated food supply mechanisms to a cold-water coral (CWC) reef at 260 m depth on the Norwegian continental shelf using data from a cabled ocean observatory equipped with Acoustic Doppler Current Profilers (ADCPs), an echosounder, and sensors for chlorophyll, turbidity and hydrography in the benthic boundary layer (BBL). Tidal currents of up to tens of cm s-1 dominated BBL hydrodynamics while residual currents were weak (~10 cm s-1), emphasizing a supply and high retention of locally produced phytodetritus within the trough. A direct connection between the reefs and surface organic matter (OM) was established by turbulent mixing and passive particle settling, but relative contributions varied seasonally. Fresh OM from a spring-bloom was quickly mixed into the BBL, but temperature stratification in summer reduced the surface-to-bottom connectivity and reduced the phytodetritus supply. A qualitative comparison among acoustic backscatter in the ADCPs (600 kHz, 190 kHz) and echosounder (70 kHz) suggests that vertically migrating zooplankton may present an alternative food source in summer. Nocturnal feeding by zooplankton in the upper water column sustains downward OM transport independent from water column mixing and may dominate as food supply pathway over sedimentation of the phytodetritus, especially during stratified conditions. In addition, it could present a concentrating mechanism for nutritional components as compensation for the deteriorating phytodetritus quality. Overall, the observed patterns suggest seasonal changes in the food supply pathways to the reef communities. The moderating role of temperature stratification in phytodetritus transport suggests stronger dependence of the cold-water corals on zooplankton for their dietary requirements with increased stratification under future climate scenarios. This study demonstrates the added value of permanent ocean observatories to research based on dedicated campaigns and regular monitoring.

Rationale: We compared gas chromatography/isotope ratio mass spectrometry (GC/IRMS) and liquid chromatography/isotope ratio mass spectrometry (LC/IRMS) for the measurement of δ13C values in carbohydrates. Contrary to GC/IRMS, no derivatisation is needed for LC/IRMS analysis of carbohydrates. Hence, although LC/IRMS is expected to be more accurate and precise, no direct comparison has been reported. Methods: GC/IRMS with the aldonitrile penta-acetate (ANPA) derivatisation method was compared with LC/IRMS without derivatisation. A large number of glucose standards and a variety of natural samples were analysed for five neutral carbohydrates at natural abundance as well as at 13C-enriched levels. Gas chromatography/chemical ionisation mass spectrometry (GC/CIMS) was applied to check for incomplete derivatisation of the carbohydrate, which would impair the accuracy of the GC/IRMS method. Results: The LC/IRMS technique provided excellent precision (±0.08 ‰ and ±3.1 ‰ at natural abundance and enrichment levels, respectively) for the glucose standards and this technique proved to be superior to GC/IRMS (±0.62 ‰ and ±19.8 ‰ at natural abundance and enrichment levels, respectively). For GC/IRMS measurements the derivatisation correction and the conversion of carbohydrates into CO2 had a considerable effect on the measured δ13C values. However, we did not find any significant differences in the accuracy of the two techniques over the full range of natural δ13C abundances and 13C-labelled glucose. The difference in the performance of GC/IRMS and LC/IRMS diminished when the δ13C values were measured in natural samples, because the chromatographic performance and background correction became critical factors, particularly for LC/IRMS. The derivatisation of carbohydrates for the GC/IRMS method was complete. Conclusions: Although both LC/IRMS and GC/IRMS are reliable techniques for compound-specific stable carbon isotope analysis of carbohydrates (provided that derivatisation is complete and the calibration requirements are met), LC/IRMS is the technique of choice. The reasons for this are the improved precision, simpler sample preparation, and straightforward isotopic calibration.

In low-nutrient, macrophyte-dominated coastal zones, benthic ammonium (NH4+) uptake may be influenced by the structural properties of plant canopies via their effect on near-bed hydrodynamics. Using a dual-tracer (uranine and 15NH4+) method that does not require enclosures, we examined how this process affects nutrient uptake rates within a tidally dominated, patchy Caulerpa prolifera–Cymodocea nodosa landscape. NH4+ uptake was determined by calculating tissue 15N excesses and correcting for 15N enrichment as derived from uranine concentration. Vertical hydrodynamic profiles were measured in the downstream flow direction from outside to inside of the C. nodosa bed by using an array of acoustic Doppler velocimeters. The transition from a C. prolifera to a C. nodosa bed included a change in both benthic canopy properties (short and dense to tall and sparse) and sediment topography (0.2-m increase in water column depth) that resulted in an increase in longitudinal advection and turbulent diffusivity within the C. nodosa canopy between 0.5 and 1.5 m from the leading edge. Vertical differences in canopy water exchange appeared to explain variations in uptake between biotic functional groups; however, no clear differences in longitudinal uptake were found. Using in situ labeling, this study demonstrated for the first time the role of hydrodynamics in structuring NH4+ uptake within an undisturbed, patchy macrophyte landscape.

Water quality of the Scheldt estuary (Belgium/The Netherlands) has considerably improved in recent years, especially in the upstream, freshwater reaches. Within the zooplankton community, the copepod Eurytemora affinis, typically abundant in brackish water and quasi-absent from freshwater before 2007, has since substantially developed in the latter, where it now represents 90% of the crustacean mesozooplankton community. Simultaneously, cyclopoid copepod abundance has greatly decreased, while cladoceran abundance did not change. The study aim was: 1) to verify if the zooplankton community described for the period 2007–2009 by Mialet et al. (2011) has stabilized until present, and 2) to look for the environmental conditions favouring E. affinis development and causing changes in the upstream freshwater zooplankton community. The 2002–2012 temporal evolution of the zooplankton distribution at three stations in the upstream freshwater Scheldt estuary was analyzed. Water quality remained better after 2007 than before, and some factors revealed continuous improvement in annual mean concentrations (e.g. increase in O2, decrease in BOD5 and NH4-N concentration). The increase in oxygen and the decrease in NH4-N concentration, together with low discharge during summer were the main environmental factors explaining the development and timing of E. affinis in the upstream freshwater reach. In this reach, E. affinis maximal abundance is shifted to higher temperatures (summer) compared to its typical maximum spring abundance peak in the brackish zone of the Scheldt estuary and in most temperate estuaries. The changes in zooplankton community followed a temporal and spatial gradient induced by the spatio-temporal evolution of water quality improvement. The most downstream station (3) allowed E. affinis development (oxygen concentration > 4 mg L-1; NH4-N concentration < 2 mg L-1, discharge (Q) < 50 m3 s-1) from 2007 onwards, and this station showed the highest E. affinis and the lowest cyclopoid abundance. At the more upstream stations E. affinis developed later and less strongly, and cyclopoids decreased less in abundance than at station 3. While there may be several explanations for the decrease in cyclopoid abundance (competitive grazing, high predation pressure, NH3-N toxicity, sensitivity to oxygen, etc.), there is no clear cause for their decline. Water quality improvement in the freshwater Scheldt estuary has led to environmental, post-heavy polluted conditions, under which no data on zooplankton populations in this estuary were available. This has indicated a plasticity in the temperature tolerance of E. affinis.

We investigated whether climate change results in long-term changes in phytoplankton biomass and phenology in a turbid eutrophic coastal plain estuary. Changes in annual mean chlorophyll a (chla) concentrations were studied for the period 1978–2006 in the eutrophic and turbid macro-tidal Western Scheldt estuary. Three stations were investigated: WS1, at the mouth of the estuary; station WS6, halfway up the estuary; and station WS11, near the Dutch–Belgian border near the upstream end of the estuary. No significant long-term changes in yearly averaged chla concentrations were observed in WS1 and WS6, but in WS11 the phytoplankton biomass decreased considerably. This is most likely due to an increase in grazing pressure as a result of an improvement in the dissolved oxygen concentrations. Spectral analyses revealed a possible periodicity of 7 years in the mean chla which was related to periodicity in river discharge. We also observed strong phenological responses in the timing of the spring/summer bloom which were related to a well-documented increase in the temperature in the estuary. The fulcrum, the center of gravity or the day at which 50% of the cumulative chla was reached during the year, advanced by 1–2 days/year. A similar trend was observed for the month in which the maximum bloom was observed, with the exception of station WS1. All stations showed an earlier initiation of the bloom, whereas the day at which the phytoplankton bloom was terminated also moved forward in time excepted for WS11. As a result, the bloom length decreased at station WS1, remained the same at station WS6, and increased at WS11. This complicated pattern in bloom phenology demonstrates the complex nature of ecosystem functioning in estuaries.

The Lofoten-Versterålen (LoVe) Ocean Observatory is a cabled observatory on the Norwegian continental shelf providing long-term environmental data, including time-lapse imagery of a cold-water coral reef, from a multisensory stationary platform. The high temporal data resolution (frequency and duration) allows new insights into the behavioural responses of reef organisms to environmental variations and interactions between the water column and the seabed community. The dominant current direction in the area is north-west, with the near seabed currents being most consistent (follows the topography). Data analyses clearly indicate increase in polyp activity of Lophelia Pertusa correlates with the spring bloom. The polyp activity increases though the summer, when after the water-column stratification reduces the concentration of phytoplankton after the spring bloom. The reduced phytoplankton concentration and increasing polyp activity seems to correlate with a colour change in L pertusa, i.e. from white to pink and back to white again in the period April – end of November 2015. We preliminary results infer that zooplankton may represent an important food source for the cold-water corals in stratified summer conditions.
