Coxiella burnetii is an intracellular bacterial pathogen that can be associated with significant reproductive disease or acute mortality in livestock and wildlife. A novel marine mammal-associated strain of C. burnetii has been identified in pinnipeds of the northwestern Pacific Ocean. Little is known about C. burnetii infection in regard to reproductive success or population status. Our objective was to characterize the severity and extent of histologic lesions in 117 opportunistically collected placentas from presumed-normal northern fur seals (Callorhinus ursinus) in July 2011 on St. Paul Island, Alaska, US, where a high placental prevalence of C. burnetii had been reported. Sections were examined by histology and immunohistochemistry and impression smears with modified acid-fast stain. The nature and frequency of histologic changes were compared with target COM1 PCR-confirmed C. burnetii positive and negative placentas. Overall, histologic changes were similar to placental lesions described in aborting ruminants; however, changes were variable within and between placentas. Vasculitis and occasional intracellular bacteria were seen only in C. burnetii PCR-positive placentas. Dystrophic mineralization, edema, and inflammation were seen in PCR-positive and negative placentas, although they were statistically more common in PCR-positive placentas. Results suggest that C. burnetti and associated pathologic changes are multifocal and variable in placentas from these presumably live-born pups. Therefore, multiple sections of tissue from different placental areas should be examined microscopically, and screened by PCR, to ensure accurate diagnosis as the genomes per gram of placenta may not necessarily represent the severity of placental disease. These limitations should inform field biologists, diagnosticians, and pathologists how best to screen and sample for pathogens and histopathology in marine mammal placental samples.

Over the past several decades there has been a precipitous decline of northern fur seals (Callorhinus ursinus; NFS) at their breeding grounds on the Pribilof Islands in the Bering Sea. The cause of this decline is likely multifactorial and could include changes in environmental parameters, prey abundance and distribution as well as exposure to pathogens and pollutants. Evaluation of inflammatory markers and antioxidant levels of the current population of fur seals in addition to hematologic and biochemical profiles could provide important information regarding health and subclinical or clinical disease in this population. Serum and plasma samples were obtained from clinically healthy adult female NFS and references intervals were determined for multiple parameters that can be altered in response to the presence of disease and environmental stressors. We established a reference interval for cytokines involved in acute inflammation and infection (TNFa, IL1, IL6, IL8, KC, IL10, C-reactive Protein) by utilizing commercially available canine cross-reactive antibodies. Reference intervals were also established for reactive oxygen species (hydrogen peroxide and malondialdehyde), as well as antioxidant levels (vitamin E and selenium) and acute phase proteins evaluated by serum electrophoresis. To improve the ability to compare and interpret indicators of health and disease in this species, we developed reference intervals for commonly utilized hematologic and biochemical tests in addition to the aforementioned markers of oxidative stress and inflammatory biomarkers. There were several animals identified as outliers indicating that they may have had subclinical illness or inflammation. Further investigation utilizing these tests in clinically ill animals and comparison to animals that exhibit normal behavior and no overt signs of illness could increase our understanding of the utility of measuring these parameters in this species.

Steller sea lion (Eumetopias jubatus) young-of-the-year (YOY) are nutritionally dependent upon their dam through the majority of their first year. Several indices of body condition were measured in YOY 1.5–9 mo of age captured in Southeast Alaska (n = 122), the Gulf of Alaska (n = 182), and the Aleutian Islands (n = 38) to test the hypothesis that nutritional stress impacted the ability of adult female Steller sea lions to adequately nourish their late gestation YOY in the central Aleutian Islands in the early 2000s. Body mass (kg) and percent total body lipid content (%TBL) increased with age in all three regions of Alaska that were sampled (P < 0.05). Young-of-the-year 7–9 mo of age were leaner in Southeast Alaska (27.6% ± 1.0%) and Gulf of Alaska (29.5% ± 0.8%) than in the Aleutian Islands (35.7% ± 1.2%, P < 0.001). Condition indices calculated from morphometric measures did not strongly predict the %TBL measured by isotope dilution. The trend for Aleutian Island YOY to have larger body mass and larger body fat reserves are counter to what would be expected if dams were unable to adequately provision their late lactation YOY due to inadequate food availability in the central Aleutian Islands.

Current climate trends resulting in rapid declines in sea ice and increasing water temperatures are likely to expand the northern geographic range and duration of favorable conditions for harmful algal blooms (HABs), making algal toxins a growing concern in Alaskan marine food webs. Two of the most common HAB toxins along the west coast of North America are the neurotoxins domoic acid (DA) and saxitoxin (STX). Over the last 20 years, DA toxicosis has caused significant illness and mortality in marine mammals along the west coast of the USA, but has not been reported to impact marine mammals foraging in Alaskan waters. Saxitoxin, the most potent of the paralytic shellfish poisoning toxins, has been well-documented in shellfish in the Aleutians and Gulf of Alaska for decades and associated with human illnesses and deaths due to consumption of toxic clams. There is little information regarding exposure of Alaskan marine mammals. Here, the spatial patterns and prevalence of DA and STX exposure in Alaskan marine mammals are documented in order to assess health risks to northern populations including those species that are important to the nutritional, cultural, and economic well-being of Alaskan coastal communities. In this study, 905 marine mammals from 13 species were sampled including; humpback whales, bowhead whales, beluga whales, harbor porpoises, northern fur seals, Steller sea lions, harbor seals, ringed seals, bearded seals, spotted seals, ribbon seals, Pacific walruses, and northern sea otters. Domoic acid was detected in all 13 species examined and had the greatest prevalence in bowhead whales (68%) and harbor seals (67%). Saxitoxin was detected in 10 of the 13 species, with the highest prevalence in humpback whales (50%) and bowhead whales (32%). Pacific walruses contained the highest concentrations of both STX and DA, with DA concentrations similar to those detected in California sea lions exhibiting clinical signs of DA toxicosis (seizures) off the coast of Central California, USA. Forty-six individual marine mammals contained detectable concentrations of both toxins emphasizing the potential for combined exposure risks. Additionally, fetuses from a beluga whale, a harbor porpoise and a Steller sea lion contained detectable concentrations of DA documenting maternal toxin transfer in these species. These results provide evidence that HAB toxins are present throughout Alaska waters at levels high enough to be detected in marine mammals and have the potential to impact marine mammal health in the Arctic marine environment.

Aerial imagery is the most effective method National Marine Fisheries Service (NMFS) uses to assess abundance of Steller sea lions (Eumetopias jubatus). These images are traditionally captured from occupied aircraft, but the long distances between airfields along the 1900 km Aleutian Island chain, inclement weather during the survey season, and dangerous winds at sites adjacent to cliffs severely limit flying opportunities. Because of the pressing need for current trend information for a population in persistent decline we turned to a small unoccupied aircraft system (UAS), an APH-22 hexacopter. Our primary objective was to supplement traditional aerial surveys during the annual abundance survey. The second objective was to test whether the resolution of images captured with the hexacopter was adequate for sighting permanently marked individuals. From June to July 2014, NMFS biologists based on a research vessel assessed sites from Attu Island to the Delarof Islands (n = 23), surveying sites from land (n = 12) and with the hexacopter (n = 11). Simultaneously, traditional aerial surveys were conducted east of the Delarof Islands (n = 172). This combined approach enabled us to conduct the most complete survey of adult, juvenile, and newborn Steller sea lions in the Aleutian Islands since the 1970s. Images collected also allowed for us to identify alpha-numeric permanent marks on individuals as small as juveniles. With this successful implementation of UAS, NMFS plans to use the hexacopter to supplement future surveys.

Otarine herpesvirus 1 (OtHV1) is strongly associated with California sea lion (CSL, Zalophus californianus) urogenital carcinoma, the most common cancer documented in marine mammals. In addition to CSL, OtHV1 has also been found in association with carcinoma in South American fur seals (Arctocephalus australis), demonstrating it can infect related species. Northern fur seals (NFS, Callorhinus ursinus) are sympatric with CSL, and copulation between these species has been observed; yet, there are no reports of urogenital carcinoma in NFS. We describe a new Otarine herpesvirus found in vaginal swabs from NFS, herein called OtHV4. Partial sequencing of the polymerase gene and the glycoprotein B gene revealed OtHV4 is closely related to OtHV1, with 95% homology in the region of polymerase sequenced, and phylogenetic analyses demonstrate that they are sister taxa. An OtHV4-specific hydrolysis probe quantitative PCR was developed and validated, and its use on vaginal swabs revealed 16 of 50 (32%) wild adult female NFS were positive for OtHV4. The identification of a virus highly similar to the carcinoma-associated OtHV1 in a sympatric species without carcinoma suggests that comparative genomics of OtHV1 and OtHV4 may identify candidate viral oncogenes.

Brucella species infect a wide range of hosts with a broad spectrum of clinical manifestations. In mammals, one of the most significant consequences of Brucella infection is reproductive failure. There is evidence of Brucella exposure in many species of marine mammals, but the outcome of infection is often challenging to determine. The eastern Pacific stock of northern fur seals (NFSs, Callorhinus ursinus) has declined significantly, spawning research into potential causes for this trend, including investigation into reproductive health. The objective of the current study was to determine if NFSs on St. Paul Island, Alaska have evidence of Brucella exposure or infection. Archived DNA extracted from placentas ( n = 119) and serum ( n = 40) samples were available for testing by insertion sequence (IS) 711 polymerase chain reaction (PCR) and the Brucella microagglutination test (BMAT), respectively. As well, placental tissue was available for histologic examination. Six (5%) placentas were positive by PCR, and a single animal had severe placentitis. Multilocus variable number tandem repeat analysis profiles were highly clustered and closely related to other Brucella pinnipedialis isolates. A single animal was positive on BMAT, and 12 animals had titers within the borderline range; 1 borderline animal was positive by PCR on serum. The findings suggest that NFSs on the Pribilof Islands are exposed to Brucella and that the organism has the ability to cause severe placental disease. Given the population trend of the NFS, and the zoonotic nature of this pathogen, further investigation into the epidemiology of this disease is recommended.

Coxiella burnetii, a zoonotic bacterium, has recently been identified in several marine mammal species on the Pacific Coast of North America, but little is known about the epidemiology, transmission, and pathogenesis in these species. We tested sera archived from northern fur seals (NFS, Callorhinus ursinus; n=236) and Steller sea lions (SSL, Eumetopias jubatus; n=72) sampled in Alaska for C. burnetii antibodies, and vaginal swabs from NFS (n=40) for C. burnetii by qPCR. The antibody prevalence in NFS samples from 2009 and 2011 (69%) was significantly higher than in 1994 (49%). The antibody prevalence of SSL samples from 2007 to 2011 was 59%. All NFS vaginal swabs were negative for C. burnetii, despite an 80% antibody prevalence in the matched sera. The significant increase in antibody prevalence in NFS from 1994 to 2011 suggests that the pathogen may be increasingly common or that there is marked temporal variation within the vulnerable NFS population. The high antibody prevalence in SSL suggests that this pathogen may also be significant in the endangered SSL population. These results confirm that C. burnetii is more prevalent within these populations than previously known. More research is needed to determine how this bacterium may affect individual, population, and reproductive health of marine mammals.

Viruses of the family Polyomaviridae infect a wide variety of avian and mammalian hosts with a broad spectrum of outcomes including asymptomatic infection, acute systemic disease, and tumor induction. In 2010, intranuclear viral inclusion bodies were identified in trophoblasts of a single northern fur seal (NFS; Callorhinus ursinus) placenta from a presumed healthy birth on St. Paul Island, Alaska. On transmission electron microscopy, virions were approximately 40 nm in diameter and were arranged in paracrystal-line arrays within the nucleus. The tissue was positive for the polyomaviral major capsid gene (VP1) by PCR, and the sequenced product revealed a novel Orthopolyomavirus. Twenty-nine additional NFS placentas, devoid of viral inclusions on histologic examination, were tested for polyomavirus by PCR; all were negative. The significance of this novel virus for the infected animal is unknown, but the virus does not appear to be very prevalent within the placentas from newborn northern fur seal pups.

We used visual observations during nine field seasons to determine the causes and death rate of Steller sea lions older than 1 year on eight rookeries in the Russian Far East. The average annual death rate was 0.48% of the total recorded Steller sea lions on shore and varied among rookeriesy. The mortality did not exceed 0.29% in six rookeries, but reached 0.80–1.19% in two rookeries. Individuals of all age and sex groups were recorded among dead animals. Bulls died as a result of territorial conflicts, and cows and young lions were crushed or suffocated by bulls during copulation. The death of nearly half of the females was attributed to 11 bulls.

The biology of the Steller sea lion (Eumetopias jubatus) has been the subject of intense scientific investigation. This is primarily due to the rapid decline of population size in the western part of the species’ range since the 1970s and the subsequent Threatened and Endangered species listings that had direct impact on the management of one of the world’s largest fisheries. The Steller sea lion has emerged as an indicator species representing the environmental health of the North Pacific Ocean and Bering Sea. In this study, to better understand the historical processes that have culminated in the extant populations of E. jubatus, a large genetic data set consisting of 3 mitochondrial regions for .1,000 individuals was analyzed from multiple phylogeographic and demographic perspectives. The results describe the role of climate change in shaping the population structure of E. jubatus. Climatically associated historical processes apparently involved differential demographic responses to ice ages (and putative glacial vicariance) dependent on population size. Ice ages during times of small effective population size promoted restricted gene flow and fragmentation, and ice ages occurring during times of large population size promoted gene flow and dispersal. These results illustrate that effective population size has a profound effect on how species respond to climate change, an observation with obvious implications for large mammals and endangered species under the present conditions of imminent anthropogenically caused climate change. In addition, the results confirm previous observations of strongly biased historic and contemporary gene flow involving dispersal from west to east. Furthermore, phylogenetic patterns in combination with available fossil data suggest the potential of an Asian origin of E. jubatus. The results of this study provide a detailed scenario for the history that has shaped contemporary populations of E. jubatus.

The 2008 IUCN (International Union for Conservation of Nature) review of the status of the world’s mammals identified marine mammals (IUCN 2008) as disproportionally threatened and data poor compared to their terrestrial counterparts, and their status was noted as a particular concern (IUCN 2008, Mace et al. 2008, Schipper et al. 2008). The threats faced by marine mammals were identified as being different, with accidental mortality and pollution being dominant threats that superseded habitat loss, which was identified as the principal concern for land mammals. It was suggested that harvesting remained a major threat for half of the marine mammal species in the world. The distribution of marine mammals was described as being concentrated in tropical and temperate coastal platforms and associated with high levels of primary productivity. Threat levels were highest for marine mammals living in the North Atlantic, North Pacific and Southeast Asia. Range size was described as generally declining toward both poles. Although these generalities are of course correct for all marine mammals, it must be recognized that the larger number of species of cetaceans compared to other marine mammal groups (85 cetaceans, the polar bear Ursus maritimus, 4 extant sirenians and 36 pinniped species) biases the conclusions in the assessment for marine mammals to such a degree that a separate summary for the pinnipeds of the world is warranted. Additionally, a species-level assessment masks some important conservation concerns for pinnipeds with species classified as Least Concern actually containing threatened subspecies ( e.g., ringed seals). Finally, and perhaps most importantly, climate change issues were not specifically addressed in the vast majority of marine mammal assessments done for the 2008 IUCN review (IUCN 2008). We present here a complete assessment of the status and threats facing pinnipeds at the subspecies level, integrating the expected impacts of changing patterns of threat due to global climate change.

Maternal attendance patterns in free-ranging wildlife can provide insight into basic biology, foraging behavior, and population dynamics. We collected detailed visual observations of attendance patterns by adult lactating Steller sea lions (Eumetopias jubatus) from 2005 to 2007 on 6 major rookeries in the Russian Far East, including those with an increasing population trend (Sea of Okhotsk), severely depleted populations that were recovering (Kuril Islands), or those that were stable (Commander Islands). Individually identifiable females were observed during the postpartum period, with special attention paid to presence and absence during the day and to departure and arrival times. Within Russia females on several Kuril Islands rookeries exhibited extremely short foraging trip durations (median 6.5–8.0 h) and spent higher proportions of time on the rookery (75–82%), whereas females in the Sea of Okhotsk population had the longest trips (median 19.8 h) and spent the least amount of time on the roo...

Steller sea lion (Eumetopias jubatus) pups (n = 366) were hot-branded at Lowrie Island, Southeast Alaska, USA, in June 2001 and 2002 for vital-rates studies. To assess potential mortality following branding, we estimated weekly survival to 12 weeks postbranding using mark–recapture models. Survival estimates ranged from 0.984/week to 0.988/week, or 0.868 over the 12-week period; varied little with sex, year, and capture area; and were higher for larger than smaller male pups and unexpectedly lower for larger than smaller female pups. Inclusion of resights at 1–3 years of age prevented a −4.5% bias in cumulative survival to 12 weeks postbranding by accounting for pups that survived but permanently emigrated from Lowrie Island during the 12-week survey. Characterization of the polygenic and polymorphic features of the Steller sea lion major histo- compatibility complex (MHC) provides an ideal window for evaluating immunologic vigor of the population and identifying emergence of new genotypes that reflect ecosystem pressures. MHC genotyping can be used to measure the potential immunologic vigor of a population. However, since ecosystem-induced changes to MHC genotype can be slow to emerge, measurement of differential expression of these genes can potentially provide real-time evidence of immunologic perturbations. MHC DRB genes were cloned and sequenced using peripheral blood mononuclear leukocytes derived from 10 Steller sea lions from Southeast Alaska, Prince William Sound, and the Aleutian Islands. Nine unique DRB gene sequences were represented in each of 10 animals. MHC DRB gene expression was measured in a subset of six sea lions. Although DRB in genomic DNA was identical in all individuals, relative levels of expressed DRB mRNA was highly variable. Selective sup- pression of MHC DRB genes could be indicative of geographically disparate environmental pressures, thereby serving as an immediate and sensitive indicator of population and ecosystem health.

Population growth typically involves range expansion and establishment of new breeding sites, while the opposite occurs during declines. Although density dependence is widely invoked in theoretical studies of emigration and colonization in expanding populations, few empirical studies have documented the mechanisms. Still fewer have documented the direction and mechanisms of individual transfer in declining populations. Here, we screen large numbers of pups sampled on their natal rookeries for variation in mtDNA (n = 1106) and 16 microsatellite loci (n = 588) and show that new Steller sea lion breeding sites did not follow the typical paradigm and were instead colonized by sea lions from both a declining (Endangered) population and an increasing population. Dispersing individuals colonized rookeries in the distributional hiatus between two evolutionarily distinct (Φ¯(st) = 0.222, R¯(st) = 0.053, K = 2) metapopulations recently described as separate subspecies. Hardy-Weinberg, mixed-stock and relatedness analysis revealed levels of interbreeding on the new rookeries that exclude (i) assortative mating among eastern and western forms, and (ii) inbreeding avoidance as primary motivations for dispersal. Positive and negative density dependence is implicated in both cases of individual transfer. Migration distance limits, and conspecific attraction and performance likely influenced the sequence of rookery colonizations. This study demonstrates that resource limitation may trigger an exodus of breeding animals from declining populations, with substantial impacts on distribution and patterns of genetic variation. It also revealed that this event is rare because colonists dispersed across an evolutionary boundary, suggesting that the causative factors behind recent declines are unusual or of larger magnitude than normally occur.

Genetic studies and differing population trends support the separation of Steller sea lions (Eumetopias jubatus) into a western distinct population segment (WDPS) and an eastern DPS (EDPS) with the dividing line between populations at 144° W. Despite little exchange for thousands of years, the gap between the breeding ranges narrowed during the past 15–30 years with the formation of new rookeries near the DPS boundary. We analyzed >22,000 sightings of 4,172 sea lions branded as pups in each DPS from 2000–2010 to estimate probabilities of a sea lion born in one DPS being seen within the range of the other DPS (either ‘West’ or ‘East’). Males from both populations regularly traveled across the DPS boundary; probabilities were highest at ages 2–5 and for males born in Prince William Sound and southern Southeast Alaska. The probability of WDPS females being in the East at age 5 was 0.067 but 0 for EDPS females which rarely traveled to the West. Prince William Sound-born females had high probabilities of being in the East during breeding and non-breeding seasons. We present strong evidence that WDPS females have permanently emigrated to the East, reproducing at two ‘mixing zone’ rookeries. We documented breeding bulls that traveled >6,500 km round trip from their natal rookery in southern Alaska to the northern Bering Sea and central Aleutian Islands and back within one year. WDPS animals began moving East in the 1990s, following steep population declines in the central Gulf of Alaska. Results of our study, and others documenting high survival and rapid population growth in northern Southeast Alaska suggest that conditions in this mixing zone region have been optimal for sea lions. It is unclear whether eastward movement across the DPS boundary is due to less-optimal conditions in the West or a reflection of favorable conditions in the East.

In 2010, Coxiella burnetii was identified in 75% of northern fur seal placentas from a single rookery in Alaska, but nothing was known about the significance of this organism in the population. Although many infectious organisms cause increased cell death, C. burnetii has been shown to suppress apoptosis of the host macrophages as an intracellular survival mechanism. To determine if infection induces a similar functional change in the placenta, immunohistochemistry for antibodies to cleaved caspase-3 (activated caspase-3) and the (TDT)-mediated dUTP-digoxigenin nick end labeling (TUNEL) technique were used to compare the amount of placental apoptosis in infected and noninfected placentas. There was a statistically significant difference in the frequency of apoptotic cells between infected and uninfected placentas, with more apoptosis identified in the uninfected placentas. This finding suggests that the survival mechanism of C. burnetii in host macrophages to reduce apoptosis may also be utilized in trophoblasts. The significance of decreased trophoblastic apoptosis for the northern fur seal fetus requires further investigation.

The decline in the number of northern fur seal (NFS; Callorhinus ursinus) pups on St. Paul Island, Alaska, has led to multidisciplinary research, including investigation into issues of reproductive health and success. Given the recent identification of Coxiella burnetii in the placenta of two other marine mammal species, NFS placentas were collected from Reef rookery on St. Paul Island, Alaska, during the 2010 pupping season, examined histologically, and tested for C. burnetii using polymerase chain reaction (PCR). Of 146 placentas examined, gram-negative intratrophoblastic bacteria that were positive for C. burnetii on immunohistochemistry were observed in 5 (3%) placentas. Placental infection was usually devoid of associated inflammation or significant ancillary pathology. One hundred nine (75%) of the placentas were positive for C. burnetii on PCR. C. burnetii is globally distributed and persists for long periods in the environment, providing ample opportunity for exposure of many species. The significance of this finding for the declining fur seal population, potential human exposure and infection, and impact on other sympatric marine mammal or terrestrial species is unclear; further investigation into the epidemiology of Coxiella in the marine ecosystem is warranted.

Despite the widely recognized incidence of homoplasy characterizing this region, the hypervariable region I (HVRI) of the mitochondrial control region is one of the most frequently used genetic markers for population genetic and phylogeographic studies. We present an evolutionary analysis of HVRI and cytochrome b sequences from a range-wide survey of 1031 Steller sea lions, Eumetopias jubatus, to quantify homoplasy and substitution rate at HVRI. Variation in HVRI was distributed across 41 variable sites in the 238-bp segment examined. All variants at HVR1 were found to be transitions. However, our analyses suggest that a minimum of 101 changes have actually occurred within HVRI with as many as 18 substitutions occurring at a single site. By including this hidden variation into our analyses, several instances of apparent long-range dispersal were resolved to be homoplasies and 8.5-12% of observed HVRI haplotypes were found to have geographic distributions descriptive of convergent molecular evolution rather than identity by descent. We estimate the rate of substitution at HVRI in Steller sea lions to be approximately 24 times that of cytochrome b with an absolute rate of HVRI substitution estimated at 27.45% per million years. These findings have direct implications regarding the utility of HVRI data to generate a variety of evolutionary genetic hypotheses.

Many studies use genetic markers to explore population structure and variability within species. However, only a minority use more than one type of marker and, despite increasing evidence of a link between heterozygosity and individual fitness, few ask whether diversity correlates with population trajectory. To address these issues, we analysed data from the Steller's sea lion, Eumetiopias jubatus, where three stocks are distributed over a vast geographical range and where both genetic samples and detailed demographic data have been collected from many diverse breeding colonies. To previously published mitochondrial DNA (mtDNA) and microsatellite data sets, we have added new data for amplified fragment length polymorphism (AFLP) markers, comprising 238 loci scored in 285 sea lions sampled from 23 natal rookeries. Genotypic diversity was low relative to most vertebrates, with only 37 loci (15.5%) being polymorphic. Moreover, contrasting geographical patterns of genetic diversity were found at the three markers, with Nei's gene diversity tending to be higher for AFLPs and microsatellites in rookeries of the western and Asian stocks, while the highest mtDNA values were found in the eastern stock. Overall, and despite strongly contrasting demographic histories, after applying phylogenetic correction we found little correlation between genetic diversity and either colony size or demography. In contrast, we were able to show a highly significant positive relationship between AFLP diversity and current population size across a range of pinniped species, even though equivalent analyses did not reveal significant trends for either microsatellites or mtDNA.

The endangered Western population of the Steller sea lion declined for three decades for uncertain reasons. We present baseline data of metal concentrations in pups as a first step towards investigating the potential threat of developmental exposures to contaminants. Seven metals were investigated: arsenic, cadmium, silver, aluminum, mercury, lead and vanadium. Vanadium was detected in only a single blubber sample. Mercury appears to be the most toxicologically significant metal with concentrations in the liver well above the current action level for mercury in fish. The concentrations of aluminum, arsenic, silver, cadmium and lead were present in one-fourth to two-thirds of all samples and were at either comparable or below concentrations previously reported. Neither gender nor region had a significant effect on metal burdens. Future work should consider metal concentrations in juveniles and adults and toxicological studies need to be performed to begin to assess the toxicity of these metals.

A young harbor seal (Phoca vitulina richardi), stranded on the coast of California, was found to have a 20-cm-diameter cranial cervical mass. Surgical excision revealed the subcutaneous mass to be covered in haired skin with multiple glabrous areas and structures resembling a jaw with tooth buds, eyelids, and a tail. The mass deformed the host pup's skull. Histologic examination revealed a complete vertebra in the tail, teeth in the jaw, and areas resembling tongue and larynx. Class 1 MHC sequences amplified by polymerase chain reaction from the mass and the host twin were identical. The mass was diagnosed as a fetus in fetu, a rare congenital anomaly in which 1 conjoined twin is completely enclosed in the body of the other twin. The host pup died, and no additional defects were found; however, blubber levels of persistent organic pollutants were high. The cause of the congenital anomaly in this pup is uncertain.

Lesions suggestive of poxvirus infection were observed in two Steller sea lions (Eu- metopias jubatus) in Alaska during live capture-and-release studies during 2000 and 2001. Both of these animals, female pups in poor body condition, were from Prince William Sound; this population is part of the declining western stock. Umbilicated, typically ulcerated dermal nodules were present, primarily on the fore flippers in one case, and over most of the body in the second case. Histologically, there were discrete masses in the superficial dermis composed of epithelial cells, some of which contained eosinophilic intracytoplasmic inclusion bodies. Negative staining of skin biopsy homogenates demonstrated the presence of orthopoxvirus-like particles. Total DNA extracted from skin biopsies were analyzed by polymerase chain reaction (PCR) using primers that targeted the DNA polymerase and DNA topoisomerase genes. These primers directed the amplification of fragments 543 base pairs (bp) and 344 bp, respectively, whose deduced amino acid sequences indicated the presence of a novel poxvirus within the Chordopoxvirinae subfamily. Comparison of these amino acid sequences with homologous sequences from members of the Chordopoxvirinae indicated highest identity with orthopoxviruses.

Information concerning mechanistic processes underlying changes in vital rates and ultimately population growth rate is required to monitor impacts of environmental change on wildlife. We estimated age-specific survival and examined factors influencing survival for a threatened population of Steller sea lions (Eumetopias jubatus) in southeastern Alaska. We used mark-recapture models and data from 1,995 individuals marked at approximately one month of age at four of five rookeries in southeastern Alaska, and resighted from Oregon to the Bering Sea. Average annual survival probability for females was 0.64 for pups and 0.77 for yearlings, and increased from 0.91 to 0.96 from age 3–7 yrs. Annual survival probability of males averaged 0.60 for pups and 0.88 by 7 yrs, resulting in probability of survival to age 7, 33% lower for males compared to females. Pups from northern southeastern Alaska (including an area of low summer population size but rapid growth) were twice as likely to survive to age 7 compared to pups from southern rookeries (including a large, historical, stable rookery). Effects of early conditions on future fitness were observed as (1) environmental conditions in the birth year equally affected first- and second-year survival, and (2) effects of body mass at approximately one month of age were still apparent at 7 yrs. Survival from 0–2 yrs varied among five cohorts by a maximum absolute difference of 0.12. We observed survival costs for long-distance dispersal for males, particularly as juveniles. However, survival was higher for non-pups that dispersed to northern southeastern Alaska, suggesting that moving to an area with greater productivity, greater safety, or lower population size may alleviate a poor start and provide a mechanism for spatial structure for sea lion populations.

Background: The northern fur seal (Callorhinus ursinus) is an important cultural and nutritional resource for the Aleut community on St. Paul Island Alaska. In recent years, an increasing number of zoonotic pathogens have been identified in the population, but the public health significance of these findings is unknown. To determine the prevalence of Coxiella burnetii and Brucella spp. in northern fur seal tissues, eight tissue types from 50 subsistence-harvested fur seals were tested for bacterial DNA by real-time polymerase chain reaction. Findings: Of the 400 samples tested, only a single splenic sample was positive for Brucella spp. and the cycle threshold (ct) value was extremely high suggesting a low concentration of DNA within the tissue. C. burnetii DNA was not detected. Conclusions: Findings suggest that the risk of humans contracting brucellosis or Q fever from the consumption of harvested northern fur seals is low.
