The aim of this study was to compare plant–soil interactions in the native range of two congeneric European species differing in their invasive success in the world: a globally invasive Cirsium vulgare and non-invasive C. oleraceum. We assessed changes in soil nutrients and soil biota following soil conditioning by each species and compared performance of plants grown in self-conditioned and unconditioned soil, from which all, some or no biota was excluded. The invasive species depleted more nutrients than the non-invasive species and coped better with altered nutrient levels. The invasive species had higher seedling establishment which benefited from the presence of unconditioned biota transferred by soil filtrate. Biomass of both species increased in soil with self-conditioned soil filtrate and decreased in soil with self-conditioned whole-soil inoculum compared to unconditioned filtrate and inoculum. However, the increase was smaller and the decrease greater for the invasive species. The invasive species allocated less biomass to roots when associated with harmful biota, reducing negative effects of the biota on its performance. The results show that in the native range the invasive species is more limited by self-conditioned pathogens and benefits more from unconditioned mutualists and thus may benefit more from loss of effectively specialized soil biota in a secondary range. Our study highlights the utility of detailed plant-soil feedback research in species native range for understanding factors regulating species performance in their native range and pinpointing the types of biota involved in their regulation.

The transformation of dewatered sewage sludge into vermicompost provides an advantageous solution in cases where the sludge is not too contaminated with inorganic pollutants, especially heavy metals. In addition to the conversion of the sludge to a product with a higher-added value, undesirable organic pollutants and micropollutants are partially eliminated. Anaerobically stabilized dewatered sewage sludge from a medium-sized WWTP was subjected to the vermicomposting process under field conditions. Straw was used as the bedding material in the form of two mixing ratios. The almost 1 year of the monitoring of the process focused on the hazardous substances present, the concentrations of which are regulated by legislation on the use of sludge on agricultural land. In addition, the contents of macro- and micro-nutrients such as N, P, K, Mo, Ca, Mg, and the wintering of the earthworm inocula were monitored. The potential of the vermicomposting process to reduce the content of emergent pollutants from the PPCP group was described with respect to 34 detected substances, including five endocrine disruptors. The study suggested that the bio-stabilization of dewatered sewage sludge using earthworms provides an effective technology for converting noxious wastewater treatment products into nutrient-rich bio-fertilizers.

The study of 16 alpine ecosystems in Europe highlights that temperature increases may lead to changes on plant and fungal communities resulting in fungal biomass and topsoil carbon content loss. Alpine tundra ecosystems suffer from ongoing warming-induced tree encroachment and vegetation shifts. While the effects of tree line expansion on the alpine ecosystem receive a lot of attention, there is also an urgent need for understanding the effect of climate change on shifts within alpine vegetation itself, and how these shifts will consequently affect soil microorganisms and related ecosystem characteristics such as carbon storage. For this purpose, we explored relationships between climate, soil chemistry, vegetation, and fungal communities across seven mountain ranges at 16 alpine tundra locations in Europe. Among environmental factors, our data highlighted that plant community composition had the most important influence on variation in fungal community composition when considered in combination with other factors, while climatic factors had the most important influence solely. According to our results, we suggest that rising temperature, associated with a replacement of ericoid-dominated alpine vegetation by non-mycorrhizal or arbuscular mycorrhizal herbs and grasses, will induce profound changes in fungal communities toward higher dominance of saprotrophic and arbuscular mycorrhizal fungi at the expense of fungal root endophytes. Our study highlights the utility of detailed PSF research in the native range of species for understanding the factors that regulate performance of invasive and non-invasive species in their native range, and for pinpointing the types of biota involved in their regulation and how this regulation changes across the plants life cycle.

Environmental context Endocrine disrupting compounds (EDCs) are among the most recently targeted micropollutants detected in wastewater treatment plant (WWTP) effluents and in aquatic environments. There is a need for the development of robust analytical methods for most relevant estrogenic EDCs. This study provides optimisation of analytical techniques and addresses several relevant aspects that are often overlooked in the literature. The method was finally successfully employed for the analysis of WWTP effluents. Abstract Two analytical approaches – liquid chromatography–tandem mass spectrometry (LC-MS/MS) and gas chromatography–tandem mass spectrometry (GC-MS/MS) methods – were compared for the simultaneous determination of the 19 most important oestrogenic endocrine disrupting chemicals (EDCs), such as 17β-oestradiol, oestrone, 17α-ethinyloestradiol, bisphenol A and triclosan in wastewater treatment plant effluents. To lower the instrument limits of detection (ILODs), a derivatisation step preceded detection in both methods. The stability, sensitivity and ease of use of dansylation (Dns) for LC-MS/MS and trimethylsilylation (TMS) for GC-MS/MS derivatives were evaluated before method validation. TMS derivatisation products were highly unstable over time. Parameters such as susceptibility to matrix effects and the stability of monodansylated and didansylated derivatisation products of phytohormones are discussed. Lower ILODs of highly potent EDCs (0.11 ng mL−1 for 17β-oestradiol, 0.01 ng mL−1 for 17α-ethinyloestradiol and 0.22 ng mL−1 for oestrone) and stability of derivatisation products within 7 days were achieved using LC–MS/MS; therefore, further validation of this method at environmentally relevant concentrations was conducted. The method limits of detection (MLODs) met the requirements of the European Union defined in Directive 2008/105/ES for 17α-ethinyloestradiol (0.035 ng L−1) and 17β-oestradiol (0.4 ng L−1). Twenty samples of wastewater treatment plant effluent from the Czech Republic were screened using LC-MS/MS. Fifteen of the EDCs were detected in at least one sample. The most abundant EDCs were bisphenol A, with a concentration up to 1107 ng L−1, and triclosan, with a concentration up to 76 ng L−1. No seasonal trend between late spring and autumn samples was observed in the frequency or quantity of analytes.

Secondary metabolites of cyanobacteria and algae released during algal blooms often exhibit toxic effects, but only a small number of the metabolites are the subject of routine analytical screenings. Alternatively, ecotoxicological assays offer a better representation of the overall negative effects. The aim of this work was to compare multiple assays in their sensitivity towards cellular algal organic matter (COM) of the toxin-producing cyanobacterium Microcystis aeruginosa. Multiple endpoints were investigated: mortality, growth inhibition, bioluminescence inhibition, genotoxicity, endocrine-disrupting effects, oxidative stress, and the induction of ethoxyresorufin-O-deethylase (EROD). Three rainbow trout (Oncorhynchus mykiss) cell lines as well as representatives of bacteria, yeasts, algae, vascular plants, and crustaceans were employed, and the results were expressed per mg of dissolved organic carbon (DOC) in the COM. M. aeruginosa COM was toxic to the RTgill-W1, RTG-2, and RTL-W1 cell lines (EC50 values ranging from 0.48 ± 0.02 to 1.9 ± 0.1 mgDOC/L), to the crustacean Thamnocephalus platyurus (LC50 = 20 ± 1 mgDOC/L), and to Lepidium sativum (IC50 = 241 ± 13 mgDOC/L). In contrast, no effect was observed for bacteria and yeasts, and the growth of the alga Desmodesmus subspicatus was even stimulated. No genotoxicity, endocrine-disrupting effects or increase in oxidative stress or EROD activity was detected. The content of six microcystins (MC-LR, MC-RR, MC-YR, MC-LY, MC-LW, and MC-LF), anatoxin-a, cylindrospermopsin, and nodularin in the M. aeruginosa COM was determined by liquid chromatography–tandem mass spectrometry. An artificially prepared mixture of the detected cyanotoxins in the corresponding concentrations did not induce response in the O. mykiss cell lines and T. platyurus, suggesting that other cyanobacterial metabolites are responsible for the toxicity of M. aeruginosa.

During production of γ-hexachlorocyclohexane (γ-HCH), thousands of tons of other isomers were synthesized as byproducts, and after dumping represent sources of contamination for the environment. Several microbes have the potential for aerobic and anaerobic degradation of HCHs, and zero-valent iron is an effective remediation agent for abiotic dechlorination of HCHs, whereas the combination of the processes has not yet been explored. In this study, a sequence of anoxic/oxic chemico-biological treatments for the degradation of HCHs in a real extremely contaminated soil (10-30 g/kg) was applied. Approximately 1500 kg of the soil was employed, and various combinations of reducing and oxygen-releasing chemicals were used for setting up the aerobic and anaerobic phases. The best results were obtained with mZVI/nZVI, grass cuttings, and oxygen-releasing compounds. In this case, 80 % removal of HCHs was achieved in 129 days, and 98 % degradation was achieved after 1106 days. The analysis of HCHs and their transformation products proved active degradation when slight accumulation of the transformation product during the anaerobic phase was followed by aerobic degradation. The results document that switching between aerobic and anaerobic phases, together with the addition of grass, also created suitable conditions for the biodegradation of HCHs and monochlorobenzene/benzene by microbes.

Soil microbial networks play a crucial role in plant community stability. However, we lack knowledge on the network topologies associated with stability and the pathways shaping these networks. In a 13-year mesocosm experiment, we determined links between plant community stability and soil microbial networks. We found that plant communities on soil abandoned from agricultural practices 60 years prior to the experiment promoted destabilising properties and were associated with coupled prokaryote and fungal soil networks. This coupling was mediated by strong interactions of plants and microbiota with soil resource cycling. Conversely, plant communities on natural grassland soil exhibited a high stability, which was associated with decoupled prokaryote and fungal soil networks. This decoupling was mediated by a large variety of past plant community pathways shaping especially fungal networks. We conclude that plant community stability is associated with a decoupling of prokaryote and fungal soil networks and mediated by plant-soil interactions.

After wildfires in Mediterranean ecosystems, ruderal mosses are pioneer species, stabilizing the soil surface previous to the establishment of vascular vegetation. However, little is known about the implication of pioneer moss biocrusts for the recovery and resilience of soils in early post-fire stages in semi-arid areas. Therefore, we studied the effects of the burgeoning biocrust on soil physicochemical and biochemical properties and the diversity and composition of microbial communities after a moderate-to-high wildfire severity. Seven months after the wildfire, the biocrust softened the strong impact of the fire in soils, affecting the diversity and composition of bacteria and fungi community compared to the uncrusted soils exposed to unfavourable environmental stress. Soil moisture, phosphorous, and enzyme activities representing the altered biogeochemical cycles after the fire, were the main explanatory variables for biocrust microbial community composition under the semi-arid conditions. High bacterial diversity was found in soils under mosses, while long-lasting legacies are expected in the fungal community, which showed greater sensitivity to the fire. The composition of bacterial and fungal communities at several taxonomical levels was profoundly altered by the presence of the moss biocrust, showing a rapid successional transition toward the unburned soil community. Pioneer moss biocrust play an important role improving the resilience of soil microbial communities. In the context of increasing fire intensity, studying the moss biocrust effects on the recovery of soils microbiome is essential to understanding the resistance and resilience of Mediterranean forests to wildfires.

Twenty-five samples of ivory artefacts were analysed using a radiocarbon dating method. These artefacts were part of a collection of 77 items seized during a wildlife law enforcement operation targeting online illegal ivory trade. Based on the opinion of a qualified expert, 14 of the 25 samples were declared antique, meaning their presumed origin is older than 1947. According to the European Union’s law, antique ivory is exempt from an otherwise total ban on commercial ivory activities since such trade does not contribute to the current decline in elephant population. The radiocarbon measurement has revealed alarming findings that 17 of the 25 processed samples (68%) are not antique as their origin is younger than 1947. The antique expert’s determination of the ivory age failed in 12 of 14 cases (86%).

Soils represent important pools of soil organic carbon (SOC) that can be greatly influenced by labile C inputs, which are expected to increase in future due to CO2 enrichment of atmosphere and a concomitant rise in plant primary productivity. Studying effects of variable labile C inputs on SOC pool helps to understand how soils respond to global change. However, this knowledge is missing for coniferous forest soils despite being widespread throughout the northern temperate zone. We conducted a 7-month field manipulation experiment to study the effects of variable labile C inputs (simulated by additions of C4 sucrose) on the C content in soil fractions and on microbial abundance in the organic (O), surface mineral (A), and subsoil mineral (B) horizons of a temperate coniferous forest soil. SOC in less-protected soil fractions and total organic C were substantially decreased by labile C additions that simulated future increases in C inputs. The SOC losses were comparable between the A and B horizon (40% vs. 30%). However, because sucrose availability estimated from its incorporation into soil fractions and microbial biomass sharply decreased with soil depth, the loss of C was higher in the B than in the A horizon when related to the amount of sucrose added. Utilization of sucrose was highest by fungi in the O horizon and by bacteria in the mineral soil horizons. The results indicate that future increases in labile C inputs to coniferous forest soils will cause rapid and substantial losses of SOC in both the surface and subsoil mineral horizons.

Biologically active micropollutants that contain diverse phenolic/aromatic structures are regularly present in wastewater effluents and are even found in drinking water. Advanced green technologies utilizing immobilized laccase and/or peroxidase, which target these micropollutants directly, may provide a reasonable alternative to standard treatments. Nevertheless, the use of these enzymes is associated with several issues that may prevent their application, such as the low activity of laccase at neutral and basic pH or the necessity of hydrogen peroxide addition as a co-substrate for peroxidases. In this study, the activity of laccase from Trametes versicolor and horseradish peroxidase was evaluated across a range of commonly used substrates (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), syringaldazine, and guaiacol). Moreover, conditions for their optimal performance were explored along with an assessment of whether these conditions accurately reflect the effectivity of both enzymes in the degradation of a mixture of bisphenol A, 17α-ethinylestradiol, triclosan, and diclofenac in tap drinking water and secondary wastewater effluent. Laccase and horseradish peroxidase showed optimal activity at strongly acidic pH if ABTS was used as a substrate. Correspondingly, the activities of both enzymes detected using ABTS in real waters were significantly enhanced by adding approximately 2.5% (v/v) of McIlvaine’s buffer. Degradation of a mixture of micropollutants in wastewater with 2.5% McIlvaine’s buffer (pH 7) resulted in a substantial decrease in estrogenic activity. Low degradation efficiency of micropollutants by laccase was observed in pure McIlvaine’s buffer of pH 3 and 7, compared with efficient degradation in tap water of pH 7.5 without buffer. This study clearly shows that enzyme activity needs to be evaluated on micropollutants in real waters as the assessment of optimal conditions based on commonly used substrates in pure buffer or deionized water can be misleading.

Copper oxide nanoparticles (CuO NPs) are widely used in industry. Once released, they can enter the soil system and endanger organisms living in this environment. Therefore, monitoring the NP impact on soil organisms and identification of suitable biomarkers associated with NP pollution are required. In this study, immune effector cells of the earthworm Eisenia andrei, amoebocytes, were exposed to environmentally relevant sublethal concentrations of CuO NPs (1, 10, and 100 μg mL−1 of Cu) and their impact on the cellular and subcellular levels, as well as on the mRNA levels of molecules involved in the defense reactions, was assessed in vitro. CuO NPs decreased the viability of both amoebocyte subpopulations by 40% at the highest concentration tested (100 μg mL−1 of Cu). Further, CuO NPs caused significant attenuation of the phagocytic function of hyaline amoebocytes after 6 and 24 hours of exposure, by 37 and 25%, respectively. The concentration of the lipid peroxidation subproduct, malondialdehyde, was 10 times elevated in cells exposed to CuO NPs (100 μg mL−1 of Cu) after 6 hours of exposure. We hypothesize that malondialdehyde may induce DNA breaks, cell cycle arrest, and subsequent cell death. Electron microscopy showed the interaction between CuO NPs and immune effector cells, amoebocytes. Moreover, aggregates of CuO NPs were shown to be engulfed and located in the cytoplasm of these cells. However, data from all experiments indicate that the observed effects of CuO NPs on earthworm coelomocytes were caused mainly by the dissolved Cu2+ ions derived from nanoparticles (NPs). The determination of effective parameters such as oxidative stress, immune reactivity, and genotoxicity would provide valuable comprehension and data for environmental assessment of NP impact on soil organisms.

Malting sludge is waste that could be used as a good soil conditioner after proper treatment. In the current study, the feasibility of vermicomposting malting sludge and its mixtures with straw pellets on the basis of physico-chemical and biological properties was verified. A vermicomposting system with continuous feeding of earthworms Eisenia andrei was used. The greatest number and biomass of earthworms was found in the variant with 25% malt house sludge + 75% straw pellets (on average of all layers: 320 earthworms/kg and 35 g/kg, respectively), followed by a variant with 50% malt house sludge + 50% straw pellets (on average of all layers: 47 earthworms/kg and 13 g/kg, respectively), indicating that a minimum of 50% (vol.) straw pellets is necessary for successful vermicomposting of malting sludge. Most earthworms lived in the youngest upper layer (42% and 52% of total number and earthworm biomass, respectively). On the contrary, the oldest bottom layers (final vermicomposts) after 180 days of vermicomposting were characterized by maturity, indicating lesser contents of microorganisms and enzyme activity. These vermicomposts had favorable agrochemical properties (pH = 7.8, EC = 1.2 mS cm−1, C/N = 11, Ptot = 1.23%, Ktot = 2.55%, Mgtot = 0.42%). The proportion of the available contents in the total contents were 10%, 59%, and 19% for P, K, and Mg, respectively.

The advantages of combining composting and vermicomposting to break down and remove pollutants from organic waste are reviewed. This mini-review aims to present the benefits of combining these methods and the outcome of specific cases of environmental remediation.

Both plant species and CO₂ concentration can potentially affect rhizodeposition and consequently soil microbial activity and community composition. However, the effect differs based on plant developmental stage. We focused on the effect of three plant species (forbs, grasses, and N₂‐fixers) at an early stage of development on root C deposition and fate, soil organic matter (SOM) mineralization and soil microbial community composition at ambient (aCO₂) and elevated (eCO₂) CO₂ levels. Plants were grown from seed, under continuous ¹³C‐labelling atmospheres (400 and 800 µmol mol⁻¹ CO₂), in grassland soil for three weeks. At the end of the growth period, soil respiration, dissolved organic C (DOC) and phospholipid fatty acid (PLFA) profiles were quantified and isotopically partitioned into root‐ and soil‐derived components. Root‐derived DOC (0.53 ± 0.34 and 0.26 ± 0.29 µg mL soil solution⁻¹) and soil‐derived CO₂ (6.14 ± 0.55 and 5.04 ± 0.44 µg CO₂‐C h⁻¹) were on average two times and 22% higher at eCO₂ than at aCO₂, respectively. Plant species differed in exudate production at aCO₂ (0.11 ± 0.11, 0.10 ± 0.18, and 0.58 ± 0.58 µg mL soil solution⁻¹ for Plantago, Festuca, and Lotus, respectively) but not at eCO₂ (0.20 ± 0.28, 0.66 ± 0.32, and 0.75 ± 0.15 µg mL soil solution⁻¹ for Plantago, Festuca, and Lotus, respectively). However, no differences among plant species or CO₂ levels were apparent when DOC was expressed per gram of roots. Relative abundance of PLFAs did not differ between the two CO₂ levels. A higher abundance of actinobacteria and G‐positive bacteria occurred in unplanted (8.07 ± 0.48 and 24.36 ± 1.18 mol%) and Festuca‐affected (7.63 ± 0.31 and 23.62 ± 0.69 mol%) soil than in Plantago‐ (7.04 ± 0.36 and 23.41 ± 1.13 mol%) and Lotus‐affected (7.24 ± 0.17 and 23.13 ± 0.52 mol%) soil. In conclusion, the differences in root exudate production and soil respiration are mainly caused by differences in root biomass at an early stage of development. However, plant species evidently produce root exudates of varying quality affecting associated microbial community composition.

Labile carbon (C) input to soils is expected to affect soil organic matter (SOM) decomposition and soil organic C (SOC) stocks in temperate coniferous forests. We hypothesized that the SOM decomposition rate, C content in soil fractions, and microbial and faunal abundance and activity were increased along the gradient in labile C input around wood ant nests. Three distances from the nest that differed in annual labile C input to soil were selected: 4 m with 6379 mg C m−2, 30 m with 9060 mg C m−2, and 70 m with 9215 mg C m−2. Soil from the organic horizon (Oe+Oa), surface mineral horizon (A), and subsoil mineral horizon (B) was analyzed for C content in soil fractions and for activity and abundance of soil microorganisms and fauna. In addition, a 1-year litter-bag and soil-bag decomposition experiment was conducted. Although the rate of soil decomposition did not differ along the labile C input gradient, the rate of litter decomposition in the B horizon increased as labile C input increased with distance from the nest. Correspondingly, the C content in bulk soil and in the labile and less-protected soil fractions in the B horizon decreased as labile C input increased. We infer that, because the O and A horizons are less C-limited than the B horizon, the changes in the labile C input along the gradient affected the B horizon more than the surface O and A horizons. However, soil microbial and faunal activity and abundances were not consistently affected by the gradient. Apparently, C stocks in soil fractions are more important for microbial and faunal communities than labile C inputs. Although the results indicate that SOC content changes very slowly in the coniferous forest soil of the current study, increases in the input of natural labile C leads to decreases in the SOC stock in the B horizon. By increasing the labile C input to temperate forest soils, future increases in atmospheric CO2 concentration may therefore lead to a significant loss of SOC in deep soil layers.

Nanomaterials have attracted research attention due to their unique properties, which have also made them increasingly useful for various applications, including remediation technologies. In this work, we tried to explore the aging phenomenon of several newly developed nanoscale zero-valent iron (nZVI) particles containing 0.25, 1, and 5% sulfur and to monitor the possible toxicity changes related to the nZVI particles before and during the aging period. We used several relevant techniques for monitoring nZVI aging, including transmission electron microscopy, X-ray powder diffraction, and particle size analysis. The toxicity of the nZVI particles was monitored using a new oxidative stress assay based on lipid peroxidation, a simple bacterial cultivation test. A test using activated sludge respiration and phospholipid fatty acid analysis of microbial populations was also employed. The results of the material analyses showed that the sulfidated nZVI particles were more stable than bare nZVI particles in terms of Fe0 content and particle diameter. The stability was more pronounced with the increasing content of sulfur in the nanoparticles; the content of α-Fe was only 35.3% in the nZVI particles compared with 78.6% in the 5% sulfur-doped nanoparticles after 60 days of aging. The results of the oxidative stress assay revealed that the toxicity was also lower with increasing sulfur content; nevertheless, the oxidative stress marker increased substantially after 7 days of aging, reaching a similar level to that caused by bare nZVI in some cases. Other results showed that even incubation in media used for the respective tests caused extensive aging; therefore, this information must be considered before the selection of toxicity assays for nanomaterial evaluation.

The formation of soil aggregates is related to the stability of soil organic carbon, which distinguishes the living conditions of microbial communities in different micro-niches. Here, we investigated fungal and bacterial community structure, networks, biomass and respiration in individual micro-niches between and within soil aggregates using seven different chronosequences (both primary and secondary successions from pioneer stages to well-developed ecosystems) on a European-scale gradient from a maritime climate in Belgium to a more continental climate in Hungary. We showed that geographical location was the most important factor (74% for bacterial community, 53% for fungal community) affecting microbial community composition, while soil aggregates (5% for bacterial community and 12% for fungal community) were the second most important factor followed by age of succession and vegetation. Moreover, aggregates (52%) were the most important factor influencing the biomass of major microbial groups, followed by location (37%) and age of succession (6%). Interestingly, macro-aggregates had less interconnected microbial networks and low microbial respiration rates compared to other aggregate sizes. We therefore hypothesize that the embedding of organic particles in the mineral matrix of macro-aggregates is a crucial point in microbial community organization, as evidenced by microbial community connectivity and microbial respiration. Our results demonstrate the importance of soil aggregates for microbial community organization and soil carbon stabilization.

The RTgill-W1 (gill), RTG-2 (gonad), and RTL-W1 (liver) cell lines derived from a freshwater fish rainbow trout (Oncorhynchus mykiss), were used to assess the toxicity of polyethylene terephthalate (PET) and two forms of polyvinyl chloride (PVC). Two size fractions (25-μm and 90-μm particles) were tested for all materials. The highest tested concentration was 1 mg/ml, corresponding to from 70 000 ± 9000 to 620 000 ± 57 000 particles/ml for 25-μm particles and from 2300 ± 100 to 11 000 ± 1000 particles/ml for 90-μm particles (depending on the material). Toxicity differences between commercial PVC dry blend powder and secondary microplastics created from a processed PVC were newly described. After a 24-h exposure, the cells were analyzed for changes in viability, 7-ethoxyresorufin-O-deethylase (EROD) activity, and reactive oxygen species (ROS) generation. In addition to the microplastic suspensions, leachates and particles remaining after leaching resuspended in fresh exposure medium were tested. The particles were subjected to leaching for 1, 8, and 15 days. The PVC dry blend (25 μm and 90 μm) and processed PVC (25 μm) increased ROS generation, to which leached chemicals appeared to be the major contributor. PVC dry blend caused substantially higher ROS induction than processed PVC, showing that the former is not suitable for toxicity testing, as it can produce different results from those of secondary PVC. The 90-μm PVC dry blend increased ROS generation only after prolonged leaching. PET did not induce any changes in ROS generation, and none of the tested polymers had any effect on viability or EROD activity. The importance of choosing realistic extraction procedures for microplastic toxicity experiments was emphasized. Conducting long-term experiments is crucial to detect possible environmentally relevant effects. In conclusion, the tested materials showed no acute toxicity to the cell lines.

A better understanding of factors controlling the distribution of soil organic matter (SOM) in differently stabi-lized pools in soils during ecosystem development is essential to more efficiently manage soil as a C sink and aid global warming mitigation. We, thus, investigated C and N stocks in bulk soil and differently stabilized soil fractions (free particulate organic matter (fPOM), micro-and macroaggregates, and mineral-associated organic matter (MAOM)), in seven chronosequences comprising primary and secondary successions at various locations in Europe and tested the influence of multiple environmental parameters on these C and N stocks.Best-subset multiple regression indicated that plant litter production and succession age were of minor importance. Above a certain threshold of organic matter inputs to the soil, changes in the quantity of such inputs may have little direct effect on the partitioning of organic matter to various soil fractions. Instead, bio-physicochemical soil properties (such as pH and PLFA and clay contents) were the best predictors for both C and N stocks. We also found a shift in the relevance of biotic and abiotic factors from fPOM and the various aggregate fractions to MAOM, the latter being exclusively associated with pH and the clay content. This indicates that sites dominated by MAOM likely more slowly react to management interventions, such as increasing or diversifying organic matter inputs, than sites dominated by fractions more responsive to biotic factors, such as fPOM. Finally, as compared to primary successions, secondary successions were associated with higher C and N stocks in aggregate fractions, pointing towards better conditions for soil structure formation in the latter. Our results highlight the need to elucidate the form in which C and N exist in the soil for selection of proper management strategies and that soil biophysicochemical parameters may have a disparately larger impact on the partitioning of C and N to variously stabilized soil fractions than the rate of organic matter inputs.

In this study, the diversity of bphA genes was assessed in a 13C-enriched metagenome upon stable isotope probing (SIP) of microbial populations in legacy PCB-contaminated soil with 13C-biphenyl (BP). In total, 13 bphA sequence variants (SVs) were identified in the final amplicon dataset. Of these, one SV comprised 59% of all sequences, and when it was translated into a protein sequence, it exhibited 87, 77.4, and 76.7% identity to its homologs from Pseudomonas furukawaii KF707, Cupriavidus sp. WS, and Pseudomonas alcaliphila B-367, respectively. This same BphA sequence also contained unusual amino acid residues, Alanine, Valine, and Serine in region III, which had been reported to be crucial for the substrate specificity of the corresponding biphenyl dioxygenase (BPDO), and was accordingly designated BphA_AVS. The DNA locus of 18 kbp containing the BphA_AVS-coding sequence retrieved from the metagenome was comprised of 16 ORFs and was most likely borne by Paraburkholderia sp. The BPDO corresponding to bphAE_AVS was cloned and heterologously expressed in E. coli, and its substrate specificity toward PCBs and a spectrum of flavonoids was assessed. Although depleting a rather narrow spectrum of PCB congeners, the efficient transformation of flavone and flavanone was demonstrated through dihydroxylation of the B-ring of the molecules. The homology-based functional assignment of the putative proteins encoded by the rest of ORFs in the AVS region suggests their potential involvement in the transformation of aromatic compounds, such as flavonoids. In conclusion, this study contributes to the body of information on the involvement of soil-borne BPDOs in the metabolism of flavonoid compounds, and our paper provides a more advanced context for understanding the interactions between plants, microbes and anthropogenic compounds in the soil.

Purpose: To compare plant-soil feedback (PSF) of invasive Cirsium vulgare and non-invasive C. oleraceum in their native range to test a hypothesis that the invasive species is more limited by specialized pathogens in the native range and/or able to benefit more from generalist mutualists, and thus may benefit more from loss of specialized soil biota in a secondary range.Methods: We assessed changes in soil nutrients and biota following soil conditioning by each species and compared performance of plants grown in self-conditioned and control soil, from which all, some or no biota was excluded. Results: The invasive species depleted more nutrients than the non-invasive species and coped better with altered nutrient levels. The invasive species had higher seedling emergence which benefited from presence of non-specific microbes. The invasive species biomass responded less positively to specialized (self-conditioned) microbiota and more negatively to specialized larger-sized biota compared to the non-specialized control biota, suggesting the species may benefit more from enemy release and suffer less from loss of specialized mutualists when introduced to a secondary range. The invasive species showed greater ability to decrease its root-shoot ratio in presence of harmful biota and thus reduce their negative effects on its performance.Conclusions: Our study highlights the utility of detailed PSF research in the native range of species for understanding the factors that regulate performance of invasive and non-invasive species in their native range, and for pinpointing the types of biota involved in their regulation and how this changes across the plants life cycle.

Despite being drastically restricted in the 1970s, polychlorinated biphenyls (PCBs) still belong among the most hazardous contaminants. The chemical stability and dielectric properties of PCBs made them suitable for a number of applications, which then lead to their ubiquitous presence in the environment. PCBs are highly bioaccumulative and persistent, and their teratogenic, carcinogenic, and endocrine-disrupting features have been widely reported in the literature. This review discusses recent advances in different techniques and approaches to remediate historically contaminated matrices, which are one of the most problematic in regard to decontamination feasibility and efficiency. The current knowledge published in the literature shows that PCBs are not sufficiently removed from the environment by natural processes, and thus, the suitability of some approaches (e.g., natural attenuation) is limited. Physicochemical processes are still the most effective; however, their extensive use is constrained by their high cost and often their destructiveness toward the matrices. Despite their limited reliability, biological methods and their application in combinations with other techniques could be promising. The literature reviewed in this paper documents that a combination of techniques differing in their principles should be a future research direction. Other aspects discussed in this work include the incompleteness of some studies. More attention should be given to the evaluation of toxicity during these processes, particularly in terms of monitoring different modes of toxic action. In addition, decomposition mechanisms and products need to be sufficiently clarified before combined, tailor-made approaches can be employed.

Mixture toxicity, including agonistic and antagonistic effects, is an unrevealed environmental problem. Estrogenic endocrine disruptors are known to cause adverse effects for aquatic biota, but causative chemicals and their contributions to the total activity in sewage sludge remain unknown. Therefore, advanced analytical methods, a yeast bioassay and mixture toxicity models were concurrently applied for the characterization of 8 selected sludges with delectable estrogenic activity (and 3 sludges with no activity as blanks) out of 25 samples from wastewater treatment plants (WWTPs). The first applied full logistic model adequately explained total activity by considering the concentrations of the monitored compounds. The results showed that the activity was primarily caused by natural estrogens in municipal WWTP sludge. Nevertheless, activity in a sample originating from a car-wash facility was dominantly caused by partial agonists - nonylphenols - and only a model enabling prediction of all dose-response curve parameters of the final mixture curve explained these results. Antiestrogenic effects were negligible, and effect-directed analysis identified the causative chemicals.

A phenotype is the expression of interactions between species genotype and environment. We quantified the contributions of ecological and phylogenetic associations to phenotypic variation in Geosmithia fungi. Geosmithia are symbiotic beetle-associated saprotrophs with a range of life histories and host specificities, including obligate nutritional beetle mutualists (ambrosia fungi) and phytopathogens. We hypothesized that: (1) species phenotypes are better explained by their ecology than by their phylogenetic relationships; (2) niche specialization was accompanied by enzymatic capability losses; and (3) ambrosia Geosmithia species have higher nutritional quality and antibiotic capabilities than species with facultative symbioses. Our results confirmed that long-term co-evolved specialists have reduced metabolic breadth in comparison to generalists. Phytopathogenic G. morbida produces unique enzyme suites with affinity to ligno-cellulose. Mycelia of ambrosia fungi contain large amounts of oleic fatty acid with nutritive and possibly allelopathic function. Overall, our results indicate that Geosmithia ecology have greater effect on species phenotype than their phylogenetic relationships.

The creamy white to beige, aerobic, non-motile, ovoid to rod-shaped, Gram-stain-negative strain, Cd-10(T), was isolated from heavy-metal-contaminated sludge from a decantation basin of a heavy metal processing factory based on its ability to tolerate CdCl2 in the cultivation medium. In the reconstruction of its phylogeny based on 16S rRNA gene sequences, strain Cd-10(T) clustered with species of the genera Gemmobacter, Xinfangfangia, Tabrizicola and Rhodobacter within the family Rhodobacteraceae. Its 16S rRNA gene sequence exhibited 96.32% pairwise similarity to the type strain of Xinfangfangia soli, 95.3 % to that of Gemmobacter intermedius, followed by Tabrizicola fusiformis (95.10 %), Rhodobacter sediminis (94.88 %), Gemmobacter nectariphilus and Rhodobacter capsulatus (both 94.81%). The major respiratory quinone was Q-10 accompanied by Q-9, the fatty acid profile consisted predominantly of C-18:1 omega 7C, C-18:0, C-16:0 and C-16:1 omega 7c, the major polar lipids were phosphatidylglycerol, phosphatidylmethylethanolamine, phosphatidylcholine and diphosphatidylglycerol. An analysis of the percentage of conserved proteins deduced from draft or complete genomic sequences of strain Cd-10(T) and representatives of its closest relatives suggested that strain Cd-10(T) is a member of a novel genus within the Rhodobacteraceae family for which we propose the name Pseudogemmobacter. Strain Cd-10(T) (=DSM 103618(T)=NCCB 100645(T)) is the type strain of Pseudogemmobacter bohemicus gen. nov., sp. nov., the type species of the genus Pseudogemmobacter gen. nov.

The removal of algal organic matter (AOM) is a growing concern for the water treatment industry worldwide. The current study investigates coagulation of non-proteinaceous AOM (AOM after protein separation), which has been minimally explored compared with proteinaceous fractions. Jar tests with either aluminum sulphate (alum) or polyaluminium chloride (PACl) were performed at doses of 0.2-3.0 mg Al per 1 mg of dissolved organic carbon in the pH range 3.0-10.5. Additionally, non-proteinaceous matter was characterized in terms of charge, molecular weight and carbohydrate content to assess the treatability of its different fractions. Results showed that only up to 25% of non-proteinaceous AOM can be removed by coagulation under optimized conditions. The optimal coagulation pH (6.6-8.0 for alum and 7.5-9.0 for PACl) and low surface charge of the removed fraction indicated that the prevailing coagulation mechanism was adsorption of non-proteinaceous matter onto aluminum hydroxide precipitates. The lowest residual Al concentrations were achieved in very narrow pH ranges, especially in the case of PACl. High-molecular weight saccharide-like organics were amenable to coagulation compared to low-molecular weight (3 kDa) substances. Their high content in non-proteinaceous matter (about 67%) was the reason for its low removal. Comparison with our previous studies implies that proteinaceous and non-proteinaceous matter is coagulated under different conditions due to the employment of diverse coagulation mechanisms. The study suggests that further research should focus on the removal of low-molecular weight AOM, reluctant to coagulate, with other treatment processes to minimize its detrimental effect on water safety.

Soil microbial communities involved in 13C-labeled cellulose utilization from two contrasting (in terms of vegetation and climatic conditions) forest sites at different elevations were studied by using a laboratory microcosm experiment and a PLFA (phospholipid fatty acid)-SIP (stable isotope probing) approach considering the effects of incubation temperature (0, 10 and 20 °C) and time (1–4 weeks). The amounts of cellulose-13C assimilated by soil microorganisms and the composition of microbial communities utilizing cellulose were site-dependent. Total, bacterial and actinobacterial biomass incorporated higher amounts of 13C in soil from the deciduous forest site (at the lower elevation), while fungal biomass contained increased amounts of 13C in soil from the coniferous forest site (at the higher elevation). Increasing temperatures resulted in increased amounts of 13C assimilated by the different PLFA-based microbial groups, except fungi, and in significant changes in the community structure of cellulose utilizers. Fungi were better adapted to cold conditions than bacteria. Longer incubation times determined an increase in the amount of 13C incorporated into total and bacterial PLFAs but had no effect on the composition of labeled microbial communities.

Factors shaping community structure of soil microbiota have been intensively studied; however, the pattern in community composition and structure of soil microbiota at large geographical scales and factors regulating its metabolic activity remains poorly understood. Here, we used a field transplantation experiments to investigate the effects of substrate and climatic conditions on basal soil respiration, microbial biomass C and diversity of soil microbiota by comparing local and transplanted soils along a latitudinal gradient. Soil samples collected in April 2008 at donor site (Sokolov, Czech Republic) in Central Europe were gamma-ray sterilized and transplanted to receptor sites in Europe and the USA in May and early June 2008. Soil samples were taken in June 2009 after one year of exposure and immediately prepared for laboratory analysis. Basal soil respiration in local soils increased from 22 to 42 mg CO2-C kg−1 h−1 with latitude while basal soil respiration in transplanted soils decreased with latitude from 32 to 19 mg CO2-C kg−1 h−1. The microbial biomass C in both transplanted and local soils decreased with latitude. Content of fungal and bacterial phospholipid fatty acids increased nearly twice with latitude in local soils. Shannon diversity index of fungal community decreased from 2.5 to 1.2 along the latitudinal gradient in transplanted soils while local soils increased from 0.9 to 2.4 with latitude. Based on our results, microbial activity is driven mainly by changes of the soil substrate along latitudinal and climatic gradients while microbial biomass is driven more by global climatic factors itself. The diversity of soil microbial communities is mostly affected by latitudinal and climatic factors while community structure is mostly shaped by substrate quality.
