Bu tez çalışmasında, 9 adet 3-alkil(aril)-4-amino-4,5-dihidro-1H-1,2,4-triazol-5-on bileşiği elde edilerek 4-(2-nitrobenzensulfoniloksi)benzaldehit, 4-(4-nitrobenzensulfoniloksi)benzaldehit ve 4-(4-bromobenzensulfoniloksi)benzaldehit ile reaksiyonları incelenmiş ve 27 adet yeni 4,5-dihidro-1H-1,2,4-triazol-5-on türevi elde edilmiştir. Sentezlenen bileşiklerden 18 tanesi formaldehit ve morfolinle reaksiyona sokularak 18 yeni N-Mannich bazı türevleri elde edilmiştir. Çalışma kapsamında sentezlenen yeni bileşiklerin yapıları IR, 1H-NMR, 13C-NMR ve kütle spektroskopisi kullanılarak aydınlatılmıştır. Sentezlenen ve yapıları aydınlatılan yeni bileşiklerin üç farklı yöntemle (total indirgeme gücü, serbest radikal giderme aktivitesi ve metal şelat aktivitesi) antioksidan aktiviteleri incelenmiş, grafikleri çizilmiş ve sonuçlar yorumlanmıştır.Yeni bileşiklerden yapısında N-H grubu bulunan 27 tanesinin izopropil alkol, tert-butil alkol, N,N-dimetilformamid ve aseton susuz çözücülerinde tetrabutilamonyum hidroksit ile potansiyometrik olarak titrasyonları yapılarak yarı nötralizasyon metodu ile yarı nötralizasyon potansiyelleri ve karşın olan pKa değerleri tayin edilmiştir. Bu bileşiklerin asitliği üzerine çözücü etkisi ve molekül yapısının etkisi tartışılmıştır.Son olarak sentezlenen yeni bileşiklerin antimikrobiyal aktiviteleri Agar Kuyucuk Difüzyon yöntemi kullanılarak Bacillus subtilis, Bacillus cereus, Klebsiella pneumoniae, Staphylococcus aureus, Pseudomonas aeruginosa ve Escherichia coli bakterilerine karşı incelenmiştir. In this study, nine 3-alkyl(aryl)-4-amino-4,5-dihydro-1H-1,2,4-triazol-5-ones were obtained. From the reactions of these compounds with 4-(2-nitrobenzenesulfonyloxy)benzaldehyde, 4-(4-nitrobenzenesulfonyloxy)benzaldehyde and 4-(4-bromobenzenesulfonyloxy) benzaldehyde 27 new 4,5-dihydro-1H-1,2,4-triazol-5-one derivatives were synthesized. These synthesized compounds were reacted with formaldehyde and morpholine to yield 18 new N-Mannich bases. The structures of the new compounds synthesized in the study were characterized by using IR, 1H-NMR, 13C-NMR and mass spectroscopy.New compounds synthesized were analyzed for their antioxidant activities in three different methods (reducing power, free radical scavenging and metal chelating activity). Their graphs were drawn and their results were interpreted.27 new compounds which contained N-H group were titrated potentiometrically with tetrabutylammonium hydroxide in non-aqueous solvents (isopropyl alcohol, tert-butyl alcohol, N,N-dimethylformamide and acetone) because of the weak acidic properties of 4,5-dihydro-1H-1,2,4-triazol-5-one ring. The pKa values were determined by the half-neutralization method for all cases. The effects of solvents and molecular structure upon acidity were investigated and the results were discussed.Finally, the antimicrobial activities of the new compounds synthesized were evaluated against to Bacillus subtilis, Bacillus cereus, Klebsiella pneumoniae, Staphylococcus aureus, Pseudomonas aeruginosa ve Escherichia coli by using Agar Well Diffusion method. 400

İlk bölüm yeni bileşiklerin sentezini kapsamaktadır. Bu tezde, 3-alkil(aril)-4-(4-metoksi-3-sinnamoiloksibenzilidenamino)-4,5-dihidro-1H-1,2,4-triazol-5-on bileşikleri, 3-alkil(aril)-4-amino-4,5-dihidro-1H-1,2,4-triazol-5-on bileşlikleri ile 4-metoksi-3-hidroksi benzaldehidin reaksiyonundan elde edilmiştir.Çalışmanın ikinci bölümünde sentezlenen 9 yeni bileşiğin yapıları IR, 1H-NMR ve 13C-NMR spektroskopik yöntemleri ile mikroanaliz kullanılarak aydınlatılmıştır. We aimed a novel electrochemical sensor based on iron nanoparticles (FeNPs) and 2-aminoethanethiol (2-AET) functionalized graphene oxide (2-AETGO). The methods such as transmission electron microscope (TEM), scanning electron microscope (SEM), x-ray photoelectron spectroscopy (XPS), reflection-absorption infrared spectroscopy (RAIRS), electrochemical impedance spectroscopy (EIS) and the x-ray diffraction (XRD) method were used for the characterizations of nanocomposites. The linearity and the detection limit of quercetin (QR) were 1.0×10-8 - 1.0×10-7 M and 2.0×10-9 M, respectively. The modified glassy carbon electrode (FeNPs/2-AETGO/GCE) was applied to the determination of QR in food sample such as apple juice. © 2015 The Authors.
