Background: Single-cell RNA sequencing techniques have emerged as effective approaches for finding the heterogeneity between cells and discovering the differentiation stage. Adaptive total variation graph regularized nonnegative matrix factorization (ATV-NMF) has been proposed to capture the inner geometric structure and determine whether to retain feature details or denoise, which is suitable for analyzing single-cell data. However, the rank of matrix factorization significantly affects clustering performance greatly, and it is still challenging to determine the optimal rank. Objective: To solve the problem, in this paper, we propose an ensemble clustering method ANMFCE to integrate several base clustering results corresponding to different parameter rank values. Methods: Firstly, we use the ATV-NMF algorithm to obtain clustering results with different dimension reduction ranks. Secondly, the consensus function based on connected-triple-based similarity is applied to obtain the similarity matrix. Finally, the spectral clustering method is used to find the final optimal partition. Results: Clustering results on six single-cell sequencing datasets show that our method is more advanced than the individual ATV-NMF method and other comparison methods, which can illustrate that our method is effective in finding the heterogeneity in single-cell datasets. Moreover, the identification of gene markers also achieves accurate results. Conclusion: In summary, our method is effective for analyzing single-cell RNA sequencing datasets.

Kur’ân-ı Kerîm sûreleri ve âyetleri arasında dikkat çekici bir insicam ve münâsebât söz konusudur. Kur’ân’ın iç yapısında görülen bu düzen, onun ilahî oluşuyla yakından alakalıdır. Bu bağlamda Münâsebâtü’l-Kur’ân isminde bir Kur’an ilmi ortaya çıkmıştır. Kur’an’ın bu yönüne dikkat çeken birçok müfessir de münâsebâta dair eserler kaleme almışlardır. Kur’ân-ı Kerîm’in mesajını doğru anlamamıza katkı sağlayacak önemli hususlardan biri de âyet ve sûreler arasındaki münâsebâtı tespit etmektir. Bizler bu çalışmada tefsirinde münâsebâtü’l-Kur’ân’a genişçe yer veren Elmalılı’nın eserini bu yönüyle ele alıp inceledik. Çalışmamızda Elmalılı’nın hem münâsebâtü’l-Kur’ân’a dair görüşlerini hem de Bakara sûresi özelinde âyet ve sûreler arasındaki münâsebâta dair tespitlerini ortaya koyduk. Çalışma bir yandan Kur’ân’ın bir i’câz yönünü ortaya koyarken bir yandan da Kur’ân’ın anlaşılmasına mütevazi bir katkı sağlamayı hedeflemektedir., There is a remarkable compatibility and relationship between the surahs and verses of the Quran. This order seen in the inner structure of the Quran is closely related to its divine being. In this context, a science of the Quran called munasabatu'l-Qur'an has emerged. Many commentators who draw attention to this aspect of the Quran have also written works on relations. One of the most important issues that will contribute to our correct understanding of the message of the Quran is to determine the relationship between the verses and the surahs. In this study, we have examined Elmalili's work, which gives extensive place to munasabatu'l-Quran in his commentary, from this aspect. In our study, we have revealed both Elmalili's views on the relationship between the Quran and the relationship between the verses and the surahs specially in the Surah Baqara. While the study reveals an i’jaz aspect of the Quran, it also aims to make a modest contribution to the understanding of the Quran.Keywords: Quran, Commentary, munasabatu'l-Quran, Elmalili, Surah al-Baqara.

Amaç: Türkiye’de bağ alanlarında Trichothecium roseum’un ilk kez teşhis edilmesi.Yöntem ve Bulgular: 2020 yaz ayları boyunca, hem bahçede hem de hasat edilen üzümler üzerinde beyazdan açık pembe renge kadar değişen renklenmeler gözlemlenmiştir. Üzümler üzerinde külleme belirtilerini andıran bu lekelenmelerin genellikle Botrytis cinerea’nın neden olduğu kurşuni küfle birlikte bulunduğu tespit edilmiştir. Hastalığın nedenini tespit etmek amacıyla Hassa ilçesine bağlı üzüm yetiştirilen alanlardan hastalıklı asma bitkilerine ait organlar toplanarak laboratuvara getirilmiştir. İzolasyonda iki farklı yöntem kullanılmış ve ayrıca ITS rDNA gene bölgeleri ITS1 ve ITS4 primer çifti ile çoğaltılarak sekans analizi yapılmıştır. Spor süspansiyonu yaprak, meyve, sürgün gibi bağ organlarına püskürtülerek patojenisite gerçekleştirilmiştir. Patojenisite testinden 7 gün sonra oluşan belirtiler bahçe şartlarında oluşanlarla aynı olduğu gözlemlenmiştir. ITS1/4 çifti ile çoğaltılan gen bölgesinden 700 bazlık bir dizi üretilmiş ve blast analizi gerçekleştirilerek Trichothecium roseum izolatına %99 oranında benzer bulunmuştur. Morfolojik ve moleküler özelliklerine göre, elde edilen fungus Trichothecium roseum olarak teşhis edilmiş ve bu fungus Türkiye’de bağlar üzerinde ilk kez bu çalışma ile tespit edilmiştir.Genel Yorum: Türkiye’de bağ alanlarında Trichothecium roseum bulunmakta ve Hassa ilçesine ait yetiştiricilik alanlarındaki Trichothecium roseum’un yoğunluğu ya da yaygınlığı bakımından bir farklılık bulunmamaktadır.Çalışmanın Önemi ve Etkisi: Trichothecium roseum Türkiye’de bağlar üzerinde ilk kez bu çalışma ile tespit edilmiştir. Bu fungus ekonomik olarak önemli kayıplar meydana getirmemekte ancak ürettiği mikotoksinler sebebiyle hem ekonomik hem de sağlık sorunlarına yol açmaktadır., Aims: Identification of Trichothecium roseum infecting grapes first time in Turkey.Methods and Results: During the summer of 2020, white and pale pink stains were observed on grapes in the vineyards and on the harvested grapes in storages. These stains resembling powdery mildew on the grapes were observed on the clusters, mostly developing secondarily to gray rot caused by Botrytis cinerea. To determine of the causal agent of disease symptoms, some symptomatic clusters were collected both from the vineyards and storages located in Hassa district of Hatay province in Turkey. Two different methods were used to isolate the fungus from the symptomatic tissues. For molecular identification, ITS rDNA gene sequence were amplified using ITS1 and ITS4 primers. Spore suspension of the fungus was sprayed on detached vine organs such as leaf, bud, stem and cluster, and after 7 days diseases symptoms were observed resembling those in the orchard. ITS1/4 primer pairs produced about 700 bp band on the agarose gel. Based on the morphologic and molecular characteristics, the fungus was identified as Trichothecium roseum and it has been identified for the first time on grapes in Turkey with this study.Conclusions: Trichothecium roseum is present on grapes in Turkey and there is no difference in the incidence or prevalence of Trichothecium roseum in the vineyards in the cultivation areas of Hassa district of Turkey.Significance and Impact of the Study: Trichothecium roseum was detected for the first time on vineyards in Turkey with this study. This fungus does not cause significant economic damage directly, but it causes both economic and health problems due to the mycotoxins which are produced by the fungus.

In the twenty three year period, the Qur'an was subordinated to build a society of individuals with belief in tawhid. In this process, each revelation came gradually and played an important role in the realization of the target in the questions. For this reason, each revelation’s passage, which is different at different time and place, and which deals with different audiences on different topics, or in every aspect, has a meaning and integrity in many ways. In this context, the data of the science of Munasebatu’l-Qur'an can be evaluated as a proof and an example of this situation. In research, we tried to understand and reveal of this issue in short terms. As the result, seems that each of the suras from the period of Asr to Nas has complete meaning of integrity with a period before and after, Although they were subjected to different periods in the Mushaf orders. This situation, on the one hand, can be seen as a proof that the order of Mushaf is withheld But, if the arrangement of suras is jurisprudence, we can talk about an integrity because using the Quran is good for the society and it has come from a single source and to achieve a single goal. The meaning and the unity of content among the suras can be mentioned as an expression of this situation., Kur’an, yirmi üç yıllık bir zaman diliminde, tevhid inancına sahip bireylerin oluşturduğu bir toplum inşa etmek üzere nazil olmuştur. Bu süreçte peyderpey (tedricen) gelen her bir vahyin, söz konusu hedefin gerçekleşmesinde önemli bir rolü olmuştur. Bu sebeple farklı zaman ve mekânlarda nazil olan, farklı konularda farklı kitleleri muhatap alan her bir vahiy pasajı ya da her bir sure bir diğeri ile birçok yündKur’ân, yirmi üç yıllık bir zaman diliminde, tevhid inancına sahip birey-lerin oluşturduğu bir toplum inşa etmek üzere nazil olmuştur. Bu süreçte peyderpey (tedricen) gelen her bir vahyin, söz konusu hedefin gerçekleşme-sinde önemli bir rolü olmuştur. Bu sebeple farklı zaman ve mekânlarda nazil olan, farklı konularda farklı kitleleri muhatap alan her bir vahiy pasajı ya da her bir sûre bir diğeri ile birçok yönden bir anlam münâsebeti, bütünlüğü içerisindedir. Bu bağlamda gelişen Münâsebetü’l-Kur’ân ilminin verileri de bu durumun bir ispatı ve örneği olarak değerlendirilebilir. Araştırmada bu husus kısa sûreler üzerinden anlaşılmaya ve ortaya konulmaya çalışıldı. Sonuçta, Mushaf tertibinde farklı dönemlerde nazil olmalarına rağmen Asr sûresinden Nas sûresine kadarki sûrelerden her birinin öncesindeki ve sonrasındaki sûreyle tam bir anlam bütünlüğüne sahip olduğu görülmektedir. Bu durum, bir yandan Mushaf’ın tertibinin tevkifî olduğunun bir delili olarak görülebilir. Fakat sûrelerin tertibinin ictihadî olması halinde de böyle bir anlam bütünlüğünden bahsedebiliriz. Zira Kur’ân, bütünüyle tek bir kaynaktan ve tek bir hedefi gerçekleştirmek üzere gelmiştir. Sûreler arasındaki anlam münâsebeti ve muhteva birliği de, bu durumun bir ifadesi olarak zikredilebilir. en bir anlam münasebeti, bütünlüğü içerisindedir. Bu bağlamda gelişen Münasebetü’l-Kur’an ilminin verileri de bu durumun bir ispatı ve örneği olarak değerlendirilebilir.
