24. A cDNA encoding feline CD4 has a unique repeat sequence downstream of the V-like region

Abstract

We have cloned, sequenced and expressed a cDNA encoding the feline CD4 glycoprotein. This clone, termed FT121, contains a unique repeat sequence downstream of the V-like region which has not been reported in other mammalian CD4 cDNA. In addition, the cysteine residues at positions 45 and 175 have an unusual pattern. On the other hand, regions other than the unique sequence share significant homology with human CD4 and mouse L3T4 cDNA in the amino acid sequence deduced from the nucleotide sequence, especially in the putative cytoplasmic region. Here, we show that FT121 has a complete open reading frame and efficiently expresses feline CD4 molecules on COS cells. Further we found that the cytoplasmic components, lymphocyte-specific protein-tyrosine kinase p56lck binding sites and phosphorylation site (serine resides), are well conserved, though the extracellular region of the feline CD4 molecule deduced from FT121 is probably different from that of human CD4 and mouse L3T4.

Abstract

Nine isolates ofCanine parvovirus(CPV) were obtained from Vietnamese dogs and cats. One canine isolate showed a unique antigenic property which indicates a novel antigenic variant of CPV-2b when examined with hemagglutination inhibition tests using our monoclonal antibodies, 21C3 and 19D7, which were recently developed. This isolate had an amino acid substitution of residue 426, Asp to Glu, and the same substitution has recently been found in CPV from Italian dogs. This study first showed that such substitution caused an antigenic difference demonstrable by monoclonal antibodies and that a similar evolution may have occurred in CPV in Vietnam. Abstract

The gag gene of a Japanese feline immunodeficiency virus (FIV) isolate, designated as FIV TM 2, was expressed in Escherichia coli as a fusion protein with TrpE. Using this expressed protein, an enzyme-linked immunosorbent assay was developed for detection of antibodies to FIV gag protein in feline sera. With serum samples from a cat experimentally infected with FIV, it was demonstrated that the period of seroconversion detected by this method corresponded to that by Western blotting. Abstract

We have cloned, sequenced and expressed a cDNA encoding the α-chain of feline CD8. This clone, named FT8-10, has an open reading frame with 720 nucleotides in length encoding a protein with 239 amino acid residues. Sequence analysis has revealed that the feline CD8 α-chain (CD8α) shares significant homology with human (T8/Leu-2), bovine (BoCD8), rat (MRC OX8) and mouse (Lyt-2) CD8α subunits. Cysteine residues as well as the tyrosine kinase p56kk binding site are well conserved. Besides, no putative N-linked glycosylation site was found. Interestingly, immunofluorescence analysis of COS-7 cells transfected with feline CD8α expression plasmid driven by SRα promoter showed that the expressed feline CD8α cross-reacted with an anti-human CD8α monoclonal antibody OKT8, but did not react with an anti-feline monoclonal antibody, FT-2, which is thought to recognize the feline analogue of the human T8/Leu-2 and murine Lyt-2 molecules expressed on cytotoxic/suppressor T cells. The in vivo pathogenicity of canine parvovirus (CPV) type 2c (strain V203) and of CPV type 2a (strain V154) against cats was investigated. Our results indicate that both types of CPV have the potential to induce disease in cats.
