9. Agrobacterium Mediated Transformation of Fld and GUS Genes into Canola for Salinity Stress

Abstract

Salinity is one of the major abiotic stress which limits wide spread canola cultivation. One way to overcome this problem could be transfection, to produce tolerable species. Cotyledonary and hypocotyls explants obtained from 4 and 7 days old seedling of Elite and RJS003 varieties were utilized in this study. Genetic transformation was implemented through Agrobacterium tumefaciens LBA4404 containing PBI121 plasmid and Agrobacterium tumefaciens C58, LBA4404, AGL0 and EHA 101 strains which contain P6u- ubi- fvt1 construct. The T-DNA region of P6u- Ubi- Fvt1 plasmid included HPT (Hygromycin phosphotransferase) plant selectable marker and Fld (flavodoxin) gene. PBI121 plasmid had NptII (Neomycin phosphotransferase) plant Selectable marker and β-glucuronidase (GUS) reporter genes. Transfected explants were analyzed by PCR and histochemical assay for Fld and Gus genes, respectively. Our data indicated that the cotyledonary explants of both cultivars were incompetent to be infected with Fld gens. However, the transformation in Elite hypocotyls explants with Agrobacterium tumefaciens C58 and LBA 4404 strains were confirmed through PCR product and histochemical evaluation for Fld and GUS genes, respectively. Therefore, the result of this manuscript may to certain degree fulfill the endeavor appointed to this oilseed.

22. Effect of Activated Charcoal on in Vitro Propagation of Lythrum salicaria L. (Lythraceae).

Abstract

Lythrum salicaria L., commonly referred to as Purple Loosestrife, is a medicinal plant that has been valued for its therapeutic properties for centuries. The aim of this study is to determine the effect of using activated charcoal on establishing the in vitro propagation of L. Salicaria L. In this study, shoot explants (1.5 cm) were excised from buds that were 30 days old with a sterile scalpel blade and then cultured on full-strength MS medium supplemented with different concentrations of activated charcoal to multiply and increase the length of shoots. Additionally, to investigate the in vitro rooting response, shoot tip explants were cut from elongated shoots and cultured on an MS medium containing different concentrations of NAA with or without activated charcoal with three replicates in each experiment. MS medium with 0.5 g/L activated charcoal had the highest mean shoot length (7.1 cm ± 0.15) and mean number of shoots per explant (2.4±0.11). The results show that 0.5 mg/L NAA and 0.2 g/L activated charcoal provide the best response for rooting. The improved protocol can be utilized to grow roots in micro shoots of L. salicaria, which is an important stage in the micro propagation of L. salicaria. [ABSTRACT FROM AUTHOR]

27. Novel candidate genes for environmental stresses response in Synechocystissp. PCC 6803 revealed by machine learning algorithms

Abstract

Cyanobacteria have developed acclimation strategies to adapt to harsh environments, making them a model organism. Understanding the molecular mechanisms of tolerance to abiotic stresses can help elucidate how cells change their gene expression patterns in response to stress. Recent advances in sequencing techniques and bioinformatics analysis methods have led to the discovery of many genes involved in stress response in organisms. The Synechocystissp.PCC 6803is a suitable microorganism for studying transcriptome response under environmental stress. Therefore, for the first time, we employed two effective feature selection techniques namely and support vector machine recursive feature elimination (SVM-RFE) and LASSO (Least Absolute Shrinkage Selector Operator) to pinpoint the crucial genes responsive to environmental stresses in Synechocystissp. PCC 6803. We applied these algorithms of machine learning to analyze the transcriptomic data of Synechocystissp. PCC 6803under distinct conditions, encompassing light, salt and iron stress conditions. Seven candidate genes namely sll1862, slr0650, sll0760, slr0091, ssl3044, slr1285, and slr1687were selected by both LASSO and SVM-RFE algorithms. RNA-seq analysis was performed to validate the efficiency of our feature selection approach in selecting the most important genes. The RNA-seq analysis revealed significantly high expression for five genes namely sll1862, slr1687, ssl3044, slr1285, and slr0650under ion stress condition. Among these five genes, ssl3044and slr0650could be introduced as new potential candidate genes for further confirmatory genetic studies, to determine their roles in their response to abiotic stresses.
