Background Rhinologists are currently trying to find evidence useful for classifying chronic rhinosinusitis with nasal polyps (CRSwNP) in terms of pathophysiology-based endotypes. This would help us to tailor targeted therapies and follow-up protocols to each patient, improving treatment efficacy and reducing relapse risk. The histopathological study of inflammatory cells in tissue and their tendency to form aggregates has shown promise in determining the pathogenesis and predicting the clinical outcome. Objective The main purpose of this study was to assess tissue eosinophil count, eosinophil aggregation, and their relationship to different CRSwNP phenotypes. Methods A consecutive series of 96 patients who underwent endoscopic sinus surgery for CRSwNP was retrospectively assessed. Our structured histopathological approach considered eosinophil count and eosinophil aggregates. Results Patients’ clinical features included allergic fungal rhinosinusitis (AFRS) in 17 cases, nonsteroidal anti-inflammatory drug-exacerbated respiratory disease (NERD) in 19 cases, intrinsic asthma in 18 cases, extrinsic asthma in 21 cases, and allergy in 21 cases. Statistical analysis revealed more eosinophil aggregates in patients with NERD than in those with AFRS ( P = .020), intrinsic asthma ( P = .019), or extrinsic asthma ( P = .029). NERD patients had a significantly higher tissue eosinophil count than the other groups considered, apart from the AFRS ones. AFRS patients had a higher tissue eosinophil count than patients with intrinsic asthma. Conclusion Our results showed in particular that the NERD patient group’s rhinosinusitis was sustained by a significantly more severe eosinophilic reaction than in the other groups. Structured histopathology can provide useful information for classifying CRSwNP on pathophysiological grounds.

Background Eosinophils play an important role in bullous pemphigoid (BP) pathogenesis. Although tissue infiltration with eosinophils has been known for a long time, there is a lack of knowledge about the relationship between tissue eosinophil levels and disease severity and clinical characteristics of the patients. Methods Fifty-nine patients diagnosed with BP between January 2008 and December 2018 were reviewed. Haematoxylin-Eosin (H&E)-stained preparations were re-evaluated in terms of tissue eosinophil levels. For disease severity, Bullous Pemphigoid Disease Area Index (BPDAI) was used. The relationship between tissue eosinophil levels and disease severity and clinical features were evaluated. Results Erosion/blister and urticaria/erythema BPDAI scores were higher in the group with high tissue eosinophil level than the group with low tissue eosinophil level. Tissue and peripheral blood eosinophil count were correlated with total urticaria/erythema BPDAI scores. There was no correlation between blood and tissue eosinophil count. The mortality rate was 64.7% vs 44.0% in the high vs low tissue eosinophil groups. Tissue eosinophil levels were high in patients with BP accompanying neurological disease. Conclusions Tissue eosinophil count and peripheral blood eosinophil count were correlated with disease severity in BP. Tissue eosinophil levels were also high in patients with BP accompanying neurological disease.

Aim: Our study aimed to evaluate the infiltration of tissue eosinophils and mast cells in oral squamous cell carcinoma (OSCC) by using special stains. Study Design: Comparative study. Place of Study: Sharad Pawar Dental College, Sawangi (Meghe), Wardha, Maharashtra. Methodology: The study was carried out with the sample size of 30 histopathologically diagnosed cases of oral squamous cell carcinoma and comparison of infiltration of these (tissue eosinophil and mast cells) inflammatory cells with control (normal) group of patients, was done by using special stains. Special stains are wonderful because they allow us to see which we cannot see with routine Haematoxylin and eosin stain. Special stains were used to demonstrate tissue eosinophils and mast cells. Carbol Chromotrope and congo red were used for tissue eosinophil and for mast cells staining toluidine blue and thionin were used. Background: In oral carcinoma much effort has been made to predict the prognosis of patients but a sound understanding of underlying cell biology is likely to need progress. Recently, attention has been directed towards tumour associated tissue eosinophils and mast cells and their role in the biologic behavior of tumours. Aim: The retrospective study was used to evaluate the influence of tumour associated tissue eosinophils and mast cells on prognosis of oral squamous cell carcinoma (OSCC). Material and methods: The follow-up, of histopathologically diagnosed thirty cases of OSCC, was carried out for minimum period of 3 years. Special stains are wonderful they allow us to see which we can not see clear with routine H&E stain.Tissue sections were stained with special stains, Carbol Chromotrope for tissue eosinophil and Toluidine blue for tissue mast cell staining. Result: The results of the present study shows that increase infiltration of tissue eosinophils and mast cells in OSCC, associated with favourable prognosis. Conclusion: We concludes that infiltration of tissue eosinophils and mast cells are indicators of favourable prognosis in OSCC. Thus quantitative assessment of eosinophils and mast cells are the most important aspects of the microscopic evaluation of OSCC.

Eighty-three 2–4-year-old intact male goats exposed to a combination of artificial and natural challenge were segregated into responders and non-responders by ranking of weekly faecal egg counts (FECs). Retrospective analysis of samples over a 15-week-period showed responders had a statistically lower mean FEC than non-responders. Estimates of repeatability between consecutive egg counts were significant in both groups. The 6 top responders and bottom non-responders were subsequently given an artificial trickle challenge with Teladorsagia circumcincta and Trichostrongylus vitrinus . Mean faecal egg output was significantly lower in responders than non-responders. Peripheral eosinophil numbers following challenge were significantly greater in responder than non-responder goats. Abomasal and intestinal worm burdens were considerably lower in responders, with evidence of retardation of worm development compared to non-responders. Both abomasal and jejunal tissue eosinophil numbers were significantly higher in responders, although there was no difference in mucosal mast cell or globule leucocyte numbers. These results suggest that under temperate climatic conditions, it is possible to segregate male goats into responders and non-responders on the basis of simple parasitological criteria.

Extramedullary eosinophilopoiesis is described as a regular pathological feature of murine and cricetine schistosome infections. Using the Dominici staining technique, colonies of developing eosinophils were commonly found in certain tissue sites of dense oviposition, particularly in the liver, and in associated lymphoreticular tissues. The relationship of these observations to the role of eosinophilic leukocytes in schistosomiasis is discussed.

Pieces of ovary and ullimobranchial gland from adult laying hens were examined by an indirect peroxidase technique which differentiated tissue eosinophil and heterophil leucocytes. The results showed that in both organs, brown-staining peroxidase-positive eosinophils were greatly outnumbered hy pink- to red-staining heterophils, thus demonstrating that eosinophils are not the dominant granulocyte present, as has been widely reported. It is suggested that the heterophils may be required in a phagocytic role.

Factors underlying the eosinophil responses in the tissue sites of immediate hypersensitivity reactions have been investigated. A quantitative assessment of the number of eosinophils appearing in skin test reactions to ragweed antigen and compound 48/80 in ragweed-sensitive humans has been compared with several other parameters. There was a moderate, statistically significant, correlation with serum levels of IgE-class antiragweed antibody; also, eosinophil responses were minimal or absent in minimally positive threshold dilution skin tests. Tissue eosinophil responses were generally limited to those with baseline blood eosinophil levels of at least 150 mm3; however, there was no correlation between blood and tissue eosinophil levels in individual subjects. These findings suggest that the pathogenesis of the observed tissue eosinophil responses may be multifactorial.

Summary Inbred rats developed an increased blood eosinophil count 2 weeks after injection of syngeneic or semi-syngeneic lymphoma cells. An eosinophilia did not occur in allogeneic recipients. Autoradiographic studies of bone marrow eosinophil production showed that additional eosinophils were not being made, and tissue eosinophil levels were also normal. Moreover injections of prednisolone did not lower blood eosinophil counts significantly in the rats with the lymphoma. Other experiments showed that the eosinophilia was probably not due to a graft-versus-host reaction. It was concluded that the mechanism of eosinophilia was a prolongation of eosinophil survival in the blood, possibly due to blockage of normal emigration pathways into tissues, by large numbers of blood-borne lymphoma cells.
