Abstract

The description of nonequilibrium processes in nano-sized objects, where the typical energies involved are a few times, is increasingly becoming central to disciplines as diverse as condensed-matter physics, materials science, and biophysics. Major recent developments towards a unified treatment of arbitrarily large fluctuations in small systems are described by fluctuation theorems that relate the probabilities of a system absorbing from or releasing to the bath a given amount of energy in a nonequilibrium process. Here we experimentally verify the Crooks Fluctuation Theorem (CFT) under weak and strong nonequilibrium conditions by using optical tweezers to measure the irreversible mechanical work during the unfolding and refolding of a small RNA hairpin and an RNA three-helix junction. We also show that the CFT provides a powerful way to obtain folding free energies in biomolecules by determining the crossing between the unfolding and refolding irreversible work distributions. The method makes it possible to obtain folding free energies in nonequilibrium processes that dissipate up to of the average total work exerted, thereby paving the way for reconstructing free energy landscapes along reaction coordinates in nonequilibrium single-molecule experiments., Comment: PDF file, 19 pages. Abstract

The compost barn system was designed to be a sustainable alternative housing system for dairy cows. In order to help producers in the region to choose the best type of facility from the point of view of the milk production of the animals, this study compared the productivity of cows confined in an open composting barn with natural ventilation and in a closed composting barn with negative pressure ventilation and evaporative cooling panels. The temperature and relative humidity of the air were monitored, as well as the milk production of the animals housed in the facilities, and THI (Temperature and Humidity Index) were calculated. During the trials, the maximum daily temperatures of the air reached values around 27 °C and the THI remained within the normal range of up to 70. The average productivity remained in the general pattern described in the literature from 23 to 44 kg of milk cow1 day-1 , with lower rates obtained in the closed house. As the variation of the index used to describe the internal environment was not significant, it can be inferred that climatic elements as temperature and air humidity, under the conditions analyzed were not the main factors influencing the productivity rates of dairy cattle. In conclusion, under the analysed conditions the use of a closed barn with negative pressure ventilation is hardly justified as a plant that favoured the productivity of the confined cows.

24. DNA dynamics in aqueous solution: opening the double helix

Abstract

The opening of a DNA base pair is a simple reaction that is a prerequisite for replication, transcription, and other vital biological functions. Understanding the molecular mechanisms of biological reactions is crucial for predicting and, ultimately, controlling them. Realistic computer simulations of the reactions can provide the needed understanding. To model even the simplest reaction in aqueous solution requires hundreds of hours of supercomputing time. We have used molecular dynamics techniques to simulate fraying of the ends of a six base pair double strand of DNA, [TCGCGA]2, where the four bases of DNA are denoted by T (thymine), C (cytosine), G (guanine), and A (adenine), and to estimate the free energy barrier to this process. The calculations, in which the DNA was surrounded by 2,594 water molecules, required 50 hours of CRAY-2 CPU time for every simulated 100 picoseconds. A free energy barrier to fraying, which is mainly characterized by the movement of adenine away from thymine into aqueous environment, was estimated to be 4 kcal/mol. Another fraying pathway, which leads to stacking between terminal adenine and thymine, was also observed. These detailed pictures of the motions and energetics of DNA base pair opening in water are a first step toward understanding how DNA will interact with any molecule.

25. Effects of carbon dioxide on turkey poult performance and behavior

Abstract

Appropriate ventilation of poultry facilities is critical for achieving optimum performance. Ventilation promotes good air exchange to remove harmful gases, excessive heat, moisture, and particulate matter. In a turkey brooder barn, carbon dioxide (CO2) may be present at higher levels during the winter due to reduced ventilation rates to maintain high temperatures. This higher CO2 may negatively affect turkey poult performance. Therefore, the objective of this study was to evaluate the effects of subjecting tom turkey poults (commercial Large White Hybrid Converters) to different constant levels of atmospheric CO2 on their growth performance and behavior. In three consecutive replicate trials, a total of 552 poults were weighed post-hatch and randomly placed in 3 environmental control chambers, with 60 (Trial 1) and 62 (Trials 2 and 3) poults housed per chamber. They were reared with standard temperature and humidity levels for 3 wks. The poults were exposed to 3 different fixed CO2 concentrations of 2,000, 4,000, and 6,000 ppm throughout each trial. Following each trial (replicate), the CO2 treatments were switched and assigned to a different chamber in order to expose each treatment to each chamber. At the end of each trial, all poults were sent to a local turkey producer to finish grow out. For each trial, individual body weight and group feed intake were measured, and mortality and behavioral movement were recorded. Wk 3 and cumulative body weight gain of poults housed at 2,000 ppm CO2 was greater (P < 0.05) than those exposed to 4,000 and 6,000 ppm CO2. Feed intake and feed conversion were unaffected by the different CO2 concentrations. No significant difference in poult mortality was found between treatments. In addition, no effect of CO2 treatments was evident in the incidence of spontaneous turkey cardiomyopathy for turkeys processed at 19 wk of age. Abstract

The tracer gas ratio method, using CO2 as natural tracer, has been suggested as a pragmatic option to measure emissions from naturally ventilated (NV) barns without the need to directly estimate the ventilation rate. The aim of this research was to assess the performance of a low-cost Non-Dispersive Infra-Red (NDIR) sensor for intensive spatial field monitoring of CO2 concentrations in a NV dairy cow house. This was achieved by comparing NDIR sensors with two commonly applied methods, a Photo-Acoustic Spectroscope (PAS) Gas Monitor and an Open-Path laser (OP-laser). First, calibrations for the NDIR sensors were obtained in the laboratory. Then, the NDIR sensors were placed in a dairy cow barn for comparison with the PAS and OP-laser methods. The main conclusions were: (a) in order to represent the overall barn CO2 concentration of the dairy cow barn, the number of NDIR sensors to be accounted for average concentration calculation was dependent on barn length and on barn area occupation; and (b) the NDIR CO2 sensors are suitable for multi-point monitoring of CO2 concentrations in NV livestock barns, being a feasible alternative for the PAS and the OP-laser methods to monitor single-point or averaged spatial CO2 concentrations in livestock barns.

Abstract

The Brazilian poultry industry is an activity in constant development due to the high indices of productive efficiency. The accelerated evolution of poultry production has allowed to obtain early and very efficient broilers able to convert different foods into animal protein. However, due to this intensive breeding system, a series of metabolic and management problems appeared, with emphasis on thermal stress. The objective of this work was to evaluate the physiological responses of broiler chickens in the final stage of breeding (21 to 42 days of life), submitted to two thermal conditions, one representative of the thermoneutrality situation (T1) and one giving a situation of cyclic stress by heat (T2). For each experimental thermal condition, the birds were submitted to different levels of metabolizable energy of 3,050, 3,125, 3,200, 3,275 kcal kg-1. At 28, 35 and 42 days, the birds and the feed leftovers were weighed to measure the performance variables: CR (feed intake), GP (weight gain) and CA (feed conversion), viability of the rearing (Vb), productive efficiency index (PEI). As conclusions, the GP was 13.6% higher for the birds maintained at the thermoneutrality situation T1. The PEI was 32.5% higher for the birds maintained in T1 condition, when compared to those kept in T2. However, both in thermoneutral and in heat stress conditions, the increase in the level of metabolizable energy in the diet did not influence the performance and the productive efficiency index of broiler chickens aged between 21 and 42 days of age. 38. A refined protocol for calculating air flow rate of naturally ventilated broiler barns based on CO2 mass balance

Abstract

This study was conducted to evaluate relatively simple protocols for monitoring ventilation rate (VR) in naturally-ventilated barns (NVB). The test protocols were first applied to a mechanically-ventilated broiler barn (MVB), where VR was estimated more accurately and then were used to calculate VR in the NVB. CO2 concentrations were measured with two different sampling schemes: (S1) the average of indoor measurements along the length of the building at two heights of 0.5 m and 1.5 m from the litter floor; and (S2) same as previous but with concentration measurements taken only at 0.5 m from litter. The dynamic metabolic CO2 production rate of the birds was predicted with two different algorithms: (A1) remaining constant throughout the dark and light periods, and (A2) varying with animal activity on an hourly basis. The results demonstrated that the combination of S2 with A1 or A2 yielded the best estimate of building VR in the NVB.

Abstract

One-year prospective observational study of meningitis diagnosed at a third level hospital.All patients with a cerebrospinal fluid (CSF) specimen with cyto-biochemical characteristics and clinical picture consistent with meningitis were included in the study. They were followed from admission to hospital up to discharge or exitus. The epidemiologic characteristics of patients, etiology, related risk factors and predisposing situations, CSF characteristics, clinical manifestations, clinical course, and antibiotic susceptibility of the causative agents were analyzed.Ninety-five cases were included. Seventy-six (69.4%) were community acquired and 29 (30.5%) nosocomially acquired meningitis. Among community acquired meningitis, 31 (46.9%) were of bacterial origin (8 N. meningitidis, 3 H. influenzae, 2 S. pneumoniae, 1 Streptococcus group B, 1 Listeria monocytogenes, 1 Staphylococcus aureus, and 1 Brucella spp.); CSF culture was negative in 14 cases (41.2%). In most cases neither risk factor nor predisposing situations were detected. Patients with purulent meningitis and negative CSF culture had a significantly lower number of complications than patients with positive CSF culture. Among patients previously treated with beta-lactam antibiotics (8 cases) the probability of a negative CSF culture was greater than among not treated patients (OR 16.00, 95% CI 1.45-764.68; p = 0.011). The remaining cyto-biochemical characteristics were similar in both groups. Thirty-five cases (53.03%) of community acquisition were lymphocytic meningitis (31 viral, 3 tuberculous, and 1 luetic meningitis). Among nosocomial cases (29 cases, 30.5%), most were caused by gram-negative bacilli and microorganisms of the Staphylococcus genus. Fourteen cases (48.2%) were related to some type of neurosurgical procedure. Overall, only two exitus cases were recorded.The etiologic agents of community acquired meningitis are mainly N. meningitidis, S. pneumoniae and Haemophilus influenzae. The previous antibiotic therapy did not influence thy cyto-biochemical characteristics of CSF but it did influence the yielding of culture. Meningitis with negative CSF culture has a significantly lower number of complications. The availability of a Neurosurgery Department at a hospital confers a change in the epidemiologic spectrum of diagnosed meningitis, with a higher incidence of nosocomial meningitis. In our environment, a substantial proportion of cases due to Staphylococcus microorganisms was observed.

Abstract

O objetivo desta pesquisa foi estudar as condicoes termicas ambientais, com sistemas de acondicionamento termico, em maternidades suinicolas, no inveno, utilizando-se os seguintes tratamentos: 1) sala convencional (SSV) e 2) sala com amplas aberturas de janelas e sistemas de cortinas (SAC). O delineamento experimental utilizado foi o inteiramente casualizado, em esquema de parcelas subdivididas, com dois tratamentos nas parcelas e oito horarios nas subparcelas, com quatro repeticoes. Foram utilizadas 52 porcas, das racas Landrace e Large White, distribuidas aleatoriamente em dois lotes de 26 matrizes. O indice de temperatura de globo e umidade (ITGU), a carga termica radiante (CTR), o indice bioclimatico (IBC) e a umidade relativa do ar (UR) foram estimados. Os tratamentos SAC e SSV proporcionaram ambientes termicamente semelhantes. As maternidades suinicolas com amplas aberturas de janelas e sistema de cortinas ou maternidades com sala convencional proporcionaram condicoes termicas ambientasis adequadas para as porcas em lactacao, no inverno.
