Abstract Karyopherin α 2 (KPNA2, importin α1), a transport factor shuttling between the nuclear and cytoplasmic compartments, is involved in the nuclear import of proteins and participates in cellular processes such as cell cycle regulation, apoptosis, and transcriptional regulation. However, it is still unclear which signaling regulates the nucleocytoplasmic distribution of KPNA2 in response to cellular stress. In this study, we report that oxidative stress increases nuclear retention of KPNA2 through alpha serine/threonine‐protein kinase (AKT1)‐mediated reduction of serine 62 (S62) phosphorylation. We first found that AKT1 activation was required for H2O2‐induced nuclear accumulation of KPNA2. Immunoprecipitation and quantitative proteomic analysis revealed that the phosphorylation of KPNA2 at S62 was decreased under H2O2‐induced oxidative stress. We showed that cyclin‐dependent kinase 1 (CDK1), a kinase responsible for KPNA2 S62 phosphorylation, contributes to the localization of KPNA2 in the cytoplasm. AKT1 knockdown increased KPNA2 S62 phosphorylation and inhibited CDK1 activation. Furthermore, H2O2‐induced AKT1 activation promoted nuclear KPNA2 interaction with nucleophosmin 1 (NPM1), resulting in attenuation of NPM1‐mediated cyclin D1 gene transcription. Thus, we infer that the AKT1‐CDK1 axis regulates the nucleocytoplasmic shuttling and function of KPNA2 through spatiotemporal regulation of KPNA2 S62 phosphorylation under oxidative stress conditions.

Abstract Uncontrolled terpenoid emissions from forest trees in Japan may have contributed to high O3 concentrations observed in urban and suburban areas. To estimate ozone formation via a series of reactions between NO x and terpenoids using atmospheric chemistry models, it is important to produce terpenoid emission inventories by collecting all reported emission data for the major tree species in Japan and examining their reliability. In this review, we first describe three different plant terpenoid emission types, i.e., isoprene-emitting type, monoterpene-emitting type with storage tissues and organs, and monoterpene-emitting type without storage tissues and organs. Second, we describe various methods for measuring plant terpenoid emissions, including a recently developed simplified method, and explain their reliability. We emphasized that applicable measurement methods depend on the terpenoid emission types. Data obtained using static chamber methods should not be considered because they have the highest uncertainty resulting from normal chamber materials that are not specific to terpenoid measurements and lack humidity control. Finally, we show the absolute values of the collected emission rates and describe their variability. The deciduous oak species, Quercus serrata and Quercus mongolica var. crispula, and bamboo species, Phyllostachys pubescens and Phyllostachys bambusoides, are strong isoprene emitters. Among the monoterpene emitters, four evergreen broadleaf trees, including three Quercus species, had the highest basal emission rate (BER). The monoterpene storage type conifers Larix kaempferi and Pinus densiflora have relatively lower BERs. Emission data are not available for Castanopsis cuspidata, and seasonal changes in emission rates have not been reported for several major tree species in the top 20 rankings. Within species, the reported emission rates of some tree species differed by threefold. These differences may be attributed to the reliability of the measurement and analytical systems, tree age, leaf morphology, environmental conditions, and genetic diversity. We emphasize the need for reliable measurements to achieve a more precise terpenoid emission inventory for major tree species in Japan.

Abstract Inaccessibility of stored memory in ensemble cells through the forgetting process causes animals to be unable to respond to natural recalling cues. While accumulating evidence has demonstrated that reactivating memory-stored cells can switch cells from an inaccessible state to an accessible form and lead to recall of previously learned information, the underlying cellular and molecular mechanisms remain elusive. The current study used Drosophila as a model to demonstrate that the memory of one-trial aversive olfactory conditioning, although inaccessible within a few hours after learning, is stored in KCαβ and retrievable after mild retraining. One-trial aversive conditioning triggers protein synthesis to form a long-lasting cellular memory trace, approximately 20 days, via creb in KCαβ, and a transient cellular memory trace, approximately one day, via orb in MBON-α3. PPL1-α3 negatively regulates forgotten one-trial conditioning memory retrieval. The current study demonstrated that KCαβ, PPL1-α3, and MBON-α3 collaboratively regulate the formation of forgotten one-cycle aversive conditioning memory formation and retrieval.

Postoperative pulmonary complications (PPCs) most commonly occur after thoracic surgery. Not only prolonged hospital stay and increased financial expenses but also morbidity and even mortality may be troublesome for those with PPCs. Herein, we aimed to conduct a comprehensive systematic review and meta-analysis of available data to examine the effectiveness of incentive spirometry (IS) to reduce PPCs and shorten hospital stay. This systematic review and meta-analysis included 5 randomized controlled trials (RCT) and 3 retrospective cohort study (10,322 patients in total) in PubMed, Embase and Cochrane Library until September 31, 2021. We assessed the clinical efficacy of IS using length of hospital stay, PPCs, postoperative pneumonia, and postoperative atelectasis with meta-analysis, meta-regression and trial sequential analysis (TSA). With this meta-analysis, the length of hospital stay in patients undergoing IS was significantly shorter (1.8 days) than that in patients not receiving IS (MD = −1.80, 95% CI = −2.95 to −0.65). Abstract Intraoperative teaching is a challenging task. The briefing–intraoperative teaching–debriefing (BID) model, which is based on guided discovery learning at limited time intervals, has rarely been investigated. This study validated the benefits of the modified BID model on medical clerks. This study involved 37 first-year medical clerks enrolled from September 2019 to May 2020. Every learner scrubbed in one the totally implantable venous access device placement surgery and completed a pre-/posttest survey on surgical procedures and associated anatomy conducted through an intraoperative teaching questionnaire. Of these participants, 15 merely observed throughout the entire procedure (observation group), whereas the remaining 22 performed simple suturing under supervision (suturing group). Abstract Background Gold nanoparticles (AuNPs) have been widely used in local surface plasmon resonance (LSPR) immunoassays for biomolecule sensing, which is primarily based on two conventional methods: absorption spectra analysis and colorimetry. The low figure of merit (FoM) of the LSPR and high-concentration AuNP requirement restrict their limit of detection (LOD), which is approximately ng to μg mL−1 in antibody detection if there is no other signal or analyte amplification. Improvements in sensitivity have been slow in recent for a long time, and pushing the boundary of the current LOD is a great challenge of current LSPR immunoassays in biosensing. Results In this work, we developed spectral image contrast-based flow digital nanoplasmon-metry (Flow DiNM) to push the LOD boundary. Comparing the scattering image brightness of AuNPs in two neighboring wavelength bands near the LSPR peak, the peak shift signal is strongly amplified and quickly detected. Introducing digital analysis, the Flow DiNM provides an ultrahigh signal-to-noise ratio and has a lower sample volume requirement. Compared to the conventional analog LSPR immunoassay, Flow DiNM for anti-BSA detection in pure samples has an LOD as low as 1 pg mL−1 within only a 15-min detection time and 500 μL sample volume. Antibody assays against spike proteins of SARS-CoV-2 in artificial saliva that contained various proteins were also conducted to validate the detection of Flow DiNM in complicated samples. Flow DiNM shows significant discrimination in detection with an LOD of 10 pg mL−1 and a broad dynamic detection range of five orders of magnitude. Conclusion Together with the quick readout time and simple operation, this work clearly demonstrated the high sensitivity and selectivity of the developed Flow DiNM in rapid antibody detection. Spectral image contrast and digital analysis further provide a new generation of LSPR immunoassay with AuNPs. Graphical Abstract

Background: The increasing lung cancer screening for asymptomatic adults via computed tomography (CT) has increased the discovery of intermediate pulmonary nodules (PNs) that are small, with a subsolid component, or/and are relatively deeply seated. Pre- or intraoperative localization could be the main modality to identify and localize these target intermediate PNs and facilitate the subsequent surgical resection accordingly. Herein, we present the learning experience of preoperative CT-guided patent blue localization (CTPBL) for PNs in a tertiary hospital and concomitantly conduct an extensive literature review. Materials and Methods: This retrospective study included all patients with PNs undergoing CTPBL before video-assisted thoracoscopic surgery (VATS) resection from January 2019 to August 2020. After completing the consultation and informed consent, preoperative CTPBL was conducted to label these intermediate PNs. Data included the patients' characteristics (sex, age, smoking status, and comorbidities), CTPBL details (PN size and related lobe, dye dilution, puncture times, and time interval from localization to surgery), CT-guided localization complications (pneumothorax, focal parenchyma hemorrhage, hemothorax, and hemoptysis), and intraoperative findings and postoperative pathologic report. The Chi-square tests, Fisher's exact test, and independent t-tests were used to compare the abovementioned variables. Multiple regression analysis was used to identify the potential risk factors for CTPBL-related complications. Results: This study included 54 consecutive patients with 58 PNs, of which 54 had single and 4 had double nodules. The average size of PNs was 0.77 ± 0.31 cm (0.3–2.1); the localization duration and time interval from initial labeling to VATS were 40.4 ± 12.1 min (24–72) and 243.4 ± 94.8 min (118–520), separately. The success rate of preoperative CTPBL was 96.6% (56/58), and all PNs were uneventfully resected via VATS after CTPBL. CTPBL-related complications accounted for 40.7% (22/54), including pneumothorax (22/54), focal parenchyma hemorrhage (11/54), hemothorax (2/54), and hemoptysis (2/54), in order. Based on our multiple regression analysis, the number of PNs is the only potential risk factor related to CTPBL-related hemoptysis (95% confidence interval = −0.545–−0.233; P < 0.001). Conclusion: Based on our learning experience, CTPBL for intermediate PNs could be a safe procedure to label intermediate PNs for thoracoscopic excisional biopsy with a satisfactory success rate. Moreover, our multiple regression analysis demonstrated that patients undergoing repetitive localizations for two PNs were prone to have hemoptysis after CTPBL.

Visual data on the geographic distribution of carbon storage help policy makers to formulate countermeasures for global warming. However, Taiwan, as an island showing diversity in climate and topography, had lacked valid visual data on the distribution of forest carbon storage between the last two forest surveys (1993–2015). This study established a model to estimate and illustrate the distribution of forest carbon storage. This model uses land use, stand morphology, and carbon conversion coefficient databases accordingly for 51 types of major forests in Taiwan. An estimation in 2006 was conducted and shows an overall carbon storage of 165.65 Mt C, with forest carbon storage per unit area of 71.56 t C ha−1, where natural forests and plantations respectively contributed 114.15 Mt C (68.9%) and 51.50 Mt C (31.1%). By assuming no change in land use type, the carbon sequestration from 2006 to 2007 by the 51 forest types was estimated to be 5.21 Mt C yr−1 using historical tree growth and mortality rates. The result reflects the reality of the land use status and the events of coverage shifting with time by combining the two forest surveys in Taiwan.

Abstract The standard size of bougie for laparoscopic sleeve gastrectomy (LSG) is not yet established. Therefore, a systematic review and network meta-analysis were conducted to assess the weight loss effects and associated complications of LSG for patients with morbid obesity, based on different bougie sizes. A total of 15 studies were reviewed in this systemic review and network meta-analysis (2,848 participants), including RCTs and retrospective studies in PubMed, and Embase until September 1, 2020. The effectiveness of different bougie calibration sizes was assessed based on excess weight loss (EWL), total complications, and staple line leak. Within this network meta-analysis, S-sized (≤ 32 Fr.) and M-sized (33–36 Fr.) bougies had similar effects and were associated with the highest EWL improvement among all different bougie sizes (S-sized: standardized mean difference [SMD], 10.52; 95% confidence interval [CI] − 5.59 to − 26.63, surface under the cumulative ranking curve [SUCRA], 0.78; and M-sized: SMD, 10.16; 95% CI − 3.04–23.37; SUCRA, 0.75). M-sized bougie was associated with the lowest incidence of total complications (M-sized: odds ratio, 0.43; 95% CI, 0.16–1.11; SUCRA, 0.92). Based on our network meta-analysis, using M-sized bougie (33–36 Fr.) is an optimal choice to balance the effectiveness and perioperative safety of LSG in the clinical practice.

ObjectiveDelayed progressive mass effect (DPME) after securing an aneurysm is uncommon following microsurgical or endovascular repair and leads to a poor clinical outcome. Patients with ruptured middle cerebral artery (MCA) aneurysms have a high risk of postoperative oedema and mass effect, which may require decompressive treatment. Because few studies have discussed the risk and predictive factors, we focused on ruptured MCA aneurysms and evaluated the outcomes of these patients and the necessity of salvage surgery when DPME presented.MethodsData on 891 patients with aneurysmal subarachnoid haemorrhage (aSAH) treated between January 2011 and February 2020 were extracted from the medical database of a tertiary referral centre. A total of 113 patients with aSAH resulting from at least one MCA aneurysm were identified. After excluding patients with several clinical confounders, we enrolled 80 patients with surgically treated aSAH. We examined the characteristics of aneurysms and hematomas, perioperative contrast pooling patterns, presence of distal hematomas, perisylvian low density, occlusive treatment modality, management strategies, the need for salvage surgical decompression, and postoperative 90-day outcomes to identify possible risk factors.ResultsDPME was observed in 27 of the 80 patients (33.7%). Abstract The metabolic changes in melanoma cells that are required for tumor metastasis have not been fully elucidated. In this study, we show that the increase in glucose uptake and mitochondrial oxidative phosphorylation confers metastatic ability as a result of aryl hydrocarbon receptor nuclear translocator (ARNT) deficiency. In clinical tissue specimens, increased ARNT, pyruvate dehydrogenase kinase 1 (PDK1), and NAD(P)H quinine oxidoreductase-1 (NQO1) was observed in benign nevi, whereas lower expression was observed in melanoma. The depletion of ARNT dramatically repressed PDK1 and NQO1 expression, which resulted in an increase of ROS levels. The elimination of ROS using N-acetylcysteine (NAC) and inhibition of oxidative phosphorylation using carbonyl cyanide m-chlorophenyl hydrazone (CCCP) and rotenone inhibited the ARNT and PDK1 deficiency-induced cell migration and invasion. In addition, ARNT deficiency in tumor cells manipulated the glycolytic pathway through enhancement of the glucose uptake rate, which reduced glucose dependence. Intriguingly, CCCP and NAC dramatically inhibited ARNT and PDK1 deficiency-induced tumor cell extravasation in mouse models. Our work demonstrates that downregulation of ARNT and PDK1 expression serves as a prognosticator, which confers metastatic potential as the metastasizing cells depend on metabolic changes.

In this work, the properties of gallium oxide (Ga2O3) and its excellent interface properties to GaN-based materials are explored as a gate insulator layer for GaN-based metal-oxide-semiconductor high-electron mobility transistors (MOSHEMTs). A novel vapor cooling condensation system was used to deposit the high quality Ga2O3 films with high insulation and low defect suitable for gate insulator layer. The characteristics of the Ga2O3 films were further explored by implementing GaN-based fin-channel array MOSHEMTs with recessed-gates and different channel widths. Compared to planar channel structure, the direct current, high frequency, and flicker noise performances were enhanced in the fin-channel MOSHEMTs with Ga2O3 gate insulator layer. For the GaN-based fin-channel array MOSHEMTs with 300-nm-wide channel, the devices exhibited superior performances of maximum extrinsic transconductance of 194.2 mS/mm, threshold voltage of −1.4 V, extrinsic unit gain cutoff frequency of 6.4 GHz, maximum oscillation frequency of 14.8 GHz, and normalized noise power of 8.45 $\times \,\, 10^{-15}$ Hz $^{-1}$ . It was also demonstrated that the associated performances were improved by reducing the width of fin-channel array.

Background: According to recent animal models for lung adenocarcinoma metastasis, cardiac function may be related to the clinical outcome. The aim of this study is to identify a predictable index for postoperative metastasis (POM) that is associated with cardiac function. Methods. Two hundred and seven consecutive patients who underwent thoracoscopic resection for stage I lung adenocarcinoma were included. Disease-free survival (DFS), overall survival (OS), and patients’ clinical and pathological characteristics were analyzed. Results. Among the 207 patients, 17 cases demonstrated metastasis, 110 cases received a preoperative echocardiogram, and six cases had POM. Mitral valve peak A velocity, which is one of the left ventricular diastolic function parameters affected by BMI (MVPABMI), was associated with a negative factor for POM (hazard ratio (HR): 2.139, p = 0.019) and a poor 5-year DFS in the above median (100% vs. 87%, p = 0.014). The predictable rate increased from 30.7% to 75% when the MVPABMI was above the median = 3.15 in the solid subtype). Conclusions. MVPABMI is a novel index for POM prediction in early-stage lung adenocarcinoma. This is a pilot study and the first attempt at research to verify that the diastole and the BMI may be associated with POM in early-stage lung adenocarcinoma.

Considerable isoprene emissions from several bamboo species have been reported. However, bamboos are highly diverse in taxonomy and have different niches or habitats among species, and the present investigation might be insufficient to conclude a representative isoprene emission trait for bamboos. In this study, isoprene flux, leaf mass per area (LMA), photosynthetic rate, and electron transport rate (ETR) observations were conducted for 18 species within five genera of bamboo species, which include different growth types (tall and dwarf) and climates of the region of origin (temperate, warm-temperate, and subtropical). As a result, we observed that dwarf bamboos exhibited very low or no emission; in contrast, tall bamboos demonstrated considerable isoprene emission fluxes mainly in August and September 2019 at temperatures greater than 30 °C. For tall bamboos, isoprene emission fluxes, photosynthetic rate, and ETR in area-based units were correlated with LMA. To exclude the systematic correlation among isoprene emission flux, photosynthetic rate, and ETR, correlations among the observations of mass-based units were also tested, and the results demonstrated significant positive correlations. The distinction in isoprene emission traits between tall and dwarf bamboos was independent of LMA, photosynthetic rate, and ETR, as there was no difference between them. This implies that the distinction in isoprene emission was caused by genetic differences. The low emission of isoprene from the dwarf species is reasonable because dwarf bamboos usually grow in areas with relatively low heat stress and low light where the production of isoprene could be futile due to carbon loss. This study suggests separating the two bamboo types into different functional types of isoprene emissions.

The fractional derivative (FD) model is one kind of numerical model of viscoelastic (VE) damper, which can describe the behavior of the frequency sensitivity of VE damper well with some empirical parameters. However, the FD model is difficult to apply to practice design because of its complex calculation. Compared with the FD model, the integer derivative (ID) models are widely used as the VE damper equipped in the high-rise building. However, ID models lack consideration of frequency sensitivity, which significantly affects their accuracy. To consider frequency sensitivity in the ID model, this study attempted to use the 4-element and 6-element models of ID models to approximate the FD model, easily describing the VE characteristic of the damper. The wind forces of 500-year-return periods were employed to analyze the influence of the frequency sensitivity of the VE damper on wind-induced responses and energy dissipation. The results of this study showed that the 4-element and 6-element models not only matched frequency sensitivities well to the FD model in the low-frequency region of wind excitation but also had good agreements with the FD model in wind-induced responses and energy dissipation. Based on the findings in this study, the 4-element and 6-element models were recommended as numerical models for the wind-resistant design of high-rise buildings with VE dampers. It helps improve the wind-induced behavior simulation of VE dampers considering their frequency sensitivity in high accuracy without massive experimental cost.

Introduction: Acne vulgaris afflicts many people, and despite the multitude of the anti-acne products on the market, there is still no effective treatment that can prevent and cure this disease. The severity of acne vulgaris is highly associated with the inflammatory response to Propionibacterium acnes (P. acnes) now referred to as Cutibacterium acnes (C. acnes), an opportunistic skin bacterium in the human skin microbiome. Areas covered: We here provide the prospects of creating acne vaccines targeting secreted virulence factors of C. acnes including secretory Christie-Atkins-Munch-Peterson (CAMP) factor. Neutralization of secreted virulence factors by either active or passive vaccination may have a lower risk of disturbing the microbial ecosystem in the human skin microbiome. Expert opinion: Major steps could be taken to start a public vaccination program at an early age to prevent the future occurrence of acne vulgaris. Future therapeutic monoclonal antibodies can be designed to specifically neutralize virulence factors of C. acnes including CAMP factors without disrupting the optimal balance of C. acnes in the human skin microbiome and lowering the risk of creating drug-resistant C. acnes. Targeting secreted virulence factors without disturbing the commensal relationship of host can be a novel gateway towards the therapeutic treatment of acne vulgaris.

The high isoprene emission capacity of palm species can decrease regional air quality and enhance the greenhouse effect when land is converted to palm plantations. Propagation of low-emitting individuals can be a strategy for reducing isoprene emission from palms. However, the identification of low-emitting individuals requires large-scale sampling. Thus, we aimed to develop a rapid method in which the isoprene emission rate of leaf segments is observed. We examined the temperature response and effect of incubation length on the isoprene emission rate of palm leaf and found that leaf temperatures at 25 to 30 °C and an incubation length of 40 min are suitable parameters. To further examine the validity of the method, we applied both the enclosure method and this method to the same sections of leaves. High coefficient of determinations (0.993 and 0.982) between the results of the two methods were detected regardless of seasonal temperature. This result proves that the method is capable of measuring the isoprene emission rate under any growth conditions if the incubation temperature is controlled. By using a water bath tank and a tested light source, we can simply implement a unified environmental control of multiple samples at once, which achieves a higher time efficiency than conventional enclosure measurements.

The majority of digital sensors rely on von Neumann architecture microprocessors to process sampled data. When the sampled data require complex computation for 24×7, the processing element will a consume significant amount of energy and computation resources. Several new sensing algorithms use deep neural network algorithms and consume even more computation resources. High resource consumption prevents such systems for 24×7 deployment although they can deliver impressive results. This work adopts a Computing-In-Memory (CIM) device, which integrates a storage and analog processing unit to eliminate data movement, to process sampled data. This work designs and evaluates the CIM-based sensing framework for human pose recognition. The framework consists of uncertainty-aware training, activation function design, and CIM error model collection. The evaluation results show that the framework can improve the detection accuracy of three poses classification on CIM devices using binary weights from 33.3% to 91.5% while that on ideal CIM is 92.1%. Although on digital systems the accuracy is 98.7% with binary weight and 99.5% with floating weight, the energy consumption of executing 1 convolution layer on a CIM device is only 30,000 to 50,000 times less than the digital sensing system. Such a design can significantly reduce power consumption and enables battery-powered always-on sensors.

Traumatic intraventricular hemorrhage (tIVH) is associated with increased mortality and disability in traumatic brain injury (TBI). However, the significance of tIVH itself remains unclear. Our goal is to assess whether tIVH affects in-hospital mortality and short-term functional outcomes. We retrospectively reviewed the records of 5048 patients with TBI during a 5-year period, and 149 tIVH patients were analyzed. Confounding was reduced using the inverse probability of treatment weighting (IPTW) based on propensity score. The association between IVH and outcomes was investigated using logistic regression in the IPTW-adjusted cohort. In our study, after adjustment for analysis, the in-hospital mortality rate (11.4% vs. 9.2%) and the poor functional outcome rate (37.9% vs.10.6%) were significantly higher in the tIVH group than in the non-tIVH group. Factors independently associated with outcomes were age ≥ 65 years, Glasgow Coma Scale (GCS) severity score, and the Graeb score. The Traumatic Graeb Score, a novel scoring system for predicting functional outcomes associated with tIVH, comprised the sum of the following components: GCS scores of 3 to 4 (=2 points), 5 to 12 (=1 point), 13 to 15 (=0 points); age ≥ 65 years, yes (=1 point), no (=0 points); Graeb score (0–12 points). A Traumatic Graeb Score ≥ 4 is an optimal cutoff value for poor short-term functional outcomes.

Objective: The chest tube drainage system (CTDS) of choice for the pleural cavity after pulmonary resection remains controversial. This systematic review and network meta-analysis (NMA) aimed to assess the length of hospital stay, chest tube placement duration, and prolonged air leak among different types of CTDS. Methods: This systemic review and NMA included 21 randomized controlled trials (3399 patients) in PubMed and Embase until 1 June 2021. We performed a frequentist random effect in our NMA, and a P-score was adopted to determine the best treatment. We assessed the clinical efficacy of different CTDSs (digital/suction/non-suction) using the length of hospital stay, chest tube placement duration, and presence of prolonged air leak. Results: Based on the NMA, digital CTDS was the most beneficial intervention for the length of hospital stay, being 1.4 days less than that of suction CTDS (mean difference (MD): −1.40; 95% confidence interval (CI): −2.20 to −0.60). Digital CTDS also had significantly reduced chest tube placement duration, being 0.68 days less than that of suction CTDSs (MD: −0.68; 95% CI: −1.32 to −0.04). Neither digital nor non-suction CTDS significantly reduced the risk of prolonged air leak. Conclusions: Digital CTDS is associated with better outcomes than suction and non-suction CTDS for patients undergoing pulmonary resections, specifically 0.68 days shorter chest tube duration and 1.4 days shorter hospital stay than suction CTDS.

Background: Pituicytes are glial cells supporting hypothalamic axons in the posterior pituitary gland. Pituicytoma arises from pituicytes, and is currently classified as a World Health Organization grade I central nervous system tumor. Clinically, some patients with pituicytoma are misdiagnosed with functioning pituitary adenoma before surgical interventions and pathological examinations. This study was designed to investigate the correlation between pituicytoma and abnormal hormone function. Methods: The records of patients who underwent pituitary surgery at Linkou Chang Gung Memorial Hospital between 2000 and 2016 were reviewed. Patients with a pathological diagnosis of pituicytoma were included. Among these patients, those with inadequate data and/or specimens were excluded. Clinical information including presenting symptoms, serum hormone level, neuroimages, and results of pathological examinations including hematoxylin and eosin stains and immunohistochemical stains were collected for analysis. Results: Between 2000 and 2016, eight patients were pathologically diagnosed with pituicytoma. Two patients were excluded due to missing data and/or specimens. Among the six remaining patients, five presented with hormone changes. The immunohistochemical stains of pituicytoma confirmed no secretory function; however, the surrounding pituitary glandular tissue showed positive results responsible for clinical hormone changes. Conclusions: Pituicytoma had a negative endocrine secretory function in this study. However, pituicytoma was associated with hyperactivity of the surrounding pituitary glandular tissue, which resulted in clinically abnormal hormone secretion. The diagnosis of pituicytoma before a thorough pathological examination is difficult when the patients present with hormone dysfunction. Therefore, maximum resection of the pituitary tumor is necessary to make the diagnosis of pituicytoma and to exclude pituitary adenoma. Neurocutaneous melanosis (NCM) is characterized by diffuse or focal proliferation of melanin-producing cells over the skin or leptomeninges. Even without malignant transformation, the prognosis is poor after symptomatic progression of NCM, resulting from either mass effect in central nervous system or hydrocephalus. We reported a 26-year-old female patient with rapid deterioration after the onset of NCM. Despite no solid tumor formation, elevated cerebrospinal fluid protein content, which complicated shunting procedure, associated with hydrocephalus played the key role in clinical deterioration. Adult onset of NCM with Dandy–Walker malformation was never reported in our literature review. Diagnosis and treatment for such disease entity are discussed.

This work develops a novel automatic irrigation system to implement the customized and accurate watering for an individual seedling. The system integrates the modules of visual recognition of the stem-leaf junction, identification of the stem-root junction as the watering point, and control of the spraying nozzle. The model of YOLOv3 is employed to screen the stem-leave junction of an orchid seedling, whose depth map then acquired by the method of Semi-Global Block Matching (SGBM) extracts the three-dimensional (3D) coordinates of the junction center. Next, the concept of leaf vector is introduced to identify the stem-root junction of the orchid seedling as the accurate watering point, which the spraying nozzle is controlled to reach for supplement of the specific amount of water. A number of experiments were conducted to verify the proposed irrigation system for orchid seedlings at different locations with various heights. The experimental results show that the rates of successful watering are 82% and 83.3% for the uni-pot and multi-pot orchid seedlings, respectively.

Developing cost-effective and fast detection methods in full-field optical coherence microscopy (FF-OCM) platform for biological applications is of prime focus today. In this paper, we report a cost-effective Linnik type FF-OCM system with a single broadband white light emitting diode (WLED) (470-850 nm, λ0 = 630 nm) light source in conjunction with a derivative-based image reconstruction algorithm. Sequential 2-D multiple phase-shifted interferograms were obtained by moving the sample stage using piezoelectric transducer and recorded via a 2-D charge complementary oxide semiconductor camera. Subsequently, the en-face sectional images were reconstructed using a derivative-based algorithm. The measured axial and lateral resolutions along with interferometric phase stability of the system were 0.9 μm, 1.4 μm, and 11 mrad, respectively. The advantages of the derivative-based algorithm over 4-step phase shifted method were demonstrated using USAF target and onion as test samples. Moreover, studies via the present system of the microstructure of skin epidermis in mouse ear with melanoma showed a significant deterioration from normal mouse skin in stratum corneum of the epidermis. This is the first study of the application of a derivative-based algorithm for biological samples in FF-OCM wherein WLED is used as a light source. The present system is stable, compact, and cost-effective compared to the conventional FF-OCM systems and provides comparable spatial resolution.

Antimicrobial peptides (AMPs) are important components of the host innate defense mechanism against invading microorganisms. Although AMPs are known to act on bacterial membranes and increase membrane permeability, the action mechanism of most AMPs still remains unclear. In this report, we found that the TP4 peptides from Nile tilapia anchored on E. coli cells and enabled them permeable to SYTOX Green in few minutes after TP4 addition. TP4 peptides existed in small dots either on live or glutaraldehyde-fixed cells. TP4 peptides were driven into oligomers either in soluble or insoluble form by a membrane-mimicking anionic surfactant, sarkosyl, depending on the concentrations employed. The binding forces among TP4 components were mediated through hydrophobic interaction. The soluble oligomers were negatively charged on surface, while the insoluble oligomers could be fused with each other or piled on existing particles to form larger particles with diameters 0.1 to 20 μm by hydrophobic interactions. Interestingly, the morphology and solubility of TP4 particles changed with the concentration of exogenous sarkosyl or trifluoroethanol. The TP4 peptides were assembled into oligomers on or in bacterial membrane. This study provides direct evidence and a model for the oligomerization and insertion of AMPs into bacterial membrane before entering into cytosol.

Antimicrobial peptides are important components of the host innate defense mechanism against invading pathogens, especially for drug-resistant bacteria. In addition to bactericidal activity, the 25 residue peptide TP4 isolated from Nile tilapia also stimulates cell proliferation and regulates the innate immune system in mice. In this report, TP4 hyperpolarized and depolarized the membrane potential of Pseudomonas aeruginosa at sub-lethal and lethal concentrations. It also inhibited and eradicated biofilm formation. The in vitro binding of TP4 to bacterial outer membrane target protein, OprI, was markedly enhanced by a membrane-like surfactant sarkosyl and lipopolysaccharide, which converted TP4 into an α-helix. The solution structure of TP4 in dodecylphosphocholine was solved by NMR analyses. It contained a typical α-helix at residues Phe10-Arg22 and a distorted helical segment at Ile6-Phe10, as well as a hydrophobic core at the N-terminus and a cationic patch at the C-terminus. Residues Ile16, Leu19 and Ile20 in the hydrophobic face of the main helix were critical for the integrity of amphipathic structure, other hydrophobic residues played important roles in hemolytic and bactericidal activities. A model for the assembly of helical TP4 embedded in sarkosyl vesicle is proposed. This study may provide valuable insight for engineering AMPs to have potent bactericidal activity but low hemolytic activity.

The emergence of antibiotic-resistant microbial strains has become a public health issue and there is an urgent need to develop new anti-infective molecules. Although natural antimicrobial peptides (AMPs) can exert bactericidal activities, they have not shown clinical efficacy. The limitations of native peptides may be overcome with rational design and synthesis. Here, we provide evidence that the bactericidal activity of a synthetic peptide, GW-Q6, against Pseudomonas aeruginosa is mediated through outer membrane protein OprI. Hyperpolarization/depolarization of membrane potential and increase of membrane permeability were observed after GW-Q6 treatment. Helical structure as well as hydrophobicity was induced by an amphipathic surfactant, sarkosyl, for binding to OprI and possible to membrane. NMR studies demonstrated GW-Q6 is an amphipathic α-helical structure in DPC micelles. The paramagnetic relaxation enhancement (PRE) approach revealed that GW-Q6 orients its α-helix segment (K7-K17) into DPC micelles. Additionally, this α-helix segment is critical for membrane permeabilization and antimicrobial activity. Moreover, residues K3, K7, and K14 could be critical for helical formation and membrane binding while residues Y19 and W20 for directing the C-terminus of the peptide to the surface of micelle. Taken together, our study provides mechanistic insights into the mode of action of the GW-Q6 peptide and suggests its applicability in modifying and developing potent AMPs as therapeutic agents.

Unlike USA300, a strain of community-acquired methicillin-resistant Staphylococcus aureus (MRSA), commensal Staphylococcus aureus (S. aureus) bacteria isolated from human skin demonstrated the ability to mediate the glycerol fermentation to produce short-chain fatty acids (SCFAs). Quantitative proteomic analysis of enzymes involved in glycerol fermentation demonstrated that the expression levels of six enzymes, including glycerol-3-phosphate dehydrogenase (GPDH) and phosphoglycerate mutase (PGM), in commensal S. aureus are more than three-fold higher than those in USA300. Western blotting validated the low expression levels of GPDH in USA300, MRSA252 (a strain of hospital-acquired MRSA), and invasive methicillin-susceptible S. aureus (MSSA). In the presence of glycerol, commensal S. aureus effectively suppressed the growth of USA300 in vitro and in vivo. Active immunization of mice with lysates or recombinant α-hemolysin of commensal S. aureus or passive immunization with neutralizing sera provided immune protection against the skin infection of USA300. Our data illustrate for the first time that commensal S. aureus elicits both innate and adaptive immunity via glycerol fermentation and systemic antibody production, respectively, to fight off the skin infection of pathogenic MRSA.

The absorption coefficient at 1.4 eV is divided by the value at 0.9 eV to obtain the factor used to judge the quality of μc-Si:H. PV device performance can be predicted by multiplying Voc with Isc when using this layer as an intrinsic layer. The results show a good relationship between the quality factor and the product of open-circuit voltage and short-circuit current. However, the final efficiency is influenced by the identities of the interface in the multilayer structure.

This article describes the care experience of an inhalation injury case successfully weaned from the ventilator. Data was collected from March 1, 2022, to March 11, 2022, using Gordon's Functional Health Patterns Assessment, including interviews, physical assessments, and medical history research. The identified health problems were inactive respiratory clearance, pain, and anxiety. During the nursing period, the author instructed the patient in diaphragmatic breathing; employed high-frequency chest wall oscillation therapy; taught effective coughing techniques to promote mucus clearance and maintain airway patency; and facilitated early separation from the ventilator through chest rehabilitation exercises. Implement prioritized pain relief and appropriate sedation. Appropriate wound dressings and securement techniques were chosen to facilitate healing and prevent dislodgment of tubes, particularly in the treatment of facial burn wounds. Utilizing body language to facilitate parent-child interaction and using electronic writing tablets to encourage the expression of anxious emotions. The health problems of critically ill patients are multifaceted, and interdisciplinary collaboration among various specialties is essential to provide the most appropriate and comprehensive care, facilitating a successful recovery.
