1. Optimizing treatment administration strategies using negative mNGS results in corticosteroid-sensitive diffuse parenchymal lung diseases

Abstract

Summary: Timely adjustments of antibiotic and corticosteroid treatments are vital for patients with diffuse parenchymal lung diseases (DPLDs). In this study, 41 DPLD patients with negative metagenomic next-generation sequencing (mNGS) results who were responsive to corticosteroids were enrolled. Among these patients, about 26.8% suffered from drug-induced DPLD, while 9.8% presented autoimmune-related DPLD. Following the report of the negative mNGS results, in 34 patients with complete antibiotics administration profiles, 79.4% (27/34) patients discontinued antibiotics after receiving negative mNGS results. Moreover, 70.7% (29/41) patients began or increased the administration of corticosteroid upon receipt of negative mNGS results. In the microbiota analysis, Staphylococcus and Stenotrophomonas showed higher detection rates in patients with oxygenation index (OI) below 300, while Escherichia and Stenotrophomonas had higher abundance in patients with pleural effusion. In summary, our findings demonstrated the clinical significance of mNGS in assisting the antibiotic and corticosteroid treatment adjustments in corticosteroid-responsive DPLD. Lung microbiota may imply the severity of the disease.

Abstract

This study investigated the effects of nitrogen form on root activity and nitrogen uptake kinetics of Camellia oleifera Abel. seedlings, providing a scientific basis for improving nitrogen use efficiency and scientific fertilization in C. oleifera production. Taking one-year-old C. oleifera cultivar ‘Xianglin 27’ seedlings as subjects, 8 mmol·L−1 of nitrogen in varied forms (NO3−:NH4+ = 0:0, 10:0, 7:3, 5:5, 3:7, 0:10) was applied in this study as the treatment conditions to investigate the effects of different nitrogen forms on root activity and nitrogen uptake kinetics in C. oleifera seedlings. Comparing the performance of nutrient solutions with different NO3−:NH4+ ratios, the results showed that a mixed nitrogen source improved the root activity of C. oleifera seedlings based on total absorption area, active absorption area, active absorption area ratio, specific surface area, and active specific surface area. When NO3−:NH4+ = 5:5, the total absorption area and active absorption area of the seedling roots reached the maximum. The results of uptake kinetic parameters showed that Vmax NH4+ > Vmax NO3− and Km NO3− > Km NH4+, indicating that the uptake potential of ammonium–nitrogen by C. oleifera seedlings is greater than that of nitrate–nitrogen. The conclusion was that compared to either ammonium– or nitrate–nitrogen, the mixed nitrogen source was better for promoting the root activity of C. oleifera seedlings, and the best nitrate/ammonium ratio was 5:5.

Abstract

Polyacrylamide (PAM) nanofibers was spun by electrospinning. In order to increase the molecular weight of the PAM, It has been cross linked by Glutaraldehyde solution. By means of scanning electron microscopy (SEM), the effect of electrospun technological parameters on the fibers’ morphology were researched. The parameters include the concentration of PAM spinning solution, the applied voltage and tip-collector distance (TCD). The PAM nanofibers of uniform thickness with the fibers’ diameter distribution of 80 ~ 160nm were got under the conditions as followers: the concentration of 16 wt.%, applied voltage of 23 kV, the TCD of 13cm.
