Abstract It has been known that resonances often occur when high‐voltage direct current (HVDC) systems are connected into the AC system via voltage‐source converters (VSCs). The resonances may lead to system failure and even damage primary equipment under some severe conditions. However, the existing damping methods are inadequate to ensure the system is always stable. To solve it, this article introduces the passive impedance adapter (PIA) in parallel with VSC to compensate the negative impedance of VSC‐HVDC system especially at mid‐ and high‐frequency ranges. A novel design and implementation method for the PIA is then proposed with the reduced volume and power loss to ensure the whole system impedance always positive. By this way, no resonance exists and the system is stable under any practical condition, such as the different cable lengths and impedances. Finally, a simulation platform is built to verify the effectiveness of the proposed PIA method in compensating the VSC‐HVDC impedance.

Abstract Chloroplast genomes, pivotal for understanding plant evolution, remain unexplored in Rutaceae, a family with key perennial crops like citrus. Leveraging next-generation sequencing data from 509 Rutaceae accessions across 15 species, we conducted a de novo assembly of 343 chloroplast genomes, unveiling a chloroplast variation map highlighting the heterogeneous evolution rates across genome regions. Notably, differences in chloroplast genome size primarily originate from large single-copy and small single-copy regions. Structural variants predominantly occurred in the single-copy region, with two insertions located at the single-copy and inverted repeat region boundary. Phylogenetic analysis, principal component analysis, and population genetic statistics confirmed the cohesive clustering of different Citrus species, reflecting evolutionary dynamics in Citrus diversification. Furthermore, a close chloroplast genetic affinity was revealed among Atalantia (previously regarded as primitive citrus), Clausena, and Murraya. Zanthoxylum formed a distinct group with heightened genetic diversity. Through expanding our analysis to include 34 published chloroplast genomes, we explored chloroplast gene selection, revealing divergent evolutionary trends in photosynthetic pathways. While Photosystem I and Photosystem II exhibited robust negative selection, indicating stability, the Nicotinamide adenine dinucleotide (NADH) dehydrogenase pathway demonstrated rapid evolution, which was indicative of environmental adaptation. Finally, we discussed the effects of gene length and GC content on chloroplast gene evolution. In conclusion, our study reveals the genetic characterization of chloroplast genomes during Rutaceae diversification, providing insights into the evolutionary history of this family.

The apolipoprotein E (APOE) ɛ4 allele is a recognized genetic risk factor for Alzheimer’s Disease (AD). Studies have shown that APOE ɛ4 mediates the modulation of intrinsic functional brain networks in cognitively normal individuals and significantly disrupts the whole-brain topological structure in AD patients. However, how APOE ɛ4 regulates brain functional connectivity (FC) and consequently affects the levels of cognitive impairment in AD patients remains unknown. In this study, we systematically analyzed functional magnetic resonance imaging (fMRI) data from two distinct cohorts: an In-house dataset includes 59 AD patients (73.37 ± 6.42 years), and the ADNI dataset includes 117 AD patients (74.91 ± 7.91 years). Experimental comparisons were conducted by grouping AD patients based on both APOE ɛ4 status and cognitive impairment levels of AD. Network-Based Statistic (NBS) method and the Graph Neural Network Explainer (GNN-Explainer) were combined to identify significant FC changes across different comparisons. Importantly, the GNN-Explainer method was introduced as an enhancement over the NBS method to better model complex high-order nonlinear characteristics for discovering FC features that significantly contribute to classification tasks. The results showed that APOE ɛ4 primarily influenced temporal lobe FCs, while it influenced different cognitive impairment levels of AD by adjusting prefrontal-parietal FCs. These findings were validated by p-values < 0.05 from NBS method, and 5-fold cross-validation along with ablation studies from the GNN-Explainer method. In conclusion, our findings provide new insights into the role of APOE ɛ4 in altering FC dynamics during the progression of AD, highlighting potential targets for early intervention.

This paper addresses the H∞ filtering problem for mean field stochastic differential systems that involve both state-dependent and disturbance-dependent noise. We assume that the state as well as the measurement output is distracted by an uncertain exogenous disturbance. Firstly, a sufficient condition for the stochastic-bounded real lemma is given. Next, H∞ filtering, which is built upon a stochastic-bounded real lemma, is put forward by two linear matrix inequalities. Furthermore, the validation of the theoretical analysis is demonstrated with two examples.

[Introduction] Due to the dense load, complex grid structure, and close electrical connections in the Guangdong power grid area, there are three major problems for a long time: the risk of large-scale power outages, the interaction between AC and DC, and the excessive short-circuit current, making it difficult to meet the needs of future power load development and flexible system regulation.[Method] To address the above issues, a high-performance VSC-HVDC (Voltage Source Converter-based High Voltage Direct Current) back-to-back project was constructed in the core area of Guangdong Power Grid, and a new generation of high-performance VSC-HVDC back-to-back technology was studied and adopted. The VSC-HVDC interconnection control of complex power grids, the development of highly reliable and high-performance converter valves, and the design of green and efficient converter stations were deeply studied. The upgrade of VSC-HVDC back-to-back technology was achieved. [Result] The project successfully achieved automatic control of asynchronous power flow, significantly reducing the link delay of the control and protection system, developed high-performance and highly reliable VSC-HVDC converter valves, and optimized the design in terms of noise reduction, loss reduction, water conservation, and other aspects of the converter station. A comprehensive perception system for the converter station was built, achieving intelligent operation and maintenance. At present, the project is running well. [Conclusion] The VSC-HVDC back-to-back project of Guangdong Power Grid has solved the three major stability problems faced by the multi DC feeder receiving end grid for a long time and enhanced the long-term safe and reliable power supply capacity of the Guangdong-Hong Kong-Macao Greater Bay Area.

A new theoretical-based constitutive model has been proposed in this study to concurrently predict the ageing and coupled creep-ageing behavior in both micro and macro ways of aluminum alloys under complex thermal and mechanical loading histories. The nucleation, growth and coarsening theories for age strengthening of aluminum alloys have been modified with the integration of dislocation-based theories raised by plastic or creep deformation to form the new theoretical-based creep-ageing modelling framework. Both the key microstructures (precipitates, solid solutes and dislocations) and macro properties (creep/stress-relaxation, yield strength and strengthening components) under various thermal and loading histories (e.g., single-step/multi-step isothermal and non-isothermal, pre-strain loaded) of a typical Al–Zn–Mg alloy have been effectively predicted and validated by the same set model. Furthermore, the capability of the developed model to reveal the detailed deformation and strengthening mechanisms with complex thermal and loading histories, and to assist the design of complex non-isothermal creep-ageing processes with optimization objectives of accurate shape and high strength in the formed alloys has been discussed and demonstrated.

Background: Lung adenocarcinoma (LUAD) stands as the foremost histological subtype of non-small-cell lung cancer, accounting for approximately 40% of all lung cancer diagnoses. However, there remains a critical unmet need to enhance the prediction of clinical outcomes and therapy responses in LUAD patients. Keratins (KRTs), serving as the structural components of the intermediate filament cytoskeleton in epithelial cells, play a crucial role in the advancement of tumor progression. This study investigated the prognostic significance of the KRT family gene and developed a KRT gene signature (KGS) for prognostic assessment and treatment guidance in LUAD. Methods: Transcriptome profiles and associated clinical details of LUAD patients were meticulously gathered from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases. The KGS score was developed based on the expression of five prognostic KRT genes (KRT7, KRT8, KRT17, KRT18, and KRT80), and the upper quartile of the KGS score was chosen as the cutoff. The Kaplan-Meier method was evaluated to compare survival outcomes between KGS-high and KGS-low groups. The underlying mechanism was further investigated by GSEA, GSVA, and other bioinformatic algorithms. Results: High expression of the KGS signature exhibited a robust association with poorer overall survival (OS) in the TCGA-LUAD dataset (HR: 1.81; 95% CI: 1.35–2.42, P = 0.00011). The association was further corroborated in three external GEO cohorts, including GSE31210 (HR: 3.31; 95% CI: 1.7–6.47, P = 0.00017), GSE72094 (HR: 1.95; 95% CI: 1.34–2.85, P = 0.00057) and GSE26939 (HR: 3.19; 95% CI: 1.74–5.84, P

Recently, the theory of semi-tensor product (STP) method of matrices has received much attention from variety communities covering engineering, economics and industries, etc. This paper describes a detailed survey on some recent applications of the STP method in finite systems. First, some useful mathematical tools on the STP method are provided. Second, many recent developments about robustness analysis on the given finite systems are delineated, such as robust stable analysis of switched logical networks with time-delayed, robust set stabilization of Boolean control networks, event-triggered controller design for robust set stabilization of logical networks, stability analysis in distribution of probabilistic Boolean networks, and how to solve a disturbance decoupling problem by event triggered control for logical control networks. Finally, several research problems in future works are predicted.

Abstract Background The detection of peritoneal metastasis (PM) is limited by current imaging tools. In this prospective study, we aimed to evaluate the sensitivity and specificity of peritoneal cell-free DNA (cfDNA) for diagnosis of PM. Methods Colorectal cancer (CRC) patients with/without PM were enrolled. The cfDNA experimental personnel and statists were blinded to the diagnosis of PM. Ultradeep sequencing covering large genomic regions (35000X, Next-generation sequencing) of cfDNA in peritoneal lavage fluid (FLD) and matched tumor tissues was performed. Results A total of 64 cases were recruited prospectively and 51 were enrolled into final analysis. In training cohort, 100% (17/17) PM patients obtained positive FLD cfDNA, comparing to 5/23 (21.7%) in patients without PM. Peritoneal cfDNA had a high sensitivity of 100% and specificity of 77.3% for diagnosis of PM (AUC: 0.95). In validation group of 11, 5/6 (83%) patients with PM obtained positive FLD cfDNA, comparing to 0/5 in non-PM (P = 0.031) with a sensitivity of 83.3% and specificity of 100%. Positive FLD cfDNA was associated with poor recurrence-free survival (P = 0.013) and was preceding radiographic evidence of recurrence. Conclusions Peritoneal cfDNA is a promising sensitive biomarker for earlier detection of PM in CRC than current radiological tools. It can potentially guide selection for targeted therapies and serve as a surrogate instead of laparoscopic explore in the future. When using ground-based synthetic aperture radar (GB-SAR) for monitoring open-pit mines, dynamic atmospheric conditions can interfere with the propagation speed of electromagnetic waves, resulting in atmospheric phase errors. These errors are particularly complex in rapidly changing weather conditions or steep terrain, significantly impacting monitoring accuracy. In such scenarios, traditional regression model-based atmospheric phase correction (APC) methods often become unsuitable. To address this issue, this paper proposes a clustering method based on the spatial autocorrelation function. First, the interferogram is uniformly divided into multiple blocks, and the phase consistency of each block is evaluated using the spatial autocorrelation function. Then, a region growing algorithm is employed to classify each block according to its phase pattern, followed by merging adjacent blocks based on statistical data. To verify the feasibility of the proposed method, both the traditional regression model-based method and the proposed method were applied to deformation monitoring of an open-pit mine in Northwest China. The experimental results show that for complex atmospheric phase scenarios, the proposed method significantly outperformed traditional methods, demonstrating its superiority.

Vulto-van Silfhout-de Vries syndrome (VSVS; MIM 615828) is an extremely rare autosomal dominant disorder with unknown incidence. It is always caused by de novo heterozygous pathogenic variants in the DEAF1 gene, which encodes deformed epidermal autoregulatory factor-1 homology. VSVS is characterized by mild to severe intellectual disability (ID) and/or global developmental delay (GDD), seriously limited language expression, behavioral abnormalities, somnipathy, and reduced pain sensitivity. In this study, we present a Chinese boy with moderate GDD and ID, severe expressive language impairment, behavioral issues, autism spectrum disorder (ASD), sleeping dysfunction, high pain threshold, generalized seizures, imbalanced gait, and recurrent respiratory infections as clinical features. A de novo heterozygous pathogenic missense variant was found in the 5th exon of DEAF1 gene, NM_021008.4 c.782G>C (p. Arg261Pro) variant by whole exome sequencing (WES). c.782G>C had not been previously reported in genomic databases and literature. According to the ACMG criteria, this missense variant was considered to be “Likely Pathogenic”. We diagnosed the boy with VSVS both genetically and clinically. At a follow-up of 2.1 years, his seizures were well controlled after valproic acid therapy. In addition, the child’s recurrent respiratory infections improved at 3.5 years of age, which has not been reported in previous individuals. Maybe the recurrent respiratory infections like sleep problems reported in the literature are not permanent but may improve naturally over time. The literature review showed that there were 35 individuals with 28 different de novo pathogenic variants of DEAF1-related VSVS. These variants were mostly missense and the clinical manifestations were similar to our patient. Our study expands the genotypic and phenotypic profiles of de novo DEAF1.

In this article, we study the Pareto optimal $ H_{2} $ /$ H_{\infty} $ filter design problem for a generalization of discrete-time stochastic systems. By constructing the estimation equation of the given systems with the estimated signal, a filter error estimation system is obtained. The aim is to obtain a gain matrix $ K^{\star} $ that optimizes both performance indicators we set. To deal with this problem, two different upper bounds for two performance indicators are given respectively. The optimal problem therefore is transformed into a Pareto optimal problem with linear matrix inequalities ($ LMIs $) which can be addressed through the $ LMI $ toolbox in $ MATLAB $.

Objectives Primary tumor tissue is often analyzed to search for predictive biomarkers and DNA-guided personalized therapies, but there is an incomplete understanding of the discrepancies in the genomic profiles between primary tumors and metastases, such as liver and lung metastases. Methods We performed in-depth targeted next-generation sequencing of 520 key cancer-associated genes for 47 matched primary and metastatic tumor samples which were retrospectively collected. Results A total of 699 mutations were detected in the 47 samples. The coincidence rate of primary tumors and metastases was 51.8% (n = 362), and compared to patients with liver metastases, patients with lung metastases had a significantly greater coincidence rate ( P = .021). The number of specific mutations for the primary tumors and liver and lung metastases was 186 (26.6%), 122 (17.5%), and 29 (4.1%), respectively. Analysis of a patient with all three occurrences, including a primary tumor, liver metastasis, and lung metastasis, indicated a possible polyclonal seeding mechanism for liver metastases. Remarkably, multiple samples from patients with primary and metastatic tumors supported a mechanism of synchronous parallel dissemination from primary tumors to metastatic tumors that were not mediated through pre-metastatic tumors. We also found that the PI3K-Akt signaling pathway significantly altered lung metastases compared to matched primary tumors ( P = .001). In addition, patients with mutations in CTCF, PIK3CA, or TP53 and LRP1B, AURKA, FGFR1, ATRX, DNMT3B, or GNAS had larger primary tumor sizes and metastases, especially patients with both LRP1B and AURKA mutations. Interestingly, CRC patients with TP53 -disruptive mutations were more likely to have liver metastases ( P = .016). Conclusion In this study, we demonstrate significant differences in the genomic landscapes of colorectal cancer patients based on the site of metastasis. Notably, we observe a larger genomic variation between primary tumors and liver metastasis compared to primary tumors and lung metastasis. These findings can be used for tailoring treatments based on the specific metastatic site.

The micro-deformation monitoring radar is usually based on Permanent Scatterer (PS) technology to realize deformation inversion. When the region is continuously monitored for a long time, the radar image amplitude and pixel variance will change significantly with time. Therefore, it is difficult to select phase-stable scatterers by conventional amplitude deviation methods, as they can seriously affect the accuracy of deformation inversion. For different regions studied within the same scenario, using a PS selection method based on the same threshold often increases the size of the deformation error. Therefore, this paper proposes a new PS selection method based on the Gaussian Mixture Model (GMM). Firstly, PS candidates (PSCs) are selected based on the pixels’ amplitude information. Then, the amplitude deviation index of each PSC is calculated, and each pixel’s probability values in different Gaussian distributions are acquired through iterations. Subsequently, the cluster types of pixels with larger probability values are designated as low-amplitude deviation pixels. Finally, the coherence coefficient and phase stability of low-amplitude deviation pixels are calculated. By comparing the probability values of each of the pixels in different Gaussian distributions, the cluster type with the larger probability, such as high-coherence pixels and high-phase stability pixels, is selected and designated as the final PS. Our analysis of the measured data revealed that the proposed method not only increased the number of PSs in the group, but also improved the stability of the number of PSs between groups.

Abstract Background Sputum biopsies offer unique advantages such as non-invasiveness and convenient collection. The one investigation so far on sputum for genome profiling in advanced non-small cell lung cancer (aNSCLC) suggested promising performance. However, it remains undefined whether clinicohistologic characteristics were associated with performance and how this knowledge could help guide choice of liquid biopsy. Methods Targeted sequencing with a 520-gene panel was performed on prospectively collected matched tumor tissue (TIS), plasma (PLA), and sputum supernatant (SPU) from 71 aNSCLC patients (NCT05034445). Genomic alteration detection was characterized in a series of aspects and interrogated for association with 14 clinicohistologic features. Nomograms were constructed with logistic regression for predicting the liquid biopsy type with greater sensitivity. Results Compared with PLA, SPU showed comparable quality control metrics, mutation detection rate (SPU: 67.6%, PLA: 70.4%), concordance with tumor tissue (67.6% vs. 73.2%), and correlation with tissue-based tumor mutation burden levels (r = 0.92 vs. 0.94). For driver alterations, detection was less sensitive with SPU (50.0%) than PLA (63.5%) in the entire cohort but similarly or more sensitive in patients with centrally located lung tumors or smoking history or for altered ALK or KRAS. Two nomograms were constructed and enabled predicting the probability of superior sensitivity with SPU with moderate to borderline high accuracy. Conclusion In addition to demonstrating comparable performance in multiple aspects, this study is the first to propose nomograms for choosing liquid biopsy based on clinicohistologic characteristics. Future research is warranted to delineate the clinical utility of sputum for genome profiling.

Cell adhesion molecules (CAM) are crucial in several pathological inflammation processes in Alzheimer’s disease (AD). However, their potential for clinical diagnostics remains unknown. The present investigation evaluated the clinical significance of ALCAM, VCAM-1, NCAM, and ICAM-1 levels in the plasma of participants with cognitive impairment (44 patients with mild cognitive impairment, 71 patients with Alzheimer’s dementia, and 18 patients with other dementia) and 28 controls with normal cognitive ability. We also detected plasma levels of multiple inflammatory factors (IFN-gamma, IL-18, IL-1beta, IL-13, IL-8, IL-7, CCL11, MCP-1, TSLP, IL-10, BDNF, IL-17, IL-5, TREM-1) using Multiplex liquid chip and plasma levels of Abeta1-42 and Abeta1-40 using liquid-phase flow cytometry (FCM). Our findings demonstrated a correlation of ALCAM and VCAM-1 with age, the severity of cognitive decline, and MTA, but no significant difference between groups for NCAM and ICAM-1. ALCAM and VCAM-1 both demonstrated a positive correlation with the degree of atrophy in the medial temporal lobe structure. Further analysis revealed no significant correlation in plasma between VCAM-1, ALCAM and Abeta1-40, Abeta1-42. Nevertheless, there was a significant correlation between VCAM-1, ALCAM and many inflammatory factors. Furthermore, the predictive value of ALCAM and VCAM-1 for AD was assessed using a multi-parameter regression model. ALCAM and VCAM-1 in combination with ApoE4, education, age, and MMSE could predict AD with high precision (AUC=0.891; AIC=146.9) without imaging diagnosis. ALCAM and VCAM-1 combination improved the predictive accuracy significantly. In a nutshell, these findings revealed ALCAM and VCAM-1 as reliable indicators of Alzheimer’s disease.

Abstract Growing efforts are being invested in investigating various molecular approaches to detect minimal residual disease (MRD) and predict disease recurrence. In our study, we investigated the utility of parallel longitudinal analysis of mutation and DNA methylation profiles for predicting MRD in postoperative non‐small‐cell lung cancer (NSCLC) patients. Tumor tissues and longitudinal blood samples were obtained from 65 patients with resected stage IA‐IIIB NSCLC. Somatic mutation and DNA methylation profiling were performed using ultra‐deep targeted sequencing and targeted bisulfite sequencing, respectively. Dynamic changes in plasma‐based mutation and tumor‐informed methylation profiles, reflected as MRD score, were observed from before surgery (baseline) to postoperative follow‐up, reflecting the decrease in tumor burden of the patients with resected NSCLC. Mutations were detected from plasma samples in 63% of the patients at baseline, which significantly reduced to 23‐25% during post‐operative follow‐ups. MRD score positive rate was 95.7% at baseline, which reduced to 74% at the first and 70% at the second follow‐up. Among the 5 relapsed patients with parallel longitudinal analysis of mutation and methylation profile, elevated MRD score was observed at follow‐up between 0.5‐7 months prior to radiologic recurrence for all 5 patients. Of them, 4 patients also had concomitant increase in allelic fraction of mutations in at least 1 follow‐up time point, but one patient had no mutation detected throughout all follow‐ups. Our results demonstrate that longitudinal profiling of mutation and DNA methylation may have potential for detecting MRD and predicting recurrence in postoperative NSCLC patients.

Abstract Background The combination of bevacizumab and epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) could prolong progression-free survival (PFS) in patients with EGFR-mutant advanced non-small-cell lung cancer (NSCLC). Our study investigated the clinical and molecular factors that affect the efficacy of first-generation EGFR-TKI with or without bevacizumab and identify the subset of patients who can benefit from combination therapy. Methods Our study included 318 patients with EGFR-mutant locally advanced/advanced NSCLC treated with either first-generation EGFR-TKI combined with bevacizumab (A+T; n = 159) or EGFR-TKI monotherapy (T; n = 159). Two nomogram models to predict PFS and overall survival (OS), respectively, were constructed using two factors that impact EGFR-TKI efficacy: metastatic site and presence of concurrent mutations. The study cohort was stratified into 2 cohorts for training (n = 176) and validation (n = 142) of the nomogram model. Using the median score from the nomogram, the patients were stratified into two groups to analyze their survival outcome. Results The A+T group had significantly longer PFS (14.0 vs. 10.5 months; p

This study has developed, ultrasound-assisted, a novel food packaging film (U-zein/SiO2) for food packaging applications. Incorporating an optimal concentration of 18 mg/mL of nano-SiO2 and subjecting the film to 10 min of ultrasonic treatment resulted in a remarkable increase of 32.89% in elongation at break and 55.86% in tensile strength. In addition, the incorporation of nano-SiO2 effectively reduces the water content and solubility of the composite film, resulting in improved water/oxygen barrier properties. These physiochemical properties were further improved with the application of ultrasound. The analysis of attenuated total reflectance-Fourier transform infrared, X-ray diffraction, differential scanning calorimetry, and scanning electronic microscope demonstrated that the ultrasound treatment improved the hydrogen bonds, improved thermal stability, molecular arrangement, structure stability, and intermolecular compatibility of the composite film, resulting in enhanced physio-mechanical properties of the film. In addition, the ultrasound treatment led to a smoother film surface and reduced the pores on the film’s cross-section. Moreover, the U-zein/SiO2 film exhibited excellent mechanical and water/oxygen barrier properties in different storage environments over a period of 30 days. These results offer sound theoretical support for the practical application of the prepared preservative film.

Photodynamic therapy (PDT) is an innovative and perspective antineoplastic therapy. Tetra-α-(4-carboxyphenoxy) phthalocyanine zinc (TαPcZn)-mediated PDT (TαPcZn-PDT) has shown antitumor activity in some tumor cells, but the manner in which caspase-1 is involved in the regulation of apoptosis and pyroptosis in the TαPcZn-PDT-treated breast cancer MCF-7 cells is unclear. Therefore, effects of TαPcZn-PDT on cytotoxicity, cell viability, apoptosis, pyroptosis, cellular reactive oxygen species (ROS), mitochondrial membrane potential (ΔΨm), caspase-1, caspase-3, and nuclear transcription factor-κB (NFκB) in MCF-7 cells was firstly examined in the present study. The findings demonstrated that TαPcZn-PDT resulted in the increase in cytotoxicity and the percentage of apoptotic and pyroptotic cells, the reduction in cell viability and ΔΨm, the production of ROS and the activation of caspase-1, caspase-3 and NFκB in MCF-7 cells. Furthermore, the results also revealed that siRNA-targeting caspase-1 (siRNA-caspase-1) attenuated the effect of TαPcZn-PDT on apoptosis, pyroptosis and the activation of caspase-1, caspase-3 and NFκB in MCF-7 cells. Abstract The aim of this study is to establish the expression of ATP-sensitive potassium channels(KATP) in human umbilical arteries with severe pre-eclampsia. Real-time quantitative PCR and western blotting were used to detect the mRNA and protein expression levels of KATP channel subunits Kir6.1 and SUR2B in human umbilical arteries from normal pregnant and those with severe pre-eclampsia, early onset severe pre-eclampsia and late onset severe pre-eclampsia. The mRNA and protein levels of SUR2B in the severe pre-eclampsia group were lower than those in the normal group (P 0.05). The mRNA and protein levels of SUR2B in early onset severe pre-eclampsia group were lower than those in late onset severe pre-eclampsia group (P 0.05). The mRNA and protein expression levels of SUR2B in pregnant women with severe pre-eclampsia were lower than those in normal pregnant women, suggesting that the expression of the SUR2B of the KATP channel may be related to the occurrence and development of severe pre-eclampsia. Compared with late onset severe pre-eclampsia, the mRNA and protein expression levels of SUR2B were lower in the umbilical arteries of women with early onset severe pre-eclampsia, suggesting that the occurrence time of severe pre-eclampsia may be related to the extent reduced expression of the SUR2B of the KATP channel.

Manufacturing process and human operational errors may cause small-sized defects, such as cracks, over-welding, and black edges, on solar cell surfaces. These surface defects are subtle and, therefore, difficult to observe and detect. Accurate detection and replacement of defective battery modules is necessary to ensure the energy conversion efficiency of solar cells. To improve the adaptability to the scale changes of various types of surface defects of solar cells, this study proposed a multi-feature region proposal fusion network (MF-RPN) structure to detect surface defects. In such a network, region proposals are extracted from different feature layers of convolutional neural networks. Additionally, considering that multiple aspect ratios and scale settings and the use of multiple RPNs, result in an overlap of candidate regions and lead to information redundancy, we designed a multiscale region proposal selection strategy (MRPSS) to reduce the number of region proposals and improve network accuracy. Owing to the complete learning of shallow-detail texture information and deep semantic information, our multiscale RPN fusion structure can effectively improve an object’s multiscale feature extraction ability for various scales and types of surface defects of solar cells. Experimental results demonstrate that our method outperforms other methods by achieving a higher detection accuracy.

This article is concerned with the problem of delay-dependent stabilization for a class of stochastic Markov systems with event-triggered feedback control. An event-triggered mechanism (ETM) is proposed with the purpose of effectively reducing the transmissions of redundant massages, in which the generation of sensor sampling and control actuation is not periodic but only when some event-driven conditions are satisfied. In the meanwhile, a novel Lyapunov-Krasovskii functional (LKF) is applied to the closed-loop systems to establish the criterion of practically exponential mean-square stability. And a positive lower bound on the inter-execution times is guaranteed, that is, the Zeno behavior will not happen under this ETM. Furthermore, the event-triggered feedback controller can be constructed by solving the relevant linear matrix inequalities (LMIs). In the end, a numerical example displays the feasibility of our results.

The precise adjustment of handedness of helical architectures is important to regulate their functions. Macroscopic chirality inversion has been achieved in organic supramolecular systems by pH, metal ions, solvents, chiral and non-chiral additives, temperature, and light, but rarely in coordination polymers (CPs). In particular, salt-assisted macroscopic chirality inversion has not been reported. In this work, we carried out a systematic investigation on the role of pH and salt in regulating the morphology of CPs based on Gd(NO3)3 and R-(1-phenylethylamino)methylphosphonic acid (R-pempH2). Without extra NO3−, the chirality inversion from the left-handed superhelix R-M to the right-handed superhelix R-P can be achieved by pH modulation from 3.2 to 3.8. The addition of NaNO3 (2.0 eq) at pH 3.8 results in an inversion of chiral sense from R-P to R-M as a pure phase. To our knowledge, this is the first example of salt-assisted macroscopic helical inversion in artificial systems.

Pine wilt disease (PWD) is one of the most destructive forest diseases that has led to rapid wilting and mortality in susceptible host pine trees. Spatially explicit detection of pine wood nematode (PWN)-induced infestation is important for forest management, policy making, and practices. Previous studies have mapped forest disturbances in response to various forest diseases and/or insects over large areas using remote-sensing techniques, but these efforts were often constrained by the limited availability of ground truth information needed for the calibration and validation of moderate-resolution satellite algorithms in the process of linking plot-scale measurements to satellite data. In this study, we proposed a two-level up-sampling strategy by integrating unmanned aerial vehicle (UAV) surveys and high-resolution Radarsat-2 satellite imagery for expanding the number of training samples at the 30-m resampled Sentinel-1 resolution. Random forest algorithms were separately used in the prediction of the Radarsat-2 and Sentinel-1 infestation map induced by PWN. After data acquisition in Muping District during August and September 2021, we first verified the ability of a deep-learning-based object detection algorithm (i.e., YOLOv5 model) in the detection of infested trees from coregistered UAV-based RGB images (Average Precision (AP) of larger than 70% and R2 of 0.94). A random forest algorithm trained using the up-sampling UAV infestation map reference and corresponding Radarsat-2 pixel values was then used to produce the Radarsat-2 infestation map, resulting in an overall accuracy of 72.57%. Another random forest algorithm trained using the Radarsat-2 infestation pixels with moderate and high severity (i.e., an infestation severity of larger than 0.25, where the value was empirically set based on a trade-off between classification accuracy and infection detectability) and corresponding Sentinel-1 pixel values was subsequently used to predict the Sentinel-1 infestation map, resulting in an overall accuracy of 87.63%, where the validation data are Radarsat-2 references rather than UAV references. The Sentinel-1 map was also validated by independent UAV surveys, with an overall accuracy of 76.30% and a Kappa coefficient of 0.45. We found that the expanded training samples by the integration of UAV and Radarsat-2 strengthened the medium-resolution Sentinel-1-based prediction model of PWD. This study demonstrates that the proposed method enables effective PWN infestation mapping over multiple scales.

Abstract Background Abaxially anthocyanic leaves of deeply-shaded understorey plants play important ecological significance for the environmental adaption. In contrast to the transient pigmentation in other plants, anthocyanins are permanently presented in these abaxially red leaves, however, the mechanism for the pigment maintenance remains unclear. In the present study, we investigated phenolic metabolites that may affect pigment stability and degradation in Excoecaria cochinchinensis (a bush of permanently abaxial-red leaves), via a comparison with Osmanthus fragrans (a bush of transiently red leaves). Results High levels of galloylated anthocyanins were identified in the Excoecaria but not in the Osmanthus plants. The galloylated anthocyanin showed slightly higher stability than two non-galloylated anthocyanins, while all the 3 pigments were rapidly degraded by peroxidase (POD) in vitro. High levels of hydrolysable tannins [mainly galloylglucoses/ellagitannins (GGs/ETs)] were identified in Excoecaria but none in Osmanthus. GGs/ETs showed inhibition effect on POD, with IC50 ranged from 35.55 to 83.27 μM, correlated to the markedly lower POD activities detected in Excoecaria than in Osmanthus. Strong copigmentation was observed for GGs/ETs and anthocyanins, with more than 30% increase in the red intensity of non-galloylated anthocyanin solutions. In the leaf tissue, the hydrolysable tannins were observed to be co-localized with anthocyanins at the abaxial layer of the Excoecaria leaves, correlated to the low POD activity, more acidity and increased red intensity of the tissue. Conclusion The results suggest that the Excoecaria leaves accumulate a distinct group of phenolic metabolites, mainly GGs/ETs, at the abaxial layer, which prevent anthocyanin degradation and increase the pigment stability, and consequently lead to the permanent maintenance of the red leaves.

This paper focuses on anti-synchronization and synchronization of a stochastic multi-coupled chaotic system with ring connection and control schemes. First, the system is simplified by means of the formula deformation technique and the controller. Based on the Lyapunov method and stochastic differential theory, some sufficient conditions are obtained by some control methods. At last, the Chen system, Lorenz system, and Lü system with stochastic perturbations are used to verify the correctness of the conclusions.

Previous studies found patients with POLE exonuclease domain mutations (EDMs) in targeted exons were related to significant better outcomes in stage II-III colorectal cancer (CRC). The detailed mutational profile of the entire POLE exonuclease domain, tumor mutation burden (TMB) and immune cell infiltration in POLE EDMs tumors, and the prognostic value of such mutations in stage II CRCs were largely unknown to us. This study was to clarify the characteristics, immune response and prognostic value of somatic POLE EDMs in stage II CRC. A total of 295 patients with stage II CRC were sequenced by next-generation sequencing with a targeted genetic panel. Simultaneous detection of the immune cells was conducted using a five-color immunohistochemical multiplex technique. The detailed molecular characteristics, tumor-infiltrating lymphocyte (TIL) and prognostic effect of POLE EDMs in stage II CRC were analyzed. BackgroundEarly onset colorectal cancer (EO CRC) is a heterogeneous colorectal cancer subtype with obvious hereditary tendencies and increasing incidence. We sought to determine the susceptibility genes and molecular characteristics of EO CRC.Methods330 EO metastatic CRC (mCRC) (≤55 years) and 110 average-onset (AO) mCRC patients (>55 years) were enrolled. Capture-based targeted sequencing was performed on tumor tissue and paired white blood cells using a sequencing panel of 520 genes. The association between molecular alterations and overall survival (OS) was analyzed.ResultsOf the 330 EO mCRC patients, 31 carried pathogenic or likely pathogenic germline mutations, with 16 of them diagnosed with lynch syndrome. Fifteen patients had germline mutations in non-mismatch repair genes, including four in MUTHY, three in RAD50, one in TP53, and eight in other genes. Twenty-nine genes were recurrently mutated in EO mCRC, including TP53, APC, KRAS, SMAD4, and BRCA2. The majority of genomic alterations were comparable between EO and AO mCRC. EO mCRC patients were more likely to have a high tumor mutation burden (p < 0.05). RNF43, RBM10, TSC, and BRAF V600E mutations were more commonly observed in EO mCRC, while APC, ASXL1, DNMT3B, and MET genes were more commonly altered in AO patients. At the pathway level, the WNT pathway was the only differentially mutated pathway between EO and AO mCRC (p < 0.0001). The wild-type WNT pathway (p = 0.0017) and mutated TGF-β pathway (p = 0.023) were associated with unfavorable OS in EO mCRC.ConclusionsApproximately one in 10 EO mCRC was associated with hereditary tumors. The spectrum of somatic alterations was largely comparable between EO and AO mCRC with several notable differences.

Abstract Background Hepatocellular carcinoma (HCC) is the one of the most common cancers and lethal diseases in the world. DNA methylation alteration is frequently observed in HCC and may play important roles in carcinogenesis and diagnosis. Methods Using the TCGA HCC dataset, we classified HCC patients into different methylation subtypes, identified differentially methylated and expressed genes, and analyzed cis- and trans-regulation of DNA methylation and gene expression. To find potential diagnostic biomarkers for HCC, we screened HCC-specific CpGs by comparing the methylation profiles of 375 samples from HCC patients, 50 normal liver samples, 184 normal blood samples, and 3780 samples from patients with other cancers. A logistic regression model was constructed to distinguish HCC patients from normal controls. Model performance was evaluated using three independent datasets (including 327 HCC samples and 122 normal samples) and ten newly collected biopsies. Results We identified a group of patients with a CpG island methylator phenotype (CIMP) and found that the overall survival of CIMP patients was poorer than that of non-CIMP patients. Our analyses showed that the cis-regulation of DNA methylation and gene expression was dominated by the negative correlation, while the trans-regulation was more complex. More importantly, we identified six HCC-specific hypermethylated sites as potential diagnostic biomarkers. The combination of six sites achieved ~ 92% sensitivity in predicting HCC, ~ 98% specificity in excluding normal livers, and ~ 98% specificity in excluding other cancers. Compared with previously published methylation markers, our markers are the only ones that can distinguish HCC from other cancers. Conclusions Overall, our study systematically describes the DNA methylation characteristics of HCC and provides promising biomarkers for the diagnosis of HCC.

Abstract Background To explore the influences of prenatal antibiotic exposure, the intensity of prenatal and postnatal antibiotic exposure on gut microbiota of preterm infants and whether gut microbiota and drug resistant strains in the neonatal intensive care unit (NICU) over a defined period are related. Methods Among 28 preterm infants, there were two groups, the PAT (prenatal antibiotic therapy) group (12 cases), and the PAF (prenatal antibiotic free) group (12 cases). Fecal samples from both groups were collected on days 7 and 14. According to the time of prenatal and postnatal antibiotic exposure, cases were divided into two groups, H (high) group (11 cases) and L (low) group (11 cases), and fecal samples on day 14 were collected. Genomic DNA was extracted from the fecal samples and was subjected to high throughput 16S rRNA amplicon sequencing. Bioinformatics methods were used to analyze the sequencing results. In this paper, the bipartite consensus problem is investigated for multiagent systems with second-order dynamics and antagonistic interactions. We consider bipartite consensus under two kinds of protocols. One is an absolute damping protocol, and the other is a relative damping protocol. When there are no time delays, necessary and sufficient conditions are derived for the system to asymptotically achieve the bipartite consensus under the given protocols. When there are communication delays, sufficient conditions are obtained for the system to asymptotically achieve the bipartite consensus. In case G(A) is structurally unbalanced, consensus can also be asymptotically achieved, while this case is trivial.

Based on the LaSalle invariant principle of stochastic differential equations and the graph theory, several sufficient conditions are obtained by adaptive feedback control and impulsive control. The error dynamical system is globally almost surely asymptotically stable. The conclusion is new and feasible. Finally, an example is given to show the validity of the results.

To investigate the changes of physiological and biochemical indexes, male mice were fed a regular diet or short time high fat diet (HFD) for 10 days with oral administration of saline, metformin, resveratrol, or injected intraperitoneally (ip) with digoxin respectively every day. Food intake and body weight were recorded simultaneously. Blood was collected after mice were sacrificed and then tested with commercial kits. The data manifested that metformin and resveratrol only ameliorate free fatty acids and glycerol in HFD-fed mice. Stabilizing multiagent systems including unstable agents shows the advantage of cooperation. This paper addresses the problem of stabilization of heterogeneous multiagent systems. Under cycle graphs, a sufficient condition for the stabilization problem via harmonic control is provided and an algorithm of designing the interconnection gains is presented. Furthermore, a sufficient and necessary condition for stabilization problem via harmonic control under cycle graphs is first given when the graph contains less than 5 nodes.

In this paper, exponential stability and robust H∞ control problem are investigated for a class of discrete-time time-delay stochastic systems with infinite Markov jump and multiplicative noises. The jumping parameters are modeled as an infinite-state Markov chain. By using a novel Lyapunov-Krasovskii functional, a new sufficient condition in terms of matrix inequalities is derived to guarantee the mean square exponential stability of the equilibrium point. Then some sufficient conditions for the existence of feedback controller are presented to guarantee that the resulting closed-loop system has mean square exponential stability for the zero exogenous disturbance and satisfies a prescribed H∞ performance level. Numerical simulations are exploited to validate the applicability of developed theoretical results.

BackgroundLysine acetylation is a crucial type of protein post-translational modification, which is involved in many important cellular processes and serious diseases. However, identification of protein acetylated sites through traditional experiment methods is time-consuming and laborious. Those methods are not suitable to identify a large number of acetylated sites quickly. Therefore, computational methods are still very valuable to accelerate lysine acetylated site finding.ResultIn this study, many biological characteristics of acetylated sites have been investigated, such as the amino acid sequence around the acetylated sites, the physicochemical property of the amino acids and the transition probability of adjacent amino acids. A logistic regression method was then utilized to integrate these information for generating a novel lysine acetylation prediction system named LAceP. When compared with existing methods, LAceP overwhelms most of state-of-the-art methods. Especially, LAceP has a more balanced prediction capability for positive and negative datasets.ConclusionLAceP can integrate different biological features to predict lysine acetylation with high accuracy.
