Purpose: Ischemia and hypoxia are the main factors limiting limb replantation and transplantation. Static cold storage (SCS), a common preservation method for tissues and organs, can only prolong limb ischemia time to 4 – 6 h. The normothermic machine perfusion (NMP) is a promising method for the preservation of tissues and organs, which can extend the preservation time in vitro by providing continuous oxygen and nutrients. This study aimed to evaluate the difference in the efficacy of the 2 limb preservation methods. Methods: The 6 forelimbs from beagle dogs were divided into 2 groups. In the SCS group (n = 3), the limbs were preserved in a sterile refrigerator at 4 °C for 24 h, and in the NMP group (n = 3), the perfusate prepared with autologous blood was used for the oxygenated machine perfusion at physiological temperature for 24 h, and the solution was changed every 6 h. The effects of limb storage were evaluated by weight gain, perfusate biochemical analysis, enzyme-linked immunosorbent assay, and histological analysis. All statistical analyses and graphs were performed using GraphPad Prism 9.0 one-way or two-way analysis of variance. The p value of less than 0.05 was considered to indicate statistical significance. Results: In the NMP group, the weight gained percentage was 11.72% ± 4.06%; the hypoxia-inducible factor-1α contents showed no significant changes; the shape of muscle fibers was normal; the gap between muscle fibers slightly increased, showing the intercellular distance of (30.19 ± 2.83) μm; and the vascular α-smooth muscle actin (α-SMA) contents were lower than those in the normal blood vessels. The creatine kinase level in the perfusate of the NMP group increased from the beginning of perfusion, decreased after each perfusate change, and remained stable at the end of perfusion showing a peak level of 4097.6 U/L. The lactate dehydrogenase level of the NMP group increased near the end of perfusion and reached the peak level of 374.4 U/L. In the SCS group, the percentage of weight gain was 0.18% ± 0.10%, and the contents of hypoxia-inducible factor-1α increased gradually and reached the maximum level of (164.85 ± 20.75) pg/mL at the end of the experiment. The muscle fibers lost their normal shape and the gap between muscle fibers increased, showing an intercellular distance of (41.66 ± 5.38) μm. The contents of vascular α-SMA were much lower in the SCS group as compared to normal blood vessels. Conclusions: NMP caused lesser muscle damage and contained more vascular α-SMA as compared to SCS. This study demonstrated that NMP of the amputated limb with perfusate solution based on autologous blood could maintain the physiological activities of the limb for at least 24 h.

In view of the prevailing insecticide resistance and increasing safety and health concerns, there is an urgent need to develop effective and environmentally friendly methods to control stored-product pests. S-methoprene, a microbial volatile compound with insecticidal properties, has been identified as a potential candidate for this purpose. However, its efficacy against storage pests is not well understood. In this study, wheat was treated with S-methoprene at a rate of 5 mg/kg and stored in ambient conditions within a grain bin. Samples were collected after 90 and 365 days, and bioassays were conducted on 20 mixed-sex developmental stages of Tribolium castaneum and T. confusum. S-methoprene showed significant ovicidal, pupicidal, and adulticidal activity against T. castaneum and T. confusum. The efficacy of S-methoprene was found to be influenced by the developmental stage of the pests and the timing of its application. In the 90-day treatment group, the hatching rate of three T. castaneum strains was zero, while that of T. confusum strains from Guangdong and Beijing was (1.67±2.04)% and 0%, respectively. S-methoprene treatment in the 90-day group resulted in effective pupicidal activity against both T. castaneum and T. confusum, with the F1 generation of adults being unable to reproduce. However, in the 365-day treatment group, the pupicidal activity significantly decreased, and the F1 generation of adults were able to reproduce but their F1 generation pupae can not hatch. The adulticidal activity of both pests had some effects in both treatments, but the mortality rate significantly decreased in the 365-day treatment group. The F1 generation of T. castaneum only consisted of larvae, while some larvae of T. confusum were able to pupate but could not further develop into adults. T. castaneum was found to be more susceptible to S-methoprene than T. confusum. After S-methoprene induction, the activity of three detoxification enzymes in Tribolium species was affected in varying degrees and had different variation patterns. The activity was positively correlated with induction time.

Abstract Background Incidental thymus region masses during thoracic examinations are not uncommon. The clinician’s decision-making for treatment largely depends on imaging findings. Due to the lack of specific indicators, it may be of great value to explore the role of radiomics in risk categorization of the thymic epithelial tumors (TETs). Methods Four databases (PubMed, Web of Science, EMBASE and the Cochrane Library) were screened to identify eligible articles reporting radiomics models of diagnostic performance for risk categorization in TETs patients. The quality assessment of diagnostic accuracy studies 2 (QUADAS-2) and radiomics quality score (RQS) were used for methodological quality assessment. The pooled area under the receiver operating characteristic curve (AUC), sensitivity and specificity with their 95% confidence intervals were calculated. Results A total of 2134 patients in 13 studies were included in this meta-analysis. The pooled AUC of 11 studies reporting high/low-risk histologic subtypes was 0.855 (95% CI, 0.817–0.893), while the pooled AUC of 4 studies differentiating stage classification was 0.826 (95% CI, 0.817–0.893). Meta-regression revealed no source of significant heterogeneity. Subgroup analysis demonstrated that the best diagnostic imaging was contrast enhanced computer tomography (CECT) with largest pooled AUC (0.873, 95% CI 0.832–0.914). Publication bias was found to be no significance by Deeks’ funnel plot. Conclusions This present study shows promise for preoperative selection of high-risk TETs patients based on radiomics signatures with current available evidence. However, methodological quality in further studies still needs to be improved for feasibility confirmation and clinical application of radiomics-based models in predicting risk categorization of the thymic epithelial tumors.

Therefore, the requirement of low power consumption is difficult to be satisfied. In order to solve this problem, we propose a low complexity cell Identity (ID) detection algorithm. Firstly, the algorithm splits the secondary synchronization signal, and reduces the multiplication operation by using the law of phase offset changing with frame number. Then it uses the symmetry of Zadoff-Chu (ZC) sequence in the secondary synchronization signal to calculate the autocorrelation value of the received signal. Combined with the cross-correlation between the local signal and the received signal, the secondary synchronization signal can be detected and the cell ID number can be calculated. By comparing with other algorithms, the proposed algorithm can greatly reduce the complexity of the algorithm while maintaining the acceptable performance.

Early life experience influences stress reactivity and mental health through effects on cognitive-emotional functions that are, in part, linked to gene expression in the dorsal and ventral hippocampus. The hippocampal dentate gyrus (DG) is a major site for experience-dependent plasticity associated with sustained transcriptional alterations, potentially mediated by epigenetic modifications. Here, we report comprehensive DNA methylome, hydroxymethylome and transcriptome data sets from mouse dorsal and ventral DG. We find genome-wide transcriptional and methylation differences between dorsal and ventral DG, including at key developmental transcriptional factors. Peripubertal environmental enrichment increases hippocampal volume and enhances dorsal DG-specific differences in gene expression. Enrichment also enhances dorsal-ventral differences in DNA methylation, including at binding sites of the transcription factor NeuroD1, a regulator of adult neurogenesis. These results indicate a dorsal-ventral asymmetry in transcription and methylation that parallels well-known functional and anatomical differences, and that may be enhanced by environmental enrichment.

Abstract Background The next-generation sequencing (NGS) technology has greatly facilitated genomic and transcriptomic studies, contributing significantly in expanding the current knowledge on genome and transcriptome. However, the continually evolving variety of sequencing platforms, protocols and analytical pipelines has led the research community to focus on cross-platform evaluation and standardization. As a NGS pioneer in China, the Beijing Genomics Institute (BGI) has announced its own NGS platform designated as BGISEQ-500, since 2016. The capability of this platform in large-scale DNA sequencing and small RNA analysis has been already evaluated. However, the comparative performance of BGISEQ-500 platform in transcriptome analysis remains yet to be elucidated. The Illumina series, a leading sequencing platform in China’s sequencing market, would be a preferable reference to evaluate new platforms. Methods To this end, we describe a cross-platform comparative study between BGISEQ-500 and Illumina HiSeq4000 for analysis of Arabidopsis thaliana WT (Col 0) transcriptome. The key parameters in RNA sequencing and transcriptomic data processing were assessed in biological replicate experiments, using aforesaid platforms. Results The results from the two platforms BGISEQ-500 and Illumina HiSeq4000 shared high concordance in both inter- (correlation, 0.88–0.93) and intra-platform (correlation, 0.95–0.98) comparison for gene quantification, identification of differentially expressed genes and alternative splicing events. However, the two platforms yielded highly variable interpretation results for single nucleotide polymorphism and insertion–deletion analysis. Conclusion The present case study provides a comprehensive reference dataset to validate the capability of BGISEQ-500 enabling it to be established as a competitive and reliable platform in plant transcriptome analysis.

Stress resilience is accompanied by broad changes in gene expression. This study shows that estrogen receptor α (ERα) is a key upstream regulator of these changes in the nucleus accumbens, and that overexpression of ERα increases behavioral resilience via a sex-specific transcriptional mechanism.

Environmental enrichment has functional and molecular effects on mammalian hippocampus. Here, Zhang and colleagues show that environmental enrichment of mice is correlated with dorsal-ventral asymmetry in transcription and DNA methylation of the dentate gyrus.

The precipitation in the forecast period influences flood forecasting precision, due to the uncertainty of the input to the hydrological model. Taking the ZhangHe basin as the example, the research adopts the precipitation forecast and ensemble precipitation forecast product of the AREM model, uses the Xin Anjiang hydrological model, and tests the flood forecasts. The results show that the flood forecast result can be clearly improved when considering precipitation during the forecast period. Hydrological forecast based on Ensemble Precipitation prediction gives better hydrological forecast information, better satisfying the need for risk information for flood prevention and disaster reduction, and has broad development opportunities.
