Abstract Diverse post-translational modifications have been shown to play important roles in regulating protein function in eukaryotes. By contrast, the roles of post-translational modifications in bacteria are not so well understood, particularly as they relate to pathogenesis. Here, we demonstrate post-translational protein modification by covalent addition of lactate to lysine residues (lactylation) in the human pathogen Staphylococcus aureus. Lactylation is dependent on lactate concentration and specifically affects alpha-toxin, in which a single lactylated lysine is required for full activity and virulence in infection models. Given that lactate levels typically increase during infection, our results suggest that the pathogen can use protein lactylation as a mechanism to increase toxin-mediated virulence during infection.

Abstract Morphogenesis requires building stable macromolecular structures from highly dynamic proteins. Muscles are anchored by long-lasting integrin adhesions to resist contractile force. However, the mechanisms governing integrin diffusion, immobilization, and activation within developing tissues remain elusive. Here, we show that actin polymerization-driven membrane protrusions form nanotopographies that enable strong adhesion at Drosophila muscle attachment sites (MASs). Super-resolution microscopy reveals that integrins assemble adhesive belts around Arp2/3-dependent actin protrusions, forming invadosome-like structures with membrane nanotopographies. Single protein tracking shows that, during MAS development, integrins become immobile and confined within diffusion traps formed by the membrane nanotopographies. Actin filaments also display restricted motion and confinement, indicating strong mechanical connection with integrins. Using isolated muscle cells, we show that substrate nanotopography, rather than rigidity, drives adhesion maturation by regulating actin protrusion, integrin diffusion and immobilization. These results thus demonstrate that actin-polymerization-driven membrane protrusions are essential for the formation of strong integrin adhesions sites in the developing embryo, and highlight the important contribution of geometry to morphogenesis.

ABSTRACTStaphylococcus aureus (S. aureus) is a notorious pathogen that cause metastatic or complicated infections. Hypervirulent ST398 clonotype strains, remarkably increased in recent years, dominated Community-associated S. aureus (CA-SA) infections in the past decade in China. Small RNAs like RNAIII have been demonstrated to play important roles in regulating the virulence of S. aureus, however, the regulatory roles played by many of these sRNAs in the ST398 clonotype strains are still unclear. Through transcriptome screening and combined with knockout phenotype analysis, we have identified a highly transcribed sRNA, RSaX28, in the ST398 clonotype strains. Sequence analysis revealed that RSaX28 is highly conserved in the most epidemic clonotypes of S. aureus, but its high transcription level is particularly prominent in the ST398 clonotype strains. Characterization of RSaX28 through RACE and Northern blot revealed its length to be 533nt. RSaX28 is capable of promoting the hemolytic ability, reducing biofilm formation capacity, and enhancing virulence of S. aureus in the in vivo murine infection model. Through IntaRNA prediction and EMSA validation, we found that RSaX28 can specifically interact with RNAIII, promoting its stability and positively regulating the translation of downstream alpha-toxin while inhibiting the translation of Sbi, thereby regulating the virulence and biofilm formation capacity of the ST398 clonotype strains. RSaX28 is an important virulence regulatory factor in the ST398 clonotype S. aureus and represents a potential important target for future treatment and immune intervention against S. aureus infections.

ABSTRACTOXA-232 is one of the most common OXA-48-like carbapenemase derivatives and is widely disseminated in nosocomial settings across countries. The blaOXA-232 gene is located on a 6-kb non-conjugative ColKP3-type plasmid, while the dissemination of blaOXA-232 into different Enterobacterales species and the polyclonal dissemination of OXA-232-producing K. pneumoniae revealed the horizontal transfer of blaOXA-232. However, it’s still unclear how this non-conjugative ColKP3 plasmid could facilitate the mobilization of blaOXA-232. Here, we observed the in vivo intraspecies transfer of blaOXA-232 during a nosocomial outbreak of OXA-232-producing K. pneumoniae. We demonstrated the presence of ColKP3 OXA-232 plasmid in the outer membrane vesicles (OMVs) derived from clinical isolates, and OMVs could facilitate the horizontal transfer of blaOXA-232 among Enterobacterales. In contrast, for the most prevalent carbapenemase genes, including blaKPC-2 and blaNDM-1, though the presence of carbapenemase genes and plasmid backbones in the vesicular lumen was observed, OMVs couldn’t promote effective transformation, probably due to the low copy number of plasmids in clinical isolates and the low number of plasmids loaded into vesicles. Conjugation assay revealed that the epidemic IncX3 NDM-1 and IncFII(pHN7A8)/IncR KPC-2 plasmids were conjugative and could be horizontally transferred via independent conjugation or with the help of a co-existent conjugative plasmid. For the large-size and low-copy number conjugative plasmids carrying carbapenemase genes, OMVs-mediated gene exchange may only serve as an alternative pathway for horizontal transfer. In conclusion, diverse mobilization strategies were employed by plasmids harbouring carbapenemase genes, and plasmids display a proper choice of mobility pathway due to their individual properties.

Abstract Background Monoterpenes are among the most important volatile aromatic compounds contributing to the flavor and aroma of grapes and wine. However, the molecular basis of monoterpene biosynthesis has not yet been fully elucidated. Results In our study, transcriptomics and gas chromatography-mass spectrometry (GC-MS) were used to mine candidate genes and transcription factors involved in monoterpene biosynthesis between high-monoterpene and zero-monoterpene table grape cultivars. We found that monoterpene biosynthesis was positively correlated by the expression of five genes encoding 1-deoxy-D-xylulose-5-phosphate synthase (VvDXSs), one encoding 4-hydroxy-3-methylbut-2-enyl diphosphate reductase (VvHDR), three hydroxy-3-methylglutaryl-CoA synthases (VvHMGSs) and one mevalonate kinase (VvMVK), whereas the expression of one isopentenyl diphosphate isomerase (VvIDI) and one 3-hydroxy-3-methylglutaryl-CoA reductase (VvHMGR) negatively correlated monoterpene biosynthesis. Of these genes, VvIDI was selected to validate its function in monoterpene accumulation through a transient overexpression experiment, and was shown to inhibit the biosynthesis of grape linalool and α-terpineol. Meanwhile, we found that a 64-amino acid extension sequence at the N-terminus can guide the VvIDI protein to target the chloroplast. Conclusions The findings of this study should help to guide future functional analysis of key genes as well as mining the potential regulatory mechanism of monoterpene biosynthesis in grapes and grape products.

The template matching method has been widely utilized in the defect detection of wafer surfaces. However, the traditional matching approaches are limited by illumination, noise, and deformation, which cannot meet the requirements of accuracy and robustness. In this paper, a novel multiple targets localization method, named Normalized Cross-correlation Adaptive Variable Step-Size Dynamic Template (NCC-AVSSDT) matching, is proposed to improve the accuracy and efficiency of image localization, which combines the advantages of NCC and AVSSDT. The AVSSDT method is utilized to dynamically adjust the scanning step size based on the NCC matching coefficients. This approach optimizes the scanning process, accelerating convergence toward the optimal matching position. Experimental results verify the accuracy and robustness of the proposed method under different conditions, especially when dealing with rotational variations and variations in noise textures. Therefore, NCC-AVSSDT can be used to perform multiple targets localization of chip image in nearly real-time. Three experiment types were used for comprehensive evaluations, including multiple targets, noise, and rotation angles. Experimental results show that NCC-AVSSDT is much better than the sequential similarity detection algorithm and mean absolute deviation methods in terms of multiple targets (0.667 vs 0.811 s, 0.832 s) and success rate (100% vs 35%, 20%).

The Sustainable Development Goals (SDGs) adopted by the United Nations in 2015 represent the current perceptions of humans regarding understanding and monitoring development. Achieving all 17 goals simultaneously is unrealistic. Considering the interconnected nature of SDGs, identifying their critical dimensions, goals, indicators, and mutual relationships is necessary. In addition, with increasing reservations about the sustainability of SDGs, it is crucial to explore consistency across different dimensions to ensure policy coherence in maximizing synergies and minimizing trade-offs. Our study employed multiple factor analysis (MFA) and hierarchical clustering on principal components (HCPC) to investigate these issues and analyze the results based on the public value (PV) theory. The results indicated that the Human Development Index (HDI) and gross domestic product per capita (GDPP) constitute the first principal component (PC) and are determinants in differentiating country clusters. However, they contradict environmental indicators such as CO2 emissions per capita and ecological footprint gha per person (EFP) and have low synergy with the Happy Planet Index (HPI). Additionally, the relationships between income level, inequality, and environmental quality correspond to a combined Kuznets curve and an environmental Kuznets curve (EKC). Moreover, governance capacity has become increasingly crucial in sustainable development, particularly in the capability to prioritize different PVs in a timely and strategic manner. Finally, despite the novelty of EFP and HPI, they cannot reveal the entire development story. SDGs require embracing more such indicators to enrich the value bases of development and achieve a sustainable future.

Staphylococcus aureus (SA) is a relatively uncommon cause of urinary tract infections (UTIs) in the general population. Although rare, S. aureus-induced UTIs are prone to potentially life-threatening invasive infections such as bacteremia. To investigate the molecular epidemiology, phenotypic characteristics, and pathophysiology of S. aureus-induced UTIs, we analyzed non-repetitive 4,405 S. aureus isolates collected from various clinical sources from 2008 to 2020 from a general hospital in Shanghai, China. Among these, 193 isolates (4.38%) were cultivated from the midstream urine specimens. Epidemiological analysis showed UTI-derived ST1 (UTI-ST1) and UTI-ST5 are the primary sequence types of UTI-SA. Furthermore, we randomly selected 10 isolates from each of the UTI-ST1, non-UTI-ST1 (nUTI-ST1), and UTI-ST5 groups to characterize their in vitro and in vivo phenotypes. The in vitro phenotypic assays revealed that UTI-ST1 exhibits an obvious decline in hemolysis of human red blood cells and increased biofilm and adhesion in the urea-supplemented medium, compared to the medium without urea, while UTI-ST5 and nUTI-ST1 did not show significant differences between the biofilm-forming and adhesion abilities. In addition, the UTI-ST1 displayed intense urease activities by highly expressing urease genes, indicating the potential role of urease in UTI-ST1 survival and persistence. Furthermore, in vitro virulence assays using the UTI-ST1 ureC mutant showed no significant difference in the hemolytic and biofilm-forming phenotypes in the presence or absence of urea in the tryptic soy broth (TSB) medium. The in vivo UTI model also showed that the CFU of the UTI-ST1 ureC mutant rapidly reduced during UTI pathogenesis 72 h post-infection, while UTI-ST1 and UTI-ST5 persisted in the urine of the infected mice. Furthermore, the phenotypes and the urease expression of UTI-ST1 were found to be potentially regulated by the Agr system with the change in environmental pH. In summary, our results provide important insights into the role of urease in S. aureus-induced UTI pathogenesis in promoting bacterial persistence in the nutrient-limiting urinary microenvironment.

The initial interface debonding damage behavior of cemented hip arthroplasty was simulated and investigated through the push-in experiment. A series of nondestructive testing techniques were carried out to detect damage to the stem-cement interface and cement. Results suggested that the initial stem-cement interface debonding and cement damage were induced at femur loading early period. The cement was subjected to a combination action of shear stress and bending from a sinking stem causing a pressure-arch effect resulting in compression creep and micro-crack on the cement successively. The stem-cement interfacial friction prevent stem from sinking due to the internal shear effect caused by the cement-stem interface micro-sliding. The interface debonding procession increased at first and then became stable, which was shown in cement's creep deformation-friction-fracture model. The initial micro-cracks released damage energy which was converted into stress waves. Further, the relationship of bond stress and slippage at different locations caused different degrees of damage belt for the interface and cement. It was concluded that the combination effect of pressure arching and shear lag was the primary reason for cement's initial damage and the matrix defect affected the failure progress.

Suboptimal light stress in grapevines is increasing worldwide with the spread of grapevine cultivation, which can affect grapevine physiology and productivity, such as in Southern China. Abscisic acid (ABA) is an important hormone in plant adaptive responses to abiotic stress, including low light stress. However, ABA signaling pathway genes (APGs) are not well characterized in the grapevine, and little is known of their potentially mitigating role in grapevine growth under weak light stress. Our study aimed to explore the potential role of the ABA signaling pathway in the response of grapevines to suboptimal light conditions. In this study, APGs were identified in the grapevine genome, and the distribution of conserved motifs was shown to reflect their phylogenetic relationships. Gene duplication analysis indicated that segmental duplication was an important driver for gene expansion in the grapevine ABA signaling pathway. Suboptimal conditions of light were shown to seriously affect the growth of grapevine leaves and berries, with the differential regulation of APGs in the grapevine. Our study summarizes the basic characteristics of APGs in grapevine, which can now be examined further for their roles in grapevine’s response to suboptimal light conditions.

Cell-fate networks are traditionally studied within the framework of gene regulatory networks. This paradigm considers only interactions of genes through expressed transcription factors and does not incorporate chromatin modification processes. This paper introduces a mathematical model that seamlessly combines gene regulatory networks and DNA methylation (DNAm), with the goal of quantitatively characterizing the contribution of epigenetic regulation to gene silencing. The ‘Basin of Attraction percentage’ is introduced as a metric to quantify gene silencing abilities. As a case study, a computational and theoretical analysis is carried out for a model of the pluripotent stem cell circuit as well as a simplified self-activating gene model. The results confirm that the methodology quantitatively captures the key role that DNAm plays in enhancing the stability of the silenced gene state.

Abstract Bacterial membrane vesicles (MVs) have recently gained much attention and have been shown to carry a wide diversity of secreted bacterial components. However, it is poorly understood whether MV carriage is an indispensable requirement for a cargo's function. Bacteriocins as weapons of bacterial warfare shape the composition of microbial communities. Many bacteriocins have pronounced hydrophobicity that is imposed by their mechanism of action, but how they diffuse through aqueous environments to reach their target competitors is not known. Here we show that antimicrobial competitive activity of an exemplary hydrophobic bacteriocin of the thiopeptide antibiotic family, micrococcin P1 (MP1), is dependent on incorporation into MVs, which were found to carry MP1 at high concentrations. In contrast, MP1 without MV association was poorly active due to low solubility. Furthermore, we provide previously unavailable evidence that MVs fuse with a Gram‐positive bacterium's cytoplasmic membrane, in this case to deliver a bacteriocin to its intracellular target. Our findings demonstrate how bacteria overcome the problem associated with secreting hydrophobic small molecules and delivering them to their target and show that MVs have a key function in bacterial warfare. Furthermore, our study provides hitherto rare evidence that MVs provide an essential rather than merely accessory function in bacterial physiology.

Summary: A design for genetically encoded counters is proposed via repressor-based circuits. An N-bit counter reads sequences of input pulses and displays the total number of pulses, modulo 2N. The design is based on distributed computation with specialized cell types allocated to specific tasks. This allows scalability and bypasses constraints on the maximal number of circuit genes per cell due to toxicity or failures due to resource limitations. The design starts with a single-bit counter. The N-bit counter is then obtained by interconnecting (using diffusible chemicals) a set of N single-bit counters and connector modules. An optimization framework is used to determine appropriate gate parameters and to compute bounds on admissible pulse widths and relaxation (inter-pulse) times, as well as to guide the construction of novel gates. This work can be viewed as a step toward obtaining circuits that are capable of finite automaton computation in analogy to digital central processing units.

Background: Bloodstream infections (BSIs) are recognized as important nosocomial infections. Klebsiella pneumoniae is one of the major causes of bacteremia. This retrospective study focused on drug susceptibility and molecular epidemiology of K. pneumoniae isolated from intensive care unit (ICU) patients with BSI in Shanghai, China.Methods: Consecutive K. pneumoniae isolates were collected from ICU patients. Antibiotic susceptibility testing was conducted by the broth microdilution method. PCR was performed to detect antimicrobial resistance genes. We also completed multilocus sequence typing (MLST) and GoeBURST was used to analyze the result of MLST.Results: A total of 78 K. pneumoniae isolates were enrolled. K. pneumoniae from ICU-BSIs were highly resistant to almost all common antibiotics. The most frequent resistance determinants responsible for extended-spectrum β-lactamase (ESBL) producers were blaCTX−M−14, blaCTX−M−15, and blaCTX−M−55. KPC was the only enzyme, which was detected by the carbapenemase producers. The most principal sequence types (STs) were ST11, ST15, and ST23.Conclusion: This study presents for the first time the antibiotic resistance phenotype and molecular epidemiology of K. pneumoniae isolated from ICU patients with BSIs in Shanghai. ICU-BSI K. pneumoniae is characteristic of a high resistance rate. The occurrence of the KPC-2 enzyme may result from nosocomial clonal dissemination of ST11 K. pneumoniae.

With the frequent occurrence of extreme natural disasters, unfavorable growth environment is a common phenomenon in the life cycle of plants. In recent years, universal stress proteins (USPs) have attracted extensive attention in the field of plant science for their expression patterns and functional analysis. However, the characterization of the USP family remains unclear in grape. In this study, we identified 30 VvUSPs in the grape genome, which could be divided into three classes according to their encoded protein sequences, and this classification was reflected by the distribution of conserved motifs. Gene duplication analysis demonstrated that segmental duplication was an important pathway in the expansion of the VvUSP family. The expression patterns of 12 VvUSPs were significantly different between tissues, implying they had different functions in various tissues. Cis-acting element and expression analysis showed that most of the promoter regions of VvUSPs contained sequences responsive to hormones and stress elements, especially the promoter region of VIT_16s0013g01920. In conclusion, our findings provide comprehensive information for the further investigation of the genetics and protein functions of the USP gene family in grape.

Abstract Background Multi-drug resistance is the major cause of chemotherapy failure in hepatocellular carcinoma (HCC). YAP, a critical effector of the Hippo pathway, has been shown to contribute to the progression, metastasis and invasion of cancers. However, the potential role of YAP in mediating drug resistance remains obscure. Methods RT-qPCR and western blot were used to assess YAP expression in HCC cell lines. CCK-8 assays, flow cytometry, a xenograft tumour model, immunochemistry and GFP-mRFP-LC3 fusion proteins were utilized to evaluate the effect of YAP on multi-drug resistance, intracellular ROS production and the autophagy of HCC cells in vitro and in vivo. Autophagy inhibitor and rescue experiments were carried out to elucidate the mechanism by which YAP promotes chemoresistance in HCC cells. Results We found that BEL/FU, a typical HCC cell line with chemoresistance, exhibited overexpression of YAP. Moreover, the inhibition of YAP by shRNA or verteporfin conferred the sensitivity of BEL/FU cells to chemotherapeutic agents through autophagy-related cell death in vitro and in vivo. Mechanistically, YAP silencing significantly enhanced autophagic flux by increasing RAC1-driven ROS, which contributed to the inactivation of mTOR in HCC cells. In addition, the antagonist of autophagy reversed the enhanced effect of YAP silencing on cell death under treatment with chemotherapeutic agents. Conclusion Our findings suggested that YAP upregulation endowed HCC cells with multi-drug resistance via the RAC1-ROS-mTOR pathway, resulting in the repression of autophagy-related cell death. The blockade of YAP may serve as a promising novel therapeutic strategy for overcoming chemoresistance in HCC.

Objective: The majority of patients with COVID-19 showed mild symptoms. However, approximately 5% of them were critically ill and require intensive care unit admission for advanced life supports. Patients in the intensive care unit were high risk for venous thromboembolism and hemorrhage due to the immobility and anticoagulants used during advanced life supports. The aim of the study was to report the incidence and treatments of the two complications in such patients. Method: Patients with COVID-19 (Group 1) and patients with community-acquired pneumonia (Group 2) that required intensive care unit admission were enrolled in this retrospective study. Their demographics, laboratory results, ultrasound findings and complications such as venous thromboembolism and hemorrhage were collected and compared. Results: Thirty-four patients with COVID-19 and 51 patients with community-acquired pneumonia were included. The mean ages were 66 and 63 years in Groups 1 and 2, respectively. Venous thromboembolism was detected in 6 (18%) patients with COVID-19 and 18 (35%) patients with community-acquired pneumonia (P = 0.09). The major type was distal deep venous thrombosis. Twenty-one bleeding events occurred in 12 (35%) patients with COVID-19 and 5 bleeding events occurred in 5 (10%) patients with community-acquired pneumonia, respectively (P = 0.01). Gastrointestinal system was the most common source of bleeding. With the exception of one death due to intracranial bleeding, blood transfusion with or without surgical/endoscopic treatments was able to manage the bleeding in the remaining patients. Multivariable logistic regression showed increasing odds of hemorrhage with extracorporeal membrane oxygenation (odds ratio: 13.9, 95% confidence interval: 4.0–48.1) and COVID-19 (odds ratio: 4.7, 95% confidence interval: 1.2–17.9). Conclusion: Venous thromboembolism and hemorrhage were common in both groups. The predominant type of venous thromboembolism was distal deep venous thrombosis, which presented a low risk of progression. COVID-19 and extracorporeal membrane oxygenation were risk factors for hemorrhage. Blood transfusion with or without surgical/endoscopic treatments was able to manage it in most cases.

Aroma is one of the most important indicators of grape quality. Girdling and foliar fertilization with K (K2O) are common agronomic practices applied to improve berry quality in grape production. However, little is known about its effect on the accumulation and biosynthesis of the entire aromatic profile. Our study was aimed to explore the influences of girdling and foliar fertilization with K (alone or in combination) on the general properties, phenolic composition, volatile free aroma compounds, spatial and temporal expression of terpene-related genes and sensory properties in ‘Hanxiangmi’ table grape. In this study, we found that girdling and foliar fertilization with K (alone or in combination) facilitated fruit enlargement and increased the accumulation of phenolic compounds in skin. The combination treatment of girdling and foliar fertilization with K significantly promoted the concentrations of total soluble solids (TSS) in the pulp and proanthocyanidins in the berry skin, and had a lower titratable acidity (TA) compared to those of the control. In contrast, girdling treatment alone increased the concentrations of titratable acidity. Volatile free aroma composition analysis revealed that the combination treatment increased the volatile compounds and concentrations significantly, most notably in terpenes, such as nerol, citronellol and linalool. Spatial and temporal expression analysis showed that the expression level of VvDXS was significantly correlated with linalool and total terpenes concentrations, as a result of which, we speculated that VvDXS is the candidate gene for the regulation of important grape terpenes. We hope that our results can direct farmers to better apply girdling and foliar fertilization with K in grape production.

Abstract Background Plant homeodomain finger protein 8 (PHF8) serves an activator of epithelial-mesenchymal transition (EMT) and is implicated in various tumors. However, little is known about PHF8 roles in hepatocellular carcinoma (HCC) and regulating E-cadherin expression. Methods PHF8 expression pattern was investigated by informatic analysis and verified by RT-qPCR and immunochemistry in HCC tissues and cell lines. CCK8, xenograft tumor model, transwell assay, and tandem mCherry-GFP-LC3 fusion protein assay were utilized to assess the effects of PHF8 on proliferation, metastasis and autophagy of HCC cells in vitro and in vivo. ChIP, immunoblot analysis, rescue experiments and inhibitor treatment were used to clarify the mechanism by which PHF8 facilitated EMT, metastasis and autophagy. Results PHF8 upregulation was quite prevalent in HCC tissues and closely correlated with worse overall survival and disease-relapse free survival. Furthermore, PHF8-knockdown dramatically suppressed cell growth, migration, invasion and autophagy, and the expression of SNAI1, VIM, N-cadherin and FIP200, and increased E-cadherin level, while PHF8-overexpression led to the opposite results. Additionally, FIP200 augmentation reversed the inhibited effects of PHF8-siliencing on tumor migration, invasion and autophagy. Mechanistically, PHF8 was involved in transcriptionally regulating the expression of SNAI1, VIM and FIP200, rather than N-cadherin and E-cadherin. Noticeably, E-cadherin degradation could be accelerated by PHF8-mediated FIP200-dependent autophagy, a crucial pathway complementary to transcriptional repression of E-cadherin by SNAI1 activation. Conclusion These findings suggested that PHF8 played an oncogenic role in facilitating FIP200-dependent autophagic degradation of E-cadherin, EMT and metastasis in HCC. PHF8 might be a promising target for prevention, treatment and prognostic prediction of HCC.

Abstract Background Holliday junction recognition protein (HJURP) has been implicated in many cancers including hepatocellular carcinoma (HCC). However, the underlying mechanism by which HJURP promotes HCC cell proliferation remains unclear. Methods RT-qPCR and immunohistochemistry were used to detect HJURP expression in HCC and adjacent tumor tissues and HCC cell lines. The localization of p21 were determined by immunofluorescence and western blot. Co-immunoprecipitation and western blot were used to validate the p21 stability and signaling pathways affected by HJURP. The effects of HJURP on HCC cell proliferation were assessed both in vivo and in vitro. The ERK1/2 pathway inhibitor U0126 and AKT pathway agonist SC-79 were used to treat HCC cell lines for further mechanistic investigations. Results HJURP expression was higher in HCC tissues than in para-tumor tissues. Moreover, ectopic HJURP expression facilitated the proliferation of HCC cells, whereas the depletion of HJURP resulted in decreased cell growth in vitro and in vivo. Furthermore, the effects of HJURP silencing were reversed by p21 knockdown. Likewise, p21 overexpression inhibited cell growth ability mediated by HJURP elevation. Mechanistically, HJURP destabilized p21 via the MAPK/ERK1/2 and AKT/GSK3β pathways, which regulated the nucleus-cytoplasm translocation and ubiquitin-mediated degradation of p21. Clinically, high HJURP expression was correlated with unfavorable prognoses in HCC individuals. Conclusions Our data revealed that HJURP is an oncogene that drives cell cycle progression upstream of p21 in HCC. These findings may provide a potential therapeutic and prognostic target for HCC.

Grape production in southern China suffers great loss due to various environmental stresses. To understand the mechanism of how the grape plants respond to these stresses is an active area of research in developing cultivation techniques. Plant stress resistance is known to rely on special proteins. Amongst them, DnaJ protein (HSP40) serves as co-chaperones of HSP70, playing crucial roles in various stress response. However, the DnaJ proteins encoded by the DnaJ gene family in Vitis vinifera L. have not been fully described yet. In this study, we identified 78 VvDnaJs in the grape genome that can be classified into three groups—namely, DJA, DJB, and DJC. To reveal the evolutionary and stress response mechanisms for the VvDnaJ gene family, their evolutionary and expression patterns were analyzed using the bioinformatic approach and qRT-PCR. We found that the members in the same group exhibited a similar gene structure and protein domain organization. Gene duplication analysis demonstrated that segmental and tandem duplication may not be the dominant pathway of gene expansion in the VvDnaJ gene family. Codon usage pattern analysis showed that the codon usage pattern of VvDnaJs differs obviously from the monocotyledon counterparts. Tissue-specific analysis revealed that 12 VvDnaJs present a distinct expression profile, implying their distinct roles in various tissues. Cis-acting element analysis showed that almost all VvDnaJs contained the elements responsive to either hormones or stresses. Therefore, the expression levels of VvDnaJs subjected to exogenous hormone applications and stress treatments were determined, and we found that VvDnaJs were sensitive to hormone treatments and shade, salt, and heat stresses, especially VIT_00s0324g00040. The findings of this study could provide comprehensive information for the further investigation on the genetics and protein functions of the DnaJ gene family in grape.

Objective: This study aims to analyze the specific impact of hip osteoarthritis on the failure and subsequent revision of total hip replacements, with a particular focus on athletic populations. Considering the unique biomechanical demands and higher physical activity levels of athletes, the study seeks to understand how these factors influence post-surgical outcomes in cases of hip osteoarthritis. Methods: The research involved collecting and analyzing data from athletes who had undergone total hip replacement surgeries due to hip osteoarthritis. Patient demographics such as age, gender, and the severity of the disease were recorded. The study performed a detailed correlation analysis to evaluate the influence of hip osteoarthritis on the failure rates of total hip replacements in this specific group. It particularly focused on hip failure caused by arthritis, cement fixation failure in aseptic arthritis cases, and cemented hip failure due to primary inflammation. Results: The findings indicate that in athletic populations, hip osteoarthritis is significantly correlated with higher failure rates after total hip replacement, with an influence rate of 30.64%. This rate is notably higher compared to general population statistics, underscoring the unique challenges faced in treating athletes. The study also found a notable correlation between the type and severity of arthritis and the failure modes of hip replacements, including issues related to cement fixation. Conclusion: This study highlights the significant impact of hip osteoarthritis on the failure of total hip replacements in athletes, a group that typically places higher physical demands on hip joints post-surgery. The correlation between arthritis severity and replacement failure underscores the need for tailored surgical and rehabilitation approaches for athletic individuals. These findings stress the importance of considering the specific biomechanical and lifestyle needs of athletes in managing hip osteoarthritis and planning total hip replacement surgeries. To facilitate the recycling and reuse of cigarette filters and oil/water separation, a superhydrophobic cigarette filter was made by coating with dodecanethiol-modified polypyrrole (Ppy) particles by a dip-coating method. SEM, FTIR, and XPS were used to analyze the surface morphology and chemical compositions. The as-prepared superhydrophobic cigarette filter can realize wettability alteration via changing the ammonium persulfate (APS) concentration from 0.15 mol/L to 3 mol/L, and the contact angle increased from 0° on the original cigarette filter to 155° with a sliding angle of 5°. The superhydrophobic cigarette filter could effectively separate various oils and organic solvents. The separation efficiency was 98.8% and the separation stability was good. Furthermore, the as-prepared superhydrophobic cigarette filter had a large oil absorption range and could absorb different oils and organic solvents, including petroleum ether, engine oil, vegetable oil, n-hexane, and chloroform, with maximum absorption capacities ranging from 9.4 g/g to 22.7 g/g. According to the above results, we believe that the as-prepared superhydrophobic cigarette filter should have great potential in the recovery of solid waste and high-efficiency oil/water separation.
