Increasing drilling speed and efficiency of hard formation for deep and ultra-deep well is one of the international recognized drilling problems and key technologies to be tackled urgently. Particle jet impact drilling technology is an efficient non-contact rock-breaking method to overcome slow drilling speed, which has great development and application potential in drilling speed-increase of hard formation and deep well. High efficiency drilling technology and rock-breaking speed-increase mechanism in high temperature, high pressure and high hardness formations of deep and ultra-deep wells were mainly focused and keynoted in this paper. With extensive investigation of domestic and foreign literature, the working principle, key technical devices, deep-well-rock mechanical characteristic, unconventional constitutive model and rock-breaking mechanism of particle jet impact drilling technology were analyzed, which proved the feasibility and high efficiency for deep and hard stratum, and also, dynamic failure mechanism of rock needs to be elaborated by constructing the constitutive model with high temperature and pressure. Meanwhile, the major problems to be solved at present and development direction future were summarized, which mainly included: miniaturization of drilling equipment and individualization of drilling bit; optimization of jet parameters and the evaluation method of rock-breaking effect; establishment of mechanical property and unconventional constitutive model of deep-well-rock; rock-breaking mechanism and dynamic response under particle jet coupling impact. The research can help for better understanding of deep-well drilling speed-increasing technology and also promote the development and engineering application of particle jet impact drilling speed-increase theory and equipment.

Phase transition is common during (de)-intercalating layered sodium oxides, which directly affects the structural stability and electrochemical performance. However, the artificial control of phase transition to achieve advanced sodium-ion batteries is lacking, since the remarkably little is known about the influencing factor relative to the sliding process of transition-metal slabs upon sodium release and uptake of layered oxides. Herein, we for the first time demonstrate the manipulation of oxygen vacancy concentrations in multinary metallic oxides has a significant impact on the reversibility of phase transition, thereby determining the sodium storage performance of cathode materials. Results show that abundant oxygen vacancies intrigue the return of the already slide transition-metal slabs between O3 and P3 phase transition, in contrast to the few oxygen vacancies and resulted irreversibility. Additionally, the abundant oxygen vacancies enhance the electronic and ionic conductivity of the Na0.9Ni0.3Co0.15Mn0.05Ti0.5O2 electrode, delivering the high initial Coulombic efficiency of 97.1%, large reversible capacity of 112.7 mAh·g−1, superior rate capability upon 100 C and splendid cycling performance over 1,000 cycles. Our findings open up new horizons for artificially manipulating the structural evolution and electrochemical process of layered cathodes, and pave a way in designing advanced sodium-ion batteries.

Chemotherapy and irradiation cause DNA damage to hematopoietic stem cells (HSCs), leading to HSC depletion and dysfunction and the risk of malignant transformation over time. Extrinsic regulation of HSC DNA repair is not well understood, and therapies to augment HSC DNA repair following myelosuppression remain undeveloped. We report that epidermal growth factor receptor (EGFR) regulates DNA repair in HSCs following irradiation via activation of the DNA-dependent protein kinase–catalytic subunit (DNA-PKcs) and nonhomologous end joining (NHEJ). We show that hematopoietic regeneration in vivo following total body irradiation is dependent upon EGFR-mediated repair of DNA damage via activation of DNA-PKcs. Conditional deletion of EGFR in hematopoietic stem and progenitor cells (HSPCs) significantly decreased DNA-PKcs activity following irradiation, causing increased HSC DNA damage and depressed HSC recovery over time. Systemic administration of epidermal growth factor (EGF) promoted HSC DNA repair and rapid hematologic recovery in chemotherapy-treated mice and had no effect on acute myeloid leukemia growth in vivo. Further, EGF treatment drove the recovery of human HSCs capable of multilineage in vivo repopulation following radiation injury. Whole-genome sequencing analysis revealed no increase in coding region mutations in HSPCs from EGF-treated mice, but increased intergenic copy number variant mutations were detected. These studies demonstrate that EGF promotes HSC DNA repair and hematopoietic regeneration in vivo via augmentation of NHEJ. EGF has therapeutic potential to promote human hematopoietic regeneration, and further studies are warranted to assess long-term hematopoietic effects.

The discovery of novel hematopoietic stem cell (HSC) surface markers can enhance understanding of HSC identity and function. We have discovered a population of primitive bone marrow (BM) HSCs distinguished by their expression of the heparan sulfate proteoglycan Syndecan-2, which serves as both a marker and a regulator of HSC function. Syndecan-2 expression was increased 10-fold in CD150+CD48–CD34–c-Kit+Sca-1+Lineage– cells (long-term HSCs [LT-HSCs]) compared with differentiated hematopoietic cells. Isolation of BM cells based solely on syndecan-2 surface expression produced a 24-fold enrichment for LT-HSCs and sixfold enrichment for α-catulin+c-kit+ HSCs, and yielded HSCs with superior in vivo repopulating capacity compared with CD150+ cells. Competitive repopulation assays revealed the HSC frequency to be 17-fold higher in syndecan-2+CD34–KSL cells compared with syndecan-2–CD34–KSL cells and indistinguishable from CD150+CD34–KSL cells. Syndecan-2 expression also identified nearly all repopulating HSCs within the CD150+CD34–KSL population. Mechanistically, syndecan-2 regulates HSC repopulating capacity through control of expression of Cdkn1c (p57) and HSC quiescence. Loss of syndecan-2 expression caused increased HSC cell cycle entry, downregulation of Cdkn1c, and loss of HSC long-term repopulating capacity. Syndecan-2 is a novel marker of HSCs that regulates HSC repopulating capacity via control of HSC quiescence.

Ionizing radiation and chemotherapy deplete hematopoietic stem cells and damage the vascular niche wherein hematopoietic stem cells reside. Hematopoietic stem cell regeneration requires signaling from an intact bone marrow (BM) vascular niche, but the mechanisms that control BM vascular niche regeneration are poorly understood. We report that BM vascular endothelial cells secrete semaphorin 3 A (SEMA3A) in response to myeloablation and SEMA3A induces p53 – mediated apoptosis in BM endothelial cells via signaling through its receptor, Neuropilin 1 (NRP1), and activation of cyclin dependent kinase 5. Endothelial cell – specific deletion of Nrp1 or Sema3a or administration of anti-NRP1 antibody suppresses BM endothelial cell apoptosis, accelerates BM vascular regeneration and concordantly drives hematopoietic reconstitution in irradiated mice. In response to NRP1 inhibition, BM endothelial cells increase expression and secretion of the Wnt signal amplifying protein, R spondin 2. Systemic administration of anti - R spondin 2 blocks HSC regeneration and hematopoietic reconstitution which otherwise occurrs in response to NRP1 inhibition. Receptor type protein tyrosine phosphatase-sigma (PTPσ) is primarily expressed by adult neurons and regulates neural regeneration. We recently discovered that PTPσ is also expressed by hematopoietic stem cells (HSCs). Here, we describe small molecule inhibitors of PTPσ that promote HSC regeneration in vivo. Systemic administration of the PTPσ inhibitor, DJ001, or its analog, to irradiated mice promotes HSC regeneration, accelerates hematologic recovery, and improves survival. Similarly, DJ001 administration accelerates hematologic recovery in mice treated with 5-fluorouracil chemotherapy. DJ001 displays high specificity for PTPσ and antagonizes PTPσ via unique non-competitive, allosteric binding. Mechanistically, DJ001 suppresses radiation-induced HSC apoptosis via activation of the RhoGTPase, RAC1, and induction of BCL-XL. Furthermore, treatment of irradiated human HSCs with DJ001 promotes the regeneration of human HSCs capable of multilineage in vivo repopulation. These studies demonstrate the therapeutic potential of selective, small-molecule PTPσ inhibitors for human hematopoietic regeneration., Protein tyrosine phosphatase sigma (PTPσ) deficient haematopoietic stem cells (HSCs) demonstrate increased engraftment following transplantation. Natural deposit of natural gas hydrate (NGH) exists in the colder regions on the earth, which is an unconventional energy and has great potential. About 97% of the NGH is located in the offshore and only 3% is located on the land. Due to its sensitivity to the external environment and easy decomposition, it is necessary to suppress the external environment changes caused by the complex vibration of the drill string during its exploitation. In this paper, the fluid-solid coupling nonlinear vibration of drill string is studied. The nonlinear dynamic model of drill string with drilling fluid is established. The relationship between variable parameters of drill string and complex dynamics of drill string is analyzed. Furthermore, the bifurcation diagram is given. The results have certain significance for discussing the non-linear dynamic mechanism of drill string under complex conditions and reasonably selecting the drilling process parameters of NGH.

As a new type of drilling method, with adding a certain proportion of steel particles into high-pressure pipe, high-velocity particle is formed to impact on rock surface to produce very large rock-breaking stress and achieve to drill rapidly. Therefore, based on cavity expansion theory, firstly, mathematical model of stress of particle impact rock was established, and the stress distribution of rock-breaking was numerical calculated. Then, the dynamical rock-breaking process under particle impact was simulated, and simulation results were compared with numerical calculation results. The results showed that there was a good agreement with each other, and rock-breaking stress could increase with increase of particle impact velocity. It would be of great significance to the development and improvement of particle impact drilling theory.

The Sodium-ion-battery, using the layered oxide material as cathodes coupled with carbon anodes, has been proposed for utilization in practice long time ago. However, the prevailing layered oxides is still hindered by serious phase transition and volume strain upon sodium release. Herein, we show a novel copper doped layered P_1 manganese-based oxide material (NCM) with unique bulk and nanoscale Na-free surface structures as a sodium host to overcome the above limitation. Our research disclosed this P_1 NCM material has a wide phase-transition-free zone between 2 V and 4.5 V with a tiny lattice volume change upon cycling. Besides, it delivers an excellent chemical stability during humid air exposure. In contrast to most environmentally sensitive layered materials, after aging for 60 days in humid air, P_1 NCM still exhibits the superior cycling performance (95.8% capacity retention after 1000 cycles) and excellent rate capability as well as an outstanding sodium storage performance in full cells.

Tyrosine kinase inhibitors (TKIs) induce molecular remission in the majority of patients with chronic myelogenous leukemia (CML), but the persistence of CML stem cells hinders cure and necessitates indefinite TKI therapy. We report that CML stem cells upregulate the expression of pleiotrophin (PTN) and require cell-autonomous PTN signaling for CML pathogenesis in BCR/ABL(+) mice. Constitutive PTN deletion substantially reduced the numbers of CML stem cells capable of initiating CML in vivo. Hematopoietic cell–specific deletion of PTN suppressed CML development in BCR/ABL(+) mice, suggesting that cell-autonomous PTN signaling was necessary for CML disease evolution. Mechanistically, PTN promoted CML stem cell survival and TKI resistance via induction of Jun and the unfolded protein response. Human CML cells were also dependent on cell-autonomous PTN signaling, and anti-PTN antibody suppressed human CML colony formation and CML repopulation in vivo. Our results suggest that targeted inhibition of PTN has therapeutic potential to eradicate CML stem cells.

Aiming at the problems of difficult observation of rock damage mechanism and many influencing factors of rock-breaking process, the damage mechanism of particle water-jet coupling impact was studied. Based on SPH-FEM coupling algorithm, the numerical model of particle water-jet impact rock was established, and the mechanical properties of rock were characterized by JH-2 (Johnson Holmquist II) damage model. Then, damage evolution and failure effect under the impact of water-jet and particle water-jet were simulated. It shows that the failure of rock is a cyclic process of “damage–cumulative damage–failure–cumulative damage–failure”, which is mainly manifested as shear powder and tensile cracks. Meanwhile, particle water-jet coupling impact rock-breaking experiment system and methods were introduced and the comparison way of experiment and simulation was used to analyze the influence law of particle ratio, water-jet impact velocity, particle diameter, impact angle on rock damage and failure depth. The propagation law of transverse and longitudinal damage inside of rock was studied by defining Gaussian points and damage would often occur instantaneously which was basically microsecond level.

Multicellular organisms have multiple homologs of the yeast ATG8 gene, but the differential roles of these homologs in autophagy during development remain largely unknown. Here we investigated structure/function relationships in the two C. elegans Atg8 homologs, LGG-1 and LGG-2. lgg-1 is essential for degradation of protein aggregates, while lgg-2 has cargo-specific and developmental-stage-specific roles in aggregate degradation. Crystallography revealed that the N-terminal tails of LGG-1 and LGG-2 adopt the closed and open form, respectively. LGG-1 and LGG-2 interact differentially with autophagy substrates and Atg proteins, many of which carry a LIR motif. LGG-1 and LGG-2 have structurally distinct substrate binding pockets that prefer different residues in the interacting LIR motif, thus influencing binding specificity. Lipidated LGG-1 and LGG-2 possess distinct membrane tethering and fusion activities, which may result from the N-terminal differences. Our study reveals the differential function of two ATG8 homologs in autophagy during C. elegans development.

Bone marrow (BM) perivascular stromal cells and vascular endothelial cells (ECs) are essential for hematopoietic stem cell (HSC) maintenance, but the roles of distinct niche compartments during HSC regeneration are less understood. Here we show that Leptin receptor-expressing (LepR+) BM stromal cells and ECs dichotomously regulate HSC maintenance and regeneration via secretion of pleiotrophin (PTN). BM stromal cells are the key source of PTN during steady-state hematopoiesis because its deletion from stromal cells, but not hematopoietic cells, osteoblasts, or ECs, depletes the HSC pool. Following myelosuppressive irradiation, PTN expression is increased in bone marrow endothelial cells (BMECs), and PTN(+) ECs are more frequent in the niche. Moreover, deleting Ptn from ECs impairs HSC regeneration whereas Ptn deletion from BM stromal cells does not. These findings reveal dichotomous and complementary regulation of HSC maintenance and regeneration by BM stromal cells and ECs.

The impact kinetic energy of particle jet determines rock-breaking effect and velocity, and as acceleration device of particle jet, the energy conversion efficiency of nozzle determines rock-breaking efficiency. According to three different types of nozzles of the straight-taper nozzle, the pressure drop model and energy conversion efficiency of particle jet was established, and the energy conversion efficiency under different type nozzles was studied. And then, the acceleration ability of different nozzles for was analyzed with simulation. The results of model calculation and simulation show that constant-acceleration nozzle has the best acceleration ability and energy conversion efficiency. The rock-breaking experiment was carried out with the adopted nozzle, and comparison analysis between experimental results and numerical calculation prove that the applicability of mathematical model.

Sodium-ion batteries (SIBs) are the popular alternative for grid-scale energy storage due to the abundant resources and wide distribution of sodium. However, NaCrO2, one of the most promising layered oxides for cathode materials, suffers from a serious capacity decrease by charging to the potential higher than 3.6 V, attributed to the irreversible chromium ions migration from CrO6 slabs to sodium layers during the initial charge process. Herein we synthesize a novel O3-type layered Na0.88Cr0.88Ru0.12O2 material, which could be well operated at elevated potential 3.8 V. The Na0.88Cr0.88Ru0.12O2 electrode delivers an extended reversible discharge capacity of 156 mAh g−1 and superior rate capability. Moreover, Na0.88Cr0.88Ru0.12O2 electrode shows excellent cycling performance, illustrated by 80.7% capacity retention over 1100 cycles. The results of high-angle annular dark-field scanning transmission electron microscopy measurement indicate that the undesired migration of Cr ions during sodium extraction can be suppressed via Ru doping, which can significantly improve the electrochemical properties of Na0.88Cr0.88Ru0.12O2 compared to NaCrO2. Our findings provide new insights for the common transition-metal migration during cycling layered cathodes, and motivate a new design strategy for high-performance SIBs.

Particle jet impact drilling technology is an efficient method to improve rock-breaking speed by high-speed particles and water-jet impact. The process and damage stage of rock-breaking by combined impact of high-speed water-jet and particles are very complicated, therefore, it is of great significance to study the rock-breaking depth in different damage stages for particle jet impact drilling. Therefore, laboratory test was carried out to study the effects of rock breaking depth on particle diameter, impact velocity and particle mixing ratio. Then, the semi-analytical model of rock-breaking depth under particle jet impact was established in stable damage condition, and the effect factors of rock-breaking depth were numerically analyzed. Based on SPH-FEM coupling algorithm, the dynamical simulation model of particle jet impact rock was established, and the simulation of rock-breaking process and damage evolution under particle jet impact was carried out. Effects of different particle jet parameters on rock-breaking depth were studied by comparing the semi-analytical mode calculation, dynamic simulation and experimental results. Analysis results showed that rock-breaking depth by particle jet impact increases with the increase of jet velocity and particle mixing ratio in high-pressure water-jet, and decreases with the increase of particle diameter. According to the requirements of particle jet impact drilling technology and the analysis findings, it is recommended to adopt water-jet velocity of 200–220 m/s, particle mixing ratio in water-jet of 0.20%, and particle diameter of 1.0 mm. Research results can provide theoretical basis for the parameters optimization and field application of particle jet impact drilling technology.

Hematopoietic myelosuppression is the principal dose-limiting toxicity of curative chemotherapy and radiotherapy in patients with cancer. Both chemotherapy and radiotherapy deplete hematopoietic stem cells (HSCs) via induction of DNA damage. DNA damage contributes to both HSC dysfunction and risk for malignant transformation over time. Extrinsic signals capable of promoting DNA repair in HSCs following injury can potentially improve HSC function and may decrease risk for dysplasia and leukemia over time. Here, we show that treatment with epidermal growth factor (EGF) decreases DNA damage in murine HSCs following irradiation via activation of the DNA-dependent protein kinase catalytic subunit (DNA-PKcs) and nonhomologous end joining (NHEJ) repair. Specifically, we observed that in vitro treatment of irradiated bone marrow (BM) ckit+sca-1+lin- (KSL) hematopoietic stem/progenitor cells (HSPCs) with 100 ng/ml EGF decreased HSPC DNA damage at 1 hour as measured by gamma-H2AX foci and Comet assay (p Conversely, doxycycline inducible, cell - specific suppression of EGFR in hematopoietic cells using SCL-tTA;EGFR-DN mice caused a significant depressed regeneration of BM SLAM+KSL cells and KSL cells at day +10 after irradiation (p=0.005, p=0.02, respectively). SCL-tTA;EGFR-DN mice also displayed significantly decreased survival at day +30 compared to EGFR wild type mice (23.1% survival vs. 58.3% survival, p=0.04). Acute myeloid leukemia (AML) is a common and potentially fatal hematologic malignancy. Allogeneic stem cell transplantation is the only curative therapy for most subtypes of AML, but carries a significant risk of transplant-related mortality. The development of novel therapies to eradicate AML remains a substantial area of unmet medical need. Growth factor receptor bound protein 10 (Grb10) is a member of the family of imprinted genes. Our laboratory demonstrated that in normal hematopoietic system, deletion of the maternal allele of Grb10 significantly increased hematopoietic stem cell long-term repopulating capacity (Yan et al. Cell Rep 2016). Grb10 has been shown to bind the intracellular domain of various tyrosine kinase receptors, e.g. KIT, FLT3 and RET, as well as low-density lipoprotein receptor-related protein 6, a negative regulator of the Wnt/β-catenin pathway. Analyzing RNAseq data from the Leucegene Project, we found that Grb10 is expressed on the majority of patient AML samples regardless of leukemia mutation profile. Radiation injury damages both the bone marrow hematopoietic stem cell and its supportive endothelial niche. In response to radiation stress, bone marrow endothelial cells (BM ECs) upregulate expression of a gene, Sema3A, which encodes for the secreted protein, Semaphorin 3A (SEMA3A), and its receptor, Neuropilin 1 (Nrp1). Commensurate with this, we observed a substantial increase in SEMA3A protein levels in the BM of mice and in NRP1 expression on VE-cadherin+ BM ECs following 500 cGy total body irradiation (TBI) (p In contrast, systemic administration of anti-NRP1 antibody (10 µg, IV every other day), which blocks SEMA3A binding to NRP1 on BM ECs, accelerated regeneration of the sinusoidal BM vasculature at day +10 following 500 cGy TBI compared to irradiated controls. Aging hematopoietic stem cells display distinct abnormalities such as myeloid skewing, decreased repopulating capacity, and leukemia predisposition. Radiation exposure potentiates aging of the hematopoietic system, as evidenced by premature and persistent senescence of HSCs in mice exposed to moderate dose irradiation (Blood, 2014. 123(20): p. 3105-15). Our laboratory demonstrated that EGF promotes hematopoietic stem cell (HSC) regeneration after radiation injury (Nat Med, 2013. 19(3): p. 295-304). Based on these results, we hypothesized that EGF and EGFR signaling may have rejuvenating effects on aging HSCs. For comparisons of young and aged mice, we utilized 2-4 month old mice and >18-24 month old female C57BL/6 mice, respectively. We discovered that aged C57BL/6 mice have decreased levels of EGF in the peripheral blood compared to young mice (p= 0.02) and decreased expression of EGFR on bone marrow (BM) ckit+sca-1+lin- (KSL) stem/progenitor cells (p= 0.03). BM KSL cells from aged mice displayed increased DNA damage in culture with thrombopoietin, SCF and Flt-3 ligand (TSF) compared to young KSL cells (p< 0.0001). Aged BM KSL cells treated with 100 ng/ml EGF had decreased DNA damage compared to aged BM KSL cells treated with TSF only (p< 0.01) Treatment with EGF for 7 days increased the colony forming capacity of aged BM KSL cells (p< 0.01). Importantly, competitive repopulation assays revealed that 7 day in vitro treatment of aged BM KSL cells with EGF significantly increased primary donor cell engraftment (p< 0.01) at 16 weeks compared to aged BM KSL cells cultured with TSF alone. Based on these in vitro observations, we next tested whether systemic administration of a recombinant, pegylated EGF (pEGF), 20 mcg three times weekly for 4 weeks could alter the hematopoietic characteristics of aged C57BL/6 mice. pEGF treatment decreased BM myeloid skewing (p< 0.01) and increased CD3 T cell content (p< 0.01) in aged mice compared to saline-injected controls. Furthermore, competitive repopulation assays revealed that pEGF treatment of aged mice caused a significant increase in functional HSCs capable of competitive engraftment in recipient congenic mice. Total primary donor cell (p< 0.01), myeloid cell (p< 0.01), T cell (p< 0.01) and B cell (p= 0.01) engraftment were significantly increased in recipient mice transplanted with BM cells from aged, pEGF-treated donors compared to recipients of aged, saline-treated donors at 20 weeks. Additionally, secondary transplantation also resulted in increased donor engraftment (p< 0.01) and increased donor myeloid engraftment (p< 0.05) at 12 weeks. These results suggested that systemic administration of pEGF rejuvenates the repopulating ability of aged HSCs. We next sought to determine if deficiency in EGFR signaling could accelerate hematopoietic aging in mice. For this purpose, we utilized a doxycycline-inducible, hematopoietic cell specific EGFR dominant negative mutant model (SCL-tTA;EGFR-DN mice). Interestingly, adult (13 month old) SCL-tTA;EGFR-DN mice displayed increased myeloid skewing in the peripheral blood (p< 0.05) and BM cells from SCL-tTA;EGFR-DN mice generated significantly decreased colony forming cells, compared to age-matched, EGFR-expressing mice (p< 0.01). Mechanistically, BM KSL cells from 13 month old SCL-tTA;EGFR-DN mice demonstrated increased senescence (p< 0.01) and increased expression of p16 (p< 0.05) compared to young mice. In contrast, BM KSL cells from aged C57BL/6 mice that were treated with 100 ng/ml EGF (plus TSF) for 7 days displayed decreased senescence and decreased p16 expression compared to BM KSL cells from aged mice treated with TSF alone. Taken together, these studies suggest that EGF/EGFR signaling declines with age and that reactivation of EGFR signaling via EGF treatment can ameliorate clinically relevant features of hematopoietic aging, including HSC self-renewal capacity.
