Background: There is uncertainty whether prior statin use (PSU) influences the outcomes of intravenous thrombolysis (IVT) in acute ischemic stroke (AIS). Therefore, we conducted a systematic meta-analysis of published clinical trials to evaluate whether statin use combined with IVT therapy may be useful and safe. Methods: Electronic databases were searched for clinical trials that involved outcomes between PSU and nonstatin use (NSU) among stroke patients treated with IVT. Functional outcome measures were 3-month favorable outcome (modified Rankin scale [mRS] 0–2) and 3-month excellent outcome (mRS 0–1). Adverse outcome measures were 3-month all-cause mortality, intracranial hemorrhage (ICH) and symptomatic ICH (sICH). Data were extracted and used to calculate odds ratios (ORs) with 95% confidence intervals (CIs) by Review Manager 5.4 software. BACKGROUND Wernicke's encephalopathy is a disease caused by thiamine deficiency. The lesions usually involve the periphery of the aqueduct, midbrain, tectum, third ventricle, papillary body, and thalamus. It is very rare to affect the vermis and cerebellar hemispheres. CASE SUMMARY We report a 77-year-old female patient admitted to the emergency department of our hospital for 2 d of unconsciousness. Brain magnetic resonance imaging showed increased diffusion weighted imaging signals in the bilateral thalamus, periventricular regions of the third ventricle, corpora quadrigemina, vermis, and cerebellar hemispheres. Wernicke's encephalopathy was considered. She was given thiamine therapy and became conscious after the treatment. CONCLUSION Wernicke's encephalopathy may have various imaging manifestations. Clinicians should keep in mind that Wernicke’s encephalopathy may occur in patients who experience prolonged periods of malnutrition.

This study aimed to explore the role of SCIRT in acute myeloid leukemia (AML) and its interaction with miR-21.This study included 66 AML patients who were diagnosed with AML and received doxorubicin (Dox) treatment. Bone marrow was isolated from all patients before and after treatment to prepare BM mononuclear cells (BMMNCs). BMMNCs from another 60 healthy controls were also collected. The expression of SCIRT and miR-21 were analyzed with RT-qPCR. Subcellular location of SCIRT was analyzed with cellular fractionation assay. RNA pull-down assay was performed to analyze the interaction between SCIRT and miR-21. The roles of SCIRT and miR-21 in regulating the expression of each other were explored with overexpression assay. The role of SCIRT and miR-21 in Dox-induced AML cell apoptosis was analyzed with cell apoptosis assay.SCIRT was downregulated in AML and further downregulated in AML patients who developed drug resistance (DR) after treatment. In contrast, miR-21 was upregulated in AML and further upregulated in AML patients with DR. SCIRT was detected in both nuclear and cytoplasm and it directly interacted with miR-21. SCIRT and miR-21 did not affect the expression of each other. In contrast, SCIRT suppressed the inhibitory role of miR-21 in the apoptosis of AML cells induced by Dox.In conclusion, SCIRT was downregulated in AML and it sponged miR-21 in cytoplasm to increase the chemosensitivity to Dox.

The aim of this study was to determine early weight loss-associated changes in subcutaneous abdominal white adipose tissue (WAT) gene expression in obese men with lowered serum testosterone by RNA next-generation sequencing. Fourteen men, mean age (IQR) 51.6 years (43.4–54.5), BMI 38.3 kg/m2 (34.6–40.8) and total testosterone 8.4 nmol/L (7.5–9.5) provided subcutaneous WAT samples at baseline and after 2 weeks of a very low energy diet. Body weight loss was similar in participants receiving testosterone (n = 6), −5.27 kg [95% CI −6.17; −4.26], and placebo (n = 8), −4.57 kg [95% CI −6.10; −3.55], p = 0.86. The experimental results demonstrate that the implementation aims to proactively control the usage of water by both companies and customers and to achieve higher levels of sustainable water supply.

While the secondary metabolites of DDT such as 2,2-bis(chlorophenyl)-1-chloroethylene (DDMU) and 2,2-bis(chlorophenyl)methane (DDM) have been detected in the environment for several decades, knowledge is extremely limited on their bioaccumulation characteristics. Here, we reported the bioaccumulation and maternal transfer of p,p'-DDMU and p,p'-DDM in two wild fishes, i.e., the northern snakehead (Channa argus) and crucian carp (Carassius auratus), from a DDT contaminated site in South China. The hepatic concentrations of p,p'-DDMU and p,p'-DDM in the fish were up to 549 and 893 ng/g lipid weight, contributing 5.3% and 3.2% in average to ΣDDXs (the sum concentrations of DDT and its 6 metabolites), respectively. The residues of p,p'-DDMU and p,p'-DDM in the fish exhibited interspecific and intraspecific variations, resulting from the differences in lipid content, sex, and body sizes (length and mass) between or within species. Both p,p'-DDMU and p,p'-DDM were consistently detected in the fish eggs, demonstrating their maternal transfer in female fish. The mean eggs to liver lipid-normalized concentration (E/L) ratios of p,p'-DDMU and p,p'-DDM were 0.98 and 1.77 in the northern snakehead, 0.35 and 0.01 in crucian carp, respectively; which were comparable to or even exceeded those of DDT and its major metabolites calculated in the same individual. Statistical analyses of the data showed that the E/L ratios were positively correlated with body sizes of the fish, but negatively correlated with the hepatic concentrations of p,p'-DDMU and p,p'-DDM in females; suggesting the influences of fish sizes and the mother body residues on their maternal transfer efficiencies.

Following cerebral infarction, activated microglia cells can release a large amount of inflammatory cytokines, thereby exacerbating neuronal damage. It has been demonstrated that the long non-coding RNA small nucleolar RNA host gene 1 (SNHG1) exerts a protective effect against cerebral infarction. However, its specific role in cerebral infarction and underlying mechanism have yet to be fully elucidated. The present study aimed to investigate the effects of the SNHG1 and microRNA (miR)-329-3p in cerebral infarction and to determine the underlying molecular mechanisms. An in vitro oxygen-glucose deprivation (OGD) model was established using the BV-2 microglial cell line. The mRNA expression levels of SNHG1 and miR-329-3p were analyzed using reverse transcription-quantitative PCR and the protein expression levels of cleaved caspase-3 and caspase-3 were detected using western blotting. The binding relationship between SNHG1 and miR-329-3p was predicted using starBase and verified using a dual luciferase reporter assay. The release of TNF-α and nitric oxide, as well as caspase-3 activity, were detected using appropriate commercial kits. Flow cytometry analysis was performed to measure cell apoptosis. The results of the present study revealed that the expression levels of SNHG1 were upregulated in the OGD-induced BV-2 cell model. miR-329-3p was discovered to directly target SNHG1, and its mRNA expression levels were downregulated in the OGD-induced BV-2 cell model. The SNHG1-plasmid downregulated miR-329-3p expression levels, while this effect was reversed by transfection with the miR-329-3p mimic. The overexpression of SNHG1 or knockdown of miR-329-3p inhibited OGD-induced BV-2 cell activation. In conclusion, the results of the present study suggested that SNHG1 may reduce microglial cell activity by regulating the expression of miR-329-3p, indicating its potential protective role in cerebral infarction.

Chinese dairy industries have developed rapidly, providing consumers with high-quality sources of nutrition. However, many problems have also appeared during the development process, especially the low quality of milk. To improve milk quality, a large amount of concentrated feed is usually added to the diet within a certain period of time, which increases the milk production to a certain extent. However, long-term feeding with high-concentration feed can lead to subacute rumen acidosis. Therefore, the present study aimed to determine the effect of adding a buffer on subacute rumen acidosis, and the improvement of milk production and milk quality. We also aimed to study the mechanism of promoting mammary gland lactation. A total of 12 healthy mid-lactating goats were randomly divided into two groups, they were high-grain diet group (Control) and buffering agent group. To understand the effects of high-grain diets with buffers on amino acids in jugular blood and the effects of amino acids on milk protein synthesis, Milk-Testing™ Milkoscan 4000, commercial kits, and high-performance liquid chromatography (HPLC) measurements were integrated with the milk protein rate, the amino acid concentration in jugular venous blood samples, quantitative real-time PCR, comparative proteomics, and western blotting to study differentially expressed proteins and amino acids in mammary gland tissues of goats fed high-grain diets. Feeding lactating goats with buffering agent increased the percentage of milk protein in milk, significantly increased the amino acid content of jugular blood (p < 0.05), and increase the amino acid transporter levels in the mammary gland. Compared with the high-grain group, 2-dimensional electrophoresis technology, matrix-assisted laser desorption/ionization-time of flight/time of flight proteomics analyzer, and western blot analysis further verified that the expression levels of beta casein (CSN2) and lactoferrin (LF) proteins in the mammary glands of lactating goats were higher when fed a high-grain diets and buffers. The mechanism of increased milk protein synthesis was demonstrated to be related to the activation of mammalian target of rapamycin (mTOR) pathway signals.

Subarachnoid haemorrhage (SAH) is a type of hemorrhagic stroke that is associated with high morbidity and mortality. New effective treatments are needed to improve outcomes. The pathophysiology of SAH is complex and includes early brain injury and delayed cerebral ischemia, both of which are characterized by blood–brain barrier (BBB) impairment. We isolated brain endothelial cells (BECs) from mice subjected to SAH by injection of blood into the prechiasmatic cistern. We used gene expression profiling to identify 707 unique genes (2.8% of transcripts, 403 upregulated, 304 downregulated, 24,865 interrogated probe sets) that were significantly differentially expressed in mouse BECs after SAH. The pathway involving prostaglandin synthesis and regulation was significantly upregulated after SAH, including increased expression of the Ptgs2 gene and its corresponding COX-2 protein. Celecoxib, a selective COX-2 inhibitor, limited upregulation of Ptgs2 in BECs. In this study, we have defined the gene expression profiling of BECs after experimental SAH and provide further insight into BBB pathophysiology, which may be relevant to other neurological diseases such as traumatic brain injury, brain tumours, ischaemic stroke, multiple sclerosis, and neurodegenerative disorders.

Aim The effectiveness of neuroprotective agents is still unclear. Here we analyzed the clinical outcomes of acute ischemic stroke (AIS) patients treated with human urinary kallidinogenase (HUK) or edaravone (Eda) combined with butylphthalide (NBP). Methods From January 2016 to December 2017, a total of 165 AIS patients were enrolled in this open‐label, randomized controlled clinical study. Patients were randomly allocated into HUK group and Eda group in a ratio of 2:1. All the patients received basic treatments and NBP (200 mg p.o. qid) while HUK group received 0.15 PNA unit of HUK injection (ivgtt. qd) and Eda group received 30 mg Eda (ivgtt. bid) for 14 consecutive days. Independency rate [12‐month modified Rankin Scale (mRS) score ≤ 1] and related factors were compared between the two groups. Results Twelve‐month mRS score of the HUK group (1, IQR 0~1) was significantly lower compared with Eda group (2, IQR 1~3, p, HUK combined with NBP can enhance AIS patients' long‐term independency rate, and the effectiveness of HUK combined therapy is better than Eda.

The crude e-waste recycling has been regulated in China since the late 2000s; however, information on the recent levels and the ecological risks of e-waste derived contaminants such as halogenated flame retardants (HFRs) in the e-waste sites are limited. We therefore examined the concentrations of several HFRs in wild, prey-sized mud carps collected from a typical e-waste site in 2006, 2011 and 2016, to understand the exposure dynamics and ecological risk of these chemicals. Several ecological and biological parameters including δ15N, δ13C, body size and lipid content of the fish were also examined, to ensure an overall uniformity of the sample set among the sampling years. Among the HFRs measured, polybrominated diphenyl ethers (PBDEs) were detected at the highest concentrations (contributing >90% to ∑HFRs), followed by Dechlorane Plus (DPs), polybrominated biphenyls (PBBs), and alternative brominated flame retardants (ABFRs). The fish concentrations of ∑PBDEs, ∑PBBs and ∑DPs significantly dropped by 65%, 57% and 53% from 2006 to 2011, and 12%, 74% and 51% from 2011 to 2016, respectively; likely reflecting the positive impact of the environmental regulations on crude e-waste recycling. The ∑ABFRs concentrations were also decreased by 80% from 2006 to 2011, but increased by 127% from 2011 to 2016; suggesting possible fresh input of these novel HFRs in recent years. In addition to the changes in the HFR concentrations, contaminant profiles in the fish were also changed, possibly due to environmental degradation of the HFRs. Despite our conservative method of risk assessment, we found that PBDEs posed an important risk both for the mud carp and for piscivorous wildlife that inhabit the e-waste site.

Accumulating reports have shown the oncogenic properties of PPMD1 (protein phosphatase, Mg2+/Mn2+ dependent 1D) in different cancer types. This study was undertaken to explore the role and therapeutic potential of PPM1D in colon cancer.HT-29 colon cancer cell line was used in this study. Expression analysis of PPMD1 was performed by qRT-PCR. Cell viability was determined by CCK-8 assay. DAPI, acridin orange/ethidium bromide (AO/EB) and propidium iodide (PI)staining assays were used for apoptosis detection. Cell cycle analysis was performed by flow cytometry. Protein expression was determined by western blot analysis.The results showed that the expression of PPMD1 was significantly upregulated in colon cancer by 3.2 to 4.8 fold. Silencing of PPMD1 caused significant decline in the proliferation rate of the HT-29 colon cancer cells that was due to induction of apoptosis as evidenced by DAPI, AO/EB and PI staining. Annexin V/PI showed a significant increase in the percentage of apoptotic of HT-29 cells upon silencing of PPMD1. The induction of apoptosis was also accompanied by increase in Bax and decrease in Bcl-2 expression. PPMD1 silencing also resulted in arrest of the HT-29 cells in the G2/M phase of the cell cycle which was also associated with upsurge of p21 and p53 and depletion of cyclin B1 expression levels. PPMD1 silencing also caused decrease in the viability of the HT-29 cells which was concomitant with suppression of MMP-2 and MMP-9 expression.These findings suggest that PPMD1 has oncogenic properties in colon cancer and exhibit therapeutic implications in colon cancer treatment.

Activity handover within engineering construction projects is time consuming, takes longer than the actual activities, and therefore, can cause duration delays. This paper presents relay chain seamless handover (RCSH) technology, which is based on relay technology theory and actor-network theory. This study systematically elaborated the RCSH processes, activity relay flows, and methods to address balancing speed under varying conditions. Our analysis results indicate that as compared to traditional activity handover, RCSH improves handover quality and guarantees the duration. RCSH technology was successfully applied in the third phase of the Three Gorges Dam Project, specifically for its concrete construction duration and quality control. Through the use of RCSH, effective overall planning, and efficient coordination of various production links, along with seamless activity handover in the third phase of the project were accomplished— with no additional time consumed by the handover process—and the construction of a concrete dam without a crack was achieved.

Phragmites australis straw (PAS) is an abundant and renewable wetland lignocellulose. Bacillus coagulans IPE22 is a robust thermophilic strain with pentose-utilizing capability and excellent resistance to growth inhibitors. This work is focused on the process study of lactic acid (LA) production from P. australis lignocellulose which has not been attempted previously. By virtue of thermophilic feature of strain IPE22, two fermentation processes (i.e., separated process and integrated process), were developed and compared under non-sterilized conditions. The integrated process combined dilute-acid pretreatment, hemicellulosic hydrolysates fermentation, and cellulose utilization. Sugars derived from hemicellulosic hydrolysates and cellulose enzymatic hydrolysis were efficiently fermented to LA in a single vessel. Using the integrated process, 41.06 g LA was produced from 100 g dry PAS. The established integrated process results in great savings in terms of time and labor, and the fermentation process under non-sterilized conditions is easy to scale up for economical production of lactic acid from PAS.

The background of this paper is the issue of how to overview the knowledge of a given query keyword. Especially, we focus on concerns of those who search for Web pages with a given query keyword. The Web search information needs of a given query keyword is collected through search engine suggests. Given a query keyword, we collect up to around 1,000 suggests, while many of them are redundant. We cluster redundant search engine suggests based on a topic model. However, one limitation of the topic model based clustering of search engine suggests is that the granularity of the topics, i.e., the clusters of search engine suggests, is too coarse. In order to overcome the problem of the coarse-grained clusters of search engine suggests, this paper further applies the word embedding technique to the Web pages used during the training of the topic model, in addition to the text data of the whole Japanese version of Wikipedia. Then, we examine the word embedding based similarity between search engines suggests and further classify search engine suggests within a single topic into finer-grained subtopics based on the similarity of word embeddings. Evaluation results prove that the proposed approach performs well in the task of subtopic clustering of search engine suggests.

Introduction: The pathophysiology of subarachnoid hemorrhage (SAH) is complex and includes disruption of the blood-brain barrier (BBB). We have previously optimized methods to freshly isolate BBB endothelial cells (BECs) and performed genome-wide expression profiling to uncover new therapeutic targets in an unbiased manner. In this study we validated 2 of the most highly upregulated targets, cyclooxygenase-2 (Cox2) and angiopoietin-2 (Angpt2). Methods: The prechiasmatic blood injection mouse model of SAH was used with experimental end-points at 24h. BBB disruption was assessed using intraperitoneal-injected cadaverine-based fluorescent dye. Global neurobehavioral assessments utilized the modified Garcia score. BECs were purified either by magnetic-based sorting for CD45-CD31+ cells or by fluorescence-activated cell sorting for Tie2+Pdgfrb- cells. Microarray results from RNA extracted from BECs were validated with real-time PCR. Immunofluorescence of coronal brain sections and enzyme-linked immunosorbent assays of brain homogenates were performed to determine protein expression. Intraperitoneal injections of the Cox2 inhibitor celecoxib (10mg/kg) were administered 30min and 12h after SAH induction. Results: CD45-CD31+ BECs derived from SAH mice demonstrated 6.6-fold and 5.4-fold increase in Cox2 and Angpt2 gene expression, respectively (n=4 per group, p Conclusions: In summary, we identified Cox2 as a potential therapeutic target in SAH. The observed beneficial effects of celecoxib treatment suggest that Cox2 may be critical to BBB disruption after SAH and its mechanism appears to be independent of Angpt2.

This paper presents techniques of retrieving useful information from a mixture of Web pages collected from either question-answer sites (Q&A sites) or Web search engines. The proposed techniques are designed to discover the maximum possible amount of know-how knowledge from such collections of Web pages, where know-how knowledge is defined as text contents qualified as information source regarding specific domain of questions. The major intent is to build a framework that selects helpful information to provide answers to various problems of interest, such as useful tips to a question. Techniques in this paper primarily attempt to complement knowledge available on Q&A sites with pages collected from search engines via topic models. In order to argue that pages collected from search engine are truly supplements to know- how knowledge on Q&A sites we verify how much extra useful information the Web search engine is able to provide by manually inspecting Web pages aggregated by the topic model.

This paper addresses the problem of identifying irrelevant items from a small set of similar documents using Web search engine suggests. Specifically, we collected volumes of Web pages through Web search engines and inspected the page contents using topic models. Among each cluster of pages sharing the same topic indicated by the topic model, our technique discovers potential content organization in the current page cluster and identifies pages that are out of focus from that topic. The metrics in our approach mainly consist of search engine suggest frequency and inter-document similarity measures. The intuition is that Web pages collected via the same search queries are more likely to share similar contents. We verify this intuition by implementing a subtopic based document selection framework and making quantitative evaluation against human made labeled data sets. Our evaluation result reveals that suggest frequency analysis along with inter-document similarity measure is effective at filtering off-topic documents in small data sets with satisfactory performance.

Protein phosphatase, Mg2+/Mn2+ dependent, 1D (PPM1D) has been associated with carcinogenesis. The present study investigated PPM1D expression as a potential biomarker in colorectal cancer (CRC). PPM1D expression was assessed using immunohistochemistry in 368 patients with CRC. The correlation between PPM1D expression, clinicopathological features and prognosis was analyzed. PPM1D small interfering (si)RNA-induced PPM1D silencing was performed in CRC cell lines to assess the effect of PPM1D on tumor cell proliferation and invasion in vitro. Selecting excellent partnering partners is crucial for project implementation．In view of the drawbacks of traditional methods which only consider the rigid index such as business index, technical data and management data, this paper establishes an evaluation system which considers soft level index and is appropriate for the project of partnering mode. From this system, this paper sets up a comprehensive evaluation mode for partners’ selection based on the entropy and vertical projection method. This mode provides a simple and scientific new way for selecting excellent partnering partners.
