CD26 is a membrane-anchored protein involved in multiple physiological and pathological processes. Here, we investigated correlations between CD26 expression and clinicopathological features in patients with pancreatic tumors.Methods: We collected 170 tumor tissue specimens and 138 paired paratumoral tissues from patients with pancreatic tumors and evaluated CD26 expression using immunohistochemistry.Results: CD26 was expressed in 79.4% of pancreatic tumors, which was significantly (P < 0.001) higher than that in paratumoral pancreatic tissues (23.2%). High expression of CD26 was correlated with ABO blood type (P = 0.035), malignancy degree (P = 0.001), CA199 (P = 0.01), and CA242 (P = 0.027). In pancreatic malignancies, CD26 expression was observed in 80.7% (130/161) of cases. Lower CD26 expression was correlated with longer disease-free survival (P = 0.048) and overall survival (P = 0.024) and was an independent predictor of overall survival (hazard ratio [HR]: 1.713; P = 0.042). Similar results were observed in pancreatic ductal adenocarcinoma (PDAC) tissues, and CD26 expression level (HR: 2.117; P = 0.008) was an independent predictor of overall survival in patients with PDAC. CD26 expression was significantly increased in pancreatic tumors and gradually increased with increasing malignancy degree, suggesting that CD26 may be involved in the tumorigenic proliferation of pancreatic tumors.Conclusion: Therefore, CD26 is a potential marker for early diagnosis and a promising therapeutic target in pancreatic tumors.Keywords: biomarker, CD26, immunohistochemistry, pancreatic cancer, prognosis

The incidence of CRC has been rapidly increasing in China. Transferrin receptor 1 (TfR1) is a key regulator of cellular iron homeostasis. Several studies have demonstrated TfR1 overexpression in a variety of human tumors, but the association between TfR1 and CRC remains unclear.Methods: TfR1 expression was evaluated in six CRC cell lines and tumor tissues. A total of 201 CRC patients were included for immunohistochemistry and 19 pairs of frozen tissues were used for real-time PCR. Cell proliferation, cell cycle, cell migration and invasion, and in vivo carcinogenesis were tested after downregulation of TfR1 by lentivirus. Protein microarray and Western blot analyses were used to explore the underlying mechanisms of TfR1 in CRC.Results: TfR1 expression was higher in CRC tissues than in normal tissues (57.2% vs 22.9%, P＜0.001). TfR1 expression was obviously higher in CRC tissues with well differentiation (P＝0.008), no lymph node metastasis (P＝0.002), no distant metastasis (P＝0.006), no vascular invasion (P＜0.001) and early TNM stage (P＝0.013). CRC patients with TfR1-positive expression had a better survival than those with TfR1-negative expression (P＝0.044). Downregulation of TfR1 expression inhibited cell proliferation, promoted cells from G1 phase to S phase and facilitated cell migration and invasion. Knockdown of TfR1 also suppressed tumor growth in BALB/C-nu mice. Protein microarray and Western blot analyses showed that the Janus protein tyrosine kinase/signal transducer and activator of transcription pathway was activated along with downregulation of TfR1 expression.Conclusion: Though TfR1 was overexpressed in colorectal cancer tissues, there was evidence that downregulation of TfR1 could promote cancer progression.Keywords: colorectal cancer, transferrin receptor 1, tumor progression, JAK/STAT pathway

In this paper, we present a high-sensitivity homogenous fused-silica low pressure sensor. It was constructed from the commercially available standard silica fiber, fused-silica diaphragm and ferrule through CO2 laser all-welding. A Fabry–Perot (FP) cavity was fully sealed between the ultra-thin fused-silica diaphragm and the ferrule end face, and the ultra-thin diaphragm acts as a pressure transducer. We proposed the laser heat conduction welding for the fused-silica diaphragm, and the laser deep penetration point welding for the fiber and the ferrule. So the monolithic structure fused-silica FP interference optical fiber sensor was achieved. The sensor has a vent hole between the fiber and the ferrule forming during the laser deep penetration point welding, which can effectively reduce temperature dependence. The sensor exhibits excellent stability and repeatability, extreme low temperature dependence with 0.025nm/ o C that is 1/76 of the theoretical value, a sensitivity of 5.15nm/kPa, and a resolution of 4.7Pa. With a monolithic fused-silica configuration, the adhesive-free bonding high-sensitivity fused-silica sensor is expected to be suitable for low pressure measurements in extreme harsh environments.

In this article, a reusable surface plasmon resonance (SPR) sensor for rapid detection of copper ions (Cu2+), based on optimized silver/gold (Ag/Au) composite film modified by modified-chitosan (MCS) thin film as an active layer, was achieved. We employed a four-layer Kretchmann semi-cylindrical prism configuration and induced a reference beam into the experimental system to improve the sensitivity and stability, and investigated the cross-linking technique of CS solution, the spin coating process of MCS film, the stability of active layer, the rebirth power of SPR sensor, and the detection for trace Cu2+. The rate of swelling or dissolution of MCS film after flushing sensor chip for four times is less than 9×10−4/min, and the decline of signal value is less than 1.5% after MCS film adsorbing Cu2+ and multiple flushing. The elution and capacity rates are above 96% after five times of elution for the sensor by EDTA solution with 0.5mmol/L. Within 1-5ppm, the SPR sensor was found to possess sensitivity of 0.7271ppm−1. The LOD is 18ppb for the trace Cu2+. The present SPR sensor not only could be used in trace monitoring of Cu2+ but also could be extended to other potential applications.

In order to improve the resolution and the measurement accuracy of optical fiber sensor based on Fabry-Perot (FP) interference, the demodulation algorithm for the FP interference spectrum is analyzed in this paper. Signal demodulation algorithm directly affects the performance of the FP sensor, such as resolution and measurement accuracy. Combined with the basic structure of the sensor, a high-resolution vernier demodulation algorithm is studied based on fast Fourier transform (FFT) algorithm and minimum mean square error algorithm, which has a resolution of 0.084nm according to the experimental results. Then, the demodulation algorithm is applied to the FP interference optical fiber pressure sensor with stainless steel diaphragm. The test results show that the performance is good in the range of 0-1MPa with the resolution of 2.10×10−4MPa and correlation coefficient of 0.99989.
