Abstract

Pyropia yezoensis is the most widely cultivated and economically important alga. Affected by climate change, the cultivation of P. yezoensis has gradually migrated to the northern coast of China, increasing from 6.8% in 2019 to 19.5% in 2023. To date, the genetic impact of northern migration on cultivation resources has not been assessed and analyzed extensively. Here, DNA barcoding (rbcL and cox1) and the presence/absence of intronic-ORFs in mitochondrial regions (rnl and cox1) were applied to investigate genetic diversity in 44 P. yezoensis specimens from 17 aquaculture farms in China, with comparisons to Korean and Japanese cultivated resources. The lower intraspecific variation was 0.31% for the cox1 gene and 0.14% for the rbcL gene, with three haplotypes, indicating that intensive selection and breeding during cultivation had narrowed the germplasm genetic variation. The intron structure of mitochondrial regions showed that the cultivated resources had 17 phenotypes, and the northern specimens shared 35.3% of genotypes with the southern specimens, indicating that the cultivated P. yezoensis is expanding its cultivation ranges through north migration. Even with lower genetic diversity, the northern area of cultivation had already developed 17.6% site-specific specimens. The genetic diversity of cultivated P. yezoensis from the Northwest Pacific is also discussed. 32. Integrated site-specific quantification of faecal bacteria and detection of DNA markers in faecal contamination source tracking as a microbial risk tracking tool in urban Lake ecosystems

Abstract

The presence of feacal-derived pathogens in water is responsible for several infectious diseases and deaths worldwide. As a solution, sources of fecal pollution in waters must be accurately assessed, properly determined and strictly controlled. However, the exercise has remained challenging due to the existing overlapping characteristics by different members of faecal coliform bacteria and the inadequacy of information pertaining to the contribution of seasonality and weather condition on tracking the possible sources of pollution. There are continued efforts to improve the Faecal Contamination Source Tracking (FCST) techniques such as Microbial Source Tracking (MST). This study aimed to make contribution to MST by evaluating the efficacy of combining site specific quantification of faecal contamination indicator bacteria and detection of DNA markers while accounting for seasonality and weather conditions’ effects in tracking the major sources of faecal contamination in a freshwater system (Donghu Lake, China). The results showed that the use of cydgene in addition to lacZand uidAgenes differentiates E. colifrom other closely related faecal bacteria. The use of selective media increases the pollution source tracking accuracy. BSA addition boosts PCR detection and increases FCST efficiency. Seasonality and weather variability also influence the detection limit for DNA markers.

Abstract

Currently, effective methods to eliminate nitrate in wastewater with a low C/N ratio are urgently needed. Considering that sediment is the main nitrogen removal carrier in lakes and that woodchip is an excellent carbon source, this study was undertaken under an integrated system of these two substances to explore the process and efficiency of the denitrification involved. Using methods of kinetics and molecular biology, denitrification processes involving a woodchip-sediment system were investigated in an up-flow bioreactor (UFBR). Performance indices, functional genes, and microbial community changes in this integrated system were explored through water quality monitoring, quantitative PCR, and high-throughput sequencing. When the hydraulic residence time reached above 3.82 days, the nitrate elimination efficiency could achieve an effective level of higher than 93.3% even under the different flow rates. It was also noted that the nitrate elimination rate in this integrated system fitted the Michaelis–Menten kinetics, a combination of zero-order equation (R2 > 0.98) and first-order equation (R2 > 0.95). A combination of zero-order kinetic equations of nitrate elimination and total organic carbon (TOC) release exhibited nitrate elimination efficiency under direct influence of TOC release rate on the C/N ratio. Microbiological analysis indicated co-existence of composting bacteria and denitrifiers as well as aerobic degradation and heterotrophic denitrification, with the former process providing effective carbon sources for the latter process. This study provided a deep insight into the denitrification process of a woodchip-sediment integrated system, which might inspire a potential wastewater treatment method for reduced eutrophication in the aquatic environment.

45. Analysis of Land Use/Cover Change in Guangdong-Hong Kong-Macao Greater Bay Area from 1979 to 2016

Abstract

This study investigated the dynamic changes of land use/cover in Guangdong-Hong Kong-Macao Greater Bay Area (GBA) from 1979 to 2016 by using transfer matrix method combined with remote sensing and GIS. The results show that during this period, Farmland reduced 19744.27 km2with a drop of 81%, 6147.76 km2of which had been designated for industrial land. The land use changed variously among the districts in the GBA, and the comprehensive dynamic index of land use of Zhongshan city was the highest; and that of Huizhou city was the lowest. Social and economic factors such as population, economy, transportation and policy all have an important impact on the land use pattern and its changes in the GBA. © 2021 SPIE. All rights reserved.
