Abstract

The relative amount of AMPA receptors expressed at the surface of neurons can be measured using superecliptic pHluorin (SEP) labeling at their N-terminus. However, the high signal variability resulting from protein overexpression in neurons and the low signal observed in intracellular vesicles make quantitative characterization of receptor trafficking difficult. Here, we establish a real-time live-cell assay of AMPAR trafficking based on fluorescence lifetime imaging (FLIM), which allows for simultaneous visualization of both surface and intracellular receptors. Using this assay, we found that elevating amyloid-beta (Aβ) levels leads to a strong increase in intracellular GluA1 and GluA2-containing receptors, indicating that Aβ triggers the endocytosis of these AMPARs. In APP/PS1 Alzheimer's disease model mouse neurons, FLIM revealed strikingly different AMPAR trafficking properties for GluA1- and GluA3-containing receptors, suggesting that chronic Aβ exposure triggered the loss of both surface and intracellular GluA3-containing receptors. Interestingly, overexpression of protein phosphatase 1 (PP1) also resulted in GluA1 endocytosis as well as depressed synaptic transmission, confirming the important role of phosphorylation in regulating AMPAR trafficking. This new approach allows for the quantitative measurement of extracellular pH, small changes in receptor trafficking, as well as simultaneous measurement of surface and internalized AMPARs in living neurons, and could therefore be applied to several different studies in the future.

Abstract

In this paper we report on the current status of studies on the expected performance for a detector designed to operate in a muon collider environment. Beam-induced backgrounds (BIB) represent the main challenge in the design of the detector and the event reconstruction algorithms. The current detector design aims to show that satisfactory performance can be achieved, while further optimizations are expected to significantly improve the overall performance. We present the characterization of the expected beam-induced background, describe the detector design and software used for detailed event simulations taking into account BIB effects. The expected performance of charged-particle reconstruction, jets, electrons, photons and muons is discussed, including an initial study on heavy-flavor jet tagging. A simple method to measure the delivered luminosity is also described. Overall, the proposed design and reconstruction algorithms can successfully reconstruct the high transverse-momentum objects needed to carry out a broad physics program., Comment: contribution to Snowmass 2021

Abstract

In the path towards a muon collider with center of mass energy of 10 TeV or more, a stage at 3 TeV emerges as an appealing option. Reviewing the physics potential of such muon collider is the main purpose of this document. In order to outline the progression of the physics performances across the stages, a few sensitivity projections for higher energy are also presented. There are many opportunities for probing new physics at a 3 TeV muon collider. Some of them are in common with the extensively documented physics case of the CLIC 3 TeV energy stage, and include measuring the Higgs trilinear coupling and testing the possible composite nature of the Higgs boson and of the top quark at the 20 TeV scale. Other opportunities are unique of a 3 TeV muon collider, and stem from the fact that muons are collided rather than electrons. This is exemplified by studying the potential to explore the microscopic origin of the current $g$-2 and $B$-physics anomalies, which are both related with muons., Comment: 73 pages, 28 figures; Contribution to Snowmass 2021

Abstract

Among the post-LHC generation of particle accelerators, the muon collider represents a unique machine with capability to provide very high energy leptonic collisions and to open the path to a vast and mostly unexplored physics programme. However, on the experimental side, such great physics potential is accompanied by unprecedented technological challenges, due to the fact that muons are unstable particles. Their decay products interact with the machine elements and produce an intense flux of background particles that eventually reach the detector and may degrade its performance. Abstract

Inflammatory bowel disease (IBD) defines two chronic inflammatory diseases of the gastrointestinal tract, ulcerative colitis and Crohn's disease. The prevalence of these diseases is increasing in the western world, and there is currently no cure for either. Biological therapy is widely used to treat IBD; however, patients often lose response to these treatments. There is a need to stratify patients into further disease subsets, which require biomarkers that could act as read-outs of disease progression and severity. Granzymes (A, B, H, K and M) are released by cytotoxic T cells and NK cells, with granzyme A and B the most abundant forms found in humans. Granzyme A has previously been reported to be elevated in tissue biopsies from IBD patients who responded well to anti-integrin therapy (etrolizumab) and subsequently achieved clinical remission, in comparison to those who did not respond. Granzyme A has also recently been reported to have a pro-inflammatory role in other inflammatory diseases including rheumatoid arthritis. Granzyme B, typically thought of as a cytotoxic protease, has been reported to be elevated in the serum of IBD patients. Based on these data, in chapter 3, we sought to characterise the expression of granzymes A and B in IBD tissue samples and to determine whether the expression of granzymes A or B could be used as blood and/or stool-based readout of IBD activity. We also investigated the source of granzyme A in IBD tissue, using fresh tissue biopsies taken from patients undergoing research colonoscopies. Granzymes typically must be activated before they exert their effects on target cells. We therefore hypothesised that activity of an enzyme would be a more informative biomarker than expression of the enzyme, as only when active are these granzymes known to have cytotoxic effects. To have a read-out of granzyme A activity, in Chapter 4, we sought to create substrate-based probes that would fluoresce when cleaved by active granzyme A but would remain intact in the presence of inactive granzyme A. Lastly, in Chapter 5, we also sought to understand the pro-inflammatory role of granzyme A and identified that it could induce the secretion of pro-inflammatory cytokines from monocytes. We investigated the receptors responsible for the pro-inflammatory effects on monocytes and used an inhibitor to block the pro-inflammatory effects of granzyme A. Finally, we synthesised a new granzyme A inhibitor, which was shown to be efficacious in vitro. We performed preliminary in vivo experiments in a mouse model of colitis using a PAR-1 receptor inhibitor and the granzyme A inhibitor but observed no efficacy at the concentrations used. Altogether, the work in this thesis characterised the expression of granzymes A and B in gut tissue from IBD patients and created new substrate-based probes for granzyme A that could act as a read-out of enzyme activity rather than expression. We also began to investigate how granzyme A could induce inflammation in an IBD setting and lastly created a granzyme A inhibitor to attenuate the pro-inflammatory effects of the enzyme.

41. An exploration of female early adolescent self-presentation and social comparisons, when engaging with social networking sites

Abstract

Background: The use of social networking sites (SNS), including Instagram and Snapchat, is an integral aspect of adolescent life (Throuvala et al., 2019). Existing research has explored the possible impact of using SNS, which is described as a 'double-edged sword' (Keles et al., 2020) due to its benefits and potential risks for adolescents. However, a significant proportion of research concerns a sample of older adolescents, this is despite findings that suggest children as young as eight years are accessing SNS every day (Children's Commissioner, 2018). Alongside being frequent, devoted users of SNS (Shankleman et al., 2021) adolescence is also a formative developmental period for identity (Erikson, 1968). One's identity is socially embedded (Davis, 2011) and individuals gain an insight into their performance through comparing themselves to others, which can be facilitated by SNS (Vogel et al., 2014). Therefore, it is necessary to further explore the development of self and social comparisons made on SNS, due to possible associations between SNS and this aspect of development. Furthermore, research utilising an early adolescent sample is required, to acknowledge the increased use of SNS, within this demographic. Method: The sample comprised of six females aged between 12 and 13 years, who all attend the same school. Semi-structured interviews were completed to explore views and experiences. The research was positioned within a social constructionist paradigm (Burr, 2015) and data analysed using Reflexive Thematic Analysis (Braun & Clarke, 2019a). Findings: Results suggest an individual's self-presentation online is a performance, where a selective or restricted self is shared. The presentation of self is underpinned by the perception of the audience, which is of great importance to adolescents. Participants spend time hypothesising possible audience views and adapt their performance accordingly, to ensure it will be accepted. Feedback can also strengthen an individual's self and participants engage in a number of actions to protect themselves online. This is in relation to online risk, alongside the protection of self from negative feedback. The sample was found to engage in a range of social comparisons against others, which can provoke a variety of emotional responses and possibly influence the development of identity, such as through individuals engaging in identity exploration.

Abstract

Proteins are intrinsically flexible macromolecules that undergo internal motions with time scales spanning femtoseconds to milliseconds. These fluctuations are implicated in the optimization of reaction barriers for enzyme catalyzed reactions. Time, temperature, and mutation dependent hydrogen-deuterium exchange coupled to mass spectrometry (HDX-MS) has been previously employed to identify spatially resolved, catalysis-linked dynamical regions of enzymes. We now extend this technique to pursue the correlation of protein flexibility and chemical reactivity within the diverse and widespread TIM barrel proteins, targeting murine adenosine deaminase (mADA) that catalyzes the irreversible deamination of adenosine to inosine and ammonia. Following a structure-function analysis of rate and activation energy for a series of mutations at a second sphere phenylalanine positioned in proximity to the bound substrate, the catalytically impaired Phe61Ala with an elevated activation energy (Ea = 7.5 kcal/mol) and the wild type (WT) mADA (Ea = 5.0 kcal/mol) were selected for HDX-MS experiments. The rate constants and activation energies of HDX for peptide segments are quantified and used to assess mutation-dependent changes in local and distal motions. Analyses reveal that approximately 50% of the protein sequence of Phe61Ala displays significant changes in the temperature dependence of HDX behaviors, with the dominant change being an increase in protein flexibility. Utilizing Phe61Ile, which displays the same activation energy for kcat as WT, as a control, we were able to further refine the HDX analysis, highlighting the regions of mADA that are altered in a functionally relevant manner. A map is constructed that illustrates the regions of protein that are proposed to be essential for the thermal optimization of active site configurations that dominate reaction barrier crossings in the native enzyme.

50. Data-Informed Course Design and the DEI Imperative

Abstract

Bay Path University in Massachusetts developed a COVID dashboard and related safety practices enabled by data (Anderson, 2020) during the pandemic. The dashboard has capabilities, in which information technology staff partnered with executive management, human resources staff, health offices, and others to identify key performance indicators (KPIs) about the pandemic's effects and safety on the campus, can be applied to academic and student success practices. In this article, the authors describe the process The American Women's College at Bay Path University took to use data to inform course designs in the most impactful ways to drive student success, both prior to and during the pandemic. They also identify the institutional student success stakeholders, whom they consider akin to a "treatment team" in a clinical setting, professionals who bring experience to bear so that the entire team can understand student learning from a holistic perspective. This approach has allowed the college to marshal their institutional resources in strong ways. They will also provide details of their iterative learning processes, as well as the next steps that their institution will take to hold themselves accountable to equitable student outcomes.
