Abstract

Background: Protein powder identification presents a challenge in quality control. There is current deliberation of the specificity of methods for the identification of milk proteins, and the consensus identification method of whey protein from the United States Pharmacopeia Food Chemical Codex relies on a combined analysis of the testing of ash, fat, lactose, loss on drying, and protein. These methods are non-specific. Milk and whey proteins both contain background DNA content. Both milk and whey proteins retain source DNA (cow), but also have bacterial DNA from natural flora, the dairy plant, and in whey protein, the cheesemaking process. The DNA in these materials is retained post-processing, even after the pasteurization process. Objective: By utilizing 16S metagenomics, the bacterial DNA in protein powders can be sequenced and cross-referenced to a curated library to ultimately create a microbiome profile of these raw materials. This microbiome can be measured for alpha and beta diversity, specifically how many and which species of bacteria are present. Method: Using 16S metagenomics, we measure alpha and beta diversity of the microbiome profile of each protein powder and use principle coordinate analysis to produce differential groupings, providing a novel identification method for raw materials. Results: In this study, we demonstrate that the microbiome of cow proteins can be used for raw material identification, as the microbiome of milk and whey proteins differ significantly. We also demonstrate that the microbiome of whey protein concentrate can differ from supplier to supplier. Conclusions: Microbiome profiling by 16S metagenomics can be an important forensic tool for quality control. 37. Relational Empowerment Among Public Safety Outreach Workers and Violence Interventionists in U.S. Cities: A Qualitative Inquiry

Abstract

The nation-wide uprisings for racial justice in the Summer of 2020 brought into mainstream awareness the calls to shift resources away from police departments and channel funds to mental health, education, and other social services. However, in the wake of recent post-pandemic surges in community violence, police budgets increased, while public opinion for grassroots, radical initiatives like “defund the police” that many activists, organizers, and abolitionists pushed into public consciousness has faltered. Studies have shown that the implementation of community-led public safety programs significantly reduce incidents of street violence in U.S. cities. Thus, research that informs community-driven violence intervention and prevention is vital to curbing this public health crisis. The current qualitative study builds on the existing literature on relational empowerment, community organizing, and community-led public safety to explore the following: (a) How do community-led public safety outreach workers, violence interventionists, and crisis responders experience relational empowerment in the context of community organizing? (b) How do they utilize relationships to enact community-level change? And (c) What are their perceptions of the barriers to relational empowerment and effective community-led violence prevention? Thematic analysis revealed unique pathways via all components of relational empowerment. Norms Built on Respect and Collective Over the Individual were identified as important themes within bridging social divides, as participants effectively utilized these principles when navigating internal disputes. The capacity to Navigate Power Structures and Channel Social Ties were themes within collaborative competence that were vital for strengthening community trust, building interorganizational partnerships, and adapting in interpersonal exchanges with those in positions of power to meet organizational goals. Facilitating others’ empowerment consisted of leadership Balancing Worker Autonomy & Guidance. Passing on legacy was identified as a relational skill consisting of Transmitting Self-Efficacy & Commitment in the form of leadership modeling, but also a competency among membership who carry legacy as they Learn, Internalize, & Apply cultural norms, values, principles, knowledge, and skills. This further demarcates earlier conceptual overlap between Passing on Legacy and Facilitating Others’ Empowerment (Christens, 2012). Participants described leadership modeling as important for increasing self-efficacy (i.e., emotional empowerment). Organization as Family, a theme within mobilizing networks was shown to increase worker motivation and engagement (i.e., behavioral empowerment). Furthermore, a reciprocal relationship was revealed between cognitive and emotional empowerment, and civic engagement. Important relational competencies not included within the construct of relational empowerment were also identified, which involved resident outreach and engagement. Lastly, relational processes tied to resident needs assessment and intervention, as well as violence intervention and mediation were also delineated. This study contributes to existing research on the practice of community-led public safety and the theoretical development of relational empowerment as a construct. A penal abolitionist framework is used to understand structural issues contributing to barriers to relational empowerment.

A new method is described for preparing controllably displaceable monolayers by using a carboxyl-functionalized self-assembled monolayer and in situ Fischer esterification, simple and reversible chemical reaction. The generality of reversible lability is examined by applying the in situ esterification reaction to the structurally distinct carboxyl-functionalized molecule 3-mercapto-1-adamantanecarboxylic acid.

Abstract

Background: The applications of deoxyribonucleic acid (DNA) barcoding methods have been extended from authenticating taxonomic provenance of animal products to identifying botanicals used as herbal medicine and in botanical dietary supplements. DNA barcoding methods for botanical identification must be adequately validated to meet regulatory compliance. Objective: The goal of this study is to provide a validation protocol for a two-tiered DNA barcoding method that aims to identify raw botanicals. Methods: A barcode database was computationally validated to define the barcode combinations that can unambiguously identify botanicals in the database. A maximum variation sampling technique was used to capture a wide range of perspectives relating to DNA barcodebased botanical identification, including plant parts and species distance, for the experimental validation. Twenty-two authenticated botanicals were purposively sampled from different plant parts--covering both closely related and distantly related species--to validate the two-tiered DNA barcoding method. The performance of the method was assessed on accuracy, precision, ruggedness, and uncertainty. Results: High accuracy (100%) and precision (1.0) were obtained from the validation samples. The method was also found to be rugged and have acceptable uncertainty. Abstract

This thesis provides a capabilities-based approach to assessing the bioterrorism threat from non-state actors. Through comparative case study, prior bioterrorism attacks are analyzed to assess capability in the three areas necessary to complete a biological weapons attack: obtaining or isolating a pathogen, weaponizing the agent, and employing or disseminating the weapon. The three cases are the Rajneeshee cult in 1984, the Aum Shinrikyo cult in the early 1990's, and the United States Postal System anthrax attacks of 2001. In contrast to current wisdom that employing biological weapons is too difficult for non-state actors, this thesis reveals a broad spectrum of capability in all studies in the areas necessary to culminate an attack. Applications of these findings must be used to assess risk generally rather than against specific groups because capability is deemed to be exptremely difficult to track. The these finds that a significant threat exists but not large enough to be over-hyped above other national security concerns. In light of this, recommendations are provided for U.S. biodefense policy emphasis in the areas of the nonproliferation regime, attribution capabilities, and defending against the changing nature of future attacks with a particular emphasis on the public health system. http://archive.org/details/thebioterrorismt109452416 Approved for public release; distribution is unlimited.
