5. Assessing α-Bisabolol as a Transmucosal Permeation Enhancer of Buccal Local Anesthetics.

Abstract

Needle-free buccal anesthesia improves dental treatment outcomes for both patients and dentists. In this study, we report on an assessment of the enhancement effects of α-bisabolol on the in vitro transmucosal permeation of prilocaine hydrochloride (PCl) and lidocaine hydrochloride (LCl) from needleless buccal films. We also evaluated the mechanical properties of the film, which consisted of Methocel™ K100 LV as the film-forming polymer (3% m·m−1), PEG 400 as a cosolvent (15% m·m−1 based on drug loading), α-bisabolol (15 and 30% m·m−1 based on drug loading), and the drugs combined at a 1:1 ratio (15 mg·unit−1). The porcine esophageal epithelium was used as a membrane barrier, and artificial saliva was the release medium. After a 1 h experiment at 25 ± 2 °C, α-bisabolol significantly decreased, rather than enhanced, the permeation fluxes (five-fold), permeability coefficients (seven-fold), and retentions (two-fold) of both PCl and LCl through the epithelium, regardless of the concentration. Moreover, the resistance and flexibility of the films markedly decreased compared to those without α-bisabolol. Therefore, under the experimental conditions, using α-bisabolol as a buccal permeation enhancer for the hydrophilic local anesthetics PCl and LCl from buccal films is not feasible. 15. Comparative homology of Pleurotus ostreatuslaccase enzyme: Swiss model or Modeller?

Abstract

AbstractLaccases stand out in the industrial context due to their versatile biotechnological applications. Although these enzymes are frequently investigated, currently, Pleurotus ostreatuslaccase structural model is unknown. Therefore, this research aims to predict and validate a P. ostreatuslaccase theoretical model by means of comparative homology. The laccase target’s primary structure (AOM73725.1) was obtained from the NCBI database, the model was predicted from homologous structures obtained from the PDB (PDB-ID: 5A7E, 2HRG, 4JHU, 1GYC) using the Swiss-Model and Modeller, and was refined in GalaxyRefine. The models were validated using PROCHECK, VERIFY 3D, ERRAT, PROVE and QMEAN Z-score servers. Moreover, molecular docking between the laccase model (Lacc4MN) and ABTS was performed on AutoDock Vina. The models that were generated by the Modeller showed superior stereochemical and structural characteristics to those predicted by the Swiss Model. The refinement made it difficult to stabilize the copper atoms which are typical of laccases. The Lacc4MN model showed the interactions between the amino acids in the active site of the laccase and the copper atoms, thereby hinting the stabilization of the metal through electrostatic interactions with histidine and cysteine. The molecular docking between Lacc4MN and ABTS showed negative free energy and the formation of two hydrogen bonds involving the amino acids ASP 208 and GLY 268, and a Pi–sulfur bond between residue HIS 458 and ABTS, which demonstrates the typical catalytic functionality of laccases. Furthermore, the theoretical model Lacc4MN presented stereochemical and structural characteristics that allow its use in silicotests.Communicated by Ramaswamy H. Sarma

Abstract

Hexachlorobenzene (HCB) is a pollutant still found in the environment despite being widely banned. Considering that basidiomycetes are useful to degrade a variety of organochlorinated pollutants, we therefore report the influence of HCB on the ligninolytic enzymatic system of Deconica castanella. The inoculum was prepared with sugarcane bagasse and soybean flour and was added in soil with and without HCB (2000 mg kg soil−1), 5% emulsion containing soybean oil and Tween 20 at proportion 9:1, v:v; with 70% moisture at 25°C. Fungal biomass was quantified by widely acknowledged growth biomarker ergosterol. The extraction of the enzymatic complex was performed and laccase, Mn‐dependent peroxidase (MnP), and lignin peroxidase (LiP) activities were determined. Furthermore, HCB and its metabolites were quantified by gas chromatography and chlorides by potentiometric titration. Results evidenced that HCB did not interfere in fungal growth, though the only detected enzymatic activity was laccase. MnP and Lip were not detected during D. castanella growth in soil. The peak of laccase enzymatic activity occurred in the presence of HCB. In addition, the laccase exhibited thermostability. Therefore, we hereby shed light on the role of laccase in the degradation of HCB by an efficient low‐cost and environmentally safe detoxification mechanism. [ABSTRACT FROM AUTHOR]

Abstract

“Arnica” extracts are widely used in folk medicine to treat acute and chronic inflammatory ailments. Nevertheless, their toxic effects upon systemic use are still not fully understood. Therefore, this work provides a systematic review on the safety of arnica extracts following preclinical trials covering their oral and intraperitoneal administration in animal models. Henceforth, PRISMA guideline was followed and the study protocol was registered in PROSPERO (CDR42020167112). Searches were performed in PubMed (MEDLINE), Scopus, Science Direct, Web of Science (Science Citation Index), and Health Virtual Library (BVS) databases; while SYRCLE’s Risk of Bias tool and CAMARADES checklist were used to assess scientific quality. From 382 articles, five studies met eligibility criteria and underwent qualitative analysis. Data of acute toxicity was reported in all the selected articles, and the treatment time was up to 14 days. Moreover, the following species were reported: Solidago chilensis (hazard categories of 4 and 5 for i.p and v.o administration, respectively); Solidago microglossa (hazard category of 3, i.p); Lychnophora trichocarpha (hazard category of ≥ 4, i.p); and Lychnophora pinaster (hazard category of ≥ 4, v.o). The alcoholic extracts showcased greater toxic potential, which increased in a dose-dependent manner after 100 mg/Kg. Concerning organ-specific toxicity, the articles reported hepatotoxicity and nephrotoxicity following histopathological analysis. However, the safety of S. chilensis, L. pinaster, L. trichocarpha, and S. microglossa following systemic administration remains unclear due to the limited experimental quality of the included papers, as well as the lack of reported chronic toxicity, pharmacokinetics, and mutagenicity studies. Los extractos de "árnica" se utilizan ampliamente en la medicina popular para tratar dolencias inflamatorias agudas y crónicas. Sin embargo, sus efectos tóxicos sobre el uso sistémico aún no se comprenden completamente. Por lo tanto, este trabajo proporciona una revisión sistemática sobre la seguridad de los extractos de árnica después de ensayos preclínicos que cubren su administración oral e intraperitoneal en modelos animales. En adelante se siguió la guía PRISMA y se registró el protocolo del estudio en PROSPERO (CDR42020167112). Las búsquedas se realizaron en las bases de datos PubMed (MEDLINE), Scopus, Science Direct, Web of Science (Science Citation Index) y Health Virtual Library (BVS); mientras que la herramienta Riesgo de sesgo de SYRCLE y la lista de verificación CAMARADES se utilizaron para evaluar la calidad científica. De 382 artículos, cinco estudios cumplieron los criterios de elegibilidad y se sometieron a análisis cualitativos. Los datos de toxicidad aguda se informaron en todos los artículos seleccionados y el tiempo de tratamiento fue de hasta 14 días. Además, se notificaron las siguientes especies: Solidago chilensis (categorías de peligro 4 y 5 para administración i.p y v.o, respectivamente); Solidago microglossa (categoría de peligro 3, i.p); Lychnophora trichocarpha (categoría de riesgo ≥ 4, i.p); y Lychnophora pinaster (categoría de riesgo ≥ 4, v.o). Los extractos alcohólicos presentaron un mayor potencial tóxico, el cual aumentó de manera dosis-dependiente a partir de los 100 mg / kg. Con respecto a la toxicidad específica de órganos, los artículos informaron sobre hepatotoxicidad y nefrotoxicidad después del análisis histopatológico. Sin embargo, la seguridad de S. chilensis, L. pinaster, L. trichocarpha y S. microglossa después de la administración sistémica sigue sin estar clara debido a la limitada calidad experimental de los artículos incluidos, así como a la falta de estudios de toxicidad crónica, farmacocinética y de mutagenicidad. Abstract

Linear furanocoumarins are skin sensitizers and anticancer agents whose appeal in skincare therapeutics is widely exploited. Owing to the need to predict the biological activities of medicines, this work aimed the investigate the predicted dermal toxicity of linear furanocoumarins through chemoinformatic approaches. Therefore, eight major linear furanocoumarins of interest in medicine were selected, and their pharmacophores / toxicophores were modelled and inputted in several databases and cheminformatic toolsets previously described in the literature. Moreover, Principal Components Analysis was performed to allow multivariable comparisons. Results showcased that the first two PCs accounted for 95.48% of all variance in the model, and molecular weight and polar surface showcased a positive correlation to Log P and Log Kp, which may be involved in skin penetration. Moreover, the pharmacophore modelling evidenced superimposition between linear furanocoumarins, ethidium bromide and acridine orange, thereby suggesting that these compounds share similar biological effects, supported by their acknowledged DNA intercalating activities. Therefore, this work showcased the application of various cheminformatic tools to screen the dermal toxicity of chemicals.

Abstract

This brief overview is intended to shed light on the current drug repositioning (also called drug repurposing) in the therapeutics of the novel coronavirus disease which emerged in 2019 (COVID-19). In this sense, the repositioning drugs for new indications can offer a better risk-versus-reward trade-off when compared to other drug development strategies, given that it makes use of drugs whose safety profile are already understood. Nonetheless, this approach allows healthcare professionals to promptly tackle the disease by investigating readily available drugs against it.

Abstract

Given the relevance of impairing reactive oxygen species (ROS) buildup in tissues, the use of exogenous antioxidants is highly regarded as a valid prophylaxis against oxidative stress and its deleterious effects on organisms. In this regard, Camelia sinensis uses as a remarkable antioxidant source have been reported in various folk and standard medicine systems around the world. In this chapter, the thermodynamics and kinetics of Camelia sinensis constituents is concisely discussed focusing on the implications of its redox profiling toward antioxidant capacity. Notwithstanding, the biological repercussion of ROS reduction as well as its therapeutic potential is also addressed to provide readers a basic background on the relevance of investigating the physicochemical features of medicinal plants.

Transition metals are highly sought after in electrode construction due their eventual electrocatalytic effects. In this context, this work presents the study of several metal oxide modified electrodes focusing in the evaluation of their electrochemical features through voltammetry and Electrochemical Impedance Spectroscopy (EIS). Moreover, a predictive statistical model was also developed using LASSO regression. Henceforth, glassy carbon electrode (GCE) and carbon paste electrode (CPE) were used to establish correlations between the herein studied modifications, namely: PdO@CPE, TiO2@CPE and LaFeO3@CPE. Results evidence that metal oxide modified electrodes may increase signal detection through electrocatalytic effects, however the herein developed model evidenced that surface area plays a major role in electrode response through voltammetry.

Abstract

This work showcased the chemoinformatic study of isoxazoline ectoparasiticides: Fluralaner (FLU) and Afoxolaner (AFO) interactions with l-glutamate-gated chloride channels (3RHW). In order to evaluate inhibition thermodynamics, computational approaches such as molecular docking were employed. Results evidenced that FLU-3RHW highest scoring pose presented lower Gibbs free energy and henceforth, lower Ki values than AFO-3RHW. The findings herein reported suggest therefore that computational methods might be useful to study the thermodynamic features of ectoparasiticides used in veterinary care, what might shed further light on their chemical and pharmacological properties.

Abstract

Cyclobenzaprine (CBP) is a centrally acting muscle relaxant whose myriad of therapeutic applications imply the need of better understanding its pharmacokinetics and thermodynamics. Henceforth, this work was concerned with an in silico investigation of CBP main metabolizers in the human organism, namely CYP1A2 and CYP3A4. For this purpose, computational methods were employed, such as molecular docking and other semi-empirical approaches. Results evidenced that the model herein depicted for CBP-CYP1A2 may not reproducibly represent the physiological interaction between CBP and this enzyme. Moreover, CBP-CYP3A4 docking results evidence thermodynamic feasibility of the molecular docking model and were further corroborated by literature, what may reproducibly represent a possible interaction between CBP and this macromolecule.
