Abstract The emotional valence of animals is challenging to assess, despite being a key component of welfare. In this study, we attempted to assess emotional valence through memory in 1- and 3-week-old piglets. It was hypothesized that piglets would spend less time in a pen where they experienced a negative event (castration) and more time in a pen where they experienced a positive event (enrichment). A testing apparatus was designed with three equally sized pens: two outer sections serving as treatment pens containing unique visual and tactile cues and a center section remaining neutral. Piglets received either negative or positive condition in one outer treatment pen and a sham treatment in the opposite. Various methods were tested (age of piglets, number and length of conditioning sessions, passive vs. active conditioning). Contrary to expectations, piglets did not decrease their time in the pen associated with the negative condition or increase their time in the pen associated with the positive condition. However, when exposed to the positive condition, results indicate older piglets developed an aversion towards the sham treatment. This study provides methodological groundwork for the application of place conditioning in piglets and highlights the nuances important for the use of cognitive tests to assess animal welfare.

Abstract Infections with Enterobacterales (E) are increasingly difficult to treat due to antimicrobial resistance. After ceftriaxone replaced chloramphenicol (CHL) as empiric therapy for suspected sepsis in Malawi in 2004, extended-spectrum beta-lactamase (ESBL)-E rapidly emerged. Concurrently, resistance to CHL in Escherichia coli and Klebsiella spp. decreased, raising the possibility of CHL re-introduction. However, many phenotypically susceptible isolates still carry CHL acetyltransferase (cat) genes. To understand the molecular mechanisms and stability of this re-emerging CHL susceptibility we use a combination of genomics, phenotypic susceptibility assays, experimental evolution, and functional assays for CAT activity. Here, we show that of 840 Malawian E. coli and Klebsiella spp. isolates, 31% have discordant CHL susceptibility genotype–phenotype, and we select a subset of 42 isolates for in-depth analysis. Stable degradation of cat genes by insertion sequences leads to re-emergence of CHL susceptibility. Our study suggests that CHL could be reintroduced as a reserve agent for critically ill patients with ESBL-E infections in Malawi and similar settings and highlights the ongoing challenges in inferring antimicrobial resistance from sequence data.

In humans, early painful experiences can increase pain sensitivity later in life, but little is known regarding this phenomenon in cattle. This study assessed if a painful event early in life affects later pain sensitivity in 40-d-old calves. Holstein calves (n = 26) were randomly assigned to control or treatment conditions, blocked within sex and birth weight. At 9.5 ± 1.8 d old, treatment calves had 1 horn bud disbudded using caustic paste, whereas control calves had 1 horn sham disbudded; in both cases, multimodal pain control was provided. All calves had the contralateral horn bud disbudded using a hot iron 4 wk later, again with multimodal pain control. Mechanical nociceptive responses were assessed weekly using an algometer applied adjacent to both horn buds and on the rump, beginning 3 d before the first disbudding and ending 30 d after the second disbudding. Following the second disbudding, both groups of calves showed evidence of increased sensitivity (i.e., algometer pressure declined 3.69 ± 0.60 √N to 2.13 ± 0.70 √N) on the contralateral bud, but there was no difference between the control group and the treatment groups. An interaction between treatment and time, likely driven by treatment differences, was found on the rump when tested 5 h after the second disbudding event. These responses are not consistent with the hypothesis that an early pain experience results in increased sensitivity to later painful experiences.

The Channa limbata fish is a type of tropical freshwater fish of the Channidae family which is relatively small as an aquarium ornamental fish with a distinctive color at the tip of its dorsal fin and has a snake-like head (dwarf snakehead). Natural snakehead fishing activities have threatened its sustainability. Breeding C. limbata fish through cultivation can increase its potential for sustainable use. This study aims to evaluate the acclimation response of wild-type dwarf snakehead fish in captivity and its sexual characteristics as a basis for domestication and hatchery technology. The fish samples used were natural catches from rivers in West Java measuring 150 mm of body length then individually acclimated indoors in an aquarium (35×20×20 cm) for 14 days. Snakehead fish live in shallow, slow-flowing river waters with a temperature of 20.2-21.3°C, TDS 16-24 mg/L at neutral pH, while the rearing water temperature and TDS are higher (temperature: 24.9-27.6°C; TDS: 88-110 mg/L). The fish mortality rate during acclimation reached 25% in fish measuring >150 mm of length on tenth day, while fish measuring 150 mm kemudian diaklimasi indoor di akuarium (35×20×20 cm) selama 14 hari. Abstract Due to their exceptional solubility and stability, nanobodies have emerged as powerful building blocks for research tools and therapeutics. However, their generation in llamas is cumbersome and costly. Here, by inserting an engineered llama immunoglobulin heavy chain (IgH) locus into IgH-deficient mice, we generate a transgenic mouse line, which we refer to as ‘LamaMouse’. We demonstrate that LamaMice solely express llama IgH molecules without association to Igκ or λ light chains. Immunization of LamaMice with AAV8, the receptor-binding domain of the SARS-CoV-2 spike protein, IgE, IgG2c, and CLEC9A enabled us to readily select respective target-specific nanobodies using classical hybridoma and phage display technologies, single B cell screening, and direct cloning of the nanobody-repertoire into a mammalian expression vector. Our work shows that the LamaMouse represents a flexible and broadly applicable platform for a facilitated selection of target-specific nanobodies.

Summary: Background: Irritable bowel syndrome (IBS) is a common and debilitating disorder manifesting with abdominal pain and bowel dysfunction. A mainstay of treatment is dietary modification, including restriction of FODMAPs (fermentable oligosaccharides, disaccharides, monosaccharides and polyols). A greater response to a low FODMAP diet has been reported in those with a distinct IBS microbiome termed IBS-P. We investigated whether this is linked to specific changes in the metabolome in IBS-P. Methods: Solid phase microextraction gas chromatography-mass spectrometry was used to examine the faecal headspace of 56 IBS cases (each paired with a non-IBS household control) at baseline, and after four-weeks of a low FODMAP diet (39 pairs). 50% cases had the IBS-P microbial subtype, while the others had a microbiome that more resembled healthy controls (termed IBS-H). Clinical response to restriction of FODMAPs was measured with the IBS-symptom severity scale, from which a pain sub score was calculated. Findings: Two distinct metabotypes were identified and mapped onto the microbial subtypes. IBS-P was characterised by a fermentative metabolic profile rich in short chain fatty acids (SCFAs). After FODMAP restriction significant reductions in SCFAs were observed in IBS-P. SCFA levels did not change significantly in the IBS-H group. The magnitude of pain and overall symptom improvement were significantly greater in IBS-P compared to IBS-H (p = 0.016 and p = 0.026, respectively). Using just five metabolites, a biomarker model could predict microbial subtype with accuracy (AUROC 0.797, sensitivity 78.6% (95% CI: 0.78–0.94), specificity 71.4% (95% CI: 0.55–0.88). Interpretation: A metabotype high in SCFAs can be manipulated by restricting fermentable carbohydrate, and is associated with an enhanced clinical response to this dietary restriction. This implies that SCFAs harbour pro-nociceptive potential when produced in a specific IBS niche. By ascertaining metabotype, microbial subtype can be predicted with accuracy. This could allow targeted FODMAP restriction in those seemingly primed to respond best. Drought is a natural phenomenon that poses a significant threat to water resources in affected regions. The detrimental effects of this extreme weather event, regardless of its type, have had an impact, necessitating concrete resource allocation projects to mitigate drought. From this perspective, our research proposes a group multicriteria sorting model to support resource allocation for drought mitigation. A multicriteria sorting model was developed based on the PROMSORT method to assist the government in evaluating 14 municipalities of the Apodi-Mossoró river basin in the Rio Grande do Norte State in Brazil. This model classifies the municipalities into high, moderate, and low-priority categories, enabling targeted attention and allocation of resources for drought mitigation efforts. The research findings demonstrate that the proposed model can effectively support strategic public policies, allocate resources, and facilitate the implementation of appropriate actions, thereby focusing efforts on the cities most severely affected by drought and alleviating the adverse effects of this natural disaster HIGHLIGHTS We identified and sorted the cities most impacted by drought through group decision-making.; Structured approach for allocating public resources.; We facilitate introduction of strategic public policies.; We consider the particularities of municipalities in the Brazilian semi-arid region.; We propose decision rules to generate a group decision.;

Abstract Signal transducer and activator of transcription 3 (STAT3) is frequently overexpressed in patients with acute myeloid leukemia (AML). STAT3 exists in two distinct alternatively spliced isoforms, the full-length isoform STAT3α and the C-terminally truncated isoform STAT3β. While STAT3α is predominantly described as an oncogenic driver, STAT3β has been suggested to act as a tumor suppressor. To elucidate the role of STAT3β in AML, we established a mouse model of STAT3β-deficient, MLL-AF9-driven AML. STAT3β deficiency significantly shortened survival of leukemic mice confirming its role as a tumor suppressor. Furthermore, RNA sequencing revealed enhanced STAT1 expression and interferon (IFN) signaling upon loss of STAT3β. Accordingly, STAT3β-deficient leukemia cells displayed enhanced sensitivity to blockade of IFN signaling through both an IFNAR1 blocking antibody and the JAK1/2 inhibitor Ruxolitinib. Analysis of human AML patient samples confirmed that elevated expression of IFN-inducible genes correlated with poor overall survival and low STAT3β expression. Together, our data corroborate the tumor suppressive role of STAT3β in a mouse model in vivo. Moreover, they provide evidence that its tumor suppressive function is linked to repression of the STAT1-mediated IFN response. These findings suggest that the STAT3β/α mRNA ratio is a significant prognostic marker in AML and holds crucial information for targeted treatment approaches. Patients displaying a low STAT3β/α mRNA ratio and unfavorable prognosis could benefit from therapeutic interventions directed at STAT1/IFN signaling.

Protected areas (PAs) such as national parks face funding issues that undermine effective management. Therefore, many PAs are exploring new financial instruments, such as visitor donations, to supplement their conservation budgets. This paper investigates visitor perceptions of one such system, a new conservation donation under consideration in Aso Kuju National Park, southwest Japan, is due to be introduced. Our on-site survey at two trailheads in autumn 2022 gauged visitors’ willingness to pay (WTP) the expected JPY 500 donation. The analysis used Bayesian linear regression to look for significant predictors of WTP. Findings show that female, older, and higher-income visitors were more likely to pay the donation collectively rather than voluntarily. Prior knowledge of the donation system was also a significant predictor of WTP, but more frequent climbers were significantly less likely to pay the donation collectively, regardless of prior Kuju climbing experience, possibly due to the perceived increase in use costs. Moreover, visitors willing to pay the cooperation donation collectively are also willing to pay higher prices than those willing to pay voluntarily. The elicited WTP values confirm that the implementation of a new conservation donation could help to improve the long-term sustainable financing of PAs such as Aso Kuju while raising issues over price fairness.

Abstract Background This cross-sectional controlled study aims to assess health-related quality of life (HRQoL) of children and adolescents with a molecular diagnosis of Marfan syndrome (MFS) or related disorders and to evaluate the factors associated with HRQoL in this population. Sixty-three children with MFS and 124 age- and sex-matched healthy children were recruited. HRQoL was assessed using the Pediatric Quality of Life Inventory (PedsQL™) generic questionnaire. The correlation between HRQoL scores and the different continuous parameters (age, body mass index, disease severity, systemic score, aortic sinus diameter, and aerobic physical capacity) was evaluated using Pearson’s or Spearman’s coefficient. A multiple linear regression analysis was performed on the two health summary self-reported PedsQL™ scores (physical and psychosocial) to identify the factors associated with HRQoL in the MFS group. Results Except for emotional functioning, all other domains of HRQoL (psychosocial and physical health, social and school functions) were significantly lower in children with MFS compared to matched healthy children. In the MFS group, the physical health summary score was significantly lower in female than in male patients (self-report: absolute difference [95%CI] = -8.7 [-17.0; -0.47], P = 0.04; proxy-report: absolute difference [95%CI] = -8.6 [-17.3; 0.02], P = 0.05) and also negatively correlated with the systemic score (self-report: R = -0.24, P = 0.06; proxy-report: R = -0.29, P = 0.03) and with the height Z-score (proxy-report: R = -0.29, P = 0.03). There was no significant difference in the physical health summary scores between the different genetic subgroups. In the subgroup of 27 patients who performed a cardiopulmonary exercise test, self- and proxy-reported physical health summary scores were highly correlated with their aerobic physical capacity assessed by peak oxygen consumption (VO2max) and ventilatory anaerobic threshold (VAT). In the multivariate analysis, the most important independent predictors of decreased physical health were increased height, decreased body mass index, decreased VAT and use of prophylactic therapy. Conclusions This study reports an impaired HRQoL in children and adolescents with MFS or related conditions, in comparison with matched healthy children. Educational and rehabilitation programs must be developed and evaluated to improve exercise capacity and HRQoL in these patients. Trial registration ClinicalTrials.gov, NCT03236571 . Abstract Stalls or crates are a very common type of housing used on pig farms that restrict an animal’s movement. How this confinement impacts the animal’s affective states is seldom investigated. We conducted a preference test over 7 days where trios of gilts (n = 10 trios, 27.4 ± 1.5 weeks old) had free access between individual self-locking stalls (~ 1.2 m2) and a shared open area allowing 2.8 m2/animal (71% of total area). Gilts had access to ad libitum feed and water both inside the crates and in the open area. After 7 days, personality traits of the animals were assessed with open field (OF) and novel object (NO) tests. Principal Component Analysis (PCA) yielded two main components, which we defined as Passivity and Engagement. Abstract Background It was hypothesized that glucose-6-phosphate dehydrogenase (G6PD) deficiency confers a protective effect against malaria infection, however, safety concerns have been raised regarding haemolytic toxicity caused by radical cure with 8-aminoquinolines in G6PD-deficient individuals. Malaria elimination and control are also complicated by the high prevalence of G6PD deficiency in malaria-endemic areas. Hence, accurate identification of G6PD deficiency is required to identify those who are eligible for malaria treatment using 8-aminoquinolines. Methods The prevalence of G6PD deficiency among 408 Thai participants diagnosed with malaria by microscopy (71), and malaria-negative controls (337), was assessed using a phenotypic test based on water-soluble tetrazolium salts. High-resolution melting (HRM) curve analysis was developed from a previous study to enable the detection of 15 common missense, synonymous and intronic G6PD mutations in Asian populations. The identified mutations were subjected to biochemical and structural characterisation to understand the molecular mechanisms underlying enzyme deficiency. Results Based on phenotypic testing, the prevalence of G6PD deficiency (T) and intronic (c.1365-13T>C and c.486-34delT) mutations was detected with intermediate to normal enzyme activity. The double missense mutations were less catalytically active than their corresponding single missense mutations, resulting in severe enzyme deficiency. While the mutations had a minor effect on binding affinity, structural instability was a key contributor to the enzyme deficiency observed in G6PD-deficient individuals. Conclusions With varying degrees of enzyme deficiency, G6PD genotyping can be used as a complement to phenotypic screening to identify those who are eligible for 8-aminoquinolines. The information gained from this study could be useful for management and treatment of malaria, as well as for the prevention of unanticipated reactions to certain medications and foods in the studied population.

Abstract Deep brain stimulation (DBS) of the supero-lateral medial forebrain bundle (slMFB) is associated with rapid and sustained antidepressant effects in treatment-resistant depression (TRD). Beyond that, improvements in social functioning have been reported. However, it is unclear whether social skills, the basis of successful social functioning, are systematically altered following slMFB DBS. Therefore, the current study investigated specific social skills (affective empathy, compassion, and theory of mind) in patients with TRD undergoing slMFB DBS in comparison to healthy subjects. 12 patients with TRD and 12 age- and gender-matched healthy subjects (5 females) performed the EmpaToM, a video-based naturalistic paradigm differentiating between affective empathy, compassion, and theory of mind. Patients were assessed before and three months after DBS onset and compared to an age- and gender-matched sample of healthy controls. All data were analyzed using non-parametric Mann-Whitney U tests. DBS treatment significantly affected patients’ affective responsiveness towards emotional versus neutral situations (i.e. affective empathy): While their affective responsiveness was reduced compared to healthy subjects at baseline, they showed normalized affective responsiveness three months after slMFB DBS onset. No effects occurred in other domains with persisting deficits in compassion and intact socio-cognitive skills. Active slMFB DBS resulted in a normalized affective responsiveness in patients with TRD. This specific effect might represent one factor supporting the resumption of social activities after recovery from chronic depression. Considering the small size of this unique sample as well as the explorative nature of this study, future studies are needed to investigate the robustness of these effects.

IntroductionSeabirds and other insular birds are an important part of marine ecosystems and are increasingly threatened worldwide. Phenology, abundance, distribution, and breeding success are important baseline parameters needed to evaluate population trends and dynamics, identify biodiversity hotspots and potential breeding sites, and assess habitat selection. In the Red Sea basin, reliable and complete data on birds are lacking for islands in most of the Important Bird Areas (IBA). Such data are now especially important since development projects have started at several of these locations. Here, we assess the distribution, abundance, phenology, and population trends of 13 bird species that breed in the Duba Islands, Al Wajh Bank, and Al Lith Islands. Breeding success and habitat selection for six species were also assessed.MethodsBetween December 2020 and August 2023, more than 90 islands were visited multiple times with different frequencies. All nests were counted, and the area occupied by the different colonies was georeferenced. Breeding success was assessed by visiting selected colonies multiple times until the chicks were ready to fledge. Habitat selection was determined by analyzing the habitat found in the georeferenced colonies.ResultsMore than 25,000 nests were counted each year, mainly in the summer, on more than 75 islands. Of the 13 species studied, the most abundant and widespread species was the White-cheeked tern, and the second most widespread was the Osprey.DiscussionWe estimate that the Al Wajh Bank hosts the following percentages of global breeding populations: Crab plover: 5%, Sooty gull: 17%–35%, White-eyed gull: 15%, Bridled tern: 1%, White-cheeked tern: 4%, and Lesser crested tern: 2%, making the area a regional and global hotspot for these species. Some of the islands occupied by breeding birds are slated for development for tourism activities as part of the Kingdom’s tourism expansion plans. At the same time, there are several ambitious conservation programs underway in these areas. To be effective, such programs require reliable and comprehensive data of the kind presented here.

On most dairy farms, calves are housed individually until weaning. However, depriving calves of an early social environment impairs behavioral development. We studied the effect of early-life social housing on calves' competitive skills. In this study, Holstein heifers were pseudorandomly assigned to either individual housing (n = 9) or pair housing (with a nonfocal companion, n = 9) at the age of 11 d. After 14 d of housing treatment, calves underwent a competition test for milk access against a group-reared calf; consisting of 2 test sessions per day for 5 d (session duration: 74.42 ± 2.29 s; mean ± standard error). Pair-housed calves performed better than individually housed calves: throughout the competition days, individually housed calves increased their latency to approach the milk bottle and decreased their time spent drinking in contrast to pair-housed calves, which exhibited stable latencies to reach the milk bottle and increased their time drinking. To control for the influence of personality on their competitive abilities, all calves were subjected to personality tests assessing boldness before being exposed to the housing treatment. Our findings indicate that calves assessed as bolder during the pretreatment personality test tended to approach the milk bottle faster. Our results provide additional evidence of the beneficial effects of social housing on dairy calves' behavioral development.

Abstract The myeloid transcription factor CEBPA is recurrently biallelically mutated (i.e., double mutated; CEBPA DM) in acute myeloid leukemia (AML) with a combination of hypermorphic N-terminal mutations (CEBPA NT), promoting expression of the leukemia-associated p30 isoform, and amorphic C-terminal mutations. The most frequently co-mutated genes in CEBPA DM AML are GATA2 and TET2, however the molecular mechanisms underlying this co-mutational spectrum are incomplete. By combining transcriptomic and epigenomic analyses of CEBPA-TET2 co-mutated patients with models thereof, we identify GATA2 as a conserved target of the CEBPA-TET2 mutational axis, providing a rationale for the mutational spectra in CEBPA DM AML. Elevated CEBPA levels, driven by CEBPA NT, mediate recruitment of TET2 to the Gata2 distal hematopoietic enhancer thereby increasing Gata2 expression. Concurrent loss of TET2 in CEBPA DM AML induces a competitive advantage by increasing Gata2 promoter methylation, thereby rebalancing GATA2 levels. Of clinical relevance, demethylating treatment of Cebpa-Tet2 co-mutated AML restores Gata2 levels and prolongs disease latency.

Abstract Negative emotional states are known to interact, potentially aggravating one another. In this study, we used a well validated paradigm (successive negative contrast, SNC) to determine if pain from a common procedure (disbudding) influences responses to a reward downshift. Holstein calves (n = 30) were trained to approach a 0.5 L milk reward. Latency to approach, number of vocalisations and pressure applied on the bottle were recorded during training. To assess how pain affected responses to reward downshift, calves were randomly assigned to one of three treatments before the downshift. Two groups were disbudded and provided the ‘gold standard’ of pain mitigation: intraoperative local anesthesia and analgesia. One of these disbudded groups was then provided supplemental analgesic before testing. The third group was sham disbudded. All calves were then subjected to the reward downshift by reducing the milk reward to just 0.1 L. Interactions were detected between test session and daily trials on pressure applied for the Disbudded group (estimate ± SEM: 0.08 ± 0.05), and on vocalisations for the Sham (0.3 ± 0.1) and Disbudding + Analgesia (0.4 ± 0.1) groups. Our results indicate that SNC is a promising paradigm for measuring negative affect in calves and suggests that pain potentially affects the response to a reward downshift.

Background/Objectives: Osteogenesis imperfecta (OI) is a rare genetic disease that is responsible for bone fragility, but also for dental malocclusions and dentinogenesis imperfecta (DI). The aim of this study was to assess whether the severity of dental malocclusion influenced the oral health-related quality of life (OHRQoL) and exposure to bullying in a paediatric OI population compared with a control group. Methods: Dental and occlusal characteristics were noted during oral and radiographic examination. The severity of malocclusion was assessed using the PAR index. P-CPQ, COHIP(34), and BCS-A questionnaires were used to evaluate, respectively, externally and self-perceived OHRQoL and bullying. Results: We included 39 patients with a mean age of 11.3 (±4.8 SD) in the OI group, and 45 patients with a mean age of 12.3 (±3.2 SD) in the control group. There were no significant differences between the two groups in terms of occlusal vertical and transverse dimensions. Patients with severe OI, presenting with bone fractures, bones deformities, and short stature, had significantly more anterior (p < 0.05) and posterior openbites (p < 0.05) and more DI (p < 0.05) compared to patients who had moderate or mild OI. Self-perceived OHRQoL was negatively impacted by the disease (p = 0.01), particularly in the domains of oral health (p < 0.05) and self-image (p < 0.001), but not by its severity. Exposure to bullying did not differ significantly between the two groups, although more patients with OI reported being teased (21.4% face to face and 7.1% online vs. 14.6% and 2.4% in the control group). Conclusion: Interventions for dental malocclusion and oral health in OI patients would help to improve their quality of life and self-image.

Abstract The novel coronavirus pandemic (COVID-19) has necessitated a global increase in the use of face masks to limit the airborne spread of the virus. The global demand for personal protective equipment has at times led to shortages of face masks for the public, therefore makeshift masks have become commonplace. The severe acute respiratory syndrome caused by coronavirus-2 (SARS-CoV-2) has a spherical particle size of ~97 nm. However, the airborne transmission of this virus requires the expulsion of droplets, typically ~0.6–500 µm in diameter (by coughing, sneezing, breathing, and talking). In this paper, we propose a face covering that has been designed to effectively capture SARS-CoV-2 whilst providing uncompromised comfort and breathability for the wearer. Herein, we describe a material approach that uses amorphous silica microspheres attached to cotton fibres to capture bioaerosols, including SARS CoV-2. Abstract Hydrogels are attracting interest in the field of bioelectronics due to their ability to serve as coatings on electrodes, improving the electrochemical interface, addressing the mechanical mismatch, and offering potential for localized drug or cell delivery. Challenges persist in integrating hydrogels with electrodes typically composed of metals and/or organic semiconductors. Here, an electrochemically driven method is introduced for direct growth of chitosan hydrogels onto poly(3,4‐ethylenedioxythiophene) (PEDOT) surfaces. The growth of ionic gelation chitosan is triggered by electrical release of a specific dopant, tripolyphosphate (TPP), from PEDOT. As a result, chitosan hydrogels grow directly from the PEDOT surface and firmly attach to it. Although this process temporarily reduces PEDOT to the benzoid structure, its unique electroactivity allows for reversible conversion to the quinoid structure after chitosan hydrogel assembly. Once assembled, the chitosan hydrogel coating can be further functionalized. The introduction of covalent cross‐links and incorporation of additional interpenetrating polymer networks (IPNs) are explored. Electrochemical characterization reveals that an interface with favorable properties is formed between PEDOT and ionic‐covalent chitosan, functionalized with a PEDOT IPN. The electroactivity of the proposed method surpasses any other PEDOT/chitosan system reported in the literature. These results underscore the potential of this material for bioelectronics applications.

Wastewater and sewage sludge treatment plants are known to be important entrance routes for microplastics (MPs) into the aquatic environment. Although traditional wastewater treatment plants with primary and secondary purification processes can remove over 99 % of MPs, the majority of these are believed to be deposited into the sewage sludge. This provides a route into the environment, especially if the treated sludge is used as a fertilizer on agricultural land. Although methods do exist for the removal of MPs from water, there are limited studies with sludge. The possibility of using electron beam treatment for the removal of microplastics from wastewater and sewage sludge has been investigated in the laboratory using both tap water and actual sewage sludge. Abstract In humans, the ‘peak-end’ rule states that recollection of an experience is most often influenced by the peak (the most intense moment) and end of the experience. We investigated whether calves followed the peak-end rule in their memory of a painful procedure: disbudding. As proxies for retrospective and ‘real-time’ reports of pain, we used conditioned place aversion, and reflex pain behaviours. In two separate trials, calves were subjected to two disbudding conditioning sessions (one horn per treatment), acting as their own control. In the first trial, calves (n = 22) were disbudded and remained in a pen for 4 h, and disbudded and left in another pen for 4 h with an additional 2 h following an analgesic treatment. In the second trial, calves (n = 22) were disbudded and left in pens for 6 h during both treatments, receiving the analgesic at either 2 h or 4 h after disbudding. Calves were then tested for place aversion. For both trials we did not observe a preference for the pens where calves received analgesic treatment towards the end of the session. We did not find an association between aversion and the sum, peak or end of pain behaviours. Results are not consistent with a peak-end effect in calves’ memory of pain.

Abstract The emergence of severe acute respiratory syndrome 2 (SARS-CoV-2) variants of concern (VOCs), with mutations linked to increased transmissibility, vaccine escape and virulence, has necessitated the widespread genomic surveillance of SARS-CoV-2. This has placed a strain on global sequencing capacity, especially in areas lacking the resources for large scale sequencing activities. Here we have developed three separate multiplex high-resolution melting assays to enable the identification of Alpha, Beta, Delta and Omicron VOCs. The assays were evaluated against whole genome sequencing on upper-respiratory swab samples collected during the Alpha, Delta and Omicron [BA.1] waves of the UK pandemic. The sensitivities of the eight individual primer sets were all 100%, and specificity ranged from 94.6 to 100%. The multiplex HRM assays have potential as a tool for high throughput surveillance of SARS-CoV-2 VOCs, particularly in areas with limited genomics facilities.
