1. High Power Electromagnetic Waves Exposure of Healthy and Tumor Bearing Mice: Assessment of Effects on Mice Growth, Behavior, Tumor Growth, and Vessel Permeabilization

Abstract

High power radiofrequencies may transiently or permanently disrupt the functioning of electronic devices, but their effect on living systems remains unknown. With the aim to evaluate the safety and biological effects of narrow-band and wide-band high-power electromagnetic (HPEM) waves, we studied their effects upon exposure of healthy and tumor-bearing mice. In field experiments, the exposure to 1.5 GHz narrow-band electromagnetic fields with the incident amplitude peak value level in the range of 40 kV/m and 150 MHz wide-band electric fields with the amplitude peak value in the range of 200 kV/m, did not alter healthy and tumor-bearing animals’ growth, nor it had any impact on cutaneous murine tumors’ growth. While we did not observe any noticeable behavioral changes in mice during the exposure to narrow-band signals when wide-band HPEM signals were applied, mice could behave in a similar way as they respond to loud noise signals: namely, if a mouse was exploring the cage prior to signal application, it returned to companion mates when wide-band HPEM signals were applied. Moreover, the effect of wide-band signals was assessed on normal blood vessels permeability in real-time in dorsal-chamber-bearing mice exposed in a pilot study using wide-band signal applicators. Our pilot study conducted within the applicator and performed at the laboratory scale suggests that the exposure to wide-band signals with the amplitude of 47.5 kV/m does not result in increased vessel permeability.

Abstract

This paper presents a reliable microwave and microfluidic miniature sensor dedicated to the measurement of glucose concentration in aqueous solution. The device; which is integrated with microtechnologies; is made of a bandstop filter implemented in a thin film microstrip technology combined with a fluidic microchannel. Glucose aqueous solutions have been characterized for concentration ranging from 80 g/L down to 0.3 g/L and are identified with the normalized insertion loss at optimal frequency. The sensitivity of the sensor has consequently been estimated at 7.6 × 10−3 dB/(g/L); together with the experimental uncertainty; the resolution of the sensor comes to 0.4 g/L. These results demonstrate the potentialities of such a sensor for the quantitative analysis of glucose in aqueous solution.

Abstract

International audience; High power electromagnetic signals can disrupt the functioning of electronic devices. As electromagnetism plays a role in cells homeostasis, such electromagnetic signals could potentially also alter some physiological processes. Herein we report on distinct biological parameters assessment after cellular spheroids exposure to high power electromagnetic signals, such as the ones used for defense applications. Signals effects were assessed in tumor cells spheroids and in normal human dermal fibroblasts spheroids, where macroscopic aspect, growth, plasma membrane integrity, induction of apoptosis, ATP content, and mitochondrial potential were investigated after spheroids exposure to high power electromagnetic signals. No significant effects were observed, indicating that 1.5 GHz narrowband electromagnetic fields with incident amplitude level of 40 kV/m, and 150 MHz moderate-band electric fields with an amplitude of 72.5 to approximately 200 kV/m, do not cause any significant alterations of assessed parameters. Electronic devices can stop operating in response to intense external electromagnetic signals. Electromagnetic environments, or more precisely intentional electromagnetic environments 1 , are among emerging approaches that could be used for defense applications. High power electromagnetic (HPEM) technology can be deployed to temporarily or permanently disrupt electronic devices or could even be used against the personnel 2. The HPEM systems that can be used to impair or destroy electronics and hardware 3 , and thus to black out the opponents electronic systems, operate in the range of tens of MHz to a few tens of GHz. Alternatively, HPEM signals (with frequencies of approximately 100 GHz and millimeter waves) could be used against personnel. This non-lethal directed-energy weapon, can heat the surface of its targets (e.g. heat people's skin) 2. Such non-lethal weapon system, also known as the Active Denial System, was developed by the U.S. military (https://jnlwp.defense. gov/Press-Room/Fact-Sheets/ArticleViewFactsheets/Article/577989/active-denial-technology-fact-sheet/ retrieved August 2018) and was designed for area denial, perimeter security and crowd control. Similar systems were also elaborated by Russia (https://www.popularmechanics.com/military/weapons/a7804/ why-russia-will-be-the-first-to-use-the-pain-ray-9833954/ retrieved August 2018) and China (http://en.people. cn/n3/2018/0205/c90000-9423875.html retrieved August 2018). High Power Microwave (HPM) and High Power Radio Frequency (HPRF) systems can radiate high power and short duration oscillatory pulses of electromagnetic fields. These pulses can exhibit different amplitudes, durations, frequency spectrums, as well as Pulse Repetition Frequencies (PRF), and can be classified into different families, according to their frequency spectrum bandwidth. The bandwidth classification distinguishes the narrowband (also known as hypo band), meso (or moderate) band, ultra-moderate (or sub-hyper) band and hyperband intentional electromagnetic environments 2 .

Abstract

International audience; This paper presents several microwave based resonant biosensors realized with a microfluidic and thin film microstrip technology. Due to the use of multiple stubs terminated by an interdigitated capacitor, this paper demonstrates that this circuit topology is suitable for multiple and simultaneous molecule concentration measurements. The accuracy study of the measurement is conducted depending on the concentration of glucose and the frequency stub loaded with an aqueous-based glucose solution.

6. High power electromagnetic pulse applicators for evaluation of biological effects induced by electromagnetic radiation waves

Abstract

The effects of electromagnetic radiation waves on health is one of the major public concerns. These waves are mainly produced at a large scale but it is important to evaluate these effects on biological samples at the laboratory scale. Here we developed a set of micro applicators, which allow evaluating the effect of electromagnetic fields on biological samples with volumes in the microliter range. The applicators can be coupled to an optical microscope and allow a real-time observation of potential structural and functional alterations of the tested sample induced by different waveforms. New design approaches are suggested to simultaneously achieve maximized electric field coupling effect and optimized electric field homogeneity in the tested sample, while minimizing the return loss when the applicators are loaded with the biological samples. These applicators allow studying the biological effect of a variety of different signals, due to their wide frequency bandwidth (beyond 1.5 GHz) and their high permissible power. In addition, different electromagnetic parameters such as the electromagnetic field magnitude, pulse repetitive factor, number of bursts or delay between bursts may be set. The efficacy of the applicators was addressed for three different signals: two types of electromagnetic waves - a damped sinusoid centered at 200 MHz (wide band signal), a radar-like signal at 1.5 GHz (the ultra-narrow band signal) and a train of millisecond square-wave monopolar electric field pulses (causing electroporation). The biological effects were thus assessed (at the microscopic scale) on two different biological models, the giant unilamellar vesicles, and tumor and normal human cells, as well as being compared to results obtained (at full scale) with signals generated by antennas.

International audience; Probing biological materials with electromagnetic waves, which has been investigated for decades [1]-[6], involves characterizing the sample with its dielectric properties that are frequency dependent. Dielectric spectroscopy is performed by placing a high-frequency circuit close to the biological species under study. Passing through the biological medium, the electric properties of the biological elements modify the electromagnetic field. 9. New efficient high power microwave applicator enabling optimal E-field coupling and homogeneity in biological sample

Abstract

A new set of applicators dedicated to the exposition of giant vesicles, cells and spheroids to high power microwave pulses have been developed in order to maximize and optimize the E-field coupling in the Device Under Test (DUT) with the intention to harness the electromagnetic experimental conditions. These applicators are simultaneously characterized by low return loss(when charged by the sample under test)thus easing illumination experimentations. We particularly focus here on the implementation of two novel ideas which enable to enhance the electrical performances of the applicators. The first is based on a volume extension to obtain a better E-field homogeneity; the second lies on the use of a half wavelength filter in the case of narrow band signals. The development of these applicators takes place in a largest project which aims at identifying beneficial and/or hazardous non thermal effects of high power microwave pulses on living organisms.

Abstract

International audience; This paper presents a technique able to detect and quantify two different solutes in aqueous solution thanks to a single shot microwave measurements of a resonant liquid sensor at two different frequencies. The principle of operation is based on isosbestic characteristics of the sensor relatively to the two considered solutes. The solutes quantification requires a calibration phase. Extracted concentration from measurements leads to root mean square error (RMSE) of only few g/L of solute (3.2 g/L for glucose and 1.7 g/L for sodium chloride). The technique then reveals the potential richness of the spectral signatures from microwave sensor for indentifying and quantifying complex liquids.

Abstract

International audience; This paper presents a miniature double-stub resonant-biosensor and associated technique to quantify the glucose concentrations of two aqueous solutions localized over the sensing area of each stub. A specific procedure and the behavior of the sensor have been developed, calibrated and validated experimentally. Seven blind filling configurations have been characterized for glucose concentrations ranging from 15 to 80 mg/L and our technique predicts the glucose concentration in each channel with a root-mean-square error of 3.2 g/L. Such a level of accuracy is the tolerance range for commercial glucometer, which outlines that the microwave technique can be used for accurate glucose analysis of multiple aqueous solutions.

Abstract

This paper deals with in vitro experiments involving biological samples exposed to free space high power pulsed electromagnetic plane waves. Biological samples under test are Giant Unilamellar Vesicles (GUV) in solutions or human mammalian tumor cells in suspensions. Different sample containers (such as Petri dishes, Eppendorf tubes or microscope glass slides) have been evaluated in terms of reachable electric coupling efficiency and reachable electric field homogeneity in the biological samples under study. Recommendations for the use of each of them are suggested. Due to the high permittivity of the cell culture medium, we show that the Device Under Test (DUT) may behave as a dielectric resonator. It is the case for the Petri Dish configuration with H field coupling in the axis direction for which the TE 01δ dielectric mode can be excited. At last we present a new idea for Field Homogenization by Volume Extension (FHVE).

Abstract

International audience; Cancer remains a leading cause of death in the world. To overcome this problem, it is necessary to develop new analysis tools, in complementarity to existing ones, to enable the early diagnostic of the diseases, personalized treatment, and further fundamental cancer mechanisms understanding. In this context, micro and millimeterwave-based dielectric spectroscopy performed at the cellular and molecular levels is progressively emerging, as it permits the non-invasive and real time probing of cells in their culture biological medium. The recent advances of this topic are given in this paper with a specific highlight of its various assets.

Abstract

International audience; A microwave resonator is presented as a microfabricated sensor dedicated to liquid characterization with perspectives for chemistry and biology. The nanolitter range aqueous solution under investigation is located on top of the planar resonator thanks to a microfluidic channel compatible with a future lab-on-a-chip integration. The interaction between the electric field and the liquid translates into a predictable relationship between electrical characteristics of the resonator (resonant frequency and associated insertion loss) and the complex permittivity of the fluid (real and imaginary parts). A prototype of the resonator has been fabricated and evaluated with de-ionized water/ethanol mixtures, with ethanol volume fraction ranging from 0 to 20 %. Good agreement has been reached between theoretical and measured electrical parameters of the resonator. The discrepancy on the resonant frequency is estimated to 0.5 %, whereas the one on the associated transmission coefficient is lower than 1 %. This translates into a maximum relative error on the real and imaginary part of the predicted relative permittivity of less than 6.5 % and 4 % respectively, validating the principle of this accurate permittivity characterization methodology.
