International audience; The structural organization of heterogeneous and multiphase natural aggregates depends on the biophysicochemical conditions prevailing in the environment, with major ions playing a crucial role. In this study, the impact of aluminum (Al) on iron-organic matter (Fe-OM) aggregates was investigated since Al can interact with OM and can be incorporated in Fe-oxyhydroxides or adsorbed on their surface. Mimetic environmental Fe-OM-Al aggregates were synthesized at various [Fe] and [Al] with a constant [OM]. At low [Al+Fe], Fe-OM-Al aggregates exhibit a colloidal behavior. Within the aggregates, Fe is present as Fe(III)-oligomers and ferrihydrite-like nanoparticles whereas Al forms monomers, oligomers and small polymers, all bound to OM. The Al and Fe phases interacted with each other. At high [Fe+Al], the Fe(III)-oligomers and Al monomers/oligomers polymerized which increases the size and quantity of the ferrihydrite-like nanoparticles and Al polymers and then branched out the OM, resulting in a large settling network. The effect of Al on the Fe-OM aggregates structure could also have an impact on the fate of pollutants. The occurrence of Al amorphous hydroxides and the increase in ferrihydrite-like nanoparticles lead to a higher availability of surface reactive sites and subsequently to an increase in the sorption capacity of the Fe-OM aggregates for pollutants that exhibit a greater affinity for minerals than for organics.

Low-molecular weight gelators (LMWGs) are small molecules (Mw < ∼1 kDa), which form self-assembled fibrillar network (SAFiN) hydrogels in water.

Controlling the viscoelastic properties of hydrogels is a challenge for many applications. Low molecular weight gelators (LMWGs) like bile salts and glycolipids and biopolymers like chitosan and alginate are good candidates for developing fully biobased hybrid hydrogels that combine the advantages of both components. Biopolymers lead to enhanced mechanics, while LMWGs add functionality. In this work, hybrid hydrogels are composed of biopolymers (gelatin, chitosan, and alginate) and microbial glycolipid bioamphiphiles, known as biosurfactants. Besides their biocompatibility and natural origin, bioamphiphiles can present chameleonic behavior, as pH and ions control their phase diagram in water around neutrality under strongly diluted conditions (5 wt%). The glycolipid used in this work behaves like a surfactant (micellar phase) at high pH or like a phospholipid (vesicle phase) at low pH. Moreover, at neutral-to-alkaline pH in the presence of calcium, it behaves like a gelator (fiber phase). The impact of each of these phases on the elastic properties of biopolymers is explored by means of oscillatory rheology, while the hybrid structure is studied by small angle X-ray scattering. The micellar and vesicular phases reduce the elastic properties of the hydrogels, while the fiber phase has the opposite effect; it enhances the hydrogel's strength by forming an interpenetrated biopolymer-LMWG network.

Hydrogels are widespread soft materials, which can be used in a wide range of applications. The control over the viscoelastic properties of the gel is of paramount importance. Ongoing environmental issues have raised the consumer's concern toward the use of more sustainable materials, including hydrogels. However, are greener materials compatible with high functionality? In a safe-by-design approach, this work demonstrates that functional hydrogels with in situ responsivity of their elastic properties by external stimuli can be developed from entirely "sustainable" components, a biobased amphiphile and biopolymers (gelatin, chitosan, and alginate). The bioamphiphile is a stimuli-responsive glycolipid obtained by microbial fermentation, which can self-assemble into fibers, but also micelles or vesicles, in water under high dilution and by a rapid variation of the stimuli. The elastic properties of the bioamphiphile-/biopolymer-interpenetrated hydrogels can be modulated by selectively triggering the phase transition of the glycolipid and/or the biopolymer inside the gel by mean of temperature or pH.

International audience; Aggregates of charged metal particles obtained by electrostatic coupling with a compound of opposite charge in the vicinity of the net zero charge ratio are of interest in the field of plasmonics because the inter-particle distance is minimal, which favours plasmonic coupling. However, these structures present a low colloidal stability limiting the development of applications. In this article we show that globally neutral aggregates formed by electrostatic complexation of citrate-stabilized gold particles and a quaternized chitosan (i.e., polycation) around the net zero charge ratio could be stabilized at a nanometric size by the subsequent addition of polyelectrolyte chains. Furthermore, the sign of the charge carried by the stabilizing chains determines the sign of the global charge carried by the stabilized complexes. The stabilization is demonstrated in saline environment on a broad pH range as well as in a cell culture media over periods of several days. Contrarily to stabilization by charged particles, our stabilized complexes are found to retain their initial characteristics (i.e. shape, size, internal structure and optical properties) after stabilization. Hence, the plasmonic coupling allows to maximize the optical absorption around the 800 nm wavelength at which the lasers used for thermoplasmonic and surface enhanced Raman scattering analysis operate.

International audience; From the 15 th century onwards, painters began to treat their oils with lead compounds before grinding them with pigments. Such a treatment induces the partial hydrolysis of the oil triglycerides and the formation of lead soaps, which significantly modify the rheological properties of the oil paint. Organization at the supramolecular scale is thus expected to explain these macroscopic changes. Synchrotron Rheo-SAXS (Small Angle X-Ray Scattering) measurements were carried out on leadtreated oils, with different lead contents. We can now propose a full picture of the relationship between structure and rheological properties of historical saponified oils. At rest, lead soaps in oil are organized as lamellar phases with a characteristic period of 50 Å. Under shear, the loss of viscoelastic properties can be linked to the modification of this organization. Continuous shear resulted in a preferential and reversible orientation of the lamellar domains which increased with the concentration of lead soaps. The parallel orientation predominates over the entire shear range (0-1000 s-1). Conversely, oscillatory shear coiled the lamellae into cylinders that oriented themselves vertically in the rheometer cell. This is the first report of such a vertical cylindrical structure obtained under shear from lamellae.

International audience; Bottom-up strategies for the production of well-defined nanostructures often rely on the self-assembly of anisotropic colloidal particles (nanowires and nanosheets). These building blocks can be obtained by delamination in a solvent of low-dimensionality crystallites. To optimize particle availability, determination of the delamination mechanism and the different organization stages of anisotropic particles in dispersion is essential. We address this fundamental issue by exploiting a recently developed system of fluorohectorite smectite clay mineral that delaminates in water, leading to colloidal dispersions of single-layer, very large (≈20 μm) clay sheets at high dilution. We show that when the clay crystallites are dispersed in water, they swell to form periodic one-dimensional stacks of fluorohectorite sheets with very low volume fraction (

The assembly and stabilization of a finite number of nanocrystals in contact in water could maximize the optical absorption per unit of material. Some local plasmonic properties exploited in applications, such as photothermia and optical signal amplification, would also be maximized which is important in the perspective of mass producing nanostructures at a lower cost. The main lock is that bringing charged particles in close contact requires the charges to be screened/suppressed, which leads to the rapid formation of micrometric aggregates. In this article, we show that aggregates containing less than 60 particles in contact can be obtained with a milli-flow system composed of turbulent mixers and flow reactors. This process allows to stop a fast non-equilibrium colloidal aggregation process at millisecond times after the initiation of the aggregation process which allows to control the aggregation number. As a case study, we considered the rapid mixing of citrate coated gold nanoparticles (NP) and AlCl3 in water to initiate a fast aggregation controlled by diffusion. Injecting a solution of polycation using a second mixer allowed us to arrest the aggregation process after a reaction time by formation of overcharged cationic aggregates. We obtained within seconds stable dispersions of a few milliliters composed of particle aggregates. Our main result is to show that it is possible to master the average aggregation number between 2 and 60 NP per aggregate by varying the a reaction time between 10 ms and 1 s.

Pentagonal packing is a long-standing issue and a rich mathematical topic, brought to the fore by recent progress in nanoparticle design. Gold pentagonal bipyramids combine fivefold symmetry and anisotropy and their section varies along the length. In this work, colloidal supercrystals of pentagonal gold bipyramids are obtained in a compact arrangement that generalizes the optimal packing of regular pentagons in the plane. Multimodal investigations reveal a two-particle unit cell with triclinic symmetry, a lower symmetry than that of the building blocks. Monte Carlo computer simulations show that this lattice achieves the densest possible packing. Going beyond pentagons, further simulations show an odd-even effect of the number of sides on the packing: odd-sided bipyramids are non-centrosymmetric and require the double-lattice arrangement to recover inversion symmetry. The supercrystals display a facet-dependent optical response that is promising for sensing, metamaterials applications, and for fundamental studies of self-assembly processes.

A microfluidic laboratory recently opened at Synchrotron SOLEIL, dedicated to in-house research and external users. Its purpose is to provide the equipment and expertise that allow the development of microfluidic systems adapted to the beamlines of SOLEIL as well as other light sources. Such systems can be used to continuously deliver a liquid sample under a photon beam, keep a solid sample in a liquid environment or provide a means to track a chemical reaction in a time-resolved manner. The laboratory provides all the amenities required for the design and preparation of soft-lithography microfluidic chips compatible with synchrotron-based experiments. Three examples of microfluidic systems that were used on SOLEIL beamlines are presented, which allow the use of X-ray techniques to study physical, chemical or biological phenomena.

International audience; Iron-organic matter (Fe-OM) aggregates are a key factor in the control of pollutant mobility. Their physical and structural organization depends on the prevailing physicochemical conditions during their formation and on subsequent exposure to variations in porewater goechemistry. Among these conditions, calcium (Ca) could be a major parameter given its high concentrations in the environment and its affinity for OM. Mimetic environmental Fe-OM-Ca associations were synthesized at various Fe/organic carbon (OC) and Ca/Fe molar ratios using Leonardite humic acid as OM model. The impact of Ca on Fe-OM aggregates was studied by a combination of X-ray absorption spectroscopy, small angle X-ray and neutron scattering and imaging techniques (TEM, cryo-TEM and cryo-TXM). Iron phases are constituted of Fe(III)-oligomers, Fe(III)-nanoparticles and ferrihydrite (Fh), all bound or embedded by OM. Iron phases exhibit a fractal organization with Fe-primary beads aggregated as Fe-primary aggregates (Fe-PA) which themselves are embedded in an OM aggregates. For Ca/OC (mol/mol) < 0.026, Fe-PA aggregate in a third level as a Fe-secondary aggregate. For Ca/OC ≥ 0.026, OM forms a large Ca-branched network in which Ca is bound as a dimer to OM carboxylic sites. In such conditions, Fe-PA are distributed in the OM network, distant from each other. All these structural transitions are driven by Ca which partially screens the Fe-OM interactions. The formation of such micrometric network should impact both the surface reactivity of the Fe phases as well as the mobility of Fe, OM and associated elements, notably in the soil porosity where they are produced under natural conditions.

Multi-responsive nanomaterials based on the self-limited assembly of plasmonic nanoparticles are of great interest due to their widespread employment in sensing applications. We present a thorough investigation of a hybrid nanomaterial based on the protein-mediated aggregation of gold nanoparticles at varying protein concentration, pH and temperature. By combining Small Angle X-ray Scattering with extinction spectroscopy, we are able to frame the morphological features of the formed fractal aggregates in a theoretical model based on patchy interactions. Based on this, we established the main factors that determine the assembly process and their strong correlation with the optical properties of the assemblies. Moreover, the calibration curves that we obtained for each parameter investigated based on the extinction spectra point out to the notable flexibility of this nanomaterial, enabling the selection of different working ranges with high sensitivity. Our study opens for the rational tuning of the morphology and the optical properties of plasmonic assemblies to design colorimetric sensors with improved performances.

International audience; Mixtures of polymer-colloid hybrids such as star polymers and microgels with nonadsorbing polymeric additives have received a lot of attention. In these materials, the interplay between entropic forces and softness is responsible for a wealth of phenomena. By contrast, binary mixtures where one component can adsorb onto the other one have been far less studied. Yet real formulations in applications often contain low molecular weight additives that can adsorb onto soft colloids. Here we study the microstructure and rheology of soft nanocomposites made of surfactants and microgels using linear and nonlinear rheology, SAXS experiments, and cryo-TEM techniques. The results are used to build a dynamical state diagram encompassing various liquid, glassy, jammed, metastable, and reentrant liquid states, which results from a subtle interplay between enthalpic, entropic, and kinetic effects. We rationalize the rheological properties of the nanocomposites in each domain of the state diagram, thus providing exquisite solutions for designing new rheology modifiers at will.

A compact active x-ray beamstop has been developed for the SWING beamline at Synchrotron SOLEIL with two main functions, blocking the x-ray beam directly transmitted by the sample to protect the Dectris EigerX4M 2D detector and monitoring its intensity. The beamstop is composed of a sensor inserted in a well of tungsten carbide. The sensor is based on a piece of free-standing single crystal chemical vapor deposited diamond used in the ionization chamber mode. The beamstop has been installed on the beamline detector stage within the detection vacuum chamber, just upstream of the large 2D detector. The intensity monitoring performance (rms noise over signal) is shown to be better than 0.06% and the linearity is shown to be better than 2% for over more than five decades. The beamstop has been calibrated between 5 and 16 keV to provide the photon flux measurements in absolute units (ph/s). The specific design of the beamstop increases the small-angle x-ray scattering q-range by a factor of 1.5 in the low angle side, as compared to the previous active beamstop, based on a more standard commercial Si diode. The beamstop has been available for three years for SWING user operation (5–17 keV). It is fully compatible with the different beamline operating modes and fluxes, except in the microbeam mode, where the very divergent beam becomes too large at the beamstop position, and the previous, larger, beamstop is then more appropriate.

International audience; The continuous production of drug delivery systems assisted by microfluidics has drawn a growing interest because of the high reproducibility, low batch-to-batch variations, narrow and controlled particle size distributions and scale-up ease induced by this kind of processes. Besides, microfluidics offers opportunities for high throughput screening of process parameters and the implementation of process characterization techniques as close to the product as possible. In this context, we propose to spotlight the GALECHIP concept through the development of an instrumented microfluidic pilot considered as a Galenic Lab-on-a-Chip to formulate nanomedicines, such as lipid nanocapsules (LNCs), under controlled process conditions. In this paper we suggest an optimal rational development in terms of chip costs and designs. First, by using two common additive manufacturing techniques, namely fused deposition modelling and multi-jet modelling to prototype customized 3D microfluidic devices (chips and connectors). Secondly, by manufacturing transparent Silicon (Si)/Glass chips with similar channel geometries but obtained by a new approach of deep reactive ion etching (DRIE) technology suitable with in situ small angle X-ray scattering characterizations. LNCs were successfully produced by a phase inversion composition (PIC) process with highly monodispersed sizes from 25 nm to 100 nm and formulated using chips manufactured by 3D printing and DRIE technologies. The transparent Si/Glass chip was also used for the small angle X-ray scattering (SAXS) analysis of the LNC formulation with the PIC process. The 3D printing and DRIE technologies and their respective advantages are discussed in terms of cost, easiness to deploy and process developments in a GALECHIP point of view.

Structural properties of plasmenyl-glycerophospholipids (plasmalogens) have been scarcely studied for plasmalogens with long polyunsaturated fatty acid (PUFA) chains, despite of their significance for the organization and functions of the cellular membranes. Elaboration of supramolecular assemblies involving PUFA-chain plasmalogens in nanostructured mixtures with lyotropic lipids may accelerate the development of nanomedicines for certain severe pathologies (e.g., peroxisomal disorders, cardiometabolic impairments, and neurodegenerative Alzheimer’s and Parkinson’s diseases). Here, we investigate the spontaneous self-assembly of bioinspired, custom-produced docosapentaenoyl (DPA) plasmenyl (ether) and ester phospholipids in aqueous environment (pH 7) by synchrotron small-angle X-ray scattering (SAXS) and cryogenic transmission electron microscopy (cryo-TEM). A coexistence of a liquid crystalline primitive cubic Im3m phase and an inverted hexagonal (HII) phase is observed for the DPA-ethanolamine plasmalogen (C16:1p-22:5n6 PE) derivative. A double-diamond cubic Pn3m phase is formed in mixed assemblies of the phosphoethanolamine plasmalogen (C16:1p-22:5n6 PE) and monoolein (MO), whereas a coexistence of cubic and lamellar liquid crystalline phases is established for the DPA-plasmenyl phosphocholine (C16:1p-22:5n6 PC)/MO mixture at ambient temperature. The DPA-diacyl phosphoinositol (22:5n6-22:5n6 PI) ester lipid displays a propensity for a lamellar phase formation. Double membrane vesicles and multilamellar onion topologies with inhomogeneous distribution of interfacial curvature are formed upon incorporation of the phosphoethanolamine plasmalogen (C16:1p-22:5n6 PE) into dioleoylphosphocholine (DOPC) bilayers. Nanoparticulate formulations of plasmalogen-loaded cubosomes, hexosomes, and various multiphase cubosome- and hexosome-derived architectures and mixed type nano-objects (e.g., oil droplet-embedding vesicles or core–shell particles with soft corona) are produced with PUFA-chain phospholipids and lipophilic antioxidant-containing membrane compositions that are characterized by synchrotron SAXS and cryo-TEM imaging. The obtained multiphase nanostructures reflect the changes in the membrane curvature induced by the inclusion of DPA-based PE and PC plasmalogens, as well as DPA-PI ester derivative, and open new opportunities for exploration of these bioinspired nanoassemblies.

International audience; A specific organization of optically active nanoscale objects can greatly affect the optical response of a system. Here, we report the controlled modification of the fluorescent emission by the assembly of water-soluble quantum rods (QRs). Our study combines optical, electron microcopy, and X-ray scattering characterizations to reveal a correlation between the self-assembly behavior of QRs into ordered 3D-arrays and the optical properties (luminescence) of formed assemblies, where the observed optical response is highly dependent on the QR aspect ratio. Specifically, shorter, 18 nm long QRs (QR18), exhibiting a well-defined smectic packing, demonstrate an enhancement of the emission intensity accompanied by a red shift and a lifetime reduction. In contrast, 40 nm long QRs (QR40), forming a columnar phase, does not show these optical properties.

International audience; Environmental iron-organic matter (Fe-OM) aggregates play a major role in the dynamic of pollutants. Nowadays, there is a lack of information about the control exerted by their structural organization on their reactivity towards metal(loid)s and in particular, the impact of major ions, such as calcium. The sorption capacity of mimetic environmental Fe-OM-Ca aggregates was investigated relative to the Fe/organic carbon (OC) and Ca/Fe ratios using As as a probe. It was shown that Fe speciation is the key factor controlling the reactivity of Fe-OM-Ca aggregates regarding the high affinity of Fe(III)-oligomers towards As and the high sorption capacity of ferrihydrite-like nanoparticles. Moreover, when it occurs at high concentration, Ca competes with Fe for OM binding leading to an increase in the amount of ferrihydrite-like nanoparticles and binding site availability. As a consequence, Ca not only impacts the ionic strength but it also has a dramatic impact on the structural organization of Fe-OM aggregates at several scales of organization, resulting in an increase of their sorption capacity. In the presence of high amounts of Ca, Fe-OM-Ca aggregates could immobilize pollutants in the soil porous media as they form a micrometric network exhibiting a strong sorption capacity.

International audience; Boehmite AlOOH is used as a precursor for the preparation of mesoporous gamma-alumina, which is of well-known interest for use as a catalyst support in petroleum refinery. The fabrication of the shaped supports is usually decomposed in several steps: synthesis of boehmite, preparation of a paste by kneading, extrusion of the paste and finally calcination to obtain the alumina phase. Mesoporous alumina is a complex hierarchical porous network, whose growth is influenced by each step of the process, and needs to be clearly understood to optimize the catalytic performances. This paper is focused on the peptization of boehmite, an important step in the fabrication which corresponds to the beginning of the kneading. Peptization is carried out in liquid phase, from two boehmites batches with different abilities to be peptized (peptization index) and with different acid/aluminum ratios. The as-obtained suspensions are characterized by coupling electron microscopy and small-angle X-ray scattering, which bring an accurate description of the samples at their different levels of structural organization. A multi-scale mechanism of peptization is then proposed. Finally, we will discuss the study of the measure of boehmites peptization index, and confirms the textural features which are relevant for the behavior of a boehmite in an acid medium.

International audience; Complex aqueous mixtures comprised of swelling clays and hydrosoluble polymers naturally occur in soils and play a major role in pedogenesis. They are also very often used for formulating oil-well drilling fluids, paints, and personal-care products. The suspensions of some natural clays, thanks to their large nanoparticle aspect ratio, spontaneously form nematic liquid-crystalline phases where the particles align parallel to each other, which affects their flow properties. We observed that adding small amounts of hydrosoluble polymers to these clay suspensions destabilizes the nematic phase with respect to the isotropic (disordered) phase. The polymers that we used (poly(ethylene oxide) and dextran) were too small to adopt particle-bridging conformations and small-angle X-ray scattering experiments showed that the structure of the nematic phase is not altered by polymer doping. However, the adsorption isotherm shows that the macromolecules adsorb onto the clay nanosheets, effectively coating them with a polymer layer. Our extension of Onsager’s theory for polymer-coated platelets properly captures the experimental phase diagram and shows how the nematic phase destabilization can be due to the polymer adsorbing more on the platelet faces than at the rim. Because the flow properties of the nematic phase are very different from those of the isotropic phase, the presence or absence of the former phase is an important factor to be determined and considered to explain the rheological behavior of these complex systems.

International audience; Lutein is a xanthophyll carotenoid provided exclusively by the diet, that has protective functions and beneficial effects on human health. Supplementation in lutein is necessary to reach the recommended daily dietary intake. However, the introduction of lutein into foods and beverages is a real challenge since this lipophilic nutrient has a poor aqueous solubility and a low bioavailability. In this study, we investigated the capacity of egg-sphingomyelin (ESM) vesicles called sphingosomes to solubilise lutein into the bilayers. The thermal and structural properties of ESM bilayers were examined in presence of various amounts of lutein by differential scanning calorimetry (DSC) and temperature-controlled X-ray diffraction (XRD), the structures of sphingosomes and lutein crystals were observed by microscopic techniques. ESM bilayers were in the fluid L α phase above the phase transition temperature T m = 39.6 • C and in the lamellar ripple P β ′ phase below T m where ESM sphingo-somes exhibited ondulations and were facetted. Lutein molecules were successfully incorporated into the ESM bilayers where they induced a structural disorganisation. For ESM/lutein 90/10 %mol (91.8/8.2 %wt; 89 mg lutein / g ESM), lutein partitioning occured with the formation of lutein crystals in the aqueous phase together with lutein-loaded ESM vesicles. This study highlighted the capacity of new lipid carriers such as egg-sphingosomes to solubilise lutein and opens perspectives for the formulation of effective lutein-fortified func-tionnal foods and beverages providing health benefits.

International audience; In recent years, microfluidic-based sample preparation techniques have emerged as a powerful tool for measurements at large scale X-ray facilities. Most often the microfluidic device was a form of hybrid system, i.e. an assembly of different materials, because a simple, versatile and inexpensive microfabrication method, on the one hand, and X-ray compatibility, on the other hand, cannot generally be achieved by the same material. The arrival of a new polymer family based on Off-Stoichiometric Thiol-Ene-epoxy (OSTE+) has recently redistributed the cards. In this context, we studied the relevance and the compatibility of OSTE+ for small-angle X-ray scattering (SAXS) studies. The material was characterized regarding its X-ray properties (transmission coefficient, attenuation coefficient, scattering pattern and polymer aging under X-ray light) and their comparison with those of the usual polymers used in microfluidics and/or for synchrotron radiation experiments. Iron-Organic Matter (Fe-OM) aggregates produced by redox alternation in wetlands are a key factor in the control of metallic pollutants mobility. Their ability to adsorb metal(loid)s depends on the size, morphology and structural arrangement between Fe and OM phases, which are mainly controlled by the OM occurrence. The physical, chemical and morphological organization of such aggregates is influenced by the physico-chemical conditions prevailing in the environment. Calcium (Ca) is a common major ion in natural waters which exhibits high affinity for OM. It can thus modify the size and the structural organization of Fe-OM aggregates and, subsequently, their ability to bind metal(loid)s. Among metal(loids), arsenic (As) is of major importance because of its high toxicity and its high affinity towards Fe(III)-oxyhydroxides. Moreover, Fe-OM aggregates are an important factor controlling the mobility of arsenic (As) in the environment.Mimetic natural Fe-OM aggregates were synthesized at various Fe/OM and Ca/Fe ratios. After a fine characterization of the size and structural organization, Fe-OM-Ca associations were used to perform As binding sorption experiments at 2 As/Fe ratios. The suspensions were stirred during 24h and subsequently filtrated ant ultra-filtrated.Our study demonstrates that Ca strongly influences the Fe-OM aggregates physical organisation. For low Ca/Fe ratio, Fe phases exhibit a fractal organization in which Fe phases are composed of oligomers, and primary nano-aggregates (around 6 nm) which aggregate in larger Fe secondary aggregates (>200 nm). Both are embedded in the OM matrix composed of isolated molecules and OM aggregates. For high Ca/Fe ratios, OM, Fe oligomers and primary nano-aggregates form a large continuous network where Fe phases are connected by OM large molecules. With the increasing Ca/Fe ratio, the amount of Fe oligomers decreases to the benefit of larger primary nano-aggregates (increase of their geometrical radius). Ultrafiltration experiments demonstrated that DOC, Fe, Ca and As follow the same size distribution. Surprisingly, As sorption increases with the increasing size and amount of primary nano-aggregates and the formation of the large network. SAXS analyses revealed that in such network, the distance between primary nano-aggregates increases as compared to their distance in secondary aggregates. All this results suggest that, with the increasing Ca/Fe ratio, although the primary nano-aggregates size increase, their structural distance allows to rise the availability of their binding site for As.This study demonstrates that Ca not only controls the Fe-OM structural organization but also its subsequent capacity to bind toxic elements such as As. These results are of major importance since such parameter was never so clearly evidence. They show that the actual representation of the physical organisation of Fe-OM aggregates and its reactivity have to be renewed as well as the geochemical models.

Various studies have shown that food structure has an impact on digestion kinetics. We focus here on the effects of gastric and intestinal enzymes (in-vitro digestion) on two canola seed storage proteins, napin and cruciferin. To monitor structure effect we conducted experiments on gels of these proteins at different pHs, yielding different structures and elastic modulus. What is new is to get information on the mechanisms at the lowest scales, using imaging and radiation scattering at large facilities: Synchrotron fluorescence microscopy, X-Ray scattering, at SOLEIL synchrotron, and Small-Angle Neutron Scattering, at Laboratoire Leon Brillouin reactor. We can identify the mechanisms at each step and in two distinct scale ranges, observed simultaneously, the one of the individual protein scale and the one of the structure connectivity: • during gelation individual canola proteins are not deeply modified in comparison with their state in solution ; larger scale gel heterogeneity appears due to connectivity or aggregation • in the gastric step (up to 40 min): ○ at short scale (large q) we see that the proteins disintegration is much slowed down in gels than in solutions, particularly in the gastric phase; ○ at larger scales (low q), we see that the gel structure is also self-resistant to the action of the enzyme (pepsin). • in the intestinal step, such kinetics differences hold until major disintegration after no more than 15 min.

Rational nanoparticle design is one of the main goals of materials science, but it can only be achieved via a thorough understanding of the growth process and of the respective roles of the molecular species involved. We demonstrate that a combination of complementary techniques can yield novel information with respect to their individual contributions. We monitored the growth of long aspect ratio silver rods from gold pentatwinned seeds by three in situ techniques (small-angle x-ray scattering, optical absorbance spectroscopy and liquid-cell transmission electron microscopy). Exploiting the difference in reaction speed between the bulk synthesis and the nanoparticle formation in the TEM cell, we show that the anisotropic growth is thermodynamically controlled (rather than kinetically) and that ascorbic acid, widely used for its mild reductive properties plays a shape-directing role, by stabilizing the {100} facets of the silver cubic lattice, in synergy with the halide ions. This approach can easily be applied to a wide variety of synthesis strategies.

The development of nanomedicines for the treatment of neurodegenerative disorders demands innovative nanoarchitectures for combined loading of multiple neuroprotective compounds. We report dual-drug loaded monoolein-based liquid crystalline architectures designed for the encapsulation of a therapeutic protein and a small molecule antioxidant. Catalase (CAT) is chosen as a metalloprotein, which provides enzymatic defense against oxidative stress caused by reactive oxygen species (ROS) such as hydrogen peroxide (H2O2). Curcumin (CU), solubilized in fish oil, is co-encapsulated as a chosen drug with multiple therapeutic activities, which may favor neuro-regeneration. The prepared self-assembled biomolecular nanoarchitectures are characterized by biological synchrotron small-angle X-ray scattering (BioSAXS) at multiple compositions of the lipid/co-lipid/water phase diagram. Constant fractions of curcumin (an antioxidant) and a PEGylated agent (TPEG1000) are included with regard to the lipid fraction. Stable cubosome architectures are obtained for several ratios of the lipid ingredients monoolein (MO) and fish oil (FO). The impact of catalase on the structural organization of the cubosome nanocarriers is revealed by the variations of the cubic lattice parameters deduced by BioSAXS. The outcome of the cellular uptake of the dual drug-loaded nanocarriers is assessed by performing a bioassay of catalase peroxidatic activity in lysates of nanoparticle-treated differentiated SH-SY5Y human cells. The obtained results reveal the neuroprotective potential of the in vitro studied cubosomes in terms of enhanced peroxidatic activity of the catalase enzyme, which enables the inhibition of H2O2 accumulation in degenerating neuronal cells.

International audience; An important aspect of cells is their shape flexibility that gives them motion but also a high adaptation versatility to their environment. This shape versatility is mediated by different types of protein−membrane interactions among which electrostatic plays an important role. In the present work we examined the interaction between a small dicationic peptide, that possesses self-assembly properties, and lipid model membranes. The peptide, lanreotide, spontaneously forms nanotubes in water that have a strictly uniform diameter. In the current work, we show that the interaction between the cationic peptide and negatively charged bilayers of lipids induces the formation of myelin sheath-like structures that we call nanoscrolls. By deciphering the different steps of formation and the molecular structure of the self-assembly, we show how electrostatics modify the spontaneous peptide and lipid way of packing.

International audience; Amorphous cerium oxalate is characterized and reported here for the first time as a primary nucleating transient precursor for a more stable crystalline hydrated phase, at high supersaturation and in strong acid solutions. Preliminary results point out an initial binodal phase separation leading to precipitation.

Post-transcriptional control of gene expression by small regulatory noncoding RNA (sRNA) needs protein accomplices to occur. Past research mainly focused on the RNA chaperone Hfq as cofactor. Nevertheless, recent studies indicated that other proteins might be involved in sRNA-based regulations. As some of these proteins have been shown to self-assemble, we describe in this chapter protocols to analyze the nano-assemblies formed. Precisely, we focus our analysis on Escherichia coli Hfq as a model, but the protocols presented here can be applied to analyze any polymer of proteins. This chapter thus provides a guideline to develop commonly used approaches to detect prokaryotic protein self-assembly, with a special focus on the detection of amyloidogenic polymers.

International audience; A new method based on the combination of small-anglescattering, reverse Monte Carlo simulations, and an aggregate recognition algorithm is proposed to characterize the structure of nanoparticle suspensions in solvents and polymer nanocomposites, allowing detailedstudies of the impact of different nanoparticle surface modifications.Experimental small-angle scattering is reproduced using simulated annealing of configurations of polydisperse particles in a simulation box compatible with the lowest experimental q-vector. Then, properties of interest likeaggregation states are extracted from these configurations and averaged. This approach has been applied to silane surface-modified silica nanoparticles with different grafting groups, in solvents and after casting into polymer matrices.It is shown that the chemistry of the silane function, in particular mono- or trifunctionality possibly related to patch formation, affects the dispersion state in a given medium, in spite of an unchanged alkylchain length. Our approach may be applied to study any dispersion or aggregation state of nanoparticles. Concerningnanocomposites, the method has potential impact on the design of new formulations allowing controlled tuning of nanoparticle dispersion.

The microstructure of polymer nanocomposites made with disordered silica filler (Zeosil(R) 1165MP) of industrial relevance and various coating agents is quantitatively analyzed using a combination of SAXS, TEM, and a recently developed structural model. The polymer matrix is formed by an end-functionalized styrene–butadiene statistical copolymer capable of covalent grafting on the silica nanoparticles. The effect of the coating agents with different alkyl chain length (C8, C12, and C18) on filler structure quantified in terms of aggregate formation, for different concentrations (up to 8 wt % with respect to silica), and the effect of a commonly added catalyzer, DPG, are studied using the structural model. As a result, we show that a strongly synergetic effect of both DPG and coating agent exists. Our findings open the road to a fundamental understanding and rational design of model and industrial nanocomposite formulation with optimized coating agents.

International audience; In the field of structural biology, Small Angle X-ray Scattering (SAXS) has undergone a tremendous evolution in the last two decades. From a craft reserved to a few experts in the late 80's, it has now turned into a high-throughput technique, following the same trend as macromolecular crystallography. Synchrotron radiation has played a key role in this evolution, by providing intense X-ray beams of high optical quality that made possible the recording of statistically meaningful data from weakly scattering biological solutions in a reasonable time. This, in turn, prompted the development of powerful and specific software for data analysis and modeling. In this mini-review, mainly addressed towards a broad readership, representing as many potential users, we try to summarize the latest aspects of evolution of BioSAXS, both conceptually and from the point of view of instrumentation. We emphasize the need for complementary experimental or computational techniques used in combination with SAXS. The great potential of these multi-pronged approaches is illustrated by a series of very recent studies covering the various ways and means of using BioSAXS.

International audience; Self-organization of fluorescent nanoparticles, using biological molecules such as phospholipids to control assembly distances, is a promising method for creating hybrid nanostructures. We report here the formation of hybrid condensed phases made of anisotropic nanoparticles and phospholipids. Such structure formation is driven by electrostatic interaction between the nanoparticles and the phospholipids, and results in the formation of a 2D rectangular liquid crystal, as confirmed by high-resolution Small-Angle X-ray Scattering (SAXS). Moreover, we show that the fluorescent properties of the NPs are not modified by the self-assembly process

International audience; The use of peptidic ligands is validated as a generic chemical platform allowing one to finely control the organization in solid phase of semiconductor nanorods originally dispersed in an aqueous media. An original method to generate, on a macroscopic scale and with the desired geometry, three-dimensional supracrystals composed of quantum rods is introduced. In a first step, nanorods are transferred in an aqueous phase thanks to the substitution of the original capping layer by peptidic ligands. Infrared and nuclear magnetic resonance spectroscopy data prove that the exchange is complete; fluorescence spectroscopy demonstrates that the emitter optical properties are not significantly altered; electrophoresis and dynamic light scattering experiments assess the good colloidal stability of the resulting aqueous suspension. In a second step, water evaporation in a microstructured environment yields superstructures with a chosen geometry and in which nanorods obey a smectic B arrangement, as shown by electron microscopy. Incidentally, bulk drying in a capillary tube generates a similar local order, as evidenced by small angle X-ray scattering.

International audience; Herein, we describe the self-assembling of gold nanorods (GNRs) induced during the ligand exchange at their surface. An exchange reaction between tricysteine PEGylated peptidic ligands and cetyltrimethylammonium bromide (CTAB)-protected gold nanorods is conducted. We demonstrated that the terminal group charge (positively or negatively charged) and the hydrophobicity of the peptidic ligands (bearing or not an undecanoyl chain) strongly affects the self-organization of the GNRs occurring in solution. Adjusting the amount of short PEGylated peptides causes a self-organization of the gold nanorods in solution, resulting in a red- or blue-shift of the plasmon bands. The decrease of their surface charge and the self-assembling in solution were first shown by zetametry, by Dynamic Light Scattering and UV-spectroscopy. Thanks to Small Angle X-ray Scattering experiments and Transmission Electron Microscopy images, the self-organization of the nanorods in solution was clearly demonstrated and correlated to the spectroscopic change in absorbance. Conversely, in the case of longer PEGylated peptidic ligands including an undecanoyl chain, the GNRs are particularly stable against aggregation for several days after purification. By controlled drying on a substrate, we showed their ability to self-organize into well-defined ordered structures making them very attractive as building blocks to design optical materials. International audience; We describe a method of controlled evaporation on a textured substrate for self-assembling and shaping gold-nanorod-based materials. Tridimensional wall features are formed over areas as large as several square millimeters. Furthermore, analyses by small-angle X-ray scattering and scanning electron microscopy techniques demonstrate that colloids are locally ordered as a smectic B phase. Such crystallization is in fact possible because we could finely adjust the nanoparticle charge, knowledge that additionally enables tuning the lattice parameters. In the future, the type of ordered self-assemblies of gold nanorods we have prepared could be used for amplifying optical signals.
