Abstract

Most genetic studies consider autism spectrum disorder (ASD) and developmental disorder (DD) separately despite overwhelming comorbidity and shared genetic etiology. Here, we analyzed de novo variants (DNVs) from 15,560 ASD (6,557 from SPARK) and 31,052 DD trios independently and also combined as broader neurodevelopmental disorders (NDDs) using three models. We identify 615 NDD candidate genes (false discovery rate [FDR] < 0.05) supported by ≥1 models, including 138 reaching Bonferroni exome-wide significance (P < 3.64e-7) in all models. The genes group into five functional networks associating with different brain developmental lineages based on single-cell nuclei transcriptomic data. We find no evidence for ASD-specific genes in contrast to 18 genes significantly enriched for DD. There are 53 genes that show mutational bias, including enrichments for missense (n = 41) or truncating (n = 12) DNVs. We also find 10 genes with evidence of male- or female-bias enrichment, including 4 X chromosome genes with significant female burden (DDX3X, MECP2, WDR45, and HDAC8). This large-scale integrative analysis identifies candidates and functional subsets of NDD genes.

Abstract

We present a sampling of recently published picturebook biographies with noteworthy endpapers. First, we highlight scholarship related to picturebook biographies and peritextual features. Second, we present examples of three ways endpapers function, using nine picturebook biographies while also incorporating pedagogical suggestions related to the endpapers. Last, we explain the importance of noticing peritextual features in picturebook biographies.

4. Preliminary Efficacy of a Daily Living Skills Intervention for Adolescents with High-Functioning Autism Spectrum Disorder

Abstract

Daily living skills deficits are strongly associated with poor adult outcomes for individuals with high-functioning autism spectrum disorder, and yet, there are no group interventions targeting daily living skills. Seven adolescents with autism spectrum disorder and their parents participated in a feasibility pilot of a 12-week manualized group treatment targeting specific daily living skills (i.e. morning routine, cooking, laundry, and money management). Outcomes included the Vineland Adaptive Behavior Scales, Second Edition (Vineland-II) age equivalence scores and four goal attainment scaling scores. Adolescents demonstrated significant improvement on two Vineland-II subdomains and on all goal attainment scaling scores at post-treatment and 6-month follow-up. The intervention has promise for improving critical daily living skills' deficits that affect independent living and employment. Limitations and implications for future studies are discussed.

9. Comparison of paleobotanical and biomarker records of mountain peatland and forest ecosystem dynamics over the last 2600 years in central Germany.

Abstract

As peatlands are a major terrestrial sink in the global carbon cycle, gaining an understanding of their development and changes throughout time is essential in order to predict their future carbon budget and potentially mitigate the adverse outcomes of climate change. With this aim to understand peat development, many studies have investigated the paleoecological dynamics by analyzing various proxies, including pollen, macrofossil, elemental, and biomarker analyses. However, as each of these proxies is known to have its own benefits and limitations, examining them in parallel allows for a deeper understanding of these paleoecological dynamics at the peatland and a systematic comparison of the power of these individual proxies. In this study, we therefore analyzed peat cores from a peatland in Germany (Beerberg, Thuringia) to (a) characterize the vegetation dynamics over the course of the peatland development during the late Holocene and (b) evaluate to what extent the inclusion of multiple proxies, specifically pollen, plant macrofossils, and biomarkers, contributes to a deeper understanding of those dynamics and interaction among factors. We found that, despite a major shift in the regional forest composition from primarily beech to spruce as well as many indicators of human impact in the region, the local plant population in the Beerberg area remained stable over time following the initial phase of peatland development up until the last couple of centuries. Therefore, little variation could be derived from the paleobotanical data alone. The combination of pollen and macrofossil analyses with the elemental and biomarker analyses enabled further understanding of the site development as these proxies added valuable additional information, including the occurrence of climatic variations, such as the Little Ice Age, and more recent disturbances, such as drainage. 16. Transformation of n-alkanes from plant to soil: a review

Abstract

Despite the importance of soil organic matter (SOM) in the global carbon cycle, there remain many open questions regarding its formation and preservation. The study of individual organic compound classes that make up SOM, such as lipid biomarkers including n-alkanes, can provide insight into the cycling of bulk SOM. While studies of lipid biomarkers, particularly n-alkanes, have increased in number in the past few decades, only a limited number have focused on the transformation of these compounds following deposition in soil archives. We performed a systematic review to consolidate the available information on plant-derived n-alkanes and their transformation from plant to soil. Our major findings were (1) a nearly ubiquitous trend of decreased total concentration of n-alkanes either with time in litterbag experiments or with depth in open plant–soil systems and (2) preferential degradation of odd-chain length and shorter chain length n-alkanes represented by a decrease in either carbon preference index (CPI) or odd-over-even predominance (OEP) with depth, indicating degradation of the n-alkane signal or a shift in vegetation composition over time. The review also highlighted a lack of data transparency and standardization across studies of lipid biomarkers, making analysis and synthesis of published data time-consuming and difficult. We recommend that the community move towards more uniform and systematic reporting of biomarker data. Furthermore, as the number of studies examining the complete leaf–litter–soil continuum is very limited as well as unevenly distributed over geographical regions, climate zones, and soil types, future data collection should focus on underrepresented areas as well as quantifying the transformation of n-alkanes through the complete continuum from plant to soil.

24. "My Name Is ...": Picturebooks Exploring Linguistically and Culturally Diverse Names.

Abstract

A child's personal name is an integral part of their identity. Names and name negotiation in children's picturebooks can explore this connection by narrativizing the impact of positive and negative experiences involving name-carriers, name-givers, and name-users. In this study, we began with a framework combining a socio-onomastic perspective with the children's literature metaphor of "mirrors and windows" (Bishop 1990) and the educational research concept of "damage and desire" narratives (Tuck 2009). Our content analysis of twelve picturebooks featuring characters with culturally and linguistically diverse names led to a coding scheme of six common episodes of name negotiation in the picturebooks' narrative arcs: (1) inflicted damage; (2) internalized damage; (3) supplying desire; (4) internalized desire; (5) asserting the desire; and (6) joining the desire. Our findings highlight how episodes of damage focus on the pain, sadness, and struggle name-carriers undergo, while episodes of desire center the support of parents and teachers as well as detailed cultural and familial information about names. We conclude that while both "damage and desire" episodes contribute to the narratives, too heavy a focus on damage could lead to the perpetuation of a "single story" (Adichie 2009) that normalizes pain and struggle as an inevitable experience for children with linguistically and culturally diverse names. 30. Transport and digestive alteration of uniformly (13)C-labeled diatoms in mudflat sediments

Abstract

The influence of deposit feeders on marine sediment was explored using uniformly (13)C-labeled diatoms as tracers. Findings show that deposit feeders responded promptly to the introduction of diatoms, and shifted phytodetritus during hoeing and feeding activities. Deposit feeders that ingested the diatoms produced fecal matter with (13)C amino acid signatures which were significantly different than that of the diatoms.

33. Transformation of n-alkanes from plant to soil: a review.

Abstract

Despite the importance of soil organic matter (SOM) in the global carbon cycle, there remain many open questions regarding its formation and preservation. The study of individual organic compound classes that make up SOM, such as lipid biomarkers including n -alkanes, can provide insight into the cycling of bulk SOM. While studies of lipid biomarkers, particularly n -alkanes, have increased in number in the past few decades, only a limited number have focused on the transformation of these compounds following deposition in soil archives. We performed a systematic review to consolidate the available information on plant-derived n -alkanes and their transformation from plant to soil. Our major findings were (1) a nearly ubiquitous trend of decreased total concentration of n -alkanes either with time in litterbag experiments or with depth in open plant–soil systems and (2) preferential degradation of odd-chain length and shorter chain length n -alkanes represented by a decrease in either carbon preference index (CPI) or odd-over-even predominance (OEP) with depth, indicating degradation of the n -alkane signal or a shift in vegetation composition over time. The review also highlighted a lack of data transparency and standardization across studies of lipid biomarkers, making analysis and synthesis of published data time-consuming and difficult. We recommend that the community move towards more uniform and systematic reporting of biomarker data. Furthermore, as the number of studies examining the complete leaf–litter–soil continuum is very limited as well as unevenly distributed over geographical regions, climate zones, and soil types, future data collection should focus on underrepresented areas as well as quantifying the transformation of n -alkanes through the complete continuum from plant to soil. Abstract

A routine 2-dose varicella vaccination program was adopted in 2007 in the US to help further decrease varicella disease and prevent varicella outbreaks. We describe trends and characteristics of varicella outbreaks reported to the Centers for Disease Control and Prevention (CDC) during 2005-2012 from 9 states.Data on varicella outbreaks collected by 9 state health departments were submitted to CDC using the CDC outbreak reporting worksheet. Information was collected on dates of the outbreak, outbreak setting and number of cases by outbreak; aggregate data were provided on the numbers of outbreak-related cases by age group, vaccination status and laboratory confirmation.Nine hundred and twenty-nine outbreaks were reported from the 6 states, which provided data for each year during 2005-2012. Based on data from these 6 states, the number of outbreaks declined by 78%, decreasing from 147 in 2005 to 33 outbreaks in 2012 (P = 0.0001). There were a total of 1015 varicella outbreaks involving 13,595 cases reported by the 9 states from 2005 to 2012. The size and duration of outbreaks declined significantly over time (P0.001). The median size of outbreaks was 12, 9 and 7 cases and median duration of outbreaks was 38, 35 and 26 days during 2005-2006, 2007-2009 and 2010-2012, respectively. Majority of outbreaks (95%) were reported from schools, declining from 97% in 2005-2006 to 89% in 2010-2012. Sixty-five percent of outbreak-related cases occurred among 5-year to 9-year olds, with the proportion declining from 76% in 2005-2006 to 45% during 2010-2012.The routine 2-dose varicella vaccination program appears to have significantly reduced the number, size and duration of varicella outbreaks in the US.

Abstract

Background In 2007, a routine second dose of varicella vaccine was recommended in the United States for children aged 4 to 6 years to better control varicella-zoster virus circulation and outbreaks. Sentinel varicella outbreak surveillance was established to assess feasibility of surveillance and describe outbreaks that are occurring. Methods Through the Centers for Disease Control and Prevention Epidemiology Laboratory Capacity funding, health departments conducted active surveillance for varicella outbreaks in schools from 2012 to 2015. Outbreaks of varicella were defined as ≥5 cases in a school within at least 1 incubation period (21 days). School nurses, healthcare providers, or laboratories reported cases and outbreaks of varicella to health departments; demographic, vaccination, and clinical data were collected. Results Georgia, Houston, Maine, Minnesota, New York City, and Philadelphia participated in all 3 years; Puerto Rico and West Virginia participated in 2012 to 2013; and Kansas and Arkansas participated in 2014 to 2015. Twenty-nine outbreaks including 262 cases were reported. The median size of the outbreaks was 7 cases (range, 5–31 cases), and the median duration was 31 days (range, 4–100 days). Of the case-patients associated with larger outbreaks (≥8 cases), 55.4% were unvaccinated, and 15.7% and 18.1% had received 1 or 2 doses of vaccine, respectively. In small outbreaks (5–7 cases), 33.3% of case-patients were unvaccinated, and 16.7% and 38.5% had received 1 or 2 doses of vaccine, respectively. Conclusions The majority of cases associated with outbreaks occurred in undervaccinated children (unvaccinated and 1-dose vaccine recipients). Outbreaks with a greater proportion of 2-dose vaccine recipients were smaller. Varicella outbreak surveillance is feasible, and continued monitoring of outbreaks remains important for describing the epidemiology of varicella during the 2-dose varicella vaccination program. Abstract

Daily living skills deficits are strongly associated with poor adult outcomes for individuals with high-functioning autism spectrum disorder, and yet, there are no group interventions targeting daily living skills. Seven adolescents with autism spectrum disorder and their parents participated in a feasibility pilot of a 12-week manualized group treatment targeting specific daily living skills (i.e. morning routine, cooking, laundry, and money management). Outcomes included the Vineland Adaptive Behavior Scales, Second Edition (Vineland-II) age equivalence scores and four goal attainment scaling scores. Adolescents demonstrated significant improvement on two Vineland-II subdomains and on all goal attainment scaling scores at post-treatment and 6-month follow-up. The intervention has promise for improving critical daily living skills’ deficits that affect independent living and employment. Limitations and implications for future studies are discussed.
