Abstract: We present new spatially resolved astrometry and photometry of the CD-27 11535 system, a member of the $\beta$ Pictoris moving group consisting of two resolved K-type stars on a $\sim$20-year orbit. We fit an orbit to relative astrometry measured from NIRC2, GPI, and archival NaCo images, in addition to literature measurements. However, the total mass inferred from this orbit is significantly discrepant from that inferred from stellar evolutionary models using the luminosity of the two stars. We explore two hypotheses that could explain this discrepant mass sum; a discrepant parallax measurement from Gaia due to variability, and the presence of an additional unresolved companion to one of the two components. We find that the $\sim$20-year orbit could not bias the parallax measurement, but that variability of the components could produce a large amplitude astrometric motion, an effect which cannot be quantified exactly without the individual Gaia measurements. The discrepancy could also be explained by an additional star in the system. We jointly fit the astrometric and photometric measurements of the system to test different binary and triple architectures for the system. Depending on the set of evolutionary models used, we find an improved goodness of fit for a triple system architecture that includes a low-mass ($M=0.177\pm0.055$\,$M_{\odot}$) companion to the primary star. Further studies of this system will be required in order to resolve this discrepancy, either by refining the parallax measurement with a more complex treatment of variability-induced astrometric motion, or by detecting a third companion., Comment: 12 pages + references and appendix, 12 figures, 5 tables

Abstract: Current analyses and predictions of spatially explicit patterns and processes in ecology most often rely on climate data interpolated from standardized weather stations. This interpolated climate data represents long-term average thermal conditions at coarse spatial resolutions only. Hence, many climate-forcing factors that operate at fine spatiotemporal resolutions are overlooked. This is particularly important in relation to effects of observation height (e.g. vegetation, snow and soil characteristics) and in habitats varying in their exposure to radiation, moisture and wind (e.g. topography, radiative forcing or cold-air pooling). Since organisms living close to the ground relate more strongly to these microclimatic conditions than to free-air temperatures, microclimatic ground and near-surface data are needed to provide realistic forecasts of the fate of such organisms under anthropogenic climate change, as well as of the functioning of the ecosystems they live in. To fill this critical gap, we highlight a call for temperature time series submissions to SoilTemp, a geospatial database initiative compiling soil and near-surface temperature data from all over the world. Currently, this database contains time series from 7,538 temperature sensors from 51 countries across all key biomes. The database will pave the way toward an improved global understanding of microclimate and bridge the gap between the available climate data and the climate at fine spatiotemporal resolutions relevant to most organisms and ecosystem processes.

Abstract: Mineral-associated organic carbon (MAOC) constitutes a major fraction of global soil carbon and is assumed less sensitive to climate than particulate organic carbon (POC) due to protection by minerals. Despite its importance for long-term carbon storage, the response of MAOC to changing climates in drylands, which cover more than 40% of the global land area, remains unexplored. Here we assess topsoil organic carbon fractions across global drylands using a standardized field survey in 326 plots from 25 countries and 6 continents. We find that soil biogeochemistry explained the majority of variation in both MAOC and POC. Both carbon fractions decreased with increases in mean annual temperature and reductions in precipitation, with MAOC responding similarly to POC. Therefore, our results suggest that ongoing climate warming and aridification may result in unforeseen carbon losses across global drylands, and that the protective role of minerals may not dampen these effects.

Abstract: Perennial plants create productive and biodiverse hotspots, known as fertile islands, beneath their canopies. These hotspots largely determine the structure and functioning of drylands worldwide. Despite their ubiquity, the factors controlling fertile islands under conditions of contrasting grazing by livestock, the most prevalent land use in drylands, remain virtually unknown. Here we evaluated the relative importance of grazing pressure and herbivore type, climate and plant functional traits on 24 soil physical and chemical attributes that represent proxies of key ecosystem services related to decomposition, soil fertility, and soil and water conservation. To do this, we conducted a standardized global survey of 288 plots at 88 sites in 25 countries worldwide. We show that aridity and plant traits are the major factors associated with the magnitude of plant effects on fertile islands in grazed drylands worldwide. Grazing pressure had little influence on the capacity of plants to support fertile islands. Taller and wider shrubs and grasses supported stronger island effects. Stable and functional soils tended to be linked to species-rich sites with taller plants. Together, our findings dispel the notion that grazing pressure or herbivore type are linked to the formation or intensification of fertile islands in drylands. Rather, our study suggests that changes in aridity, and processes that alter island identity and therefore plant traits, will have marked effects on how perennial plants support and maintain the functioning of drylands in a more arid and grazed world.

Abstract: Biological soil crusts, or biocrusts, are microbial communities found in soil surfaces in drylands and in other locations where vascular plant cover is incomplete. They are functionally significant for numerous ecosystem services, most notably in the C fixation and storage due to the ubiquity of photosynthetic microbes. Whereas carbon fixation and storage have been well studied in biocrusts, the composition, function and characteristics of other organisms in the biocrust such as heterotrophic bacteria and especially fungi are considerably less studied and this limits our ability to gain a holistic understanding of biocrust ecology and function. In this research we characterised the fungal community in biocrusts developed on Kalahari Sand soils from a site in southwest Botswana, and combined these data with previously published bacterial community data from the same site. By identifying organisational patterns in the community structure of fungi and bacteria, we found fungi that were either significantly associated with biocrust or the soil beneath biocrusts, leading to the conclusion that they likely perform functions related to the spatial organisation observed. Furthermore, we showed that within biocrusts bacterial and fungal community structures are correlated with each other i.e., a change in the bacterial community is reflected by a corresponding change in the fungal community. Importantly, this correlation but that this correlation does not occur in nearby soils. We propose that different fungi engage in short-range and long-range interactions with dryland soil surface bacteria. We have identified fungi which are candidates for further studies into their potential roles in biocrust ecology at short ranges (e.g., processing of complex compounds for waste management and resource provisioning) and longer ranges (e.g., translocation of resources such as water and the fungal loop model). Yet its impacts on ecosystem services remain uncertain because pervasive interactions between grazing pressure, climate, soil properties, and biodiversity may occur but have never been addressed simultaneously. Using a standardized survey at 98 sites across six continents, we show that interactions between grazing pressure, climate, soil, and biodiversity are critical to explain the delivery of fundamental ecosystem services across drylands worldwide. 26. Institutional Bias and the Degree Class System

Abstract: Over the last decade the proportion of students in higher education obtaining aggregate marks over 60 per cent has steadily increased. Whilst standards of learning and teaching may have improved due to the existence of quality assurance agencies and the efforts of individual higher education institutions a significant factor remains that could also have influenced this rise. The degree of autonomy allowed to individual higher education institutions regarding the systems in place to decide on the degree class awarded to a student can significantly alter the cohort degree class profile. Most institutions operate an aggregate mean marking system tied to algorithms designed to reward borderline students who have performed well across all of their modules. These algorithms vary between institutions and their effect on a cohort of 120 geography students has been investigated using the classification systems operated by 10 geography departments in England. The results indicate that a significant number of students receive higher degree classes that they would under the strict aggregate mean rules. More significantly, there is evidence that award levels vary between institutions as a result of the variety of algorithms that exist. In several instances over 10 per cent of students received elevated degree class awards with an inter-institutional variation of several per cent. These discrepancies are significant enough to warrant a review of the present system with a view to introducing operational standards across the university sector. (Contains 5 tables and 2 figures.)

Abstract: Research in global change ecology relies heavily on global climatic grids derived from estimates of air temperature in open areas at around 2 m above the ground. These climatic grids do not reflect conditions below vegetation canopies and near the ground surface, where critical ecosystem functions occur and most terrestrial species reside. Here, we provide global maps of soil temperature and bioclimatic variables at a 1-km2 resolution for 0-5 and 5-15 cm soil depth. These maps were created by calculating the difference (i.e. offset) between in situ soil temperature measurements, based on time series from over 1200 1-km2 pixels (summarized from 8519 unique temperature sensors) across all the world's major terrestrial biomes, and coarse-grained air temperature estimates from ERA5-Land (an atmospheric reanalysis by the European Centre for Medium-Range Weather Forecasts). We show that mean annual soil temperature differs markedly from the corresponding gridded air temperature, by up to 10°C (mean = 3.0 ± 2.1°C), with substantial variation across biomes and seasons. Over the year, soils in cold and/or dry biomes are substantially warmer (+3.6 ± 2.3°C) than gridded air temperature, whereas soils in warm and humid environments are on average slightly cooler (-0.7 ± 2.3°C). The observed substantial and biome-specific offsets emphasize that the projected impacts of climate and climate change on near-surface biodiversity and ecosystem functioning are inaccurately assessed when air rather than soil temperature is used, especially in cold environments. The global soil-related bioclimatic variables provided here are an important step forward for any application in ecology and related disciplines. Nevertheless, we highlight the need to fill remaining geographic gaps by collecting more in situ measurements of microclimate conditions to further enhance the spatiotemporal resolution of global soil temperature products for ecological applications.

Abstract: Research in global change ecology relies heavily on global climatic grids derived from estimates of air temperature in open areas at around 2 m above the ground. These climatic grids do not reflect conditions below vegetation canopies and near the ground surface, where critical ecosystem functions occur and most terrestrial species reside. Here, we provide global maps of soil temperature and bioclimatic variables at a 1-km² resolution for 0–5 and 5–15 cm soil depth. These maps were created by calculating the difference (i.e., offset) between in-situ soil temperature measurements, based on time series from over 1200 1-km² pixels (summarized from 8500 unique temperature sensors) across all the world’s major terrestrial biomes, and coarse-grained air temperature estimates from ERA5-Land (an atmospheric reanalysis by the European Centre for Medium-Range Weather Forecasts). We show that mean annual soil temperature differs markedly from the corresponding gridded air temperature, by up to 10°C (mean = 3.0 ± 2.1°C), with substantial variation across biomes and seasons. Over the year, soils in cold and/or dry biomes are substantially warmer (+3.6 ± 2.3°C) than gridded air temperature, whereas soils in warm and humid environments are on average slightly cooler (-0.7 ± 2.3°C). The observed substantial and biome-specific offsets emphasize that the projected impacts of climate and climate change on near-surface biodiversity and ecosystem functioning are inaccurately assessed when air rather than soil temperature is used, especially in cold environments. The global soil-related bioclimatic variables provided here are an important step forward for any application in ecology and related disciplines. Nevertheless, we highlight the need to fill remaining geographic gaps by collecting more in-situ measurements of microclimate conditions to further enhance the spatiotemporal resolution of global soil temperature products for ecological applications.

Abstract: Grazing represents the most extensive use of land worldwide. Yet its impacts on ecosystem services remain uncertain because pervasive interactions between grazing pressure, climate, soil properties, and biodiversity may occur but have never been addressed simultaneously. Using a standardized survey at 98 sites across six continents, we show that interactions between grazing pressure, climate, soil, and biodiversity are critical to explain the delivery of fundamental ecosystem services across drylands worldwide. Increasing grazing pressure reduced ecosystem service delivery in warmer and species-poor drylands, whereas positive effects of grazing were observed in colder and species-rich areas. Considering interactions between grazing and local abiotic and biotic factors is key for understanding the fate of dryland ecosystems under climate change and increasing human pressure.

Abstract: Research in global change ecology relies heavily on global climatic grids derived from estimates of air temperature in open areas at around 2 m above the ground. These climatic grids do not reflect conditions below vegetation canopies and near the ground surface, where critical ecosystem functions occur and most terrestrial species reside. Here, we provide global maps of soil temperature and bioclimatic variables at a 1-km2 resolution for 0-5 and 5-15 cm soil depth. These maps were created by calculating the difference (i.e. offset) between in situ soil temperature measurements, based on time series from over 1200 1-km2 pixels (summarized from 8519 unique temperature sensors) across all the world's major terrestrial biomes, and coarse-grained air temperature estimates from ERA5-Land (an atmospheric reanalysis by the European Centre for Medium-Range Weather Forecasts). We show that mean annual soil temperature differs markedly from the corresponding gridded air temperature, by up to 10°C (mean = 3.0 ± 2.1°C), with substantial variation across biomes and seasons. Over the year, soils in cold and/or dry biomes are substantially warmer (+3.6 ± 2.3°C) than gridded air temperature, whereas soils in warm and humid environments are on average slightly cooler (-0.7 ± 2.3°C). The observed substantial and biome-specific offsets emphasize that the projected impacts of climate and climate change on near-surface biodiversity and ecosystem functioning are inaccurately assessed when air rather than soil temperature is used, especially in cold environments. The global soil-related bioclimatic variables provided here are an important step forward for any application in ecology and related disciplines. Nevertheless, we highlight the need to fill remaining geographic gaps by collecting more in situ measurements of microclimate conditions to further enhance the spatiotemporal resolution of global soil temperature products for ecological applications.

THOMAS (1), PATRICIA E LINTON (1) Keywords: soil CO flux; drylands; photosynthetically active radiation; soil respiration chambers; cyanobacterial soil crusts; Kalahari Abstract: Abstract This paper describes the design features and capabilities of a portable automated in-situ closed chamber (ISCC) for the quantification of CO.sub.2 fluxes in dryland soils where both photosynthetic and respiratory components may be associated with a cyanobacterial crust. The processes of CO.sub.2 flux in dryland soils are briefly described in order to clarify the conditions that make quantification of these fluxes problematic. The instrumentation currently available for in-situ soil CO.sub.2 flux measurements is then reviewed demonstrating their inadequacies for the dryland environment. The ISCC described here is a member of the closed or enrichment class of soil respiration chambers. The ISCC, however, features an optical window possessing high (>90%) transmission in the photosynthetic active region (PAR) of the solar irradiance spectrum, permitting observations of photosynthesis. The ISCC possesses automatic venting and purging so that gaseous concentrations inside the chamber do not change from ambient sufficiently to significantly affect diffusion. The ISCC features both active and passive cooling employing internal solid-state Peltier coolers and external aluminised Mylar respectively. This avoids severe disturbance of the microclimate within the chamber due to admission of high fluxes of PAR and permits in-situ operation under a wide range of ambient field temperatures (~ -5 to 40[degrees]C). Sensors internal to the chamber monitor temperature, relative humidity, irradiance and pressure. In this implementation the ISCC is coupled to a portable gas chromatograph (Agilent GC-3000) to sample the chamber atmosphere. Indicative data for Kalahari Sand soils of Botswana are presented as an illustration of the general performance characteristics. Abstract: © 2020 John Wiley & Sons Ltd Current analyses and predictions of spatially explicit patterns and processes in ecology most often rely on climate data interpolated from standardized weather stations. This interpolated climate data represents long-term average thermal conditions at coarse spatial resolutions only. Hence, many climate-forcing factors that operate at fine spatiotemporal resolutions are overlooked. This is particularly important in relation to effects of observation height (e.g. vegetation, snow and soil characteristics) and in habitats varying in their exposure to radiation, moisture and wind (e.g. topography, radiative forcing or cold-air pooling). Since organisms living close to the ground relate more strongly to these microclimatic conditions than to free-air temperatures, microclimatic ground and near-surface data are needed to provide realistic forecasts of the fate of such organisms under anthropogenic climate change, as well as of the functioning of the ecosystems they live in. To fill this critical gap, we highlight a call for temperature time series submissions to SoilTemp, a geospatial database initiative compiling soil and near-surface temperature data from all over the world. Currently, this database contains time series from 7,538 temperature sensors from 51 countries across all key biomes. The database will pave the way toward an improved global understanding of microclimate and bridge the gap between the available climate data and the climate at fine spatiotemporal resolutions relevant to most organisms and ecosystem processes.

Abstract: Seasonally-flooded várzea forests of Western Amazonia are one of the most productive and biodiverse wetland forests in the world. However, data on their soil CO2emissions, soil organic matter decomposition rates, and soil C stocks are scarce. This is a concern because hydrological changes are predicted to lead to increases in the height, extent, and duration of seasonal floods, which are likely to have a significant effect on soil C stocks and fluxes. However, with no empirical data, the impact of altered flood regimes on várzea soil C cycles remains uncertain. This study quantified the effects of maximum annual flood height and soil moisture on soil CO2efflux rate (Rs) and soil organic matter decomposition rate (k) in the várzea forests of Pacaya Samiria National Reserve, Peru. The study was conducted between May and August 2017. The results showed that Rs (10.6–182.7 mg C m−2h–1) and k(0.016–0.078) varied between and within sites, and were considerably lower than the values reported for other tropical forests. In addition, Rswas negatively affected by flood height (P <0.01) and soil moisture (P< 0.001), and it decreased with decreasing river levels post flooding (P< 0.001). In contrast, k was not affected by any of the above-mentioned factors. Soil moisture was the dominant factor influencing Rs, and it was significantly affected by maximum flood height, even after the floods had subsided (P< 0.001). Consequently, we concluded that larger floods will likely lead to reduced Rs, whilst kcould remain unchanged but with decomposition processes becoming more anaerobic.

50. Expanded human blood-derived gamma delta T cells display potent antigen-presentation functions

Abstract: Cell-based immunotherapy strategies target tumors directly (via cytolytic effector cells) or aim at mobilizing endogenous anti-tumor immunity. The latter approach includes dendritic cells (DC) most frequently in the form of in vitro cultured peripheral blood monocytes-derived DC. Human blood γδT cells are selective for a single class of non-peptide agonists (“phosphoantigens”) and develop into potent antigen-presenting cells (APC), termed γδT-APC within 1–3 days of in vitro culture. Availability of large numbers of γδT-APC would be advantageous for use as a novel cellular vaccine. We here report optimal γδT cell expansion (>107 cells/ml blood) when peripheral blood mononuclear cells (PBMC) from healthy individuals and melanoma patients were stimulated with zoledronate and then cultured for 14 days in the presence of IL-2 and IL-15, yielding γδT cell cultures of variable purity (77 ± 21 and 56 ± 26%, respectively). They resembled effector memory αβT (TEM) cells and retained full functionality as assessed by in vitro tumor cell killing as well as secretion of pro-inflammatory cytokines (IFNγ, TNFα) and cell proliferation in response to stimulation with phosphoantigens. Importantly, day 14 γδT cells expressed numerous APC-related cell surface markers and, in agreement, displayed potent in vitro APC functions. Day 14 γδT cells from PBMC of patients with cancer were equally effective as their counterparts derived from blood of healthy individuals and triggered potent CD8+ αβT cell responses following processing and cross-presentation of simple (influenza M1) and complex (tuberculin purified protein derivative) protein antigens. Of note, and in clear contrast to peripheral blood γδT cells, the ability of day 14 γδT cells to trigger antigen-specific αβT cell responses did not depend on re-stimulation. We conclude that day 14 γδT cell cultures provide a convenient source of autologous APC for use in immunotherapy of patients with various cancers.
