Objective: This study assessed the status of dynamic thiol/disulfide balance (Dt/dB) among pregnant women throughout pregnancy in order to examine the possible interaction between iron deficiency anemia (IDA) and oxidative stress (OS). Methods: The investigation was designed as a prospective cohort study at the Obstetric Care Service of the Haseki Training and Research Hospital (Istanbul). Ninety pregnant women were divided into 2 groups: 45 with IDA and 45 without this condition. They were also placed into first-, second-, and third-trimester groups, with 15 participants in each group. The study gathered sociodemographic information and conducted analyses on blood samples to assess circulating (Dt/dB) status, ferritin, iron, total iron-binding capacity, and other pertinent hematological and biochemical factors. Results: There were no notable disparities observed in the thiol/disulfide equilibrium between pregnant women with or without IDA. Nevertheless, notable disparities were determined when comparing circulating disulfide values, as well as reduced thiol ratio (RTR) and thiol oxidation-reduction ratio (TORR) in relation to ferritin levels. Women with ferritin levels <20 µg/L exhibited considerably lower circulating disulfide values and significantly higher RTR and TORR values. Conclusions: Pregnant women with low levels of circulating ferritin show signs of heightened OS, as demonstrated by changes in Dt/dB measures. Tracking ferritin levels offers a more precise evaluation of OS, indicating that ferritin is a crucial indicator for OS in this specific group. It is crucial to conduct additional studies using larger and more similar groups in order to confirm these findings and enhance clinical results for both mothers and their infants. Objective: We aimed to compare Total Oxidant Level (TOL), Total Antioxidant Level (TAL), Oxidative Stress Index (OSI), Thiol/Disulfide levels, Plasma Malondialdehyde (MDA) levels in both plasma and erythrocyte patients with OCD and healthy controls. Method: Our study included 47 patients with OCD and 49 healthy controls. Sociodemographic data form was applied to all participants, CGI and Y-BOCS were applied to the patient group. TAL and TOL measurements were made in both plasma and erythrocytes, and Malondialdehyde (MDA) and Thiol/Disulfide measurements were made only in plasma. Results: TAL, TOL, OSI and MDA values in the plasma of the patient and control groups were compared, no statistically significant difference was found. However, erythrocyte TAS level was lower in the patient group (p<0.05) and OSI level was higher (p<0.05) in the patient group. While no difference was observed in plasma total thiol level in the patient group compared to the controls, plasma native thiol levels were significantly higher (p<0.001) and plasma disulfide levels were significantly lower (p<0.05). Discussion: The high level of native thiol, an antioxidant molecule in plasma, in OCD patients can be interpreted as an effort to compensate for the decreased antioxidant capacity in erythrocytes. According to these results, we think that when evaluating oxidative stress parameters in psychiatric diseases such as OCD, it is important to study the thiol/disulfide ratio as well as the total oxidant capacity of the plasma, and it would be appropriate to measure these parameters not only in the plasma but also in the erythrocyte. Background: Vitamin B12 is an important vitamin for metabolism and affects many mechanisms in the body including neuronal migration, DNA synthesis, neurotransmitter synthesis, brain and cognitive development. Increased oxidative stress in the body leads to the damage of the child development, but also plays a crucial role in the pathogenesis of many diseases encountered in the childhood period. Our aim is to investigate whether or not B12 deficiency is associated with dynamic thiol/disulfide homeostasis in adolescent patients. Methods: This is a case-controlled observational study consisting of 45 adolescent patients with vitamin b12 deficiency and a control group consisting of 45 healthy adolescent. Patients between 11 and 18 ages who applied to the outpatient clinic for the first time with one of the complaints of headache were selected due to their decreased school performance, dizziness, and fatigue. Hemogram, vitamin B12, homocysteine levels and oxidative stress parameters such as native and total thiol disulfide levels and ratios of disulfide/native thiol, disulfide/total thiol, and native thiol/total thiol were measured from the patients. Results: Vitamin B12 level was found to be significantly lower in vitamin B12 deficiency group (p < 0.001). The serum disulfide level was found to be 27.5 ± 8.38 in the case group and 20.5 ± 8.36 in the control group (p < 0.001). In the multiple linear regression analysis, it was determined that the independent variables of native thiol, homocysteine and disulfide levels effected of vitamin B12 levels (p < 0.001, p < 0.001, p < 0.005 respectively; R2 = 0.62). Conclusion: The results obtained in terms of the effect of vitamin B12 deficiency on oxidative stress in adolescents are remarkable. The increase in oxidative stress parameters in the patient group may also suggest that oxidative stress plays a vital role in vitamin B12 deficiency in adolescence. [ABSTRACT FROM AUTHOR]

Purpose: The aim of this study was to investigate the thiol/disulfide balance as a marker of oxidative stress in children with vitamin B12 deficiency. Materials and Methods: Sixty one pediatric patients with vitamin B12 deficiency and 62 healthy children as a control group was included in the study. Vitamin B12, homocysteine, complete blood count and thiol/disulfide balance parameters of the participants were measured. The relationship between thiol/disulfide balance parameters and vitamin B12 and homocysteine was investigated. Results: The groups were similar in terms of age and gender distribution. Median vitamin B12 level was 179 (98- 199) pg/ml in the patient group and 298 (201-965) pg/ml in the control group. Median homocysteine level was 11.2 (0.08-64.3) μmol/L in the patient group and 12 (4.5-26.6) μmol/L in the control group. Median values of homocysteine levels were within the normal range in both groups. Between the patient and control groups; no significant difference was found in terms of homocysteine, native thiol, total thiol, disulfide, disulfide/native thiol ratio, disulfide/total thiol ratio and native thiol/total thiol ratio. There was no significant correlation between thiol/disulfide balance parameters and vitamin B12 and homocysteine levels in the patient and control groups. Conclusion: There was no significant increase in homocysteine level, since the vitamin B12 level was not significantly low in our patient group. It can be predicted that a vitamin B12 deficiency that does not increase homocysteine will not increase oxidative stress and therefore will not increase antioxidant capacity. [ABSTRACT FROM AUTHOR]

Purpose: To evaluate thiol/disulfide homeostasis in both maternal and fetal compartment in the presence of fetal growth restriction (FGR).Materials and Methods: A prospective case-control study was carried out in women with FGR (n: 40) or normally growing fetus (n: 40). FGR was defined as estimated fetal weight below the 10th percentile for the gestational age. Maternal serum and fetal cord blood samples were collected from all participants and native thiol-disulfide exchanges were examined with automated method enabling the measurement of both sides of thiol-disulfide balance.Results: Native thiol, total thiol and disulfide amounts were decreased in the maternal serum and fetal cord blood of babies born to women with FGR (p < .05). The most sensitive marker was maternal native thiol (82.5, 95% confidence interval, 67.22-92.66%), while the maternal total thiol had highest specificity value (77.5, 95% confidence interval, 61.55-89.16%).Conclusions: Maternal and fetal serum thiol/disulfide profiles may use prediction of FGR severity and its neonatal outcome. Cyproheptadine is first-generation antihistamine drug, that is, H 1 receptor antagonist, with a drug being anesthetic, anti-serotonergic and anti-cholinergic and started to be used clinically in the 1960s. As firstly utilized as an anti-allergic drug, usage of cyproheptadine was expanded to other cases including serotonin syndrome, appetite increasing, migraines and insomnia. However, there are almost few studies seeking to explore the association between cyproheptadine and cancer in general. In the present study, we sought to determine the impact of cyproheptadine on C6 glioblastoma cells by morphological, biochemical and cytotoxic analyzes. We searched the effective doses of cyproheptadine for C6 glioblastoma cells and examined the cells under an inverted microscope. Next, we determined the protein levels of SIRT1, NFκB and IL-6 protein. Then, we measured and calculated the levels of thiols, disulfide bonds and related parameters. After that, we evaluated apoptotic activity by Annexin V and caspase 3 assays. As a result, we detected a dose-dependent increase in apoptosis and SIRT 1 protein levels, and a decrease in inflammatory proteins. Furthermore, we have detected a drop in thiol and disulfide content. Our study suggests that Cyproheptadine causes apoptosis and decreases inflammation by disrupting thiol/disulfide balance and enhancing the levels of SIRT1, offering the potential for being an anti-cancer drug.
